Mass Spectrometry-Based Evaluation of the Bland-Altman Approach: Review, Discussion, and Proposal.

Reliable quantification in biological systems of endogenous low- and high-molecular substances, drugs and their metabolites, is of particular importance in diagnosis and therapy, and in basic and clinical research. The analytical characteristics of analytical approaches have many differences, including in core features such as accuracy, precision, specificity, and limits of detection (LOD) and quantitation (LOQ). Several different mathematic approaches were developed and used for the comparison of two analytical methods applied to the same chemical compound in the same biological sample. Gener

Occurrence of anabolic agents in plants and their importance.

More than 40 plant species have been shown to contain substances that are active in biological assays for estrogenic activity. Such substances may be constitutive metabolic products of a plant, or be formed adaptively in response to environmental factors, such as fungal attack (e.g. coumestrol synthesis in alfalfa infected with Pseudopeziza medicagensis); in other instances estrogens may arise from microbial attack on plant material during storage (e.g. zearalenone formation from corn by Fusarium spp.) Phyto-estrogens may reach man through direct consumption of fresh fruit, vegetables and proc

Proteomic profiling of bull spermatozoa and seminal plasma to inform the rational development of functionally targeted semen extenders for tropical cattle.

BACKGROUND AND AIM: Artificial insemination (AI) is a cornerstone technology for genetic improvement in livestock; however, the fertility outcomes of cryopreserved semen often remain inconsistent, particularly in tropical production systems where heat stress and oxidative damage compromise sperm function. Conventional semen extenders are largely developed through empirical approaches and may not adequately reflect the molecular characteristics of locally adapted cattle breeds. Proteomic profiling offers an opportunity to identify endogenous proteins involved in sperm function and resilience, t

Exploring Lachancea thermotolerans biodiversity: insights into lactic acid production and carbon metabolism in response to oxygen availability.

Lactic acid production by Lachancea thermotolerans offers a promising biological strategy to restore wine acidity often compromised by climate change, yet this metabolic trait remains highly variable and poorly controlled. This study systematically evaluated the impact of controlled oxygenation regimes on fermentation performance and primary metabolism in three genetically distinct L. thermotolerans strains. Oxygen availability drastically altered metabolic fluxes. Under continuous aerobic conditions, carbon was redirected toward biomass formation (9- to 13-fold increase), with reduced ethanol

A conceptual blueprint for "turning cold to hot" in Osteosarcoma: from TME stratification hypotheses to adaptive therapeutic prospects.

Osteosarcoma (OS) is a quintessential “cold tumor,” and outcomes for patients with metastatic or recurrent disease have remained poor for decades. The failure of immune checkpoint inhibitors (ICIs) in OS reflects a multilayered immunosuppressive architecture rather than a single dominant lesion. This review deconstructs three principal barriers within that architecture: (1) physical T-cell exclusion driven by a dense fibrotic stroma and aberrant vasculature; (2) a myeloid-dominant suppressive network enriched for Tumor-Associated Macrophages (TAMs) and Myeloid-Derived Suppressor Cells (MDSCs);

Novel Gradient-based Approach to Spatially Fractionated Radiotherapy: Implementation and Preliminary Clinical Results.

Background and purpose Bulky tumors often present limited treatment options and poor response to conventional radiotherapy due to hypoxia, radioresistance, and dose delivery challenges near organs at risk. Spatially fractionated radiation therapy (SFRT) has shown promising results by creating heterogeneous intratumoral dose distributions that enhance local control while minimizing toxicity and stimulating immune response. We introduce a novel gradient-based approach, spatial stepwise gradient radiation therapy (2SG-RT), designed to simplify planning, preserve the biological advantages of dose

Phytoplankton Cell Death Under the Global Change Scenario.

Phytoplankton are aquatic unicellular eukaryotic (i.e. protists) or prokaryotic photosynthetic organisms thriving in the world oceans. They are at the base of aquatic food webs and play a key role in determining the effects of the environmental change on the ocean surface. Phytoplankton are responsible for around 50% of the net amount of carbon assimilated annually by photoautotrophs. The removal of atmospheric CO2 does not only depend on CO2 fixation, but long-term carbon sequestration also requires the export of the organic matter fixed by phytoplankton in the surface layer (euphotic zone) i

Development of a new high-yield integration site assay reveals disease-specific patterns across HTLV-1-associated pathologies.

Human T lymphotropic virus type 1 (HTLV-1) chronic infection is maintained through mitotic proliferation of the infected CD4+ T cells, where the viral genome is integrated as a provirus in its host genome. HTLV-1 integration sites (ISs) have a part in HTLV-1-associated pathologies, with distinct IS patterns associated with malignant proliferation or inflammatory diseases. However, IS determination remains challenging because most assays rely on complex biological and biocomputing protocols. We present an IS assay that solely relies on PCR and Sanger sequencing, which allowed HTLV-1 IS determin

Antiviral action of aqueous extracts of propolis from Scaptotrigona aff. postica (Hymenoptera; Apidae) against Zica, Chikungunya, and Mayaro virus.

The limited availability of antivirals for new highly pathogenic strains of virus has become a serious public health. Therefore, news products against these pathogens has become an urgent necessity. Among the multiple sources for news antibiotics and antivirals, insect exudates or their products has become an increasingly frequent option. Insects emerged 350 million years ago and have showed a high adaptability and resistance to the most varied biomes. Their survival for so long, in such different environments, is an indication that they have a very efficient protection against environmental i

Glycan clock of ageing-analytical precision and time-dependent inter- and i-individual variability.

Ageing is a complex biological process with variations among individuals, leading to the development of ageing clocks to estimate biological age. Glycans, particularly in immunoglobulin G (IgG), have emerged as potential biomarkers of ageing, with changes in glycosylation patterns correlating with chronological age.For precision analysis, three different plasma pools were analysed over 26 days in tetraplicates, 312 samples in total. In short-term variability analysis, two cohorts were analysed: AstraZeneca MFO cohort of 26 healthy individuals (median age 20) and a cohort of 70 premenopausal Ch

Tumor-Acidity-Cleavable Maleic Acid Amide (TACMAA): A Powerful Tool for Designing Smart Nanoparticles To Overcome Delivery Barriers in Cancer Nanomedicine.

Over the past few decades, cancer nanomedicine has been under intensive development for applications in drug delivery, cancer therapy, and molecular imaging. However, there exist a series of complex biological barriers in the path of a nanomedicine from the site of administration to the site of action. These barriers considerably prevent a nanomedicine from reaching its targets in a sufficient concentration and thus severely limit its therapeutic benefits. According to the delivery process, these biological delivery barriers can be briefly summarized in the following order: blood circulation,

High throughput proteomic analysis and a comparative review identify the nuclear chaperone, Nucleophosmin among the common set of proteins modulated in Chikungunya virus infection.

UNLABELLED: Global re-emergence of Chikungunya virus (CHIKV) has renewed the interest in its cellular pathogenesis. We subjected CHIKV-infected Human Embryo Kidney cells (HEK293), a widely used cell-based system for CHIKV infection studies, to a high throughput expression proteomics analysis by Liquid Chromatography-tandem mass spectrometry. A total of 1047 differentially expressed proteins were identified in infected cells, consistently in three biological replicates. Proteins involved in transcription, translation, apoptosis and stress response were the major ones among the 209 proteins that

Enhancement of Plant Productivity in the Post-Genomics Era.

Obtaining high plant yield is not always achievable in agricultural activity as it is determined by various factors, including cultivar quality, nutrient and water supplies, degree of infection by pathogens, natural calamities and soil conditions, which affect plant growth and development. More noticeably, sustainable plant productivity to provide sufficient food for the increasing human population has become a thorny issue to scientists in the era of unpredictable global climatic changes, appearance of more tremendous or multiple stresses, and land restriction for cultivation. Well-establishe

[Methods and applications of single-cell proteomics analysis based on mass spectrometry].

The cell is the smallest unit of living organisms. Although cells often assemble to serve a common function, intercellular heterogeneity often exists due to different genetic and environmental effects. Therefore, single-cell analysis has been regarded as an indispensable means to investigate cell heterogeneity, especially when researching cell differentiation, disease diagnosis, and therapy. As the chief factors influencing cell and biological activities, proteins have long been a major concern in biochemistry. However, due to their intrinsic lack of amplification characteristics, wide species

Fast, adaptive evolution at a bacterial host-resistance locus: the PFam54 gene array in Borrelia burgdorferi.

Microbial pathogens have evolved sophisticated mechanisms for evasion of host innate and adaptive immunities. PFam54 is the largest paralogous gene family in the genomes of Borrelia burgdorferi, the Lyme disease bacterium. One member of PFam54, the complement-regulator acquiring surface proteins 1 (BbCrasp-1), is able to abort the alternative pathway of complement activation via binding human complement-regulator factor H (FH). The gene coding for BbCRASP-1 exists in a tandem array of PFam54 genes in the B. burgdorferi genome, a result apparently of repeated gene duplications. To help elucidat

The molecular basis of mouse adaptation by human enterovirus 71.

A mouse-adapted strain of human enterovirus 71 (HEV71) was selected by serial passage of a HEV71 clinical isolate (HEV71-26M) in Chinese hamster ovary (CHO) cells (CHO-26M) and in newborn BALB/c mice (MP-26M). Despite improved growth in CHO cells, CHO-26M did not show increased virulence in newborn BALB/c mice compared with HEV71-26M. By contrast, infection of newborn mice with MP-26M resulted in severe disease of high mortality. Skeletal muscle was the primary site of replication in mice for both viruses. However, MP-26M infection induced severe necrotizing myositis, whereas CHO-26M infection
