Computational exploration of the electrochemical oxidation mechanism of thiocyanate catalyzed by cobalt-phthalocyanines.

In this study, we focused on the mechanism of the electrocatalytic oxidation of thiocyanate, which in traditional electrodes typically requires high overpotentials. As models for reducing these overpotentials and catalyzing the reaction, we used a set of modified cobalt phthalocyanines (CoPc), known as electrocatalysts. Using DFT calculations, we explored how modifications to CoPc by adding electron-donating and withdrawing groups and the coordination of 4-amino thiophenol impact the oxidation process. The reaction mechanism for the electrooxidation of thiocyanate has remained elusive, where o

The role of the anaesthetist in mitigating the environmental impacts of nitrous oxide: a narrative review.

Nitrous oxide (N2O) is a greenhouse gas (GHG) with ozone-depleting effects. It is a relatively minor, but important, measurable, and remediable part of healthcare's contribution to global warming. Anaesthetists became aware of the GHG emissions associated with their daily work practices, particularly inhalation agents including N2O, well before other clinicians. However, only relatively recent research indicates that a significant amount of N2O is leaked before patient care, representing a much larger proportion of associated GHG emissions than clinical use. In addition, there are other source

Plant-based antioxidant peptides: impact on oxidative stress and gut microbiota.

Plant-based peptides can be obtained from natural and climate-friendly sources. These peptides show various bioactivities including antioxidant activity. Oxidative stress has an impact on the gut microbiota causing inflammation, insulin resistance, osteoporosis, cancer, and several chronic diseases like type 2 diabetes, arthritis, hypertension, and atherosclerosis. Therefore, antioxidant peptides may significantly affect oxidative stress as a potential alternative to conventional medication. The production of antioxidant peptides from plant-based protein sources through conventional and innova

Inhibition of GRK2 mitigates renal fibrosis via oxidative stress pathway.

BACKGROUND: Studies have shown that G protein-coupled receptor kinase 2 (GRK2) undergoes functional changes in various diseases. Therefore, in this study, we examined the role of GRK2 in renal fibrosis induced by unilateral ureteral occlusion (UUO) and ischaemia-reperfusion (I/R), and evaluated the protective effect of pharmacological inhibition of GRK2. METHODS: UUO or I/R models were generated in GRK2 knockdown (GRK2-KD), GRK2 conditional-knockdown (GRK2-CKd), and wild-type (WT) mice. Biochemical markers of renal function and histopathological features of renal tissues were evaluated to ass

Systemic oxidative stress associates with delirium in a geriatric population with hip fracture.

BACKGROUND: Oxidative stress ensues in patients undergoing surgery for hip fracture and has been implicated in the pathophysiology of delirium. Circulating free thiols (R-SH, sulfhydryl groups) serve as biomarker of systemic oxidative stress since they are rapidly oxidized by reactive species, acting as potent antioxidants. We aimed to investigate the relationship between delirium and systemic oxidative stress in older patients hospitalized with hip fracture. METHODS: Patients aged 65 years or more, acutely admitted due to hip fracture, were included between 2005 and 2008. Delirium was diagno

Host oxidative stress primes mycobacteria for rapid antibiotic resistance evolution.

The rapid emergence of multidrug-resistant Mycobacterium tuberculosis (Mtb) threatens global TB control, yet the mechanisms enabling rapid evolution of drug resistance in Mtb remain poorly understood. Here we reveal that pre-existing mutations in oxidative stress response genes create permissive genomic backgrounds that accelerate high-level isoniazid resistance (INHR) without fitness costs, challenging the paradigm that resistance mutations always precede their fitness compensatory adaptations. Using M. smegmatis mc2155 (Msm) as a model, we show that brief exposure to sublethal INH (2× IC50)

HNGF6A ameliorates oxidative stress-mediated mitochondrial dysfunction in degenerative meniscus.

AIMS: Meniscus injury can lead to knee synovitis and cartilage deterioration, ultimately resulting in osteoarthritis (OA). Mitochondrial dysfunction has been identified as an induction factor in OA development, owing to an imbalance between the production of reactive oxygen species (ROS) and the antioxidant capacity of cells. However, the contribution of mitochondrial function in the meniscus to OA remains unclear. The purpose of this work was to elucidate the impact of HNGF6A, a mitochondrial-derived protective peptide, on meniscus senescence and degeneration to clarify the underlying mechani

NOTCH3 drives fatty acid oxidation and ferroptosis resistance in aggressive meningiomas.

PURPOSE: NOTCH3 is increasingly implicated for its oncogenic role in many malignancies, including meningiomas. While prior work has linked NOTCH3 expression to higher-grade meningiomas and treatment resistance, the metabolic phenotype of NOTCH3 activation remains unexplored in meningioma. METHODS: We performed single-cell RNA sequencing on NOTCH3 + human meningioma cell lines. Using the CH157-MN meningioma cell model, we overexpressed NOTCH3 intracellular domain (ICD) and performed untargeted metabolomic, lipidomic, and bulk RNA sequencing analyses as well as functional metabolic assays. Exploring Triptolide's Dual Role in Inflammation and Oxidative Stress: Responses of Organelles.

Triptolide (TPL), isolated from the traditional Chinese herb Tripterygium wilfordii Hook. f., is a natural compound with complex pharmacological activities. TPL has significant therapeutic effects in rheumatoid arthritis, Alzheimer's disease, diabetic nephropathy, and other diseases through anti-inflammatory and antioxidant effects. However, its pro-inflammatory and antioxidant effects produce toxicity to multiple organs such as kidney, liver, and the reproductive organs. Thus, the effects of TPL on inflammation and oxidative stress broaden the therapeutic scope of TPL, but also limit its clin

Lactate induces oxidative phosphorylation in osteoblasts via Gpr81-Stat3 signaling.

Lactate has long been regarded as an end product of glycolysis and a metabolic "waste product" under hypoxic conditions, but recent studies have revealed that lactate plays a central role in energy metabolism reprogramming and intercellular communication. However, it remains unknown whether lactate promotes osteogenic differentiation through metabolic reprogramming. Here, we showed that lactate significantly increased the cellular ATP content, activated succinate dehydrogenase activity, and enhanced oxygen consumption rate in pre-osteoblast MC3T3-E1 cells. Moreover, lactate treatment increased

Oxidation-dependent effects of alpha-1 antitrypsin on wound healing and inflammation.

Wound healing requires a delicate balance between cellular and molecular factors, all affected by reactive oxygen species (ROS). While ROS decontaminate, they also might lead to impaired wound healing, as evident in radiation-exposed skin and in venous insufficiency. Human alpha-1 antitrypsin (hAAT) is a circulating antiprotease that is anti-inflammatory and tissue-protective. Accordingly, tissue repair is enhanced in hAAT-rich conditions. hAAT undergoes oxidative modification in high-ROS environments, which alters its functional properties. While its antiprotease function is lost, the consequ

Associations between asprosin, insulin resistance, and oxidative stress in adults with obesity.

The global increase in obesity is strongly associated with insulin resistance (IR) and related metabolic impairments. Asprosin (ASP), a glucogenic adipokine induced by fasting, has recently emerged as a potential biomarker of IR and abnormal body composition. However, its physiological role in obesity remains incompletely understood. To evaluate the associations between serum ASP levels, IR indices, oxidative stress markers (OS), liver function and inflammation parameters, and body composition in overweight and obese adults during a standardized 4-hour oral glucose tolerance test (OGTT). MeCP2 at the crossroads of hypoxia, oxidative stress, and gene regulation in Rett syndrome.

Rett syndrome (RTT) is a severe neurodevelopmental disorder primarily affecting females, caused by mutations in the X-linked gene MECP2. This gene encodes methyl CpG binding protein 2 (MeCP2), a multifunctional epigenetic regulator critical for neuronal gene regulation. In addition to well-characterized neurological symptoms, such as seizures and motor abnormalities, RTT patients frequently present with irregular breathing patterns that induce intermittent hypoxia, suggesting that MeCP2 contributes to respiratory regulation as well as the brain's cellular and molecular response to hypoxia. Mec

The interplay between homocysteine, oxidative stress, and butyrylcholinesterase activity in the pathogenesis of schizophrenia.

BACKGROUND: Schizophrenia (SCZ) is a serious mental illness linked to neurobiological problems, such as hyperhomocysteinemia (HHC), increased butyrylcholinesterase (BChE) activity, and a pro-oxidant status. However, it remains unclear how these different markers interact with each other. This study aimed to analyze oxidative status, Hcys, and BChE activity variations in male drug-free SCZ patients compared to healthy controls. METHODS: We conducted a case-control study on male drug-free patients with SCZ. Metabolic parameters, oxidative status, Hcys, BChE activity, and ultrasensitive C-reacti

Algal and Fungal Antioxidants Alleviate Oxidative Stress-Induced Reproductive Defects.

The intrinsic and extrinsic factors (natural aging, diseases, infections, chemicals, radiation, heavy metals etc.) create oxidative stress, thereby causing reproductive defects in males and females. Similarly, oxidative stress causes the abnormalities in sperm and oocytes, and thus reduces the success of in vitro fertilization. Fungi and/or algae-derived metabolites (polysaccharides, carotenoids, ergothioneine, phycobiliproteins, phlorotannins, and cordycepin) alleviate the damages in ovaries and testis and correct the impaired reproductive functions (spermatogenesis, ovulation, fertilization
