Barium-stimulated chemosensory activity may not reflect inhibition of background voltage-insensitive K+ channels in the rat carotid body.

To test the hypothesis that the voltage-insensitive background leak K+ channel is responsible for the oxygen-sensitive properties of glomus cells in the rat carotid body (CB) we used Ba2+, a non-specific inhibitor of K+ currents. In vitro changes in cytosolic calcium ([Ca2+]c) and chemosensory discharge were studied to measure the effect of Ba2+. In normal Tyrode buffer, Ba2+ (3 and 5 mM) significantly increased carotid sinus nerve (CSN) discharge over baseline firing rates under normoxia (PO2 approximately 120 Torr) from approximately 150 to approximately 600 imp/0.5 s. However, addition of 2

The use of autogenous vein grafts for inferior alveolar and lingual nerve reconstruction.

PURPOSE: This study evaluated whether an autogenous vein graft forms a satisfactory conduit for nerve regeneration. PATIENTS AND METHODS: Fifteen patients received a total of 16 autogenous vein grafts to repair continuity defects of the inferior alveolar (6) and lingual nerves (10) nerves. All were treated between 4 and 10 months after injury. At surgery, the postresection defects ranged from 2 to 14 mm. All lingual nerves were repaired with saphenous vein grafts from an intraoral approach and all inferior alveolar nerves were repaired with facial vein grafts inserted from an extraoral approa

Functional characterization of three mutations of the endothelin B receptor gene in patients with Hirschsprung's disease: evidence for selective loss of Gi coupling.

BACKGROUND: Hirschsprung's disease (HSCR) is one the most common congenital intestinal disease. It leads to aganglionic megacolon in the early childhood. Several susceptibility genes have been identified : RET protooncogene and its ligand, glial cell derived neutrophic factor (GDNF), Sox 10, Endothelin-3 (EDN3) and its receptor B (EDNRB). EDNRB mutations are found in 5% of familial or sporadic HSCR. Only few EDNRB mutations found in HSCR have been explored and some of them seem to be non fonctional variants. MATERIALS AND METHODS: The properties of three mutant human endothelin B receptor (hE

Interfacing the body's own sensing receptors into neural prosthesis devices.

Functional Electric Stimulation (FES) is today available as a tool in muscle activation used in picking up objects, in standing and walking, in controlling bladder emptying, and for breathing. Despite substantial progress over nearly three decades of development, many challenges remain to provide a more efficient functionality of FES systems. The most important of these is an improved control of the activated muscles. Instead of artificial sensors for feedback, new developments in electrodes to do long-term and reliable recordings from peripheral nerves emphasize the use of the body's own sens

Hyperalgesia during naloxone-precipitated withdrawal from morphine is associated with increased on-cell activity in the rostral ventromedial medulla.

Hyperresponsiveness to noxious stimulation (hyperalgesia) is observed with naloxone-precipitated morphine withdrawal in several experimental models, and may be due to changes in central nervous system neurons. Previous studies have demonstrated that certain neurons in the rostral ventromedial medulla (on-cells) discharge just prior to nocifensive withdrawal reflexes and are inhibited by morphine. Because the tail flick latency (TFL) is shorter when on-cells are active, it has been proposed that on-cells facilitate nocifensive reflexes. The present study examined the hypothesis that the hyperal

Enkephalin modulation of neural transmission in the cat stellate ganglion: pharmacological actions of exogenous opiates.

Neural ganglionic transmission was studied in vivo in the cat, using closed chest anesthetized preparations. The right stellate ganglion and its branches were exposed retropleurally and prepared for electrical stimulation of pre- and postganglionic nerve fibers. The axillary artery was cannulated allowing direct administration of drugs in the arterial blood supplying the ganglion. Stimulation of postjunctional receptors could thus be obtained by local administration of selective agents. Local administration of nicotinic, muscarinic or histaminergic agents increased heart rate and blood pressur

Distinctive pharmacological profile of a nonadrenergic inhibitory system in bullfrog lung.

1 Bullfrog hemilungs, pretreated with atropine, are markedly relaxed on addition of carbachol. Since the relaxant effect is inhibited by tetrodotoxin or hexamethonium, it is neurally mediated and involves stimulation of nicotinic receptors with release of an unknown inhibitory transmitter.2 Carbachol-induced relaxation is nonadrenergic since: (a) it considerably exceeds the maximal effects of isoprenaline or the effect of 10(-3)M adrenaline or noradrenaline; (b) it elicits marked further relaxation in preparations already relaxed by high concentrations of catecholamines; (c) it is not attenuat

Clarke's column neurons as the focus of a corticospinal corollary circuit.

Proprioceptive sensory signals inform the CNS of the consequences of motor acts, but effective motor planning involves internal neural systems capable of anticipating actual sensory feedback. Just where and how predictive systems exert their influence remains poorly understood. We explored the possibility that spinocerebellar neurons that convey proprioceptive sensory information also integrate information from cortical command systems. Analysis of the circuitry and physiology of identified dorsal spinocerebellar tract neurons in mouse spinal cord revealed distinct populations of Clarke's colu

Dynamic Field Theory of Executive Function: Identifying Early Neurocognitive Markers.

In this Monograph, we explored neurocognitive predictors of executive function (EF) development in a cohort of children followed longitudinally from 30 to 54 months of age. We tested predictions of a dynamic field model that explains development in a benchmark measure of EF development, the dimensional change card sort (DCCS) task. This is a rule-use task that measures children's ability to switch between sorting cards by shape or color rules. A key developmental mechanism in the model is that dimensional label learning drives EF development. Data collection began in February 2019 and was comp

IGF-1 signaling pathway activation promotes axonal regeneration and repair: A mechanism study on catalpol-induced functional recovery after ischemic stroke.

ETHNOPHARMACOLOGICAL RELEVANCE: In traditional Chinese Medicine (TCM) theory, there is a concept of "tonifying the kidney and generating marrow, and marrow enriches and nourishes the brain ", which believes that tonifying the kidney and generating marrow can promote brain marrow repair and neurological function recovery. This theoretical framework has been substantiated by multiple modern medical studies. Catalpol, a bioactive iridoid glycoside extracted from Rehmannia glutinosa (Gaertn.) DC. has been traditionally employed in TCM for "kidney tonification, marrow generation, and brain nourishm

Duloxetine ameliorates cerebral ischemic injury by inhibiting autophagy.

Ischemic stroke is a severe medical condition characterized by diminished blood flow to the brain, resulting in a shortage of oxygen and nutrients. During ischemia, neurons surrounding the cerebral infarct initiate macroautophagy. However, the implications of this activation for neuronal cell survival are still debated. The identification of new autophagy modulators could aid in understanding autophagy's role in brain ischemia and lay the groundwork for innovative therapeutic strategies aimed at minimizing brain damage in this life-threatening neurological emergency. In this study, we develope

Triadimefon causes branchial arch malformations in Xenopus laevis embryos.

BACKGROUND, AIMS AND SCOPE: Triazole-derivatives are potent antifungal agents used as systemic agricultural fungicides and against fungal diseases in humans and domestic animals. They act by inhibiting the cytochrome P-450 conversion of lanosterol to ergosterol, thus resulting in faulty fungal cell wall synthesis. Some data have been published about the teratogenic activity of triazoles on rodent embryos: Hypoplasias, abnormal shape, agenesis of the branchial arches, for example, were reported as typical induced malformations. Unfortunately, no data are available on the embryotoxicity of these

Buyang Huanwu decoction promotes neurological recovery after ischemic stroke by activating the Akt/PAK5/SNPH axis to enhance mitochondrial recruitment and axonal remodeling.

ETHNOPHARMACOLOGICAL RELEVANCE: Buyang Huanwu Decoction (BHD), a classic formula in Chinese medicine, has demonstrated neuroprotective properties in numerous studies on ischemic stroke. However, the specific mechanisms through which BHD promotes neurological recovery during the recovery phase, particularly those linked to axonal remodeling, have yet to be fully elucidated. AIM OF THE STUDY: This study is designed to investigate potential molecular and cellular mechanisms responsible for the beneficial effects of BHD in ischemic stroke. MATERIALS AND METHODS: Characterization of the major che

Aperiodic and periodic neural activity during sleep in autism spectrum disorders.

BACKGROUND: Influential theories proposed that the complex and heterogeneous clinical and behavioral manifestations of autism spectrum disorder (ASD) arise from dysregulation of neural circuits driven by an imbalance between excitatory (glutamatergic) and inhibitory (GABAergic) neurotransmission. Building on this framework, and considering the role of homeostatic regulation of neuronal excitability in shaping sleep stages, we hypothesized that dysregulation of neuronal network activity-including altered ratios of excitatory drive to interneuronal inhibitory control-might manifest in sleep arch

Beyond single-person paradigms: N400 alterations in a social context in schizophrenia.

INTRODUCTION: Impairments in social cognition and context-dependent information processing constitute prominent features of schizophrenia. Among event-related potentials (ERPs), the N400 is known to be sensitive to semantic and social contextual information. However, its role in social contexts has received limited attention in schizophrenia research, as previous studies have largely examined the N400 in isolated, single-person paradigms or have focused on the P300 component. METHODS: In this study, we examined the N400 ERP component in a real-world social context using EEG hyperscanning. Pat

Alpha-adrenergic mediated changes in blood pressure variability after hypoxia-ischaemia in preterm fetal sheep.

Brain injury in preterm infants is common but often difficult to detect in the early stages. The present study investigated whether evolving preterm brain injury after exposure to hypoxia-ischaemia (HI) is associated with dynamic changes in arterial blood pressure variability (BPV) and tested the hypothesis that it was mediated by alpha-adrenoreceptor activity. Chronically instrumented 0.7 gestation fetal sheep received sham-HI (n = 8) or HI induced by 25 min of umbilical cord occlusion, followed by i.v. infusion of either the alpha-adrenergic blocker phentolamine (n = 6) or saline (n = 8) fro

From lipid dynamics to precision predictions: A new approach methodology for precision modeling of phosphoinositide signaling.

Precision medicine requires models that can translate rich molecular measurements into individualized predictions of biological response. This challenge is particularly acute for phosphoinositide signaling disorders that often exhibit cell-type-specific responses to identical genetic or pharmacological perturbations. Here, we develop a New Approach Methodology (NAM) demonstrating that basal phosphoinositide pool composition, determined by the size of the PI(4)P reserve, determines the robustness of lipid signaling. The NAM comprises a core kinetic model of phosphatidylinositol (PI), phosphatid

Impaired WNT3/IGF-1 Signaling in Dorsal Dentate Gyrus Contributes to Chronic Pain-Related Cognitive Impairment.

BACKGROUND: Neuropathic pain is frequently accompanied by cognitive deficits, but the neural circuits and molecular mechanisms linking nociceptive hypersensitivity to cognitive dysfunction remain incompletely understood. The hippocampal dentate gyrus (DG), a critical hub for adult neurogenesis and memory encoding, is emerging as a key substrate integrating pain processing and cognitive impairment, but how peripheral nerve injury disrupts DG homeostasis remains unclear. METHODS: In this study, we used the spared nerve injury (SNI) mouse model to investigate the cellular and molecular mechanism

Recent updates on cold adaptation in population and laboratory studies, including cross-adaptation with nonthermal factors.

This review aims to update our understanding of human cold adaptation. First, an overview of the thermoregulatory response to cold is provided, with some recent updates in human brown adipose tissue (BAT). Variation in BAT activity and multiorgan contributions to cold-induced thermogenesis were introduced. We found that individuals with less BAT activity rely more on shivering to compensate for less non-shivering thermogenesis (NST). The mechanisms of cold-induced vasoconstriction are summarized, including the role of arteriovenous anastomoses, adrenergic neural function, and inhibition of the
