Endocardial fibroelastosis is caused by aberrant endothelial to mesenchymal transition.

RATIONALE: Endocardial fibroelastosis (EFE) is a unique form of fibrosis, which forms a de novo subendocardial tissue layer encapsulating the myocardium and stunting its growth, and which is typically associated with congenital heart diseases of heterogeneous origin, such as hypoplastic left heart syndrome. Relevance of EFE was only recently highlighted through the establishment of staged biventricular repair surgery in infant patients with hypoplastic left heart syndrome, where surgical removal of EFE tissue has resulted in improvement in the restrictive physiology leading to the growth of th

Potentiating effects of MPL on DSPC bearing cationic liposomes promote recombinant GP63 vaccine efficacy: high immunogenicity and protection.

BACKGROUND: Vaccines that activate strong specific Th1-predominant immune responses are critically needed for many intracellular pathogens, including Leishmania. The requirement for sustained and efficient vaccination against leishmaniasis is to formulate the best combination of immunopotentiating adjuvant with the stable antigen (Ag) delivery system. The aim of the present study is to evaluate the effectiveness of an immunomodulator on liposomal Ag through subcutaneous (s.c.) route of immunization, and its usefulness during prime/boost against visceral leishmaniasis (VL) in BALB/c mice. The influence of liposome-encapsulated prostaglandin E1 on hydrogen peroxide concentrations in the exhaled breath of patients with the acute respiratory distress syndrome.

UNLABELLED: Hydrogen peroxide (H2O2) levels are increased in the exhaled breath of patients with the acute respiratory distress syndrome (ARDS). Because liposome-encapsulated prostaglandin E1 (PGE1) downregulates the CD11/CD18 receptor of the neutrophil, thereby limiting endothelial adhesion, the use of this drug should decrease the excretion of H2O2 in the expiratory condensate of patients with ARDS. Patients > 11 yr of age with ARDS (diffuse, patchy infiltrates by chest radiograph; Pao2/fraction of inspired oxygen [P/F] ratio < or = 200 mm Hg; pulmonary capillary wedge pressure < or = 18 mm

Nanoscale therapeutics for erectile dysfunction: a meta-analysis of stem cell-derived extracellular vesicles as natural nanoparticles in diabetic rat models.

BACKGROUND: Erectile dysfunction (ED), a prevalent male sexual disorder, severely impacts quality of life. Extracellular vesicles (EVs), natural nanoparticles (30-200 nm) secreted by stem cells, represent a novel nanomedicine platform for ED treatment due to their ability to encapsulate bioactive cargo (e.g., miRNAs, proteins) and target damaged tissues. Stem cell-derived extracellular vesicles (SC-EVs) have emerged as a promising therapeutic strategy for multiple diseases. This meta-analysis evaluates the therapeutic efficacy of SC-EVs in rat ED models and explores their translational potenti

Ionizing radiation-mediated dendritic cell maturation exacerbates inflammatory response of bone marrow mesenchymal stem cells and impairs osteogenesis in radiation-induced jaw injury.

BACKGROUND: Radiation-induced jaw injury is one of the most severe complications after radiotherapy for head and neck cancer, which can disrupt patients' health and quality of life. Although the direct target of inflammation and suppressed bone regeneration activity by ionizing radiation (IR) has been phenomenally observed, the underlying mechanisms and potential therapeutic targets remain blurred. Osteoimmunology emphasizes that dendritic cells (DCs) may contribute to bone diseases. METHODS: In this study, we assessed phenotypic and functional alterations of DCs in a radiation-induced jaw in

Enhancing paracellular and transcellular permeability using nanotechnological approaches for the treatment of brain and retinal diseases.

Paracellular permeability across epithelial and endothelial cells is, in large part, regulated by apical intercellular junctions also referred to as tight junctions (TJs). These junctions contribute to the spatial definition of different tissue compartments within organisms, separating them from the outside world as well as from inner compartments, with their primary physiological role of maintaining tissue homeostasis. TJs restrict the free, passive diffusion of ions and hydrophilic small molecules through paracellular clefts and are important for appropriate cell polarization and transporter

Pre-Conditioned Bone Marrow Mesenchymal Stromal Cell-Derived Secretome Exerts an Anti-Inflammatory Effect on Degenerative Nucleus Pulposus Cells In Vitro.

BACKGROUND: Intervertebral disc degeneration (IDD) is frequently associated with chronic low back pain (LBP), which contributes significantly to disability, psychological distress, and reduced work capacity. Mesenchymal stromal cell (MSC)-based treatments may offer a promising, less invasive alternative to conventional treatments for IDD. The MSC secretome has shown regenerative potential through paracrine and anti-inflammatory mechanisms. In this study, we investigated the effects of the secretome isolated from interleukin-1β (IL-1β)-preconditioned bone marrow-derived MSCs (BM-MSCs) on human

Mesenchymal stem cells-derived exosomes carrying microRNA-30b confer protection against pulmonary fibrosis by downregulating Runx1 via Spred2.

Idiopathic pulmonary fibrosis (IPF) is a chronic pulmonary fibrosis disease that is fatal. Mesenchymal stem cells (MSCs)-secreted exosomes (exos) have been linked to improving PF. Moreover, exosomal microRNAs (miRs) can control the growth of numerous diseases, including lung disorders. Our bioinformatics analysis showed that miR-30b was downregulated in tissue samples from surgical remnants of biopsies or lungs explanted from patients with IPF who underwent pulmonary transplantation. This suggests that miR-30b plays an important role in both the pathogenesis and treatment of IPF. Herein, this

Liposomes in investigative dermatology.

Liposomes are microscopic spheres, usually composed of amphiphilic phospholipids. They may be useful without skin penetration if they simply protect or sequester compounds that would otherwise be unstable in the formulation. Liposomes that remain on the skin surface are useful as light-absorbers, agents to deliver color or sunscreens, or as depots for timed-release. Liposomes that penetrate the stratum corneum have the potential to interact with living tissue. Topically applied liposomes can either mix with the stratum corneum lipid matrix or penetrate the stratum corneum by exploiting the lip

Sequential KIM-1 targeting and SMOX-responsive release enhance renal delivery of dihydroartemisinin to restore endogenous Klotho and attenuate renal fibrosis.

Restoration of endogenous Klotho represents a promising strategy for suppressing renal fibrosis, but efficient delivery of Klotho-restoring therapeutics to injured renal tubules remains challenging. Here, we developed dihydroartemisinin (DHA)-loaded phosphatidylserine liposomes coated with spermine-modified chitosan (DHA-PSLPs@SC) to achieve sequential kidney injury molecule-1 (KIM-1) targeting and spermine oxidase (SMOX)-responsive release, thereby enhancing renal delivery of DHA to restore endogenous Klotho and attenuate renal fibrosis. Decellularized Extracellular Matrix-Based Tunable 3D Hydrogel: An Alternative Methodology for the Development of a Doxorubicin-Independent 3D Breast Cancer Microphysiological Chemoresistance Model.

Breast cancer is the most prevalent malignancy among women and the second leading cause of cancer-related mortality worldwide, with 2.26 million cases and 685,000 deaths in 2020. Its complex nature complicates treatment, and emerging evidence shows that extracellular matrix (ECM) biomechanics, particularly stiffness, dynamically change in response to physiological and pathological cues. These alterations influence malignant cell morphology and promote chemoresistance by regulating tumor growth, invasion, and metastasis. Clinically, breast tumor ECM stiffness (2-100 kPa) is markedly higher than

Characterization of andrographolide-loaded exosome-like nanoparticles from Nauclea officinalis and their modulation of inflammatory and lipid metabolic pathologies in WRL68 cells.

Andrographolide (AG), a natural diterpenoid compound derived from Andrographis paniculata, exhibits potential against non-alcoholic steatohepatitis (NASH). However, its therapeutic utility is limited by poor solubility, short half-life, and low bioavailability. This study aimed to enhance AG's efficacy by encapsulating it into exosome-like nanoparticles (ELNs) isolated from Nauclea officinalis (N. officinalis, Rubiaceae family) and evaluating its anti-NASH activity in WRL68 cells. This study isolated Nauclea officinalis-derived ELNs (N-ELNs) via ultracentrifugation. Loganin, andrographolide, a

Sequential Mannose-Targeted Nanotherapy Orchestrates Tumor-Associated Macrophage Repolarization and Chemotherapeutic Response in Triple-Negative Breast Cancer.

Recently, explorations are being undertaken involving chemo-immunotherapy based on targeting cancer cells and tumor-associated macrophages (TAMs). However, these strategies mainly focus on delivering TAM modulators and chemotherapy simultaneously, without considering the effect of chemotherapy on TAMs and which may compromise TAM-modulatory action. This necessitates investigations involving sequential treatment wherein pretreatment would enable efficient M2-to-M1 repolarization before administration of chemotherapy. In this study, mannose-targeted liposomes encapsulating resiquimod (M-LIPO-RES

Mitochondria-targeting pseudo-stealthy nanophotosensitizer as a potent immunogenic cell death inducer to unleash the cancer-immunity cycle for melanoma therapy.

Immunotherapy with immune checkpoint blockade (ICB) has revolutionized cancer treatment for patients with unresectable melanoma; however, the majority of patients who harbor immunologically "cold" tumors exhibit poor outcomes. Mitochondria serve as a central hub for enhancing antitumor immune responses. Thus, selective intervention strategies targeting them can rewire mitochondrial metabolism and induce robust immunogenicity in immune-desert tumors. This study describes the development of a pseudo-stealthy nanophotosensitizer platform that effectively induces immunogenic cell death (ICD) in tu

Mitochondrial DNA-Mediated Immune Activation After Resuscitation From Cardiac Arrest.

BACKGROUND: Postcardiac arrest syndrome is characterized by systemic inflammation that contributes to poor outcomes after resuscitation from sudden cardiac arrest. Mitochondrial DNA (mtDNA) has been implicated as a proinflammatory stimulus in other contexts, but its role in postcardiac arrest syndrome is unclear. We determined if postcardiac arrest syndrome is characterized by a rise in circulating mtDNA, how mtDNA activates immune cells, and if targeting mtDNA-sensing pathways attenuates leukocyte activation. METHODS: Plasma mtDNA and nuclear DNA levels were measured ~4-hours after return of

Antioxidants in cancer therapy mitigating lipid peroxidation without compromising treatment through nanotechnology.

BACKGROUND: Cancer treatments often exploit oxidative stress to selectively kill tumour cells by disrupting their lipid peroxidation membranes and inhibiting antioxidant enzymes. However, lipid peroxidation plays a dual role in cancer progression, acting as both a tumour promoter and a suppressor. Balancing oxidative stress through antioxidant therapy remains a challenge, as excessive antioxidant activity may compromise the efficacy of chemotherapy and radiotherapy. AIM: This review explores the role of antioxidants in mitigating lipid peroxidation in cancer therapy while maintaining treatmen

Exosomal Communication Between Cumulus-Oocyte Complexes and Granulosa Cells: A New Molecular Axis for Oocyte Competence in Human-Assisted Reproduction.

Exosomal microRNAs (ex-miRs), encapsulated in extracellular vesicles (EVs), play a vital role in facilitating paracrine communication among granulosa cells (GCs), cumulus cells (CCs), and the oocyte inside follicular fluid (FF). These small non-coding RNAs are crucial for regulating folliculogenesis, oocyte maturation, and early embryonic development via modulating intracellular signaling networks. Dysregulation o has been associated with reproductive disorders such as polycystic ovarian syndrome (PCOS), diminished ovarian reserve (DOR), and inadequate ovarian response (POR), impacting oocyte
