Anterior placental location influences onset and progress of labor and postpartum outcome.

INTRODUCTION: The aim of the study is to evaluate whether placental location at term is associated with delivery outcome. METHODS: A prospective study including 2354 patients with singleton pregnancy at term admitted for vaginal delivery was conducted. Placental position was determined before delivery by ultrasonographic examination performed transabdominally with women in the supine position. Maternal characteristics and delivery outcome such as premature rupture of membranes, induction of labor, mode and gestational age at delivery, indication for cesarean section, duration of the third sta…

Increased angiotensin II in the mesometrial triangle of a transgenic rat model of preeclampsia.

The pregnant female human angiotensinogen (hAGN) transgenic rat mated with the male hrenin (hREN) transgenic rat is a model of preeclampsia with increased blood pressure, proteinuria, and placenta alterations of edema and necrosis. The reverse mating of female hRENxmale hAGN does not show preeclamptic features. Because the placenta is well-recognized to be a key contributor to the preeclamptic syndrome, our hypothesis is that local angiotensin peptide concentrations found in the placenta and its associated mesometrial triangle of the preeclamptic transgenic rat differ from the reverse mating. …

Suppressors of cytokine signaling proteins in human preterm placental tissues.

Decreased suppressors of cytokine signaling (SOCS) activity in human gestational tissues may play a part in the onset/progression of term labor. Since SOCS proteins negatively regulate cytokine-mediated inflammatory processes, we hypothesized that SOCS proteins are elevated in gestational tissues from spontaneous preterm deliveries with intrauterine infection. SOCS1, -2 and -3 mRNAs and proteins were detectable by RT-PCR and immunoblotting respectively, in preterm amnion, choriodecidua and placenta, irrespective of infection status. Immunoperoxidase staining localized SOCS1, -2 and -3 to all c…

A second case of "chorangiocarcinoma" presenting in a term asymptomatic twin pregnancy: choriocarcinoma in situ with associated villous vascular proliferation.

We report a case of "chorangiocarcinoma" (choriocarcinoma in situ in association with a villous vascular proliferation) presenting in one of dichorionic diamniotic twin placentae at term in an asymptomatic pregnancy. This case is the second reported of chorangiocarcinoma in the literature and the first case of choriocarcinoma (in situ or otherwise) in a twin pregnancy. The lesion had the gross appearance of an infarct in the presenting twin placenta. Histologic sections revealed malignant villous trophoblast in eight microscopic fields within the placental infarct. The remainder of the placent…

Rupture of fetal vessels on placental surface.

Six placentae with subamniotic haematomata were studied and the lesions shown to be derived from ruptured varices of surface fetal veins. However, the changes in the vessel walls suggested that these fetal varices had been subjected to an increase of pressure for some time. Furthermore, the fetal veins of the affected placentae were lined by thick intimal cushions that were derived from organizing thrombi. Analysis of the clinical features of these pregnancies suggested an association of the vascular abnormalities with infants of low birthweight score, an opinion that was strengthened by an as…

Molecular pathways of senescence regulate placental structure and function.

The placenta is an autonomous organ that maintains fetal growth and development. Its multinucleated syncytiotrophoblast layer, providing fetal nourishment during gestation, exhibits characteristics of cellular senescence. We show that in human placentas from pregnancies with intrauterine growth restriction, these characteristics are decreased. To elucidate the functions of pathways regulating senescence in syncytiotrophoblast, we used dynamic contrast-enhanced MRI in mice with attenuated senescence programs. This approach revealed an altered dynamics in placentas of p53-/- , Cdkn2a-/- , and Cd…

Steroid concentrations in human myometrial and placental tissues at week 39 of pregnancy.

Endogenous levels of P, E1, and E2 were determined by radioimmunoassay in human myometrium and placenta at week 39 of pregnancy. In the myometrium, P and estrogens were higher in the inner layer near the placenta than in other zones. Higher E2/P ratios were found in placental sites. A large difference in the E2/P ratios was observed between placenta and corresponding adjacent myometrial area, indicating an easier diffusion of E2 from the site of its production.

[Quantitative changes of the cytotrophoblast in underweight and normalweight newborn after risk pregnancy and antepartal therapy (author's transl)].

A histochemical-morphometric method was used to determine at the cytotrophoblast of 73 placentas, changes in the composition of the syncytium and in the Langhans' cell count in underweight and normalweight newborn with or without antepartal maternal therapy in comparison to newborn after uneventful pregnancy. The reduction of the percentage share of mature syncytium when the share of predegenerative and degenerative syncytium is increased, as weil as the increase in the average Langhans' cell count in the placentas of premature infants, hypotrophic and eutrophic infants delivered at term after…

Trophoblast-specific knockdown of CSPG4 expression causes pregnancy complications with poor placentation in mice.

The multifunctional molecule chondroitin sulfate proteoglycan 4 (CSPG4/NG2) plays key roles in organogenesis and tumorigenesis. However, its roles in placentation remain unclear. In this study, CSPG4 expression in human and mouse placentas was investigated through immunohistochemistry (IHC), qPCR and western blotting. The theoretical structure and function of CSPG4 were assessed using bioinformatic tools, and the functions of CSPG4 in fetal and placental development were investigated using a mouse model established by trophoblast-specific CSPG4 knockdown and a trophoblast cell line with CSPG4 …

Precision-cut rat placental slices as a model to study sex-dependent inflammatory response to LPS and Poly I:C.

INTRODUCTION: Maternal inflammation in pregnancy represents a major hallmark of several pregnancy complications and a significant risk factor for neurodevelopmental and neuropsychiatric disorders in the offspring. As the interface between the mother and the fetus, the placenta plays a crucial role in fetal development and programming. Moreover, studies have suggested that the placenta responds to an inflammatory environment in a sex-biased fashion. However, placenta-mediated immunoregulatory mechanisms are still poorly understood. METHODS: Therefore, we have developed a model of ex vivo preci…

Placental weight centiles adjusted for age, parity and fetal sex.

INTRODUCTION: The weight of the placenta can be indicative of efficacy in nutrient and oxygen supply. Furthermore, it has been suggested that a measure of the placenta's ability to adequately supply nutrients to the fetus can be found in the relationship between birth weight and placental weight expressed as a ratio. Our aim was to develop age adjusted placenta weight and birth weight to placenta weight ratio reference curves that are stratified by maternal parity and fetal sex. METHODS: We included singleton, non-anomalous births with a gestational age inclusive of 28 + 0 weeks to 42 + 6 wee…

The Placental Response to Guinea Pig Cytomegalovirus Depends Upon the Timing of Maternal Infection.

Human cytomegalovirus (HCMV) infects the placenta, and these placental infections can cause fetal injury and/or demise. The timing of maternal HCMV infection during pregnancy is a determinant of fetal outcomes, but how development affects the placenta's susceptibility to infection, the likelihood of placental injury post-infection, and the frequency of transplacental HCMV transmission remains unclear. In this study, guinea pig cytomegalovirus (GPCMV) was used to model primary maternal infection and compare the effects of infection at two different times on the placenta. When guinea pigs were i…

Gestational protein restriction induces alterations in placental morphology and mitochondrial function in rats during late pregnancy.

The placenta acts a regulator of nutrient composition and supply from mother to fetus and is the source of hormonal signals that affect maternal and fetal metabolism. Thus, appropriate development of the placenta is crucial for normal fetal development. We investigated the effect of gestational protein restriction (GPR) on placental morphology and mitochondrial function on day 19 of gestation. Pregnant dams were divided into two groups: normal (NP 17 % casein) or low-protein diet (LP 6 % casein). The placentas were processed for biochemical, histomorphometric and ultrastructural analysis. The …

Relationship between placental morphology and histological findings in an unselected population near term.

Whilst individual histological features are well described, there are no universally agreed criteria as to what constitutes a clinically significant histological lesion of the placenta in an uncomplicated pregnancy, nor has the presence of such histological findings been systematically related to quantitative morphological characteristics of the placenta (such as placental shape, cord insertion and cord coiling). This study aims to explore this relationship and further to describe the incidence of predefined categories of histological lesions of the placenta in an unselected obstetric populati…

Maternal obesity alters placental lysophosphatidylcholines, lipid storage, and the expression of genes associated with lipid metabolism‡.

Dyslipidemia is a characteristic of maternal obesity and previous studies have demonstrated abnormalities in fatty acid oxidation and storage in term placentas. However, there is little information about the effect of pre-pregnancy obesity on placental lipid metabolism during early pregnancy. The objective of this study was to determine the relationship between lipid profiles and markers of metabolism in placentas from obese and lean dams at midgestation. Mice were fed a western diet (WD) or normal diet (ND) and lysophosphatidylcholines (LPCs) and/or phosphatidylcholines (PCs) were measured in…

Decidual/placental and first trimester plasma levels of hsa-miR-199a-3p|hsa-miR-199b-3p and hsa-miR-3150b-3p are associated with insulin secretion in pregnancy.

BACKGROUND/AIMS: The placenta expresses and releases specific microRNAs (miRNAs) into the maternal circulation that may influence insulin secretion during pregnancy. We hypothesized that specific decidual/placental miRNAs are associated with maternal insulin secretion during pregnancy. METHODS: In the Genetics of Glucose regulation in Gestation and Growth (Gen3G) prospective cohort, we estimated maternal insulin secretion using the Stumvoll first phase index derived from an oral glucose tolerance test at ~26 weeks of gestation. We quantified miRNAs by small RNA sequencing in placenta (N=435) …

Metagenetic Analysis of the Pregnant Microbiome in Horses.

Placentitis is the leading cause of infectious abortion in the horse. Additionally, it can result in weak and/or growth restricted offspring. While the etiology of ascending placentitis is well described in mares, less is known regarding the pathogenesis of other types, such as nocardioform placentitis. This study aims to identify the microbial communities in different body sites of the pregnant mare in early gestation to establish a core microbiome that may be perturbed in pathologic pregnancies such as placentitis. We hypothesize that the equine placenta harbors a distinct resident microbiom…
