Development and characterization of NILK-2301, a novel CEACAM5xCD3 κλ bispecific antibody for immunotherapy of CEACAM5-expressing cancers.

BACKGROUND: T-cell retargeting to eliminate CEACAM5-expressing cancer cells via CEACAM5xCD3 bispecific antibodies (BsAbs) showed limited clinical activity so far, mostly due to insufficient T-cell activation, dose-limiting toxicities, and formation of anti-drug antibodies (ADA). METHODS: We present here the generation and preclinical development of NILK-2301, a BsAb composed of a common heavy chain and two different light chains, one kappa and one lambda, determining specificity (so-called κλ body format). RESULTS: NILK-2301 binds CD3ɛ on T-cells with its lambda light chain arm with an affin

Tissue- and cell-specific properties of enterochromaffin cells affect the fate of tumorigenesis toward nonendocrine adenocarcinoma of the small intestine.

Small intestinal neuroendocrine tumors (SI-NETs) are serotonin-secreting well-differentiated neuroendocrine tumors of putative enterochromaffin (EC) cell origin. However, EC cell-derived tumorigenesis remains poorly understood. Here, we examined whether the gain of Myc and the loss of RB1 and Trp53 function in EC cells result in SI-NET using tryptophan hydroxylase 1 (TPH1) Cre-ERT2-driven RB1fl Trp53fl MycLSL (RPM) mice. TPH1-Cre-induced gain of Myc and loss of RB1 and Trp53 function resulted in endocrine or neuronal tumors in pancreas, lung, enteric neurons, and brain. Lineage tracing indicat

Dietary High Dose of Iron Aggravates the Intestinal Injury but Promotes Intestinal Regeneration by Regulating Intestinal Stem Cells Activity in Adult Mice With Dextran Sodium Sulfate-Induced Colitis.

The effects of excessive dietary iron intake on the body have been an important topic. The purpose of this study was to investigate the effects of high-dose iron on intestinal damage and regeneration in dextran sodium sulfate (DSS)-induced colitis model mice. A total of 72 8-week-old adult C57BL/6 mice were randomly divided into two dietary treatment groups: the basal diet supplemented with 45 (control) and 450 mg/kg iron (high-iron) from ferrous sulfate. The mice were fed different diets for 2 weeks, and then 2.5% DSS was orally administered to all mice for 7 days. Samples of different tissue

Nested stromal epithelial tumor of the liver: six cases of a distinctive pediatric neoplasm with frequent calcifications and association with cushing syndrome.

Rare cases of nonhepatocytic mixed stromal and epithelial tumors of the liver with associated calcification and ossification have been described previously. We report 6 similar cases in children, including 2 cases associated with ectopic ACTH production. The patients were between 2 and 14 years of age at diagnosis. All tumors presented as a solitary liver mass with no extrahepatic involvement. Two adolescent females with palpable abdominal tumors presented with Cushing syndrome that abated after excision of the tumors. The other children had tumors identified incidentally on imaging studies or

A review of advances in research on mechanisms inhibiting kidney stone formation from a network pharmacology perspective.

Nephrolithiasis is a prevalent, highly recurrent urological disorder affecting 1%-15% of the global population, with 5-year recurrence rates reaching up to 50%. While surgical interventions achieve acute stone clearance, they do not resolve underlying metabolic derangements, crystal-induced tubular injury, or chronic inflammatory cascades. Natural products from global ethnomedicinal systems (including Traditional Chinese Medicine, Indian Ayurveda, Persian Unani medicine, European phytotherapy, and African ethnopharmacology) demonstrate potent multi-target anti-urolithic properties. However, de

A modern reinterpretation of the "kidney governs bone" theory: systemic regulation of the neuro-endocrine-immune network via the brain-bone axis and traditional chinese medicine intervention strategies.

The traditional Chinese medicine (TCM) theory of "Kidney Governs Bone' has been progressively validated in modern medical research. However, a purely endocrinological perspective cannot fully explain its regulatory implications under complex conditions such as stress, inflammation, and nerve injury. As bone metabolism research shifts toward a systemic regulatory paradigm, the "brain-bone axis' provides a new framework for reinterpreting "Kidney Governs Bone.' Based on current evidence, the core scientific proposition of this article is that we propose the hypothesis that the therapeutic effect

Perfluorobutanesulfonic acid (PFBS) elicits compartment-specific divergent responses in prostate homeostasis: Wnt/β-catenin oncogenic activation in epithelial cells versus mitochondria-mediated ferroptosis susceptibility in stromal cells.

Perfluorobutanesulfonic acid (PFBS), a short-chain per- and polyfluoroalkyl substance (PFAS) adopted as a replacement for perfluorooctane sulfonate (PFOS), is ubiquitously detected in environmental matrices and human biological samples. Despite growing regulatory concern, the molecular mechanisms by which PFBS affects prostate tissue at environmentally relevant concentrations remain poorly characterized. In the present study, we exposed human prostate epithelial (RWPE-1) and stromal (WPMY-1) cells to PFBS (0-100 nM, 24 h) and integrated in vitro mechanistic analyses with genome-wide RNA sequen

Liquiritigenin targets transferrin receptor to potentiate ferroptosis sensitivity in colorectal cancer cells.

BACKGROUND: Ferroptosis, a type of regulated cell death driven by iron-dependent lipid peroxidation, has emerged as a promising strategy for cancer therapy. However, the efficacy of conventional ferroptosis inducers is often limited by poor tumor responsiveness. High-throughput screening offers a promising approach for the rapid discovery of safe and efficient ferroptosis sensitizers. PURPOSE: This study aimed to systematically identify a natural compound capable of sensitizing colorectal cancer (CRC) cells to ferroptosis via high-throughput screening and delineate the underlying molecular me

Proof-of-Concept Evaluation of Primary Human FAP-CAR-NK Cells Targeting Activated Fibroblasts in Pulmonary Fibrosis.

In recent years, the feasibility of immunotherapy targeting activated fibroblasts in pulmonary fibrosis has received further support. Recent studies have shown that transient FAP-targeted immunotherapy can alleviate pulmonary fibrosis by eliminating excessively activated fibroblasts, improving the aberrant extracellular matrix environment, and promoting alveolar cell lineage remodeling, suggesting that FAP-associated pathological stromal cells are amenable to therapeutic intervention. Based on this, research on FAP-centered engineered cell therapies is being gradually extended from settings su

Therapeutic pressure drives the evolution of a protective ecotype characterized by AR-loss-induced senescence in prostate cancer.

RATIONALE: Prostate cancer treatment relies heavily on androgen deprivation therapy, yet the progression to a lethal treatment-resistant state presents a nearly universal clinical challenge. While tumor-intrinsic changes are well documented, the manner in which the broader tumor microenvironment dynamically reorganizes into distinct macroscopic ecological states under therapeutic pressure remains elusive. This underscores that dismantling specific therapy-induced TME niches may represent a promising strategy for CRPC treatment. METHODS: We constructed a comprehensive single-cell atlas compris

Molecular and Chromatin Accessibility Programs Underlying Epithelial Injury and Impaired Regeneration in Neonatal Necrotizing Enterocolitis.

BACKGROUND & AIMS: Neonatal necrotizing enterocolitis (NEC) is a severe gastrointestinal disorder with high mortality, characterized by epithelial cell injury and compromised epithelial repair. The mechanisms underlying defective epithelial regeneration remain poorly understood despite advances in single-cell omics. Addressing these challenges is essential for elucidating the pathogenesis of NEC and identifying therapeutic targets to restore epithelial regeneration and replace the damaged epithelial layer. METHODS: Using a well-established neonatal mouse model of NEC induced by formula feedin

Metabolic Reprogramming in Male Infertility: Mechanistic Integration and Translational Perspectives From Mouse Models.

Spermatogenesis is a highly energy-dependent and tightly regulated differentiation process in the male reproductive system, characterized by dynamic, stage-specific metabolic adaptations during spermatogonial proliferation, meiosis and sperm maturation. Accumulating evidence suggests that successful spermatogenesis is associated with a coordinated program of metabolic reprogramming, which involves the sequential and context-dependent utilization of distinct bioenergetic pathways rather than dependence on a single energy source. Importantly, metabolic reprogramming represents a physiological an

Case Report: Carotid body paraganglioma in a 20-year-old woman: multimodal imaging, vessel-preserving surgical excision, and long-term clinical follow-up.

Carotid body paraganglioma is a rare hypervascular neuroendocrine tumor of the head and neck, and presentation in a 20-year-old woman is uncommon. We report a young woman with a painless left lateral neck mass present for more than 5 years, with more noticeable enlargement during the preceding year. On admission, a firm, mobile, non-tender mass was palpated along the anterior border of the left sternocleidomastoid muscle, without an audible bruit. Initial and targeted ultrasonography demonstrated a hypervascular lesion centered at the left carotid bifurcation. Preoperative contrast-enhanced co

Clinical features and imaging diagnostic challenges of fetus-in-fetu associated with abdominal cryptorchidism: a case report.

BACKGROUND: Fetus-in-fetu (FIF) is a rare congenital anomaly in which a malformed monozygotic twin is incorporated within its host twin. FIF is usually retroperitoneal, whereas association with an intra-abdominal undescended testis is exceptionally uncommon. Because imaging may show a heterogeneous mass containing soft tissue, fat and calcified elements, FIF can be difficult to distinguish preoperatively from mature teratoma, especially when cryptorchidism is also present. CASE PRESENTATION: A 3-day-old male neonate was admitted because of jaundice and a palpable right abdominal mass. Physica

Oncolytic virotherapy potentiates chemo-PD-1 immunotherapy by engaging chemo-resistant bystander CD8+ T cells.

BACKGROUND: The efficacy of combined chemotherapy and programmed cell death protein-1 (PD-1) immune checkpoint blockade (ICB) is constrained by the collateral cytotoxicity of chemotherapy toward proliferating tumor-specific CD8+ T (TTST) cells, a population indispensable for antitumor immunity. This study aimed to overcome this limitation by targeting a potential chemotherapy-resistant immune cell reservoir. METHODS: By using the murine acutely resolved lymphocytic choriomeningitis virus (LCMV) infection and tumor models (colorectal cancer and melanoma), we characterized the susceptibility of

Smyca-FOXM1 ribonucleoprotein complex promotes homologous recombination and tumor immune evasion to define a therapeutic target of triple-negative breast cancer.

BACKGROUND: Triple-negative breast cancer (TNBC) is the most aggressive subtype of breast cancer with limited treatment options. Although PARP inhibitor (PARPi) offers great promise in treating TNBC with deficiency in homologous recombination (HR), most TNBC patients are HR-proficient. Furthermore, acquired resistance to PARPi remains as a challenge. Thus, there is an unmet need to identify new therapeutic target for developing advanced TNBC treatment strategy. METHODS: I-SceI reporter assay and alkaline comet assay were used to analyze the role of Smyca in HR repair. Ingenuity pathway analys

Pharmacological targeting of mitophagy via ALT001 improves herpes simplex virus 1 (HSV1)-mediated microglial inflammation and promotes amyloid β phagocytosis by restricting HSV1 infection.

Rationale: One of the hallmarks of Alzheimer's disease (AD) is the accumulation of dysfunctional mitochondria. Herpes simplex virus type 1 (HSV1) may be a risk factor for the neuropathology linked to amyloid β (Aβ) accumulation. However, the mechanisms underlying HSV1-associated mitochondrial dysfunction in AD remain unclear. ALT001 is a novel drug that ameliorates AD-related cognitive impairment via ULK1/Rab9-mediated alternative mitophagy. In this study, we investigated the effects of ALT001 on the neurodegeneration-related microglial signatures associated with HSV1 infection. Methods: Molec

A novel human specific lncRNA MEK6-AS1 regulates adipogenesis and fatty acid biosynthesis by stabilizing MEK6 mRNA.

BACKGROUND: Obesity is becoming one of the major non-communicable diseases with increasing incidence and risks that cannot be ignored. However effective and safe clinical treatment strategies still need to be deeply explored. Increased number and volume of adipocytes lead to overweight and obesity. The aim of our work is to identify lncRNAs that have important regulatory in differentiation of human mesenchymal stem cells (MSCs) into adipocytes, and to provide effective targets for clinical prevention and treatment of obesity and related metabolic disorders. METHODS: We extracted primary MSCs

Recombinant Adeno-Associated Virus Vector Mediated Gene Editing in Proliferating and Polarized Cultures of Human Airway Epithelial Cells.

Cystic fibrosis (CF) is caused by mutations in the cystic fibrosis transmembrane conductance regulator (CFTR) gene. While CRISPR-based CFTR editing approaches have shown proof-of-concept for functional rescue in primary airway basal cells, induced pluripotent stem cells, and organoid cultures derived from patients with CF, their efficacy remains suboptimal. Here, we developed the CuFiCas9(Y66S)eGFP reporter system by integrating spCas9 and a non-fluorescent Y66S eGFP mutant into CuFi-8 cells, an immortalized human airway epithelial cell line derived from a patient with CF with homozygous F508d

Cancer-associated fibroblast derived CXCL14 drives cisplatin chemoresistance by enhancing nucleotide excision repair in bladder cancer.

BACKGROUND: A significant challenge in bladder cancer treatment is primary chemoresistance, in which cancer-associated fibroblasts (CAFs) in the tumor microenvironment (TME) play a pivotal role. While the contributions of CAFs to tumor progression and drug resistance are well established, the precise molecular mechanisms by which they induce chemoresistance remain unclear. A comprehensive understanding of the effect of TME modulation-particularly through CAFs-on the chemotherapeutic response is crucial for developing effective strategies to overcome chemoresistance and improve patient survival
