Isolation, Culture, and Osteogenic Induction of Cardiac Fibroblasts from Adult Mouse Hearts.

Cardiac fibroblasts (CFs) are the predominant non-myocyte population in the heart and play essential roles in maintaining myocardial architecture and mediating tissue remodeling and fibrosis. Beyond their well-established fibrogenic functions, CFs exhibit substantial phenotypic plasticity and can acquire osteogenic characteristics, potentially contributing to pathological cardiac calcification following severe myocardial injury. Despite growing interest in the osteogenic potential of CFs, standardized and reproducible methods for their isolation and osteogenic induction remain limited. Here, w

Dual-function oral nanotherapeutic mitigates sepsis-like multi-organ failure by targeting inflammatory and fibrotic pathways.

Sepsis, a life-threatening condition, often leads to multi-organ failure and has limited treatment options. We developed a novel dual-function nanoparticle, P2Ns-NAR(NAR), which utilizes naringenin (NAR) as both a targeting ligand for gut folate receptors and an encapsulated therapeutic agent to overcome its poor oral bioavailability. Here, we investigated the efficacy of this oral formulation in a mouse model of lipopolysaccharide-induced sepsis. We observed a significant reduction in the mRNA expression of pro-inflammatory (Tlr4, NF-κB, and IL-18), apoptotic (p53, and Fas), fibrosis (TGFβ1 a

Applied Anatomy of Perimembranous Ventricular Septal Defect for Transcatheter Device Closure.

Perimembranous ventricular septal defect is a common congenital heart disease effectively treated by transcatheter device closure with high success rates and minimal complications. This review categorizes perimembranous ventricular septal defects into 7 morphologic variants based on anatomical extension and correlates these with echocardiographic classifications: type 1 is central perimembranous; type 2 is superior-anterior extension; type 3 is superior-posterior extension; type 4 is inferior-anterior extension; type 5 is inferior-posterior extension; type 6 is confluent extensions with more t

Pathogenesis, histopathology, and mechanisms of pulmonary pathology secondary to chronic left heart failure in animals.

The pulmonary consequences of congestive heart failure in domestic animals, particularly from clinical and gross morphologic perspectives, have been described. However, the full spectrum of mechanistic and microscopic alterations, especially at the molecular and cellular levels, remains less well integrated in the context of veterinary pathology. We examine the pathogenesis and consequences of pulmonary hypertension secondary to left heart failure, in which chronic elevation of hydrostatic pressure triggers complex molecular, physiologic, and morphologic responses. These include the epithelial

Integrated omics identifies molecular networks of brain-body interactions of brain injury.

The molecular mechanisms of brain-body interactions in the progression of brain diseases remain unknown. Through integrative analysis of multi-organ proteomic, metabolomic, and transcriptomic data following ischemic stroke, we identified dynamic molecular signatures across organs. The heart exhibited the highest number of differentially expressed proteins (DEPs), followed by spleen and intestine. Ten DEPs were shared among three organs post-stroke, with eight in heart, six in spleen, and five in intestine. Notably, organs displayed accelerated biological aging, particularly the intestine, ‌whi

RNA Therapies in Cardio-Kidney-Metabolic Syndrome: Advancing Disease Management.

Cardio-Kidney-Metabolic (CKM) Syndrome involves metabolic, kidney, and cardiovascular dysfunction, disproportionately affecting disadvantaged groups. Its staging (0-4) highlights the importance of early intervention. While current management targets hypertension, heart failure, dyslipidemia, and diabetes, RNA-based therapies offer innovative solutions by addressing molecular mechanisms of CKM Syndrome. Emerging RNA treatments, including antisense oligonucleotides (ASOs) and small interfering RNAs (siRNAs), show promise in slowing disease progression across CKM stages. For example, ASOs and siR

Role of Perilipins in Oxidative Stress-Implications for Cardiovascular Disease.

Oxidative stress is the imbalance between the production of reactive oxygen species (ROS) and antioxidants in a cell. In the heart, oxidative stress may deteriorate calcium handling, cause arrhythmia, and enhance maladaptive cardiac remodeling by the induction of hypertrophic and apoptotic signaling pathways. Consequently, dysregulated ROS production and oxidative stress have been implicated in numerous cardiac diseases, including heart failure, cardiac ischemia-reperfusion injury, cardiac hypertrophy, and diabetic cardiomyopathy. Lipid droplets (LDs) are conserved intracellular organelles tha

Reversing Immune Checkpoint Inhibitor-Associated Cardiotoxicity via Bioorthogonal Metabolic Engineering-Driven Extracellular Vesicle Redirecting.

The cardiotoxicity induced by immune checkpoint inhibitors (ICIs) is associated with high mortality rates. T cells play an important role in ICI-induced cardiac injury. The inhibition of local T-cell activity is considered an effective strategy for alleviating ICI-related cardiotoxicity. Tumor-derived extracellular vesicles (EVs) contribute to immunosuppression via PD-L1 overexpression. In this study, a bioorthogonal metabolic engineering-driven EV redirecting (Biomeder) strategy for in situ engineered EVs with myocardial-targeting peptides is developed. Accumulated tumor-derived EV (TuEVs) re

Role and mechanism of miRNA in cardiac microvascular endothelial cells in cardiovascular diseases.

The occurrence and development of myocardial dysfunction are associated with damage in the cardiac microvascular endothelial cells (CMECs), which can regulate nutrient exchange and oxy-gen-carbon cycling to protect cardiomyocytes. Interventions targeting microRNAs (miRNAs) can effectively mitigate CMEC injury and thus improve cardiovascular diseases. MiRNAs are a class of noncoding single-strand RNA molecules typically 21-23 nucleotides in length that are encoded by endogenous genes. They are critical regulators of organism development, cell differentiation, metabolism, and apoptosis. Current

Neurorehabilitation in a Pediatric Stroke Patient Supported on a CentriMag.

Patients supported on ventricular assist devices (VADs) benefit from rehabilitation while awaiting heart transplantation to recover from surgery, prevent deconditioning, and, in most cases, optimize transplant candidacy. With bleeding and neurological dysfunction as the most common VAD complications, the importance of rehabilitation dramatically increases when a patient on a VAD also suffers from a neurological injury. The rehabilitation needs for cardiac conditioning and neurological reeducation are not the same. Patients with severe neurological deficits require intense rehabilitation that o

Revisiting Biomarkers of Cardiac Allograft Vasculopathy: Addressing the Achilles Heel of Heart Transplantation.

Nearly half of heart transplant recipients will be diagnosed with cardiac allograft vasculopathy (CAV) within five years after transplantation. Advanced CAV can lead to worsening heart failure as well as arrhythmias and sudden cardiac death. The only curative therapy for end-stage CAV is re-transplantation. Current diagnostic methods are invasive and limited by poor sensitivity in early disease. Despite its high prevalence in the post-transplantpopulation, the underlying pathophysiology of this condition has yet to be fully described. It is thought to be primarily related to endothelial dysfun

The Review of Current Knowledge on Neutrophil Gelatinase-Associated Lipocalin (NGAL).

Neutrophil gelatinase-associated lipocalin (NGAL) is a 25-kDa protein that is secreted mostly by immune cells such as neutrophils, macrophages, and dendritic cells. Its production is stimulated in response to inflammation. The concentrations of NGAL can be measured in plasma, urine, and biological fluids such as peritoneal effluent. NGAL is known mainly as a biomarker of acute kidney injury and is released after tubular damage and during renal regeneration processes. NGAL is also elevated in chronic kidney disease and dialysis patients. It may play a role as a predictor of the progression of r

Fatality in the perioperative period: A case report on refractory hypoxemia in an infant following open inguinal herniotomy.

INTRODUCTION AND IMPORTANCE OF THE CASE: Post-operative adverse respiratory events are not uncommon among infants. PRESENTATION OF CASE: A 2-month-old male infant with an acyanotic heart disease underwent an elective open inguinal herniotomy under general anaesthesia. The intraoperative period was uneventful. The infant developed intermittent respiratory apnea and low oxygen saturation, followed by bradycardia in the post-anaesthesia care unit. Despite continued resuscitative efforts, the baby succumbed. The autopsy did not reveal new pathology. A period of interrupted monitoring during the r

A Novel Role of Semaphorin 3C in Modulating Systemic and Renal Hemodynamics.

BACKGROUND: Alterations of renal hemodynamics play an essential role in renal homeostasis and kidney diseases. Recent data indicated that semaphorin 3C (SEMA3C), a secreted glycoprotein involved in vessel development, can modulate renal vascular permeability in acute kidney injury, but whether and how it might impact systemic and renal hemodynamics is unknown. OBJECTIVES: The objective of the study was to explore the effect of SEMA3C on systemic and renal hemodynamics. METHODS: SEMA3C recombinant protein was administered intravenously in two-month-old wild-type mice, and the variations of me

Vagally Associated Second Degree Atrio-Ventricular Block in a Dog with Severe Azotemia and Evidence of Sympathetic Overdrive.

A 14 years old, 6 kg, mix-breed male dog with severe azotemia due to urinary bladder herniation was presented to our Veterinary Teaching Hospital (VTH). Electrocardiography revealed normal heart rate of 100 bpm, evidence of sinus respiratory arrhythmia (SRA) and frequent second degree atrio-ventricular block following peak inspiratory phase suggestive of vagally-induced atrio-ventricular conduction delay. Echocardiographic examination showed mild mitral regurgitation without any other cardiac changes, and systolic (SAP) and diastolic (DAP) blood pressure values were 185/90 mmHg (SAP/DAP). Card

Combining stem cells in myocardial infarction: The road to superior repair?

Myocardial infarction irreversibly destroys millions of cardiomyocytes in the ventricle, making it the leading cause of heart failure worldwide. Over the past two decades, many progenitor and stem cell types were proposed as the ideal candidate to regenerate the heart after injury. The potential of stem cell therapy has been investigated thoroughly in animal and human studies, aiming at cardiac repair by true tissue replacement, by immune modulation, or by the secretion of paracrine factors that stimulate endogenous repair processes. Despite some successful results in animal models, the outcom

Single-cell transcriptomics defines heterogeneity of epicardial cells and fibroblasts within the infarcted murine heart.

In the adult heart, the epicardium becomes activated after injury, contributing to cardiac healing by secretion of paracrine factors. Here, we analyzed by single-cell RNA sequencing combined with RNA in situ hybridization and lineage tracing of Wilms tumor protein 1-positive (WT1+) cells, the cellular composition, location, and hierarchy of epicardial stromal cells (EpiSC) in comparison to activated myocardial fibroblasts/stromal cells in infarcted mouse hearts. We identified 11 transcriptionally distinct EpiSC populations, which can be classified into three groups, each containing a cluster o
