Marine antifouling biocide 4,5-dichloro-2-n-octyl-4-isothiazolin-3-one disrupts sediment microbiome structure and function: insights from absolute quantification and enzyme activity dynamics.

The organic booster biocide DCOIT (4,5-dichloro-2-n-octyl-4-isothiazolin-3-one) is widely used in marine antifouling systems, yet its ecological impacts on sediment microbiomes remain poorly understood. Here, we integrated absolute quantitative 16S rRNA gene sequencing, metagenomics, and enzyme activity assays to examine microbial responses to DCOIT exposure (0-50 μg/g sediment) over 30 days. DCOIT induced oxidative stress and bioenergetic impairment, accompanied by reduced microbial activity and inhibition of key enzyme-mediated processes involved in organic matter turnover and nitrogen trans

Achieving Steroid-Sparing Remission in Refractory Pyoderma Gangrenosum With an Integrative Meta-Therapeutic System Based on Traditionally Engineered Nanomercury Sulfide: A Case Series.

The management of pyoderma gangrenosum (PG) remains a formidable clinical challenge due to its complex, multipathway immunopathogenesis. Although conventional steroid therapy and advanced biologic agents have improved outcomes, a substantial proportion of patients, particularly those with severe, progressive ulceration, exhibit treatment-refractory disease, highlighting a critical unmet need for novel strategies capable of network-level immune modulation. To address this, we designed and implemented a self-regulating, integrative meta-therapeutic system centered on a nanostructured, herbo-meta

The Gut Microbiota-Host Epigenetic Axis: An Emerging Biological Framework for Understanding Ethnic Disparities in Type 2 Diabetes Mellitus Susceptibility.

Persistent racial and ethnic disparities in type 2 diabetes mellitus (T2DM) are incompletely explained by genetic susceptibility, obesity, lifestyle, and socioeconomic factors, suggesting that additional biological mechanisms may link environmental exposures to metabolic disease risk. Increasing evidence indicates that the exposome, encompassing environmental exposures across the life course, may influence long-term metabolic health through molecular mechanisms that integrate environmental signals with host biology. This Review synthesizes epidemiological, experimental, and mechanistic evidenc

Mechanistic insights into bisphenol A - Induced liver fibrosis: Evidence of PPARγ downregulation and AKT1/FN1 signaling from multi-level analysis.

Non-alcoholic fatty liver disease (NAFLD) has rapidly ascended to become the foremost chronic liver disorder globally, yet the precise molecular mechanisms by which pervasive environmental endocrine-disrupting chemicals (EDCs) contribute to its pathogenesis remain largely unelucidated. This study presents a robust, multi-scale analytical framework, integrating human population genetics with cell-type-resolved molecular pathology, to definitively establish bisphenol A (BPA) as a causal accelerant of NAFLD progression. Utilizing two-sample Mendelian randomization (MR) with publicly available GWA

Proposed unified model of late-onset Alzheimer's disease: Chronic astrocytic and neuronal bioenergetic failure.

Late-onset Alzheimer's disease (LOAD) is framed here as progressive astrocyte-neuronal metabolic and neurovascular uncoupling initiated by astrocytic bioenergetic collapse. In genetically or environmentally predisposed brains, a self-reinforcing loop of lipid accumulation, inflammation, vascular impairment, glucose-handling defects, and mitochondrial dysfunction erodes astrocytic functional capacity. Subsequent cerebrovascular dysfunction and loss of blood-brain barrier (BBB) integrity perpetuate the neuroinflammatory response and drive amyloid-β deposition. Astrocytic failure then disrupts as

The Tudor-domain protein TDRD7, mutated in congenital cataract, controls the heat shock protein HSPB1 (HSP27) and lens fiber cell morphology.

Mutations of the RNA granule component TDRD7 (OMIM: 611258) cause pediatric cataract. We applied an integrated approach to uncover the molecular pathology of cataract in Tdrd7-/- mice. Early postnatal Tdrd7-/- animals precipitously develop cataract suggesting a global-level breakdown/misregulation of key cellular processes. High-throughput RNA sequencing integrated with iSyTE-bioinformatics analysis identified the molecular chaperone and cytoskeletal modulator, HSPB1, among high-priority downregulated candidates in Tdrd7-/- lens. A protein fluorescence two-dimensional difference in-gel electro

Time course determination of the effects of rapid and gradual cooling after acute hyperthermia on body temperature and intestinal integrity in pigs.

Rapid cooling after acute hyperthermia may cause a sustained increase in body temperature and exacerbate intestinal damage in pigs. Therefore, the study objective was to evaluate the temporal effects of rapid and gradual cooling on body temperature response and intestinal integrity after acute hyperthermia in pigs. In three repetitions, 54 pigs [83.3 ± 6.7 kg initial body weight (BW)], balanced by sex were exposed to thermoneutral conditions for 6 h (TN; n = 6 pigs/repetition; 21.1 ± 2.0°C), or heat stress conditions (HS; 39.3 ± 1.6°C) for 3 h, followed by a 3 h recovery period of gradual cool

Immunological differences in atopic dermatitis across age groups: insights from single-cell multi-omics.

BACKGROUND: Atopic dermatitis (AD) occurs across all ages but presents distinct clinical and immunologic features between children, adults, and older adults. The molecular programs underlying these age-specific immune differences remain poorly understood. METHODS: We performed single-cell multi-omics profiling of peripheral blood mononuclear cells (PBMCs) from 29 AD patients and 29 matched healthy controls (HC), spanning pediatric (0-17 years), adult (18-59 years), and geriatric (≥60 years) groups. Using Cellular Indexing of Transcriptomes and Epitopes by sequencing (CITE-seq), we simultaneou

The Prognostic Nutritional Index and Glycemic Status Synergistically Predict Early Renal Function Decline in Type 2 Diabetes: A Community-Based Cohort Study.

Background/Objectives: The Prognostic Nutritional Index (PNI), which integrates serum albumin and lymphocyte count, reflects both nutritional and inflammatory status. However, its role in early renal function decline among patients with type 2 diabetes (T2D), particularly in relation to glycemic control, remains unclear. This study aimed to: (1) characterize the dose-response relationship between PNI and early renal function decline in type 2 diabetes using restricted cubic splines; (2) identify whether glycemic control (HbA1c) modifies the PNI-renal decline association; and (3) evaluate the c

Emergency Medicine Resident Perceptions of Their Own Throughput Metrics.

Background Operational throughput metrics, such as emergency department (ED) length of stay (LOS) and time to disposition, are frequently and routinely tracked in EDs, but they are not usually integrated into resident education as structured formative feedback. The educational impact of routinely sharing individualized metrics with emergency medicine (EM) residents remains unclear. Methods We conducted a prospective pre-post quality improvement (QI) study at a single academic EM residency program between May and September 2025. Post-graduate year two and three residents received weekly individ

Multi-Omics Analysis Defines Endotypes and Systemic Inflammation in Primary Ciliary Dyskinesia: A Comparison with Healthy Controls.

INTRODUCTION: Primary ciliary dyskinesia (PCD) is a rare genetic disorder characterized by chronic airway inflammation and progressive lung injury. The inflammatory profile and systemic involvement remain poorly defined. We applied integrated multi omics (transcriptomics, proteomics, and metagenomics) to characterize inflammatory signatures and explore saliva as a noninvasive marker of systemic inflammation. These findings may support improved disease characterization and inform therapy and monitoring. METHODS: This cross sectional, multicenter study included participants with PCD and healthy

Integrated transcriptomic and tissue-validation analyses identify palmitoylation-associated biomarkers linked to sphingolipid metabolism and immune remodeling in psoriasis.

BACKGROUND: Psoriasis is a chronic immune-mediated inflammatory skin disease characterized by epidermal dysfunction, aberrant keratinocyte activation, and complex immune remodeling. Although lipid dysregulation has increasingly been implicated in psoriasis, the role of palmitoylation-associated genes in cutaneous inflammation remains insufficiently defined. METHODS: We integrated bulk transcriptomic datasets (GSE14905, GSE13355, and GSE121212) and a single-cell RNA-sequencing dataset (GSE151177) to identify palmitoylation-associated biomarkers in psoriasis. Consensus-Based Expert Recommendations for Diagnosis and Clinical Management of Vanishing White Matter.

Vanishing white matter (VWM) is a rare disorder, characterized by degeneration of CNS white matter, clinically often exacerbated by stressors such as fever and minor head trauma. VWM is caused by biallelic pathogenic variants in the EIF2B1-5 genes, causing reduced activity of eukaryotic initiation of translation factor 2B, resulting in dysregulation of the integrated stress response (ISR). New scientific insights and increased clinical trials in experimental therapies highlight the need for clinical guidelines to improve and standardize care for patients with VWM worldwide. Standardized care i…

Polymorphism-driven transcriptomic changes in anthelmintic metabolism pathways of Anisakis simplex s.s. L3 larvae.

BACKGROUND: Helminth infections continue to pose major challenges in human and veterinary medicine, with additional complications arising from the emergence of anthelmintic resistance. Anisakis simplex sensu stricto (A. simplex s.s.), a zoonotic nematode transmitted through the consumption of fish, is of growing concern due to its allergenic potential and clinical relevance. However, the molecular mechanisms underlying the parasite's response to anthelmintic treatment remain poorly defined. METHODS: Third-stage larvae (L3) of A. simplex s.s. were exposed to three widely used anthelmintics: al

Effects of diuron and two of its metabolites in biochemical markers and behavior of zebrafish larvae.

Diuron (3-(3,4-dichlorophenyl)-1,1-dimethylurea) is an herbicide used in many crops, including sugar cane cultivation. It is commonly found in aquatic ecosystem and is of high concern due to its ability to persist in the environment. Diuron metabolites include DCA (3,4-dichloroaniline) and DCPMU (3-(3,4-dichlorophenyl-1-methylurea). The objective of this study was to evaluate the effects of diuron and two of its metabolites in zebrafish (Danio rerio) developing embryos, from biochemical to individual level. Activities of the enzymes acetylcholinesterase (AChE), catalase (CAT), glutathione-S-tr

Agnostic identification of plasma biomarkers for postpartum hemorrhage risk.

BACKGROUND: Postpartum hemorrhage is difficult to predict, is associated with significant maternal morbidity, and is the leading cause of maternal mortality worldwide. The identification of maternal biomarkers that can predict increased postpartum hemorrhage risk would enhance clinical care and may uncover mechanisms that lead to postpartum hemorrhage. OBJECTIVE: This retrospective case-control study employed agnostic proteomic profiling of maternal plasma samples to identify differentially abundant proteins in controls and postpartum hemorrhage cases. STUDY DESIGN: Maternal plasma samples w

What is the role of the neutrophil extracellular traps in the cardiovascular disease burden associated with hemodialysis bioincompatibility?

Despite significant progress in dialysis modalities, intermittent renal replacement therapy remains an "unphysiological" treatment that imperfectly corrects uremic disorders and may lead to low-grade chronic inflammation, neutrophil activation, and oxidative stress due to repetitive blood/membrane interactions contributing to the "remaining uremic syndrome" and cardiovascular disease burden of hemodialysis patients. Understanding dialysis bioincompatibility pathways still remains a clinical and biochemical challenge. Indeed, surrogate biomarkers of inflammation including C-reactive protein cou

The Integrative Temporal Identity Model (TIM-E) for complex psychotrauma: A randomized comparative pilot study.

BACKGROUND: Complex post-traumatic stress disorder (CPTSD) is distinguished from PTSD by enduring disturbances in emotion regulation, self-concept, and relationships (disturbances in self-organization, DSO). Validated PTSD treatments (trauma-focused CBT, EMDR) can reduce core symptoms, but the persistence of DSO, adherence difficulties, and partial non-response in certain polytraumatized patients justify the exploration of integrative, phased, and process-based approaches. In this perspective, the TIM-E model proposes a mechanistic target: restoring temporal continuity of the self (past/presen

Molecular signaling in coinfection: how M. tuberculosis and respiratory viruses rewire host immunity and alter TB outcomes.

Tuberculosis (TB) caused by Mycobacterium tuberculosis (M. tuberculosis) and respiratory viral infections remain major, intersecting global health challenges, and their co-occurrence imposes a disproportionate burden in high-HIV/high-TB regions such as sub-Saharan Africa. Coinfection biology is heterogeneous and dynamic, driven by viral diversity including severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), influenza A/B, Respiratory Syncytial Virus (RSV), parainfluenza, metapneumovirus, rhinovirus, adenovirus, and bocavirus, and by the underlying TB stage, from latent and subclinica
