Genetic analysis of lice supports direct contact between modern and archaic humans.

Parasites can be used as unique markers to investigate host evolutionary history, independent of host data. Here we show that modern human head lice, Pediculus humanus, are composed of two ancient lineages, whose origin predates modern Homo sapiens by an order of magnitude (ca. 1.18 million years). One of the two louse lineages has a worldwide distribution and appears to have undergone a population bottleneck ca. 100,000 years ago along with its modern H. sapiens host. Phylogenetic and population genetic data suggest that the other lineage, found only in the New World, has remained isolated fr

[Relationship of the flea Citellophilus tesquorum altaicus (Siphonaptera: Ceratophyllidae) with the plague agent].

Some aspects of relationships of the flea Citellophilus tesquorum altaicus and bacterium Yersinia pestis of two strains isolated from different parts of the Tuva natural plague focus were studied. Peculiarities of elimination and blood meal activity of fleas infected with two strains of the plague agent were not revealed. Differences in mortality and alimentary activity are considerably determined by the sex of insects. The ability of examined strains to form a proventriculus block was not identical in the strains examined. This ability was expressed higher in the strain I-3428, which originat

Hydrogen peroxide gas plasma sterilization is effective against Cryptosporidium parvum oocysts.

The aim of this work was to evaluate in an immunosuppressed rat cryptosporidiosis model a new method that combines vacuum and low-temperature hydrogen peroxide gas plasma for sterilization of endoscopic material contaminated by Cryptosporidium parvum. Rats were challenged with oocysts either air-dried or air-dried and treated with vacuum alone or associated with gas plasma. No rat was found infected after gas plasma exposure of oocysts, whereas vacuum or air-drying alone resulted only in a decreased infectivity.

Interferon-gamma in 388 immunocompromised and immunocompetent patients for diagnosing pleural tuberculosis.

The level of interferon-gamma (IFN-gamma) in pleural fluid has been reported to be increased in pleural tuberculosis. Nevertheless, its diagnostic value has not yet been well-established, and immunocompromised patients have not previously been evaluated. The aim of this study was to determine the value of the IFN-gamma level in pleural fluid for diagnosing tuberculous pleurisy in immunocompetent and immunocompromised patients. Three hundred and eighty eight consecutive patients were studied prospectively (73 with tuberculous pleural effusions, including nine with concurrent human immunodeficie

Onset of resistance to light Hymenolepis nana infection in mice of different strains.

Rapidity in onset of resistance against Hymenolepis nana egg infection after a light primary infection was studied in low and high responder mice challenged at different time intervals. A very rapid acquisition of protection was observed in C57 and a delayed response in C3H mice. In both cases the effect of resistance on weight or worm number was related to the time of challenge infection, suggesting a "race against time" involving host response and parasite development, the outcome varying according to host genetic background.

Mining Phage Genomes for Regulatory Elements to Develop a SynBio Toolbox for Non-Model Pseudomonads.

Bacteriophages represent a largely untapped reservoir of regulatory elements that can expand the genetic toolbox for engineering non-model hosts. Using ONT-cappable-sequencing, we mapped full-length primary transcripts across five Pseudomonas phages and identified 232 transcription start sites (TSSs) and 176 transcription termination sites (TTSs), including 38 intrinsic factor-independent terminators. Motif discovery revealed 59 σ70-like promoters in KIL5, 37 in KIL3b, and unique phage-encoded RNA polymerase (RNAP) promoter motifs for KNP, ϕ2, and pphageB21, enabling discrimination between hos

Male Reproductive Phenotype Heterogeneity in Varicocele: A Hypothesis-Generating Threshold-Resilience Framework and Research Agenda.

BACKGROUND: Varicocele is common in male infertility, yet many affected men retain conventional reproductive measures, and the anatomical grade is an imperfect proxy for phenotype. Established pathways explain injury more clearly than between-person heterogeneity. METHODS: This structured, critical narrative review searched PubMed from 1 January 2015 to 26 August 2026, with targeted hand-searching. There was no preregistered protocol, dual screening, or risk-of-bias assessment, and coverage is not exhaustive. MAIN FINDINGS: The threshold-resilience framework is a hypothesis-generating model

Identification, Pathogenicity, and Chemical Control of Nigrospora oryzae, a New Pathogen Causing Mid-Late Soybean Root Rot in Heilongjiang, China.

Soybean root rot, a soil-borne fungal disease, severely impacts soybean production. Limited research has specifically addressed this disease during the mid-to-late growth stages, which represent a critical yield-limiting period for soybean. In the autumn of 2024, a total of 478 pathogenic isolates were obtained and validated following Koch's postulates. These isolates were classified into eight fungal species, among which Diaporthe longicolla was the predominant species, accounting for 72.0% of the total isolates. Notably, Nigrospora oryzae, an unreported causal agent of soybean root rot in Ch

Chemoproteomic profiling of adaptive ATP-binding protein activity in Enterococcus faecium in response to host-derived cues.

Enterococcus faecium has become a significant nosocomial and opportunistic pathogen, increasingly implicated in severe infections such as urinary tract infections, bacteraemia, and endocarditis. Due to acquired resistance to vancomycin and ampicillin and high intrinsic resistance to other antibiotics, E. faecium infections are often challenging to treat. However, relatively little is known about the molecular factors used by E. faecium during infection. Here, we employed desthiobiotin-ATP as an activity-based probe to investigate the activity profile of ATP-binding proteins in E. faecium upon

A defined synthetic algal medium enables lettuce-free culturing of unfed Paramecium bursaria while preserving host-associated microbiome composition.

Paramecium bursaria is widely cultured using undefined plant-based infusions such as lettuce extract, yet the variable composition of these media remains a major obstacle to experimental reproducibility and microbiome research. Here, we tested whether a chemically defined synthetic algal medium (AF-6) can replace conventional lettuce infusion while maintaining host physiology and associated microbial communities. An unfed clonal strain of P. bursaria, established in 2023 and capable of growth without external nutrient supplementation, proliferated comparably in AF-6 and lettuce media. To confi

Combination therapy for fungal diseases: the role of immunotherapy in the present and the future.

PURPOSE OF REVIEW: Invasive fungal infections remain associated with high morbidity and mortality despite advances in antifungal therapy. Current treatments are largely pathogen-directed and fail to address the dysregulated host immune responses that drive severe disease. Together with expanding at-risk populations and environmental changes that promote fungal adaptation, spread, and emergence resistance of fungal pathogens, this underscores the need for complementary immunotherapy. RECENT FINDINGS: Disease outcome in invasive fungal infections is determined not only by fungal burden but also

Profiling Selectivity for the Shigella Virulence Factor OspF.

Phosphothreonine lyases are a small family of bacterial virulence factors that are secreted into host cells during infection by some Gram-negative pathogens. Phosphothreonine lyases catalyze an irreversible phosphate β-elimination that converts phosphothreonine (pThr) to dehydrobutyrine (Dhb) on host proteins. Here, we explore the substrate profile for OspF, a phosphothreonine lyase secreted during Shigella flexneri infection, which has been previously reported to modify mitogen-activated protein kinases (MAPKs) to silence the host immune response. In this work, we use a combination of in vitr

Immunogenomic landscape of the T cell repertoire after human lung transplantation and its clinical relevance.

Long-term lung transplant survival is limited by rejection and infection, processes governed by T cells, the posttransplant dynamics of which are poorly defined. Using flow cytometry and T cell receptor (TCR) sequencing of serial bronchoalveolar lavage (BAL) and peripheral blood samples, we monitored T cell chimerism, phenotypes, and clonotypic dynamics in a small cohort of lung transplant recipients. We identified alloreactive clones via mixed lymphocyte reaction/TCR sequence pairing and microbial-reactive clones via database matching. We found a recipient-derived tissue-resident memory T cel

Universal CAR-T cell therapy: mechanisms and evasion strategies of bidirectional immune rejection.

While autologous chimeric antigen receptor (CAR)-T cell therapies have revolutionized the treatment of hematological malignancies, their widespread clinical application remains constrained by lengthy manufacturing cycles, prohibitive costs, and variable patient-derived cell quality. Consequently, "off-the-shelf" universal allogeneic CAR-T cells have emerged as a highly anticipated alternative. However, the clinical translation of these allogeneic therapies faces formidable immunological bottlenecks, principally driven by bidirectional immune rejection: graft-versus-host disease (GVHD) and host

The essential role of heparan sulfate in the entry of PDCoV and other porcine coronaviruses.

Porcine enteric coronaviruses, including porcine deltacoronavirus (PDCoV), porcine epidemic diarrhea virus (PEDV), swine acute diarrhea syndrome coronavirus (SADS-CoV), and transmissible gastroenteritis coronavirus (TGEV), can cause acute diarrhea, vomiting, dehydration, and high mortality in suckling piglets. Recent studies revealing human PDCoV infections and the potential of SADS-CoV to penetrate human cell lines have heightened apprehensions about the zoonotic transmission risks of these viruses. While heparan sulfate (HS) serves as a receptor in PDCoV binding, the key host genes involved

Brucella-infected macrophage-derived microparticles enhance the host anti-Brucella acquired immune response.

Microparticles (MPs) serve as critical mediators of intercellular communication by shuttling bioactive molecules, holding significant potential for clinical diagnostics and next-generation vaccine development. Brucella, an intracellular pathogen responsible for severe zoonosis, evades host immunity through persistent infection; however, the role of Brucella-induced MPs in modulating adaptive immunity remains poorly defined. In this study, we demonstrate that the infection of RAW264.7 macrophages with the attenuated Brucella melitensis strain M5 triggers robust MP release. These MPs enhance hos

NSP1 of Alongshan virus antagonizes type I interferon responses by promoting STUB1-mediated degradation of RIG-I.

Emerging tick-borne viruses have become a significant public health concern due to their association with severe human illnesses. Among these, Alongshan virus (ALSV), a segmented RNA virus first identified in northeastern China, has been linked to febrile illness, presenting with fever, headache, myalgia, and, in severe cases, multi-organ failure. Despite its clinical relevance, the mechanisms by which ALSV evades host immune defenses remain poorly understood. Here, we identify NSP1, a nonstructural protein of ALSV, as a potent inhibitor of type I interferon (IFN-I) production, specifically ta

The oral-respiratory interface: modulation of Streptococcus pneumoniae serotype adhesion by the periodontal pathogen Porphyromonas gingivalis W83.

BACKGROUND: Porphyromonas gingivalis, a keystone periodontal pathogen synergizes with Streptococcus pneumoniae by upregulating host receptors like the platelet-activating factor receptor (PAFR). This study investigated whether P. gingivalis W83 differentially modulates the adhesion of four pneumococcal serotypes (3, 4, 6A, and 7F) to alveolar epithelial cells, hypothesizing a strictly serotype-dependent synergy. METHODS: An in vitro model used A549 alveolar epithelial cells conditioned with P. gingivalis W83 supernatant (PgSup) for 24 h. PAFR upregulation was verified via RT-qPCR. Cells were

Aspergillus fumigatus coinfection facilitates Pseudomonas aeruginosa chronicity within an ex-vivo pig lung model.

Pseudomonas aeruginosa and Aspergillus fumigatus represent the dominant bacterial and fungal pathogens in the lungs of adults with cystic fibrosis. Understanding how these species interact with each other and the host may provide insight into pathology and microbial succession in the lung. The ex vivo pig lung model is suitable for studying host responses to pathogens in an ethical and cost-effective manner due to its rich cell complexity and anatomical and immunological similarities to humans. Metagenomic analysis demonstrated that A. fumigatus promoted the proliferation of Pseudomonadota and
