The recent approval in the United States of the first adeno-associated viral (AAV) vector for the treatment of an inherited retinal degeneration validates this approach for the treatment of many other diseases. A major limiting factor continues to be the size restriction of the AAV transgene at under 5 kb. Stargardt disease is the most prevalent form of recessively inherited blindness and is caused by mutations in ABCA4, the gene that codes for ATP-binding cassette transporter protein family member 4, which has a coding sequence length of 6.8 kb. Dual vector approaches increase the capacity of

Antibodies (Abs) play a central role in human immunodeficiency virus (HIV) protection due to their multiple functional inhibitory activities. W614A-3S Abs recognize a specific form of a highly conserved motif of the gp41 envelope protein and can elicit viral neutralization to protect CD4+ T cells. Here, we describe in detail the neutralizing profile of W614A-3S Abs in untreated long-term non-progressor (LTNP) HIV-infected patients. W614A-3S Abs were detected in 23.5% (16/68) of untreated LTNP patients compared with <5% (5/104) of HIV-1 progressor patients. The W614A-3S Abs had efficient neutra

Malignant melanoma represents a neoplasm stemming from melanocytes or the cells that develop from melanocytes. Melanocytes, pigment-producing cells, arise from the neural crest and migrate to their final destinations in the skin, uveal tract, meninges, and mucosa. Most melanocytes are found at the epidermal-dermal junction of the skin, and the vast majority of melanocytes arise from cutaneous sites. Cancerous growths develop when unrepaired DNA damage to skin cells (most often caused by ultraviolet radiation from sunshine or tanning beds) triggers mutations (genetic defects) that lead the skin

A high resistance and heterogeneous response to conventional anti-cancer chemotherapies characterize malignant cutaneous melanoma, the most aggressive and deadly form of skin cancer. Withaferin A (WFA), a withanolide derived from the medicinal plant Withania somnifera, has been reported for its anti-tumorigenic activity against various cancer cells. For the first time, we examined the death-inducing potential of WFA against a panel of four different human melanoma cells and investigated the cellular mechanisms involved. WFA induces apoptotic cell death with various IC50 ranging from 1.8 to 6.1

Radiotherapy for head and neck tumors often results in persistent loss of function in salivary glands. Patients suffering from impaired salivary function frequently terminate treatment prematurely because of reduced quality of life caused by malnutrition and other debilitating side-effects. It has been previously shown in mice expressing a constitutively active form of Akt (myr-Akt1), or in mice pretreated with IGF1, apoptosis is suppressed, which correlates with maintained salivary gland function measured by stimulated salivary flow. Induction of cell cycle arrest may be important for this pr

For cloning the cytokine human Midkine (MK) gene, we designed by PCgene program and synthesized a pair of PCR specific primers according to the reported human MK cDNA sequence. Total cellular RNA was extracted from a human hepatoblastoma cell line HepG2, and then the target DNA fragment was obtained by RT-PCR and subcloned into plasmid pUC118. Checked with radioisotope sequencing and ABI 377A sequencer, the nucleotide sequence of the cloned MK cDNA was identical with the reported one. A prokaryotic expression vector, named pBV220, was used to express the MK protein efficiently in E. coli strai

Thanatophoric dysplasia (TD) is the most frequent form of neonatal lethal skeletal dysplasia. Recently. mutations in the fibroblast growth factor receptor 3 (FGFR3) gene that cause two subtypes of this disorder, type I (TDI) and type II (TDII), have been identified. This discovery has now made it possible to make a definite diagnosis of TD by molecular methods. To date, prenatal diagnosis of TD has been accomplished by ultrasonography in the second trimester. However, it is not always possible to distinguish TD fetuses it utero from the other osteochondrodysplasias by ultrasonography or radiog

Anti-p80-coilin antibody produces a unique pattern of immunofluorescence staining called nuclear dots characterized by the presence of up to six discrete nuclear bodies in interphase cell nuclei. The distribution of this antibody and its clinical relevance have not been established. We studied the prevalence of anti-p80-coilin antibody in the sera from 810 patients with collagen diseases and various skin diseases. Five sera showed the nuclear dot pattern by indirect immunofluorescence on an HEp-2 cell substrate, and reacted by immunoblotting with 80-kDa protein in a nuclear extract from HeLa c

Genetic predisposition to development of the idiopathic inflammatory myopathies is probably multifactorial. Major histocompatibility complex associations with these diseases provide the strongest evidence for a genetic component. In Caucasoids, haplotypes marked by B8/DR3 are associated with each of the clinical subgroups, except mixed connective tissue disease (DR4). The strongest associations are with inclusion body myositis, polymyositis in the presence of anti-Jo-1, and with antibodies to PM-Scl in overlap syndromes. The underlying mechanisms of these associations are probably different. U

African trypanosomes (Trypanosoma brucei species) are parasitic protozoa that cause lethal diseases in humans and cattle. Previous studies showed that camptothecin, a potent and specific inhibitor of DNA topoisomerase I, is cytotoxic to African trypanosomes and related pathogenic hemoflagellates (Bodley AL and Shapiro TA, Proc Natl Acad Sci USA 92: 3726-3730, 1995). In this study, a series of camptothecin analogs was tested against axenically cultured, bloodstream form, T. brucei. Modifications to the pentacyclic nucleus of camptothecin ablated antiparasitic activity. In contrast, activity cou

Genomic DNA can adopt noncanonical secondary structures, such as G-quadruplexes (G4s), which serve as critical regulatory elements beyond the traditional double helix. Whether promoter G4s also participate in controlling innate immune signaling and host-pathogen interactions, however, remains unknown in arthropods. In this study, we identify a G4 structure within the promoter of the NEMO (NF-κB essential modulator) gene in the deer tick Ixodes scapularis. Using a combination of circular dichroism, isothermal difference spectra, and fluorescence-based assays, we demonstrate that this sequence f

R-loops are three-stranded nucleic acid structures consisting of an RNA:DNA hybrid and a displaced single-stranded DNA, typically formed during transcription. Emerging evidence indicates that R-loops are not merely transcriptional byproducts, but serve as functional regulatory structures that influence chromatin organization, transcriptional pausing, and RNA processing. However, dysregulated accumulation of R-loops can induce DNA damage and genomic instability, necessitating precise mechanisms for their regulation. This study aims to elucidate the role of the RNA-binding protein FUS (Fused in

Ornithinibacillus liuyingii sp. nov. was proposed, with type strain 27F3T (= GDMCC 1.5626T = JCM 38003T) isolated from the lung of Apodemus chevrieri collected on the Guizhou Plateau, China. Phylogenetic distinctiveness from its closest relative, Ornithinibacillus scapharcae TW25T, was confirmed by 98.5% 16S rRNA gene similarity, 76.98% average nucleotide identity, and 21.9% digital DNA–DNA hybridization value. Phenotypically, strain 27F3T was characterized as a motile, rod-shaped bacterium exhibiting a Gram-stain-negative reaction, an atypical feature within the otherwise Gram-positive genus

Bloom-forming cyanobacteria frequently exhibit "luxury phosphorus uptake" and polyphosphate (PolyP) accumulation under nitrate-limited conditions, yet the underlying molecular regulatory mechanisms remain unclear. Our previous study quantitatively demonstrated PolyP accumulation in Microcystis aeruginosa under nitrate limitation. Building on this foundation, the present study investigated the molecular responses of M. aeruginosa to nitrate limitation using integrated transcriptomic and physiological approaches. Nitrate limitation markedly suppressed nitrogen assimilation, as evidenced by reduc

The taxonomic boundaries of Pseudomonas moraviensis remain unclear despite the increasing availability of genomic data. In this study, we performed a comprehensive polyphasic taxonomic reassessment of 30 publicly available strains, together with 2 newly isolated wheat rhizosphere strains, S9433 and S9602, which exhibited genomic and phenotypic traits indicative of plant growth-promoting potential. Phylogenetic and phylogenomic analysis consistently revealed that strains currently assigned to P. moraviensis are distributed across several distinct lineages. Overall genomic relatedness indices, i

Genome analysis of the genus Pseudonocardia was performed based on 92 single-copy core genes of 56 genome sequences including that of an environmental strain, DLS-67T. The genus formed a well-defined monophyletic clade but was further divided into two clusters confirmed by coherent tree topology and amino acid identity values. Most of the species were assigned to the Pseudonocardia I (core cluster), while three species (Pseudonocardia acaciae DSM 45401T, Pseudonocardia eucalypti CM 18303T and Pseudonocardia spinosispora DSM 44797T) were included in the second cluster (Pseudonocardia II). Leaf senescence, the final stage of leaf development, is regulated by complex interplays of intrinsic genetic programs and environmental cues. Throughout their lifetimes, all living organisms encounter various endogenous and environmental challenges, many of which can cause potentially fatal DNA damage. Among these, DNA-protein crosslinks (DPCs) are particularly deleterious, as they obstruct essential processes such as replication and transcription, thereby compromising genome integrity and ultimately leading to premature aging across species. However, the biological significance of DPCs and t

Mitochondria are involved in electron transport for energy production, generating cellular ATP through oxidative phosphorylation, which in turn generates reactive oxygen species (ROS). When mitochondrial ROS exceeds the ability of antioxidant enzymes to scavenge free radicals, it leads to mitochondrial dysfunction. In this study, we used astaxanthin, a well-known natural anti-aging compound, but its unique role in protecting yeast cells with mitochondrial dysfunction is poorly understood. Pre-treatment of astaxanthin significantly increased the viability of these mutant strains (bcs1∆, cox10∆,

The metal-organic framework (MOF) material ZIF-8 was used as a nanocarrier to construct a platform capable of simultaneously delivering L-arginine and functional nucleic acid molecules. This system enables real-time monitoring of the spatial displacement of apoptosis-related enzymes as well as monitoring of the early stages of apoptosis during NO-mediated tumor therapy. ZIF-8 exhibits excellent biocompatibility and acid sensitivity, allowing it to degrade in the tumor microenvironment and release its functional molecules. In this platform, L-arginine and functional nucleic acid molecules that
