Phosphoinositide 3-kinase δ (PI3Kδ) in respiratory disease.

Defining features of chronic airway diseases include abnormal and persistent inflammatory processes, impaired airway epithelial integrity and function, and increased susceptibility to recurrent respiratory infections. Phosphoinositide 3-kinases (PI3K) are lipid kinases, which contribute to multiple physiological and pathological processes within the airway, with abnormal PI3K signalling contributing to the pathogenesis of several respiratory diseases. Consequently, the potential benefit of targeting PI3K isoforms has received considerable attention, being viewed as a viable therapeutic option

Influence of Temperature on the Efficacy of Homologous and Heterologous DNA Vaccines against Viral Hemorrhagic Septicemia in Pacific Herring.

Homologous and heterologous (genogroup Ia) DNA vaccines against viral hemorrhagic septicemia virus (genogroup IVa) conferred partial protection in Pacific Herring Clupea pallasii. Early protection at 2 weeks postvaccination (PV) was low and occurred only at an elevated temperature (12.6°C, 189 degree days), where the relative percent survival following viral exposure was similar for the two vaccines (IVa and Ia) and higher than that of negative controls at the same temperature. Late protection at 10 weeks PV was induced by both vaccines but was higher with the homologous vaccine at both 9.0°C

γδT Cells and Related Diseases.

Discovered 30 years ago, γδT cells remain an enigmatic T-cell subset. Although they account for a small portion of the total human circulating T-cell pool, their associations with other immune cells and their potential regulatory roles in related diseases have been explored but still require further investigation. γδT cells which are MHC-unrestricted innate-like lymphocytes with more unique antigen receptors than αβT cells and B cells are considered to bridge innate and adaptive immunity. They have APC functions and initiate adaptive immunity. Due to their distribution in specific tissues, sec

Ethanolamine Signaling Promotes Salmonella Niche Recognition and Adaptation during Infection.

Chemical and nutrient signaling are fundamental for all cellular processes, including interactions between the mammalian host and the microbiota, which have a significant impact on health and disease. Ethanolamine is an essential component of cell membranes and has profound signaling activity within mammalian cells by modulating inflammatory responses and intestinal physiology. Here, we describe a virulence-regulating pathway in which the foodborne pathogen Salmonella enterica serovar Typhimurium (S. Typhimurium) exploits ethanolamine signaling to recognize and adapt to distinct niches within

Differential susceptibility of Aedes aegypti to infection by the American and Southeast Asian genotypes of dengue type 2 virus.

Outbreaks of dengue hemorrhagic fever have coincided with the introduction of the Southeast (SE) Asian genotype of dengue type 2 virus in the Western Hemisphere. This introduced genotype appears to be rapidly displacing the indigenous, American genotype of dengue 2 virus throughout the region. These field observations raise the possibility that the SE Asian genotype of dengue 2 is better adapted for vector transmission than its American counterpart. To evaluate this hypothesis, we compared the ability of viral strains of the SE Asian and American genotypes to infect, replicate, and disseminate

[The role of the nasal microbiome in regulating upper airway immune homeostasis].

The nasal microbiome serves as a pivotal regulator of immune homeostasis in the upper airway. Under physiological conditions, it maintains immune equilibrium through mechanisms such as reinforcing the epithelial barrier and modulating innate and adaptive immune responses. Microbial dysbiosis, however, is recognized as a key trigger in the pathogenesis of chronic rhinosinusitis (CRS) and allergic rhinitis (AR). This review summarizes how the nasal microbiome regulates immune homeostasis by influencing epithelial barrier function and immune responses in both health and disease states, with a foc

How mitochondrial heterogeneity fuels a dynamic organelle ecosystem.

Mitochondria are traditionally viewed as a homogeneous network supporting cellular energy production. However, increasing evidence reveals substantial heterogeneity across biological scales, from intramitochondrial microdomains to specialised mitochondrial populations within cells and tissues. These subpopulations differ in morphology, metabolism, bioenergetics, and spatial organisation, reflecting adaptation to local functional demands. In this Review, we discuss the mechanisms underlying mitochondrial heterogeneity, the emerging concept of mitochondrial subclasses, their functional integrati

Mitochondria reactive oxygen species signaling-dependent immune responses in macrophages and T cells.

Mitochondria are key regulators of immune cell function, going beyond their traditional role in ATP and metabolite production to support anabolic processes and act as hubs for intracellular signaling. A key aspect of this signaling function is the production of mitochondrial reactive oxygen species (mtROS), which act as critical second messengers in both adaptive and innate immune regulation. Immune cells maintain an optimal concentration of mtROS to maintain physiological responses, and excessive or lack of mtROS production contributes to chronic inflammation, autoimmunity, and cancer. Here,

Progress and challenges of in vivo flow cytometry and its applications in circulating cells of eyes.

Circulating inflammatory cells in eyes have emerged as early indicators of numerous major diseases, yet the monitoring of these cells remains an underdeveloped field. In vivo flow cytometry (IVFC), a noninvasive technique, offers the promise of real-time, dynamic quantification of circulating cells. However, IVFC has not seen extensive applications in the detection of circulating cells in eyes, possibly due to the eye's unique physiological structure and fundus imaging limitations. This study reviews the current research progress in retinal flow cytometry and other fundus examination technique

Structures, responsible for acceptive (comensaling relation) and protective (pathogenic defense) immunity, were studied and compared in small intestine - to ileum mucosa. Data shown, that main application of the both domains of immune system is to support the correlation between body and foreign microbes, but they response is different. Most significant differences are as follows: in acceptive reactions presented only in aseptic animals - gnotobionts, inflammatory changes absent, so immune reaction complex develops into physiological condition. Symbiotic reactions release in mucosa epithelial

Osteoarthritis of the temporomandibular joint organ and its relationship to disc displacement.

To overcome disagreements with regard to the relationship between disc displacement and osteoarthritis of the temporomandibular joint (TMJ), the evidence for suggested disease mechanisms and clinical course of these disorders is reviewed. The TMJ behaves as a complex organ in which biochemical and biomechanical processes regulate the physiology of cartilage, bone, synovium, ligaments, and synovial fluid. In this concept, TMJ osteoarthritis is an organ failure involving all its structures. The development of as well as recovery from disease appears to be intimately related to exceeding and supp

Helping them cope.

1. Self-help groups for individuals with sarcoidosis (a noncontagious multisystem disorder characterized by granulomas in many organs) are rare despite the physical and emotional problems associated with this disease. 2. The stimulus for the development of the group arose when staff in a pulmonary clinic identified that sarcoidosis patients were helping each other cope with problems and concerns related to their disease and feelings of isolation. 3. Nurses play a significant role in creating vehicles of help and service to facilitate and support patients experiencing fear and anxiety about the

Distinct root apoplastic barrier distribution and conserved peroxidase function in desert trees.

The formation of extracellular barriers in roots plays a critical role in maintaining water balance and nutrient homeostasis; however, their molecular composition and physiological roles in desert-adapted plants remain largely unexplored. Here, we examine apoplastic barrier formation in the roots of two arid-adapted, ecologically and economically important tree species: date palm (Phoenix dactylifera) and grey mangrove (Avicennia marina). In date palm, lignin and suberin showed complementary distribution patterns within the outer root layers. In mangrove roots, lignin accumulated in the epider

Stress controls heterochromatin inheritance via histone H3 ubiquitylation.

Heterochromatin, marked by histone H3 lysine 9 methylation, can be epigenetically inherited through cell division1-3, maintaining gene repression that preserves cell identity and enables adaptation to environmental challenges2-6. Studies on Schizosaccharomyces pombe have shown that heterochromatin propagation depends on the read-write mechanism, wherein a sufficient density of H3K9me3-modified nucleosomes, stabilized by histone deacetylases, concentrates Clr4SUV39H on chromatin to promote further deposition of H3K9 methylation7-9. Whether other mechanisms control heterochromatin propagation by

Iron need attention: a review on the role of iron in plants.

Plants must continuously adapt to complex dynamic environmental conditions throughout their entire life cycle. Abiotic stresses, such as drought, salinity, heavy metal toxicity, extreme temperatures significantly impair plant growth and cause substantial reductions in crop yield. Iron (Fe), an essential micronutrient, serves as a cofactor for numerous metalloenzymes and a critical component of the mitochondrial and chloroplast electron transport chains. It plays an indispensable role in fundamental physiological processes such as photosynthesis, respiration, antioxidant defense, and nitrogen m

Analog habitat health and safety protocol: replicable standard system for analyzing data related to human factors in simulations.

As space exploration emerges as the next frontier for humanity, challenges related to health and safety in isolated, confined, and extreme (ICE) environments become increasingly relevant. This study examines the development of integrated monitoring protocols to establish a replicable standard for collecting and analyzing human factors indicators in both analogous and real space missions, adapting existing approaches from polar environments. A one-month analog mission was conducted with a six-member crew, during which protocols were designed for pre-, during-, and post-mission phases. The metho

Dermatophyte speciation processes reconciled with current phylogenetic species concepts.

The family Arthrodermataceae, the dermatophytes and allies, ancestrally began with Ascomycetous bifactorial sexual cycles built into an ecology that also featured considerable clonal propagation via conidia. When keratinolytic capabilities made ecological crossover to dermatopathogenicity possible, that conventional cycle, requiring moist, deposited keratinous material, could only be maintained by pathogens infecting animals burrowing or denning in habitats with soil. Lineages adapted to animals not nesting in soil became established clonally from representatives of single mating types. They b

Cell Culture Adaptation of Porcine Group A Rotavirus: Advances and Challenges for Vaccine Development.

Porcine group A rotavirus (PoRVA) is a significant cause of viral diarrhea in piglets, necessitating urgent global implementation of effective control strategies. This review assesses advancements in PoRVA in vitro cultivation and amplification, crucial for PoRVA vaccine development. Traditional PoRVA cultivation commonly employs primary porcine kidney cells or finite cell lines like MA-104, posing well-documented challenges in scalability, production cost, and their ability to recapitulate the natural intestinal microenvironment. Consequently, research has increasingly focused on adapting PoR

Adaptation or Maladaptation? Rethinking How Energy Restriction Outcomes Are Monitored.

Energy restriction (ER) serves as both a corrective health measure in metabolically compromised individuals and as a strategically implemented tool to enhance athletic performance and/or meet sporting weight divisions. Individuals with overweight/obesity who embark on a weight loss intervention often experience temporal improvements in metabolic health, only to be undermined by the phenomenon known as adaptive thermogenesis, and subsequent weight regain. ER in elite sport settings, whether a deliberate strategy to manipulate body composition or an unintentional failure to meet the high energy

Endoplasmic Reticulum-Mitochondria Calcium Drives Salivary Bioenergetics.

Saliva secretion requires continuous energy supply throughout the day. Mitochondria dynamically adapt to fluctuating energy demands, yet the mechanisms underlying the adaptions remain poorly understood. Here, we employed real-time intravital imaging and fluorescence lifetime imaging microscopy (FLIM) to monitor mitochondria functions in submandibular glands. We revealed distinct mitochondrial distribution patterns; in acinar cells, mitochondria were predominantly distributed near the cell membranes or scattered throughout the cytoplasm with extensive endoplasmic reticulum (ER)-mitochondria con
