A recently isolated Lassa virus from Mali demonstrates atypical clinical disease manifestations and decreased virulence in cynomolgus macaques.

The virulence of Soromba-R, a Lassa virus strain recently isolated from southern Mali, was assessed in 2 animal models of Lassa fever: inbred strain 13 guinea pigs and cynomolgus macaques. In both models, the Malian isolate demonstrated tissue tropism and viral titers similar to those of historical Lassa virus isolates from Sierra Leone (Josiah) and Liberia (Z-132); however, the Soromba-R isolate was found to be less pathogenic, as determined by decreased mortality and prolonged time to euthanasia in macaques. Interestingly, in addition to the classic indicators of Lassa fever, Soromba-R infec

Cannabidiol reduces host immune response and prevents cognitive impairments in Wistar rats submitted to pneumococcal meningitis.

Pneumococcal meningitis is a life-threatening disease characterized by an acute infection affecting the pia matter, arachnoid and subarachnoid space. The intense inflammatory response is associated with a significant mortality rate and neurologic sequelae, such as, seizures, sensory-motor deficits and impairment of learning and memory. The aim of this study was to evaluate the effects of acute and extended administration of cannabidiol on pro-inflammatory cytokines and behavioral parameters in adult Wistar rats submitted to pneumococcal meningitis. Male Wistar rats underwent a cisterna magna t

Elevated serum interleukin-10 at time of hospital admission is predictive of mortality in patients with Staphylococcus aureus bacteremia.

BACKGROUND: Staphylococcus aureus bacteremia (SaB) carries considerable morbidity and mortality. We examined the predictive value of serum concentrations of interleukin (IL)-10, proinflammatory cytokines, and terminal complement on patient survival and SaB duration. METHODS: Clinical information on consecutive patients with SaB at a tertiary medical center were collected prospectively. Patient serum samples obtained at the day of clinical presentation were assayed for tumor necrosis factor-α, IL-1β, IL-10, and complement membrane attack complex C5b-9 concentrations using enzyme-linked immunoa

Lipid ingredients in moisturizers can modulate skin responses to UV in barrier-disrupted human skin in vivo.

BACKGROUND: Chemicals with a molecular weight <500 and adequate lipid solubility can penetrate the intact human skin. As many lipid ingredients in moisturizers have molecular weights <500, the lipid ingredients may penetrate into the skin and affect skin responses to UV; however, little is known about this phenomenon. OBJECTIVE: To evaluate the effects of major lipid ingredients in moisturizers on skin responses to UV in tape-stripped human skin in vivo. METHODS: We evaluated the effects of three major lipid ingredients in moisturizers (cholesterol, linoleic acid, and a synthetic ceramide, N

Perioperative infliximab application ameliorates acute rejection associated inflammation after intestinal transplantation.

As we have shown in the past, acute rejection-related TNF-α upregulation in resident macrophages in the tunica muscularis after small bowel transplantation (SBTx) results in local amplification of inflammation, decisively contributing to graft dysmotility. Therefore, the aim of this study is to investigate the effectiveness of the chimeric-monoclonal-anti-TNF-α antibody infliximab as perioperative single shot treatment addressing inflammatory processes during acute rejection early after transplantation. Orthotopic, isogenic and allogenic SBTx was performed in rats (BN-Lewis/BN-BN) with inflixi

Activation of innate immune responses through Toll-like receptor 3 causes a rapid loss of salivary gland function.

BACKGROUND: Recent studies have demonstrated the expression of Toll-like receptor 3 (TLR3) in salivary glands and epithelial cell lines derived from Sjögren's syndrome (SS) patients. As viral infections are considered to be a trigger for SS, in this study we investigated whether in vivo engagement of TLR3 affects salivary gland function. METHODS: Female New Zealand Black/WF1 mice were repeatedly injected with polyinosinic:polycytidylic acid [poly(I:C)]. TLR3 expression within submandibular glands was studied using immunohistochemistry. RNA levels of inflammatory cytokines in the submandibular

Hepatic platelet accumulation in Fas-mediated hepatitis in mice.

Platelets are reported to be causally involved in experimental hepatitis. Jo2, an agonistic anti-Fas antibody, induces hepatitis in mice. We examined the in vivo behaviors of platelets in mice injected with this antibody (analyzed by measuring 5-hydroxytryptamine, a constituent of platelets). We found that Jo2 induces platelet accumulation predominantly in the liver, and that this hepatic platelet accumulation (HPA) precedes the increases in hepatitis markers (alanine- and asparagine-aminotransferases [ALT and AST]). By electron microscopy, we detected entry of platelets into hepatocytes, and

Association of inflammatory cytokine gene polymorphisms with platelet recovery in idiopathic thrombocytopenic purpura patients after the eradication of Helicobacter pylori.

BACKGROUND: Idiopathic thrombocytopenic purpura (ITP) is associated with the cytokine response and dysregulation of the cytokine network. Gene polymorphisms of proinflammatory cytokines are associated with several diseases including ITP. Recently, the successful eradication of Helicobacter pylori has been reported to improve the platelet counts in some patients with ITP. The aim of this study was to elucidate the relationship between cytokine gene polymorphisms and platelet recovery in ITP patients after the eradication of H. pylori. MATERIALS AND METHODS: Gastric H. pylori infection was conf

Multiple Nod-like receptors activate caspase 1 during Listeria monocytogenes infection.

Listeria monocytogenes escapes from the phagosome of macrophages and replicates within the cytosolic compartment. The macrophage responds to L. monocytogenes through detection pathways located on the cell surface (TLRs) and within the cytosol (Nod-like receptors) to promote inflammatory processes aimed at clearing the pathogen. Cytosolic L. monocytogenes activates caspase 1, resulting in post-translational processing of the cytokines IL-1beta and IL-18 as well as caspase 1-dependent cell death (pyroptosis). We demonstrate that the presence of L. monocytogenes within the cytosolic compartment i

Mitochondrial DNA drives NLRP3-IL-1β axis activation in microglia by binding to NLRP3, leading to neurodegeneration in Parkinson's disease models.

Dysregulated mitochondrial DNA (mtDNA) promotes inflammatory response and disease progression. However, the mechanism and role of mtDNA-mediated inflammatory activation in the pathogenesis of Parkinson's disease (PD) are not yet clear. This study demonstrates that the injection of mtDNA into the substantia nigra pars compacta induces PD pathology in mice, characterized by the loss of dopaminergic (DA) neurons and the activation of microglia. Transcriptomic profiling of magnetic-activated cell sorting (MACS)-sorted cells reveals a pronounced upregulation of genes associated with the NLRP3 infla

Prognostic significance of inflammatory cytokine polymorphisms in AML: a study of IFNG, TNFA, and IL1B polymorphisms.

BACKGROUND: Inflammatory cytokines influence the pathogenesis and progression of acute myeloid leukaemia (AML), not only by shaping the leukemic microenvironment, but also by supporting leukaemia stem cell survival and resistance to therapy. We, therefore, investigated the associations between the cytokine polymorphisms IFNG+874 A/T, TNFA-857 C/T, and IL1B -31 C/T and AML outcomes, as well as their influence on clinical features. RESULTS: Ninety-three patients with AML and 117 healthy controls were analysed. The T allele of TNFA - 857 C/T (i.e., the high-expression allele) was observed at a s

Genetic and inflammatory interplay between IL1B (-511C/T) polymorphism and diabetic nephropathy in type 2 diabetes.

BACKGROUND: Inflammatory cytokines, particularly interleukin-1β (IL-1β), have a main role in diabetic nephropathy (DN) pathogenesis. Genetic variations within the IL1B promoter region may modulate cytokine expression and affect vulnerability to renal injury in type 2 diabetes mellitus (T2DM). We aimed to assess the correlation between the IL1B (-511C/T;rs16944) polymorphism, IL-1β serum levels, and renal function among Egyptian cases with T2DM. METHODS: This study was conducted on 150 subjects, who were divided into 50 T2DM cases with DN, 50 T2DM patients without DN, and 50 healthy control su

Itaconate ameliorates cardiovascular inflammation in a mouse model of Kawasaki disease vasculitis - brief report.

BACKGROUND: Kawasaki disease (KD), a systemic vasculitis and the leading cause of acquired heart disease in children, stems from an uncontrolled inflammatory response that fails to resolve in up to 20% of IVIG-treated patients. This treatment resistance increases the risk of cardiovascular complications and highlights the need for more targeted therapeutics. METHODS: We used the Lactobacillus casei cell wall extract (LCWE) murine model of KD vasculitis to investigate the therapeutic potential of itaconate, an anti-inflammatory metabolite, in the pathogenesis of KD. The expression of aconitate

OCTN1 variants shape innate immunity and predict individual response to anti-TNFα in ulcerative colitis patients: An exploratory study.

BACKGROUND: Ulcerative colitis (UC) exhibits substantial heterogeneity in inflammatory pathways and therapeutic response. The organic cation transporter OCTN1 (SLC22A4) and its rs1050152 missense variant (L503F) have been implicated in innate immune activation and disease susceptibility. We investigated whether OCTN1 variants modulate monocyte inflammatory responses and predict clinical outcomes in UC patients treated with anti-tumor necrosis factor α (TNFα) agents. METHODS: We combined preclinical and clinical approaches. In THP-1 cells engineered for OCTN1 knockdown or variant-specific over

Navigating the maze of Alzheimer's: nimodipine and pioglitazone combination in the spotlight through inhibition of P2X7 dependent NLRP3 inflammasome activation.

Neuroinflammation is considered to be a most vibrant molecular event in Alzheimer’s disease (AD) which is mainly triggered by accumulation of misfolded amyloid-β (Aβ) in neuronal cells. The Aβ mediated neuroinflammatory cascades further initiates the downstream neurodegenerative events through acceleration of microglial immune responses and oxidative stress persuaded mitochondrial dysfunction. The purinergic receptor, or P2X7 receptor, is measured as a pattern recognition element that is instantly activated by Aβ induced ATP release. This activation of P2X7 receptor further facilitates the NEK

MIT-001, a Mitochondria-Targeted ROS Scavenger, Ameliorates DSS-Induced Colitis and Is Associated with Reduced HMGB1 and IL-1β Expression.

Inflammatory bowel disease (IBD) is characterized by chronic intestinal inflammation in which excessive cell death and the release of damage-associated molecular patterns (DAMPs) such as high-mobility group box 1 (HMGB1) amplify mucosal injury. Although necrosis-particularly regulated forms including necroptosis and ferroptosis-has emerged as a contributor to IBD pathogenesis, the therapeutic potential of targeting necrotic cell death remains incompletely explored. We investigated whether MIT-001 (previously known as NecroX-7), a mitochondria-targeted reactive oxygen species (ROS) scavenger wi

Preclinical Analysis of Bone Marrow-Derived Stem Cell Therapy Response and Transcriptomic Overlap Analysis in a Severe Alcoholic Hepatitis Mouse Model.

BACKGROUND/AIMS: Severe alcoholic hepatitis (SAH) is a life-threatening form of alcoholic liver disease resulting in high short-term mortality. Mesenchymal stem cells (MSCs) have potent immunomodulatory effects and have been evaluated in various clinical trials for the treatment of chronic liver diseases. However, clinical evidence in patients with alcoholic hepatitis remains scarce, and the underlying mechanisms of MSCs in this population are not yet fully understood. METHODS: An integrative meta-analysis identified conserved transcriptomic signatures of alcoholic hepatitis. These signatures

Involvement of monocyte/macrophage NLRP3-IL-1β signaling in the pathogenesis of myasthenia gravis.

BACKGROUND: Myasthenia gravis is an autoimmune disorder mediated by pathogenic autoantibodies targeting the neuromuscular junction. Although the role of adaptive immunity is well-established, the involvement of innate immune cells, particularly monocytes and macrophages, in myasthenia gravis pathogenesis remains unclear. This study investigated whether the NLRP3 inflammasome-caspase-1-IL-1β pathway in monocytes and macrophages contributes to disease development in myasthenia gravis and its experimental model. METHODS: Experimental autoimmune myasthenia gravis was induced in Lewis rats by immu
