This study evaluates the added value of using enrichment broths for improving the detection of multidrug-resistant (MDR) bacteria-VRE, ESBL producers, and carbapenemase-producing organisms (CPOs)-from rectal swabs. Comparing direct plating to enrichment-based methods significantly increased detection sensitivity, especially for CPOs (from 17 to 27 cases, a 58.8% increase). Detection rates for ESBL producers and VRE also improved significantly (by 20%). Pre-enrichment is therefore a valuable addition to routine MDR bacteria screening protocols, which is particularly useful when high accuracy is

Globally, food safety and security are negatively impacted by antimicrobial resistance (AMR). The emergence of resistance caused by extended-spectrum β-lactamases (ESBLs) during the AMR era has resulted in increased morbidity, longer hospital stays, and more costly treatment alternatives. This study aimed to detect the contamination of ESBL-producing Enterobacterales antibiotic-resistant bacteria and ESBL genes in food materials and clinical samples in Chiang Mai, Thailand. Microbial culture-based, molecular and nucleotide sequencing methods were used in this study. Of 279 food samples, chicke

Multidrug-resistant Enterobacterales isolates carrying extended-spectrum beta-lactamases (ESBLs) and mobile colistin resistance (mcr) genes pose a significant healthcare threat as they can lead to difficult-to-treat infections. This study investigates the prevalence of isolates co-harbouring ESBL and mcr genes and characterizes the plasmids co-harbouring those genes. ESBL-producing Enterobacterales (ESBL-E) isolates identified during point prevalence surveys (PPS) in a Dutch hospital were screened for mcr genes. Plasmids co-harbouring mcr and ESBL genes were identified using long- and short-re…

Increased incidence of infections caused by carbapenem-resistant Acinetobacter baumannii (CRAB) in intensive care units has necessitated the development of rapid diagnostics. Resistance is primarily caused by OXA-type carbapenemases, of which OXA-23 is the most widespread. This study describes the development of recombinant monoclonal antibodies targeting OXA-23 and their application in a paper-based assay for rapid CRAB detection. Six scFv binders were identified from an immunized phage-displayed mouse antibody library against the OXA-23 protein, of which scFv 11 and 13 exhibited high sensiti

BACKGROUND: The soil environment is a natural reservoir of antibiotic resistance genes (ARG). It plays a crucial role in the emergence and dissemination of antibiotic resistance determinants. Anthropogenic activities, such as using antibiotics in healthcare facilities, aggravate the proliferation of ARG in soil. Exploring the soil environment can help in gaining insight about the presence, prevalence, and distribution of antibiotic resistance determinants. In the current study, we tried to isolate antibiotic-resistant bacteria from soil samples collected from around the incinerator at Shri Mat

The human-oral associated Capnocytophaga species are low-grade pathogens, more commonly affecting immunocompromised patients. Bloodstream infections are the most common clinical presentation. Microbiological diagnosis and antibiotic susceptibility testing are challenging due to the hard-to-growth character of these species, which complicates the initiation of effective antibiotic therapy. We present the case of an immunocompromised 60-year-old female receiving cefepime who developed febrile neutropenia. Blood cultures remained positive for a Capnocytophaga gingivalis isolate producing the CSP-

Acquisition of multiple carbapenemase genes by Klebsiella pneumoniae (Kp) is an emerging public health threat. Here, we aim to elucidate the population structure of Kp blood isolates carrying two different carbapenemase genes and identify the mechanism facilitating their dissemination. The study was conducted in a tertiary healthcare center between 2014 and 2022. Twenty-four patients with bacteremia caused by Kp carrying two different carbapenemase genes were identified. All 24 blood isolates were analyzed by short-read genome sequences supplemented by long reads in a selected number of isolat

E. coli is an important source of β-lactam resistance. Extended Spectrum β-Lactamase (ESBL) producing E. coli and their phylogenetic transmission are an important problem for health. On the other hand, MALDI-TOF MS allows sensitive and specific applications in food safety and many clinical studies, and it is also a method approved by the Food and Drug Administration (FDA) for microbial identification. We thought that the protein phenotypic character of the bacteria might provide data on resistance. This study aims to investigate β-lactamases in E. coli and to examine the differences in protein

The rapid emergence and spread of antimicrobial resistance have become critical global health issues, leading to significant morbidity and mortality worldwide. With the increase in resistance to multiple drugs, especially frontline clinical antibiotics, there is an urgent need for novel and effective alternative strategies. Herein, we developed a vaccine targeting the antimicrobial resistance enzyme NDM-1, which was first identified in Klebsiella pneumoniae and has quickly spread to other gram-negative bacteria. Our results demonstrate that NDM-1 primarily triggers a humoral immune response an

BACKGROUND: Antimicrobial resistance (AMR) is a critical threat to human health. Escherichia coli and Klebsiella pneumoniae are clinically the most important species associated with AMR and are the most common carbapenemase-producing (CP) Enterobacterales detected in human specimens in Finland. Wastewater surveillance has emerged as a potential approach for population-level surveillance of AMR, as wastewater could offer a reflection from a larger population with one sample and minimal recognized ethical issues. In this study, we investigated the potential of wastewater surveillance to detect C

Interpreting the phenotypes of bla SHV alleles in Klebsiella pneumoniae genomes is complex. Whilst all strains are expected to carry a chromosomal copy conferring resistance to ampicillin, they may also carry mutations in chromosomal bla SHV alleles or additional plasmid-borne bla SHV alleles that have extended-spectrum β-lactamase (ESBL) activity and/or β-lactamase inhibitor (BLI) resistance activity. In addition, the role of individual mutations/a changes is not completely documented or understood. This has led to confusion in the literature and in antimicrobial resistance (AMR) gene databas

The global threat of antibiotic resistance has increased the importance of the detection of antibiotics. Conventional methods to detect antibiotics are time-consuming and require expensive specialized equipment. Here, we present a simple and rapid biosensor for detecting ampicillin, a commonly used antibiotic. Our method is based on the fluorescent properties of chitosan-coated Mn-doped ZnS micromaterials combined with the β-lactamase enzyme. The biosensors exhibited the highest sensitivity in a linear working range of 13.1-72.2 pM with a limit of detection of 8.24 pM in deionized water. In ad

BACKGROUND: Extended-spectrum cephalosporins (ESCs) are third and fourth generation cephalosporin antimicrobials used in humans and animals to treat infections due to multidrug-resistant (MDR) bacteria. Resistance to ESCs (ESC-R) in Enterobacterales is predominantly due to the production of extended-spectrum β-lactamases (ESBLs) and plasmid-mediated AmpC β-lactamases (AmpCs). The dynamics of ESBLs and AmpCs are changing across countries and host species, the result of global transmission of ESC-R genes. Plasmids are known to play a key role in this dissemination, but the relative importance of
