Hemolytic anemia in the mouse. Report of a new mutation and clarification of its genetics.

A new mutation causing spherocytic, hemolytic anemia has been discovered in the house mouse. It is inherited as a single autosomal recessive gene, allelic with both sph and ha, which, in turn, were shown to be allelic with each other. A revised nomenclature for the three apparent alleles is proposed: sph (formerly sph), sphha (formerly ha), and sph2Bc (the new mutation). Like the other murine hemolytic anemias, sph2Bc involves a defect in the red blood cell membrane protein, spectrin.

Association of rs429358 and rs7412 polymorphism in apolipoprotein E gene with the severity of neuropathy among type 2 diabetes patients.

BACKGROUND: Genetic factors might influence the pathophysiology of diabetic peripheral neuropathy (DPN) which leads to the variability in its severity. The present study aimed to find the association of apolipoprotein E (APOE) gene polymorphism with the severe DPN in patients with type 2 diabetes mellitus (T2DM). METHODS: Type 2 diabetes patients were divided into group I (no neuropathy) and group II (mild/moderate/severe neuropathy). Further, the subgroup analysis of group II was done to determine the difference in symptomatic severity. The nerve conduction study was done to measure various

Evaluation of Genetic Marker Polymorphisms (MTHFRC677T and ABCC2 C) for Predicting Methotrexate Toxicity in Psoriasis Patients in Egypt: A Case-Control Study.

Methotrexate is an antineoplastic agent prescribed in various treatment protocols, particularly for psoriasis. Studies on toxicogenomic and clinical data suggest that polymorphism in genes involved in folate metabolism are linked to varying individual responses to methotrexate (MTX). This study aimed to investigate the single nucleotide polymorphisms (SNPs) of two genetic DNA markers (MTHFR 677 C and ABCC2 C), their correlation to protein expression, and laboratory investigations as potential predictors of susceptibility to MTX toxicity among patients. This is a case-control study involving 90

Polymorphisms in rs2069845 are associated with IL-6 and soluble IL-6 receptor levels during total joint replacement.

As the number of patients undergoing total joint replacement (TJR) surgery increases, so does the number of revision surgeries. One driver of implant failure and subsequent revision surgery is peri-implant osteolysis, which is driven by inflammation-mediated bone loss. IL-6 is an inflammatory cytokine that is elevated during the peri-operative period. Early elevations in IL-6 levels have been linked to osteolysis development. The current study asked whether there is genetic contribution to the IL-6-related peri-operative inflammatory reaction to TJR surgery. Patients undergoing primary TJR (to

Distribution of the Duffy genotypes in Malaysian Borneo and its relation to Plasmodium knowlesi malaria susceptibility.

The Duffy blood group plays a key role in Plasmodium knowlesi and Plasmodium vivax invasion into human erythrocytes. The geographical distribution of the Duffy alleles differs between regions with the FY*A allele having high frequencies in many Asian populations, the FY*B allele is found predominately in European populations and the FY*Bes allele found predominantly in African regions. A previous study in Peninsular Malaysia indicated high homogeneity of the dominant FY*A/FY*A genotype. However, the distribution of the Duffy genotypes in Malaysian Borneo is currently unknown. In the present st…

Evidence GDF15 Plays a Role in Familial and Recurrent Hyperemesis Gravidarum.

Introduction Hyperemesis gravidarum (HG), a pregnancy complication characterized by severe nausea and vomiting in pregnancy, occurs in up to 2% of pregnancies. It is associated with both maternal and fetal morbidity. HG is highly heritable and recurs in approximately 80% of women. In a recent genome-wide association study, it was shown that placentation, appetite, and the cachexia gene GDF15 are linked to HG. The purpose of this study was to explore whether GDF15 alleles linked to overexpression of GDF15 protein segregate with the condition in families, and whether the GDF15 risk allele is ass…

Dietary Intake, FTO Genetic Variants, and Adiposity: A Combined Analysis of Over 16,000 Children and Adolescents.

The FTO gene harbors variation with the strongest effect on adiposity and obesity risk. Previous data support a role for FTO variation in influencing food intake. We conducted a combined analysis of 16,094 boys and girls aged 1-18 years from 14 studies to examine the following: 1) the association between the FTO rs9939609 variant (or a proxy) and total energy and macronutrient intake; and 2) the interaction between the FTO variant and dietary intake, and the effect on BMI. We found that the BMI-increasing allele (minor allele) of the FTO variant was associated with increased total energy intak

The insulin-sensitivity sulphonylurea receptor variant is associated with thyrotoxic paralysis.

Thyrotoxicosis is the most common cause of the acquired flaccid muscle paralysis in adults called thyrotoxic periodic paralysis (TPP) and is characterised by transient hypokalaemia and hypophosphataemia under high thyroid hormone levels that is frequently precipitated by carbohydrate load. The sulphonylurea receptor 1 (SUR1 (ABCC8)) is an essential regulatory subunit of the β-cell ATP-sensitive K(+) channel that controls insulin secretion after feeding. Additionally, the SUR1 Ala1369Ser variant appears to be associated with insulin sensitivity. We examined the ABCC8 gene at the single nucleoti

Modulation of PF10_0355 (MSPDBL2) alters Plasmodium falciparum response to antimalarial drugs.

Malaria's ability to rapidly adapt to new drugs has allowed it to remain one of the most devastating infectious diseases of humans. Understanding and tracking the genetic basis of these adaptations are critical to the success of treatment and intervention strategies. The novel antimalarial resistance locus PF10_0355 (Pfmspdbl2) was previously associated with the parasite response to halofantrine, and functional validation confirmed that overexpression of this gene lowered parasite sensitivity to both halofantrine and the structurally related antimalarials mefloquine and lumefantrine, predomina

Genetic and epigenetic mechanisms collaborate to control SERPINA3 expression and its association with placental diseases.

SERPINA3 (Serpin peptidase inhibitor clade A member 3), also known as a1-antichymotrypsin, is a serine protease inhibitor involved in a wide range of biological processes. Recently, it has been shown to be up-regulated in human placental diseases in association with a hypomethylation of the 5' region of the gene. In the present study, we show that the promoter of SERPINA3 is transcriptionally activated by three transcription factors (TFs) (SP1, MZF1 and ZBTB7B), the level of induction being dependent on the rs1884082 single nucleotide polymorphism (SNP) located inside the promoter, the T allel

Early influence of the rs4675690 on the neural substrates of sadness.

BACKGROUND: CREB1 has previously been implicated in mood disorders, suicide, and antidepressant response. There is some evidence that the T allele in rs4675690, a single-nucleotide polymorphism near the CREB1 gene, is involved in the modulation of neural responses to negative stimuli. It is not known whether differential brain activity during negative mood state appears early in life in T allele carriers. METHODS: Functional magnetic resonance imaging (fMRI) was used to measure brain activity, during a transient state of sadness, in children homozygous for the T allele or the C allele. This p

Functional variant disrupts insulin induction of USF1: mechanism for USF1-associated dyslipidemias.

BACKGROUND: The upstream transcription factor 1 (USF1) gene is associated with familial combined hyperlipidemia, the most common genetic dyslipidemia in humans, as well as with various dyslipidemic changes in numerous other studies. Typical of complex disease-associated genes, neither the explicit mutations have been described nor the functional consequences for risk allele carriers been reported at the cellular or tissue level. METHODS AND RESULTS: In this study, we aimed at describing the molecular mechanism through which the strongest associating intronic single-nucleotide polymorphism var
