Proliferative lupus nephritis and leukocytoclastic vasculitis during treatment with etanercept.

Tumor necrosis factor-alpha (TNF-alpha) is a proinflammatory cytokine. Agents that neutralize TNF-alpha are effective in the treatment of disorders such as rheumatoid arthritis, juvenile rheumatoid arthritis (JRA), spondyloarthropathies, and inflammatory bowel disease. TNF-alpha antagonist therapy has been associated with the development of antinuclear antibodies (ANA) and double-stranded DNA (dsDNA) antibodies, as well as the infrequent development of systemic lupus erythematosus (SLE)-like disease. We describe the first case of biopsy-confirmed proliferative lupus nephritis and leukocytoclas

Role of interleukin-6 in T-cell activation during primary and secondary infection with Listeria monocytogenes.

Injection of recombinant interleukin-6 (IL-6) into mice enhances recovery from infection with Listeria monocytogenes. In this study, the role of IL-6 during primary and secondary Listeria infection was further tested. Neutralization of IL-6 by polyclonal antibody exacerbated primary infection and significantly delayed gamma interferon production by cultured spleen cells. In contrast, administration of anti-IL-6 antibody at the time of secondary infection did not affect the recovery of mice from infection or gamma interferon production, showing that activated T cells are not dependent on IL-6.

[An arbovirus study of pregnant women in Southern Moravia].

The authors revealed by means of the haemagglutination-inhibition test (HIT) and the plaque reduction neutralization test (PRNT) that in 1983 to 1985 pregnant women in the Breclav district became infected with the Tahyna virus. HI antibodies against the virus were detected in 96 (28.4%) women from a total of 338 examined. A new infection was proved in 15 (4.4%) women. The majority of infections was contracted in August to October. Positive results were supported by the PRNT. Antibodies against arboviruses Sindbis, Calovo, West Nile, tick-borne encephalitis, Bhandza were not detected in any of

A Test-Negative Design for Immune Correlates Approximates a Traditional Exposure-Proximal Design but Requires Far Fewer Blood Samples.

Traditional vaccine clinical trials sample blood from all participants. In contrast, the test-negative immune correlates (TNIC) design only samples blood from participants who develop symptoms. We compared traditional to test-negative immune correlates methods in the mRNA-1273 severe acute respiratory syndrome coronavirus 2 vaccine efficacy clinical trial. Using a neutralizing antibody assay, hazard ratios were 0.48 (95% confidence interval [CI], .29-.73) and 0.55 (95% CI, .28-1.06) for traditional and test-negative methods, respectively. Analogous ratios for binding antibody assay were 0.69 (

Exposure history shapes SARS-CoV-2 viral dynamics in non-human primates and provides insights into correlates of protection against infection and transmission.

In the SARS-CoV-2 Omicron-variant era, the efficacy of vaccination in preventing viral replication is poorly understood and is further complicated by heterogeneous and hybrid immunity in the population. Here, we analysed by mathematical models the kinetics of both viral load and antibodies in non-human primates with different exposure histories, conferred by infection or vaccination with mono- or bi-valent vaccines. The model describes the control of viral replication by antibodies that accelerate the clearance and reduce the infectivity of viral particles, and suggests that a previous infecti

Protective immunity in hamsters from an oral Nipah vaccine correlates with pseudovirus neutralising antibody titre.

Nipah virus (NiV) is a deadly zoonotic pathogen with a high mortality rate and no approved vaccines, creating an urgent need for new solutions. We developed a novel, thermostable, orally administered NiV vaccine using a human serotype 5 adenovirus (AdHu5) vector to express NiV glycoproteins G and F. This vaccine, delivered via an enteric-coated capsule using the OraPro platform, eliminates the cold chain and allows for self-administration. In a Syrian hamster model, a prime-and-pull strategy—an intramuscular prime followed by an oral booster—provided 100% protection against a lethal NiV challe

Humoral immunity induced by XEC monovalent vaccines against SARS-CoV-2 variants including XEC, LP.8.1, NB.1.8.1, XFG, and BA.3.2.

The Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) Omicron variant has continuously diverged, and multiple of its sublineages are circulating worldwide. To reduce the coronavirus disease 2019 (COVID-19) with infection of the SARS-CoV-2 Omicron sublineages, some types of variant-matched vaccines have been developed and approved, such as Omicron JN.1-based vaccines and Omicron LP.8.1-based vaccines, in multiple countries, including Japan. Since, at the beginning of 2025, the Omicron XEC variant was predominant over other variants in Japan, two Japanese pharmaceutical companies have

The Dengue NS1 epitope landscape: a blueprint for next-generation vaccines and therapeutics.

The dengue virus (DENV) nonstructural protein 1 (NS1) is a critical 'double-edged sword' in dengue pathogenesis and immunity. While a key driver of vascular pathology, it is also a major target for protective immune responses that lack the risk of antibody-dependent enhancement. Anti-NS1 antibodies can neutralize NS1's pathogenic functions or clear infected cells; however, pathogenic responses can be driven by cross-reactive antibodies, causing cellular dysfunction or by NS1-antibody immune complexes triggering inflammatory damage. Concurrently, T-cell responses target conserved epitopes to dr

Convalescent Plasma Therapy in Immunocompromised Patients Infected With the BA.1 or BA.2 Omicron SARS-CoV-2.

The emergence of SARS-CoV-2 Omicron variant has led to a complete reconfiguration of the therapeutic landscape, with all monoclonal antibodies having lost any neutralization activity. We report here a case series of 75 immunocompromised patients infected by the Omicron variant who benefited from COVID-19 convalescent plasma (CCP). At Day 28, the overall survival was 76% (95% CI 67-86) with no significant difference in the clinical outcome between patients with hematological malignancies, solid organ transplantation or autoimmune diseases. No safety concern was reported during the course of the

Evaluation of bird-adapted self-amplifying mRNA vaccine formulations in chickens.

Each year, millions of poultry succumb to highly pathogenic avian influenza A virus (AIV) and infectious bursal disease virus (IBDV) infections. Conventional vaccines based on inactivated or live-attenuated viruses are useful tools for disease prevention and control, yet, they often fall short in terms of safety, efficacy, and development times. Therefore, versatile vaccine platforms are crucial to protect poultry from emerging viral pathogens. Self-amplifying (replicon) RNA vaccines offer a well-defined and scalable option for the protection of both animals and humans. The best-studied replic

Antibody targeting of conserved sites of vulnerability on the SARS-CoV-2 spike receptor-binding domain.

Given the continuous emergence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variants of concern (VoCs), immunotherapeutics that target conserved epitopes on the spike (S) glycoprotein have therapeutic advantages. Here, we report the crystal structure of the SARS-CoV-2 S receptor-binding domain (RBD) at 1.95 Å and describe flexibility and distinct conformations of the angiotensin-converting enzyme 2 (ACE2)-binding site. We identify a set of SARS-CoV-2-reactive monoclonal antibodies (mAbs) with broad RBD cross-reactivity including SARS-CoV-2 Omicron subvariants, SARS-CoV-1, an

Generation of a thermostable, oral Zika vaccine that protects against virus challenge in non-human primates.

Here we report the development of a thermally stable, orally administered, candidate Zika vaccine using human serotype 5 adenovirus (AdHu5). We engineered AdHu5 to express the genes for the envelope and NS1 proteins of Zika virus. AdHu5 was formulated using a proprietary platform, OraPro, comprising a mix of sugars and modified amino acids that can overcome elevated temperatures (37 C), and an enteric coated capsule that protects the integrity of the AdHu5 from the acid in the stomach. This enables the delivery AdHu5 to the immune system of the small intestine. We show that oral delivery of Ad

Propagation and immunological characterization of coxsackievirus A10 in a serum-free HEK293A cell culture system.

Coxsackievirus A10 (CVA10) is one of enteroviral pathogens that cause the hand, foot, and mouth disease (HFMD). Since CVA10 was reported to be not easily propagated in the Vero cell culture, a feasible manufacture process for producing formalin-inactivated CVA10 vaccine is urgently needed. Several cell lines that commonly used for viral vaccine production was tested for CVA10 (M2014 strain) culture in this study, and our result showed that CVA10 could be easily propagated in the HEK293A cells. A serum-free HEK293A cell culture system was developed for CVA10 production and the yields have reach

Act Early and at the Right Location: SARS-CoV-2 T Cell Kinetics and Tissue Localization.

The emergence of new SARS-CoV-2 lineages able to escape antibodies elicited by infection or vaccination based on the Spike protein of the Wuhan isolates has reduced the ability of Spike-specific antibodies to protect previously infected or vaccinated individuals from infection. Therefore, the role played by T cells in the containment of viral replication and spread after infection has taken a more central stage. In this brief review, we will discuss the role played by T cells in the protection from COVID-19, with a particular emphasis on the kinetics of the T cell response and its localization

Evaluation of Zika virus DNA vaccines based on NS1 and domain III of E.

BACKGROUND: A large-scale outbreak of Zika virus (ZIKV) has occurred in Brazil and other South American countries, and has rapidly spread to 60 countries and regions worldwide since 2015, but no approved anti-ZIKV vaccines are available as of 2021. METHODS: We developed four types of anti-ZIKV DNA vaccine candidates: VPC-NS1, VPC-prME, VPC-prME-NS1, and VPC-EIII-NS1. They were developed against the structural proteins prM and E, and non-structural protein 1 (NS1) of ZIKV using the mammalian cell expression vector pcDNA3.1(+) as the backbone. For immunization, we intramuscularly injected mice

The National COVID-19 Clinical Evidence Taskforce: pregnancy and perinatal guidelines.

INTRODUCTION: Pregnant women are at higher risk of severe illness from coronavirus disease 2019 (COVID-19) than non-pregnant women of a similar age. Early in the COVID-19 pandemic, it was clear that evidenced-based guidance was needed, and that it would need to be updated rapidly. The National COVID-19 Clinical Evidence Taskforce provided a resource to guide care for people with COVID-19, including during pregnancy. Care for pregnant and breastfeeding women and their babies was included as a priority when the Taskforce was set up, with a Pregnancy and Perinatal Care Panel convened to guide cli
