Evaluation of synergistic effect of biodegradable polymeric nanoparticles and aluminum based adjuvant for improving vaccine efficacy.

Aluminum based adjuvants have been used widely to induce long lasting protective immunity through vaccination. But reported incidences of toxicity and side effects of aluminum have raised concerns regarding their safety in childhood vaccines. The present study demonstrates the synergistic effect of admixture of polylactic acid-polyethylene glycol (PLA-PEG) based biodegradable nanoparticles (NPs) and aluminum phosphate as a potential adjuvant system using tetanus toxoid (TT) as a model antigen. The immunological activity of the admixture formulation was maintained up to 180 days of storage at 5

Sylysia 350/Eudragit S100 solid nanomatrix as a promising system for oral delivery of cyclosporine A.

Cyclosporine A (CyA) is a poorly soluble peptide and the available preparation is the surfactant based Sandimmum Neoral(®). In this study, we developed a novel pH-sensitive nanomatrix system of CyA with medical-grade nanoporous silica and Eudragit(®) to enhance the oral absorption of CyA as well as to improve the potential nephrotoxicity caused by the pronounced initial plasma peak of Neoral(®). The nanomatrixs were prepared by an absolutely simple conventional process. The scanning electron microscopy (SEM), X-ray powder diffraction analysis (XRD), differential thermal analysis (DSC) and spec

Incidence of the core composition on the stability, the ROS production and the toxicity of CdSe quantum dots.

Mercaptosuccinic acid-capped CdSe and alloyed CdSe(S) QDs were prepared in aqueous solution at 100 and 170°C, respectively. These dots were characterized by transmission electron microscopy (TEM), X-ray diffraction (XRD), and UV-vis and photoluminescence spectroscopies. The dots were found to be of similar size (ca. 2nm) but differ in their composition and surface chemistry. The photostability of the QDs was found to correlate with their ability to produce reactive oxygen species (ROS) upon light activation. CdSe QDs produce hydroxyl radicals immediately after irradiation due to their modest p

Arrhythmogenic role of the border between two areas of cardiac cell alignment.

The goal of this study is to develop experimental and computational models of the excitation transition between areas of cardiac tissue with different anatomical anisotropy. Alignment of seeded neonatal rat cardiomyocytes was achieved with the aid of guiding polymer (PMGI) nanofibers, and two areas with orthogonal alignment were placed into a contact. It was found that the excitation wave crossing border between the areas with different alignment direction experiences substantial perturbation, up to the complete conduction block. In addition to the experimental study, this effect was analyzed

A nanoscale ridge/groove pattern arrayed surface enhances adipogenic differentiation of human supernumerary tooth-derived dental pulp stem cells in vitro.

OBJECTIVE: The aim of this study was to establish human dental pulp stem cells (hDPSCs) from supernumerary teeth and determine the effects of a 350-nm nano-patterned surface on adipogenic and osteogenic differentiation of hDPSCs. DESIGN: Several surface markers were analysed by FACS to confirm the isolated cells as hDPSCs. To demonstrate the effects of a nano-patterned surface on the differentiation of hDPSCs, the cells were cultured on a nano-patterned surface with or without adipogenic or osteogenic induction factors. Cells were then stained with Oil red O or Alizarin red, and the lineage s

Solution NMR analysis of the interaction between the actinoporin sticholysin I and DHPC micelles--correlation with backbone dynamics.

Sticholysin I (StI), an actinoporin expressed as a water-soluble protein by the sea anemone Stichodactyla helianthus, binds to natural and model membranes, forming oligomeric pores. It is proposed that the first event of a multistep pore formation mechanism consists of the monomeric protein attachment to the lipid bilayer. To date there is no high-resolution structure of the actinoporin pore or other membrane-bound form available. Here we evaluated StI:micelle complexes of variable lipid composition to look for a suitable model for NMR studies. Micelles of pure or mixed lysophospholipids and o

Exploring the influence of carbon nanoparticles on the formation of β-sheet-rich oligomers of IAPP₂₂₋₂₈ peptide by molecular dynamics simulation.

Recent advances in nanotechnologies have led to wide use of nanomaterials in biomedical field. However, nanoparticles are found to interfere with protein misfolding and aggregation associated with many human diseases. It is still a controversial issue whether nanoparticles inhibit or promote protein aggregation. In this study, we used molecular dynamics simulations to explore the effects of three kinds of carbon nanomaterials including graphene, carbon nanotube and C₆₀ on the aggregation behavior of islet amyloid polypeptide fragment 22-28 (IAPP₂₂₋₂₈). The diverse behaviors of IAPP₂₂₋₂₈ peptid

Osteoinductive biomaterial geometries for bone regenerative engineering.

Worldwide, more than 2.2 million patients undergo bone graft procedures annually. In each of these procedures an interface is formed between the host tissue and the graft material. Synthetic implants exhibit an interface with the host tissue and the formation of a homogenous interface consisting of bone and void of intervening soft tissue is desired (osseointegration); recent developments have highlighted the benefit of incorporating nanostructures at that interface. Autograft and allograft bone are frequently used for bone loss, nonunion fractures, and spinal fusions; however, both are plague

Characterization of early stage intermediates in the nucleation phase of Aβ aggregation.

Alzheimer's disease (AD) is a common form of dementia, which is characterized by the presence of extracellular amyloid plaques comprising the amyloid β peptide (Aβ). Although the mechanism underlying AD pathogenesis remains elusive, accumulating evidence suggests that the process of amyloid fibril formation is a surface-mediated event, which plays an important role in AD onset and progression. In this study, the mechanism of Aβ aggregation on hydrophobic surfaces was investigated with dual polarization interferometry (DPI), which provides real-time information on early stages of the aggregatio

Osseointegration of zirconia implants with different surface characteristics: an evaluation in rabbits.

PURPOSE: Zirconia ceramics are a viable alternative to titanium for use as dental implants. However, the smooth surface of zirconia means that longer healing periods are needed to accomplish osseointegration compared to roughened titanium surfaces. Surface modifications can be used to increase the roughness of zirconia. The aim of this study was to assess histologically and compare the degree of early bone apposition around zirconia dental implants with sandblasted, sintered, or laser-modified surfaces to that seen around surface-modified titanium implants. Removal torque was also measured and

Preparation and properties of electrospun soy protein isolate/polyethylene oxide nanofiber membranes.

Soy protein isolate (SPI) and polyethylene oxide (PEO) were dissolved in 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) and nonwoven nanofiber membranes were prepared from the solution by electrospinning. PEO functioned as a cospinning polymer in the process to improve the spinnability of SPI. The ratio of SPI to PEO was varied and the rest spinning conditions remained unchanged. The morphology of the nanofiber membranes, SPI and PEO distribution and phase structure in the fiber, crystallization and interaction between SPI and PEO, thermal properties and wettability of the membranes were studied. Characterization of protein immobilization on nanoporous gold using atomic force microscopy and scanning electron microscopy.

Nanoporous gold (NPG), made by dealloying low carat gold alloys, is a relatively new nanomaterial finding application in catalysis, sensing, and as a support for biomolecules. NPG has attracted considerable interest due to its open bicontinuous structure, high surface-to-volume ratio, tunable porosity, chemical stability and biocompatibility. NPG also has the attractive feature of being able to be modified by self-assembled monolayers. Here we use scanning electron microscopy (SEM) and atomic force microscopy (AFM) to characterize a highly efficient approach for protein immobilization on NPG u

Nanostructured silicon particles for medical applications.

Porous silicon (PS) which has different properties from the bulk material due to the quantum confinement effects is beside other physical properties (e.g., light emitting) bioactive or even bioresorbable. The aim of this paper is to optimise the experimental conditions for the fabrication of nanostructured Si particles and to find the best methods for attaching on its surface molecules of therapeutic interest. The selective porosification has been performed using (i) a dielectric/metallic masking layer micropatterned with corresponding etching windows; (ii) a controlled diffusion process leadi
