Post-translational modification signature shapes the tumor immune microenvironment and predicts clinical outcomes in melanoma.

Post-translational modifications (PTMs) play a pivotal role in the initiation and progression of melanoma. To systematically evaluate their clinical significance, we integrated 10 widely used machine learning algorithms and 101 combinations to construct a PTM-related risk score model (PTMRS) based on 54 prognostic PTM genes. The model demonstrated robust prognostic predictive performance and stability across multiple melanoma cohorts (TCGA, GSE19234, GSE22153, GSE54467, and GSE65904). Immune profiling revealed that patients in the low-PTMRS group exhibited a more active immune microenvironment

Exploring Treatment Paradigms in Type 2 Diabetes: A Comparison of Independent and Combined Therapeutic Approaches.

Type 2 diabetes mellitus continues to pose a substantial global health burden due to its rising prevalence and complex metabolic nature. Multifactorial defects, including insulin resistance, β-cell dysfunction, and altered incretin activity, limit the effectiveness of single-target therapies. Current treatment strategies often fail to achieve sustained glycemic control, and delays in treatment intensification persist, highlighting the need for more effective and timely therapeutic approaches. This review evaluates independent and combined therapeutic strategies, focusing on their mechanisms, c

Retrospective Analysis of the Recurrence of Atypical Fibroxanthoma When Treated With Curettage and Cautery at a Single Centre.

Atypical fibroxanthoma (AFX) is a rare spindle cell neoplasm that arises within the dermis of chronically sun-exposed skin. Although wide local excision is standard practice, patients are regularly elderly and highly comorbid and therefore, curettage may offer a suitable alternative as a same day treatment with minimal aftercare. A retrospective case series was conducted analysing the files of all patients diagnosed with histopathological AFX at a single dermatology practice in Fremantle, Western Australia between the years 2016 and 2025. Kaplan-Meier analyses estimated recurrence rate with su

Enhanced but not superior: Performance outcomes from the inaugural enhanced games.

The Enhanced Games (TEG) emerged as an athletic event engineered to profit from biotechnological enhancement, such as performance-enhancing drugs (PEDs) utilization, relying on three premises: shattering world records (WR), enabling athletes to surpass their personal bests (PB), and utilizing PEDs to offset age-related physical decline. The effectiveness of TEG's PED protocols in achieving these three premises was evaluated. This is a retrospective analysis from 42 TEG athletes across Track, Swimming, and Weightlifting. TEG's performances were compared to current WR and athletes' PB previous t

Tracking Genetic Parkinson's Disease with Molecular Imaging: A Systematic Review.

BACKGROUND: Parkinson's disease (PD) is a worldwide, complex neurodegenerative disorder influenced by both genetic and environmental factors. Around 15-20% of PD cases are linked to genetic mutations, providing insights into the disease's pathogenesis. OBJECTIVE: The current review aims to summarize molecular neuroimaging findings in genetic forms of PD, also considering the pre-symptomatic phase of the disease to identify markers distinguishing genetic PD from sporadic forms of the disease as well as early markers of disease conversion. METHODS: A literature search on PubMed and Scopus was

Unveiling the molecular mechanisms of burn injury through integrated single-cell and bulk transcriptomic analysis.

PURPOSE: Burn injuries are prevalent and have a significant effect on patients' quality of life and healthcare costs. Current treatment modalities, such as wound care and surgical interventions, often face challenges due to complications like infection and inadequate healing. METHODS: This study adopted single-cell RNA sequencing (scRNA-seq) to investigate the cellular landscape of the burn microenvironment. After rigorous quality control filtering, 9,248 cells were retained for analysis. Using UMAP dimensionality reduction, these cells were annotated into 14 subpopulations, including Neutrop

Youth mental health and religiously framed radicalization in the Middle East and North Africa region: a review of evidence and gaps.

This narrative mini-review examines how youth mental health intersects with religiously framed radicalization in the Middle East and North Africa (MENA), conceptualizing radicalization as a potential maladaptive response to unmet psychological needs in highly religious, rapidly changing societies. It addresses three questions: how youth mental health relates to religious radicalization, what research gaps concern religious fanaticism as a response to lack of proper mental health care, and which conceptual, clinical, and policy developments are most needed. Available evidence indicates that the

The clinical usefulness of knowing CHRNA5 polymorphism genotype: paving the way for personalized therapy.

BACKGROUND: Tobacco smoking is a leading cause of global mortality, with cessation being the primary prevention strategy. Nicotine addiction has a genetic component; the rs503464 single nucleotide polymorphism (SNP) in the CHRNA5 gene is associated with smoking cessation therapy success. However, the impact of communicating genetic risk to patients remains unclear. This study evaluated whether knowledge of the rs503464 genotype influences smoking cessation rates. METHODS: 270 smokers were enrolled and randomized into two groups: informed and uninformed of their rs503464 genotype. All particip

Real-time multimodal phenotyping reveals distinct tumour cell dynamics and immune escape mechanisms in T cell therapies.

Adoptive T cell transfer therapy remains limited by the inability to monitor live tumour cell dynamics during treatment. Here we introduce a real-time, label-free phenotyping system that integrates electrical impedance spectroscopy, Raman spectroscopy and microscopy to analyse live tumour cells undergoing therapy. This system enables simultaneous tracking of metabolic activity, membrane integrity and cytoplasmic properties at single-cell resolution. First, analysis of glycolysis reveals that tumour-infiltrating lymphocytes suppress lactate production early, reducing tumour aggressiveness, whil

Evaluating the Use of Tumor Bank DNA to Validate Genetic Factors Impacting Opioid Response in Patients with Advanced Cancer.

Opioids are first-line therapy for cancer pain, yet up to 30% of patients fail to achieve adequate control at standard doses. Opioid response is partly genetically mediated, and understanding these factors may improve symptom management. This project aimed to assess the feasibility of using tumor bank DNA for pharmacogenetic analyses and to validate previously identified genetic variants associated with opioid response using existing genetic and clinical data. In this retrospective cohort study, clinical data (morphine equivalent daily dose, demographics) and genetic data (single-nucleotide po

Predictors and contributors of burnout among hospitalists in central Michigan.

Burnout among hospitalists is a growing focus of quality improvement for hospitals around the United States. Burnout has been associated with decreased patient care quality, which may lead to decreased system sustainability. A gap remains however regarding burnout factors in certain region-specific populations such as central Michigan. The objective of this study is to examine the associations between burnout and a collection of potentially impactful variables including gender, age, marital status, years at work, and dependents. Four domains of burnout were investigated among hospitalists at M

Intellectual humility in the relationship between religious or spiritual conflict and psychological outcomes.

We examined intellectual humility as a factor related to interpersonal religious or spiritual conflict. Among U.S. adults (N = 459), religious/spiritual conflict was associated with more perceived stress and psychological distress. Intellectual humility about religious/spiritual conflict was associated with less perceived stress, more forgiveness of the conflict partner(s), and more personal growth in response to the conflict. Intellectual humility about religious/spiritual conflict moderated links between religious/spiritual conflict and psychological distress. Contrary to expectations, the l

Linearizing and Forecasting: A Reservoir Computing Route to Digital Twins of the Brain.

Exploring the dynamics of complex systems such as the human brain is challenging due to inherent uncertainties and the limited availability of high-quality data. Here, we develop a mathematical theory for noisy linear recurrent neural networks (lRNNs) within the reservoir computing framework and demonstrate their effectiveness in constructing autonomous in silico replicas - digital-twins - of brain activity. We show that the Laplace-transform poles of high-dimensional inferred lRNNs directly encode the spectral properties of observed systems and are linked to the kernels of auto-regressive mod

Ultrathin Carbon Nanofiber Metafabric with a Hollow Aerogel Architecture for High-Efficiency Thermoregulation.

The preservation of physiological thermal homeostasis serves as a fundamental prerequisite for human endurance at all times. However, conventional single-purpose thermoregulatory materials frequently lack highly efficient integrated strategies to cope with diverse environmental demands. Here, an interface-confined assembly strategy that leverages interfacial rheology is developed to synthesize a carbon nanofiber metafabric with a hollow aerogel architecture for high-efficiency thermoregulation. By manipulating the interfacial viscosity gradient to suppress the inward radial diffusion of the sh

Multi-channel microfluidic chip with bilateral stripe for reconstruction of tissue barrier models in vitro.

Organ-on-a-chip systems can replicate human physiological functionsin vitroby simulating the dynamicin vivomicroenvironment, therefore offering great potential for applications in drug screening, disease research, and personalized medicine. Multi-channel microfluidic chips are the core physical components of organ-on-a-chip systems, which often incorporate structures such as stripes, micro-pillars, and porous membranes to confine gels within specific channels, thereby providing a three-dimensional extracellular matrix environment for reconstruction of tissue barrier modelsin vitro. However, cu

Three-way multi-criteria decision-making with linguistic information using prospect-regret theory and generalised TODIM for heart failure data analysis.

Cardiovascular disease is a leading global cause of mortality, and timely monitoring can help assess its severity. Electronic medical records contain information on patients’ symptoms, physical characteristics, and clinical laboratory test results. These data can be used in biostatistical analysis to identify patterns and relationships that may not be apparent to medical practitioners. However, when experts confront risk in complex settings, their bounded rationality may influence their decision-making behaviour. Moreover, the irrational behaviour of a single expert can substantially affect de

LAFOV PET as an enabling platform for pharmacokinetics-informed digital twins.

Digital twins (DTs) have emerged as a promising framework for precision medicine and are a key part of current initiatives to digitalize and innovate the healthcare sector. Medical DTs have shown potential to support more effective and efficient patient care, reduce healthcare costs, and enable novel approaches in drug development. Currently, DTs are primarily informed by structural imaging modalities, genetic information, clinical history, and lifestyle factors. These anatomy-driven twins can capture, for example, tumor burden and morphology in cancer, but lack the pharmacological dimension t

Understanding the landscape of hereditary breast and ovarian cancer testing in Brazil.

Next-generation sequencing (NGS) is essential for hereditary cancer diagnostics, as it detects single-nucleotide variants (SNVs), INDELs, and structural variants (SVs). Following ASCO recommendations for comprehensive sequence and structural analysis, we retrospectively analyzed a 25-gene NGS panel from 5,000 unrelated individuals, with or without personal history of cancer, to characterize the germline mutational spectrum associated with hereditary breast and ovarian cancer (HBOC). Among the participants, 12.7% carried pathogenic or likely pathogenic (P/LP) variants, predominantly comprising

Genetic Markers of Tumor Multiplicity in Non-melanoma Skin Cancer: Associations of 19 SNPs in an Italian Cohort.

INTRODUCTION: Ultraviolet exposure is the main environmental risk factor for non-melanoma skin cancers (NMSCs), but genetic susceptibility also contributes to variability in disease burden among individuals. METHODS: This study analyzed selected single-nucleotide polymorphisms (SNPs) in genes related to vitamin D and nicotinamide adenine dinucleotide metabolism, DNA repair, inflammation, and pigmentation as potential biomarkers for NMSC risk in an Italian cohort. Participants were stratified into low- and high-risk groups on the basis of tumor burden. RESULTS: No significant differences were
