Rich-club phenomenon in the interactome of P. falciparum--artifact or signature of a parasitic life style?

Recent advances have provided a first experimental protein interaction map of the human malaria parasite P. falciparum, which appears to be remotely related to interactomes of other eukaryotes. Here, we present a comparative topological analysis of this experimentally determined web with a network of conserved interactions between proteins in S. cerevisiae, C. elegans and D. melanogaster that have an ortholog in Plasmodium. Focusing on experimental interactions, we find a significant presence of a "rich-club," a topological characteristic that features an "oligarchy" of highly connected protei

Plants attract parasitic wasps to defend themselves against insect pests by releasing hexenol.

BACKGROUND: Plant volatiles play an important role in defending plants against insect attacks by attracting their natural enemies. For example, green leaf volatiles (GLVs) and terpenoids emitted from herbivore-damaged plants were found to be important in the host location of parasitic wasps. However, evidence of the functional roles and mechanisms of these semio-chemicals from a system of multiple plants in prey location by the parasitoid is limited. Little is known about the potential evolutionary trends between herbivore-induced host plant volatiles and the host location of their parasitoids

Malaria treatment efficacy among people living with HIV: the role of host and parasite factors.

Identification of an effect of HIV-associated immunosuppression on response to antimalarial therapy would help guide management of malaria i … 0.3-1.2). Among children, the risk of treatment failure increased with infection with SP-resistant parasites and anemia. Decreased CD4 cell count was not associated with impaired response to antimalarial therapy or diminished ability to clear SP-resistant parasites, suggesting that acquired immunity to malaria is retained in the face of HIV-associated immu …

Infection of wild fishes by the parasitic copepod Caligus elongatus on the south east coast of Norway.

Natural Caligus elongatus Nordmann infections of wild coastal fishes on the Norwegian south east coast were monitored at various times of the year from 2002 to 2004. The prevalence for all coastal fish (n = 4427) pooled was 15%, and there were great differences between fish species and seasons. Lumpfish Cyclopterus lumpus L. spawners were the most infected fish, with a prevalence of 61% and a median intensity of 4 lice fish(-1), whereas gadids had a mean prevalence of 19% and a median infection of 1 to 2 lice fish(-1). Sea trout Salmo trutta L. and herring Clupea harengus L. carried C. elongat

Identification of a molecular marker for genotyping human lymphatic filarial nematode parasite Wuchereria bancrofti.

In India, Mass Drug Administration is on going towards elimination of lymphatic filariasis in many areas, which might lead to intense selection pressure on the parasite populations and their genetic restructuring. This calls for molecular finger printing of Wuchereria bancrofti parasite populations at national level and monitoring genetic changes in the future. For this purpose a reliable, less expensive, rapid, and reproducible molecular tool is necessary, which is not available for W. bancrofti at this time. We identified robust molecular markers based on the comparison of random amplified p

Molecular cloning, purification and characterisation of myosin of human lymphatic filarial parasite Brugia malayi.

Global efforts have been made towards development of vaccine for prevention of lymphatic filariasis. However, lack of thorough knowledge about developmental biology and pathogenesis of filarial parasite restricts us from developing an effective vaccine. A limited number of immunodominant antigens of human lymphatic filariid Brugia malayi have been characterised; however, none of these recombinant antigens so far induced significant degree of protective immunity to challenge infection. In the present study, we identified a approximately 2.0 Kb cDNA clone by immunoscreening of cDNA library of ad

The importance of parasite life history and host density in predicting the impact of infections in red deer.

We studied a macroparasite (Elaphostrongylus cervi, Nematoda) and a microparasite infection (tuberculosis, TB) in red deer (Cervus elaphus) across different populations where managers manipulated host condition, density and aggregation by providing supplemental food. We aimed to test whether and, if so, how persistence and transmission of both parasites differentially varied as host body condition and population density varied. We took account of sex, as red deer life history greatly concerns sex-related traits. Changes in host factors had different consequences for the spread of each parasite…

[Paragonimiasis westermani with repeated pleural effusion and diagnosed by detecting parasite eggs in pleural effusion].

A 73-year-old man was referred to our hospital because of left chest pain, left pleural effusion and peripheral blood eosinophilia. After three months, left pleural effusion resolved without treatment and the etiology remained unclarified. Three years later, he was admitted for evaluation of right pleural effusion. Peripheral blood eosinophil count and serum IgE value were elevated. Eosinophilia was also observed in the pleural effusion. The diagnosis of paragonimiasis westermani was confirmed by immunoserological examination of the serum and by detection of Paragonimius eggs in the pleural ef

Tungiasis: high prevalence, parasite load, and morbidity in a rural community in Lagos State, Nigeria.

BACKGROUND: Tungiasis is common in resource-poor populations throughout Latin America, the Caribbean, and sub-Saharan Africa, but epidemiologic data from Africa on this ectoparasitosis are scarce. METHODS: A cross-sectional study was carried out in a representative sample of a rural community in Lagos State, 54 km west of Lagos (Nigeria). In the dry season, 142 households of the community were randomly selected and visited. Family members were examined for the presence of tungiasis. The localization, number, and stage of penetrated fleas, as well as the associated morbidity, were documented.

Immunomodulatory therapy associated to anti-parasite drugs as a way to prevent severe forms of malaria.

Malaria is an important problem of public health. It is estimated that 350 to 500 million clinical cases occur annually, which cause 1.1 and 1.3 million deaths every year. The excessive activation of the immune system plays an important role in the pathogenesis of the disease. The cells of the immune system of Plasmodium-infected individuals not only produce large amounts of cytokines, which have anti-parasite effects, but also participate in the pathogenesis of the severe complications of malaria. A central feature of P. falciparum infection is the sequestration of parasitized erythrocytes wi

A novel high-affinity arginine transporter from the human parasitic protozoan Leishmania donovani.

We describe the first functional and molecular characterization of an amino acid permease (LdAAP3) from the human parasitic protozoan Leishmania donovani, the causative agent of visceral leishmaniasis in humans. This permease contains 480 amino acids with 11 predicted trans-membrane domains. Expressing LdAAP3 in Saccharomyces cerevisiae mutants revealed that LdAAP3 codes for a high-affinity arginine transporter (Km 1.9 microM). LdAAP3 is highly specific for arginine as its transport was not inhibited by other amino acids or arginine-related compounds. Using green fluorescence protein (GFP) fus

The Trypanosoma brucei cAMP phosphodiesterases TbrPDEB1 and TbrPDEB2: flagellar enzymes that are essential for parasite virulence.

Cyclic nucleotide specific phosphodiesterases (PDEs) are pivotal regulators of cellular signaling. They are also important drug targets. Besides catalytic activity and substrate specificity, their subcellular localization and interaction with other cell components are also functionally important. In contrast to the mammalian PDEs, the significance of PDEs in protozoal pathogens remains mostly unknown. The genome of Trypanosoma brucei, the causative agent of human sleeping sickness, codes for five different PDEs. Two of these, TbrPDEB1 and TbrPDEB2, are closely similar, cAMP-specific PDEs conta

No evidence for specificity between host and parasite genotypes in experimental Strongyloides ratti (Nematoda) infections.

A key requirement for several theories involving the evolution of sex and sexual selection is a specificity between host and parasite genotypes, i.e. the resistance of particular host genotypes to particular parasite genotypes and the infectivity of particular parasite genotypes for particular host genotypes. Determining the scope and nature of any such specificity is also of applied relevance, since any specificity for different parasite genotypes to infect particular host genotypes may affect the level of protection afforded by vaccination, the efficacy of selective breeding of livestock for

Intestinal parasitic infections in HIV/AIDS patients: experience at a teaching hospital in central Brazil.

In order to verify the occurrence of intestinal parasitic infections in human immunodeficiency virus (HIV)/acquired immunodeficiency syndrome (AIDS) patients, 100 HIV/AIDS patients (Group 1) and 85 clinically healthy individuals (Group 2) were submitted to coproparasitological examination. Intestinal parasites were detected in 27% of patients from Group 1 and in 17.6% from Group 2. In Group 1 the most frequent parasites were Strongyloides stercoralis (12%), with 2 cases of hyperinfection; Isospora belli, 7%; Cryptosporidium sp., 4%; with 1 asymptomatic case and hookworm, 4%. Of the infected pa…

Structure of the major cytosolic glutathione S-transferase from the parasitic nematode Onchocerca volvulus.

Onchocerciasis is a debilitating parasitic disease caused by the filarial worm Onchocerca volvulus. Similar to other helminth parasites, O. volvulus is capable of evading the host's immune responses by a variety of defense mechanisms, including the detoxification activities of the glutathione S-transferases (GSTs). Additionally, in response to drug treatment, helminth GSTs are highly up-regulated, making them tempting targets both for chemotherapy and for vaccine development. We analyzed the three-dimensional x-ray structure of the major cytosolic GST from O. volvulus (Ov-GST2) in complex with…

Identification of novel genes in intestinal tissue that are regulated after infection with an intestinal nematode parasite.

Infection of resistant or susceptible mice with Trichuris muris provokes mesenteric lymph node responses which are polarized towards Th2 or Th1, respectively. These responses are well documented in the literature. In contrast, little is known about the local responses occurring within the infected intestine. Through microarray analyses, we demonstrate that the gene expression profile of infected gut tissue differs according to whether the parasite is expelled or not. Genes differentially regulated postinfection in resistant BALB/c mice include several antimicrobial genes, in particular, intele

Histochemistry of glycoconjugates in mucous cells of Salmo trutta uninfected and naturally parasitized with intestinal helminths.

Mucus secreted onto the surface of the intestine forms a physical barrier to invading parasites so that a possible attachment of helminths to the surface is prevented and their expulsion by peristalsis facilitated. In mammals, intestinal parasites induce hyperplasia and hypertrophy of intestine goblet cells and provoke changes in the mucus composition. In fish, this topic has received less attention. In the present investigation, histochemical methods were employed to compose intestinal mucous cell numbers and their glycoconjugate composition were compared by uninfected brown trout Salmo trutt…

Murine malaria parasite sequestration: CD36 is the major receptor, but cerebral pathology is unlinked to sequestration.

Sequestration of malaria-parasite-infected erythrocytes in the microvasculature of organs is thought to be a significant cause of pathology. Cerebral malaria (CM) is a major complication of Plasmodium falciparum infections, and PfEMP1-mediated sequestration of infected red blood cells has been considered to be the major feature leading to CM-related pathology. We report a system for the real-time in vivo imaging of sequestration using transgenic luciferase-expressing parasites of the rodent malaria parasite Plasmodium berghei. These studies revealed that: (i) as expected, lung tissue is a majo…
