Memory B cell Development in Response to mRNA SARS-CoV-2 and Nanoparticle Immunization in Mice.

UNLABELLED: Nanoparticle immunogens excel at rapidly inducing high levels of circulating antibodies and are being deployed as part of several novel vaccines. However, their ability to elicit memory B cell responses is less well understood. Here we compared serologic and memory B cell responses to prime boost vaccination with either SARS-CoV-2 Wuhan-Hu-1 mRNA vaccine, or protein nanoparticles: SARS-CoV-2 B.1.351 homotypic containing a single receptor binding domain (RBD); (homotypic beta) or a combination of different Sarbecovirus RBDs (mosaic 8b), respectively. The memory B cells elicited by t…

The virology of Omicron: pathophysiology, immune regulation, and clinical impact of SARS-CoV-2 sub variants.

Since its emergence in late 2019, SARS-CoV-2 has evolved into multiple variants with distinct genetic and clinical features. Among them, the Omicron variant (B.1.1.529) and its sublineages BA.2.75, JN.1.8, and KP.2 have shown enhanced transmissibility and immune evasion, while generally exhibiting reduced lower respiratory tract pathogenicity compared to earlier variants, thereby continuing to pose significant challenges to public health. In India, these variants have significantly shaped the trajectory of the pandemic, necessitating focused evaluation of their biological and clinical impact. …

Timing of surgery after fever resolution in children with SARS-CoV-2: a prospective cohort study.

BACKGROUND: Fever in children with SARS-CoV-2 infection may increase the risk of intraoperative oxygen desaturation during surgery. This study seeks to find the optimal surgery timing by examining oxygen desaturation rates after fever resolution. METHODS: A prospective cohort study from March to August 2023 included children with SARS-CoV-2 infection who were scheduled for surgery after fever resolution. The primary outcome was the incidence of intraoperative oxygen desaturation. Logistic regression models were used to calculate the adjusted incidence of oxygen desaturation, stratified by tim…

Discovery of thiosemicarbazone derivatives as promising SARS-CoV-2 Mpro inhibitors by spectroscopy and microscale thermophoresis.

Since the outbreak of COVID-19, one key strategy has been to screen inhibitors of the SARS-CoV-2 main protease (Mpro). Mpro is critical for viral replication and a key target for therapeutic development. In this work, we report the design and synthesis of 29 thiosemicarbazone derivatives. Enzymatic inhibition assays showed that 27 compounds exhibited inhibitory activity against Mpro, among which 4 compounds (1e, 2c, 2f, 3a) demonstrated significant inhibitory potency, with IC50 values of 5.97 ± 0.98 μM, 4.43 ± 1.67 μM, 10.99 ± 2.39 μM, and 28.88 ± 3.57 μM, respectively. Antibody escape of SARS-CoV-2 variants of concern on receptor-binding domain: A computational approach.

The receptor-binding domain (RBD) of the spike protein is a critical functional component responsible for binding between the SARS-CoV-2 and the ACE2 receptor, as well as monoclonal antibodies. This research focuses on evaluating the ability of SARS-CoV-2 variants to reduce or evade neutralizing antibody responses. The RBD structures of wild type, Delta, and Omicron structures along with nine RBD-directed antibodies downloaded from the Protein Data Bank were subjected to docking simulations via the HADDOCK 2.4 server to calculate Haddock score, binding affinity (ΔG) and dissociation constant (…

Disinfection of Simulated Residual SARS-CoV-2 in Air by Chlorine Dioxide Gas in an Ambulance Setting.

INTRODUCTION: Disinfecting contaminated ambulances presents a challenge to emergency medical services. The process relies on a manual cleaning step or the use of foggers and gas generators. Equipment requires training, power supply, and access to reagent refills. Chlorine dioxide (ClO2) gas is an affordable, easy-to-use alternative. METHODS: We field-tested ClO2 dry inside an ambulance against a surrogate for SARS-CoV-2. MS2 bacteriophage was aerosolized inside of an enclosed. Airborne MS2 concentrations were measured before and after ClO2 applications to compare reductions in viable virus. C…

Booster effect of the fourth dose of the SARS-CoV-2 mRNA vaccine in kidney transplant recipients.

BACKGROUND: Solid organ transplant recipients taking immunosuppressive drugs are at greater risk of severe COVID-19 than the general population. In particular, kidney transplant recipients (KTRs) are known to have lower seropositivity after basal doses of SARS-CoV-2 vaccines, and the strategy of administering booster doses in these immunocompromised individuals has been promoted worldwide. METHODS: This study evaluated the effect of a fourth dose (D4) of SARS-CoV-2 vaccine in Japanese KTRs. Anti-spike (anti-S) IgG antibody titers at 1 and 3 months after D4 of SARS-CoV-2 vaccine were evaluated…

No evidence of immune exhaustion after repeated SARS-CoV-2 vaccination in vulnerable and healthy populations.

Frequent SARS-CoV-2 vaccination in vulnerable populations has raised concerns that this may contribute to T cell exhaustion, which could negatively affect the quality of immune protection. Herein, we examined the impact of repeated SARS-CoV-2 vaccination on T cell phenotypic and functional exhaustion in frail older adults in long-term care (n = 23), individuals on immunosuppressive drugs (n = 10), and healthy adults (n = 43), in Canada. Spike-specific CD4+ and CD8+ T cell levels did not decline in any cohort following repeated SARS-CoV-2 vaccination, nor did the expression of exhaustion marker…

Vascular endothelial growth factor A: friend or foe in the pathogenesis of HIV and SARS-CoV-2 infections?

This review article discusses the role of vascular endothelial growth factor A (VEGF-A) in the pathogenesis of SARS-CoV-2 and HIV infection, both conditions being renowned for their impact on the vascular endothelium. The processes involved in vascular homeostasis and angiogenesis are reviewed briefly before exploring the interplay between hypoxia, VEGF-A, neuropilin-1 (NRP-1), and inflammatory pathways. We then focus on SARS-CoV-2 infection and show how the binding of the viral pathogen to the angiotensin-converting enzyme 2 receptor, as well as to NRP-1, leads to elevated levels of VEGF-A an…

Strong evidence for the evolution of decreasing compositional heterogeneity in SARS-CoV-2 genomes during the pandemic.

The rapid evolution of SARS-CoV-2 during the pandemic was characterized by the fixation of a plethora of mutations, many of which enable the virus to evade host resistance, likely altering the virus' genome compositional structure (i.e., the arrangement of compositional domains of varying lengths and nucleotide frequencies within the genome). To explore this hypothesis, we summarize the evolutionary effects of these mutations by computing the Sequence Compositional Complexity (SCC) in random stratified datasets of fully sequenced genomes. Phylogenetic ridge regression of SCC against time revea…

Cracking the code of a correlate of protection against SARS-CoV-2 breakthrough infection in cancer patients.

The level of protection against SARS-CoV-2 breakthrough infections conferred by the presence of anti-S1 SARS-CoV-2 antibodies (IgGs) in cancer patients is still understudied. This work examines the existence of an anti-S1 immunoglobulin G (IgG) -based correlate of protection (CoP) established by prospectively collected observational data about breakthrough infections with different SARS-CoV-2 variants in a large cohort study with vaccinated cancer patients. 760 cancer patients were longitudinally followed-up, starting before first vaccination until six months after second booster. Anti-S1 SARS…

Vaccination prior to SARS-CoV-2 infection does not affect the neurologic manifestations of long COVID.

Persistent symptoms after COVID-19 constitute the long COVID syndrome, also called post-acute sequelae of SARS-CoV-2 infection (PASC). COVID-19 vaccines reduce the gravity of ensuing SARS-CoV-2 infections. However, whether vaccines also have an impact on PASC remain unknown. We investigated whether vaccination prior to infection alters the subsequent neurologic post-acute sequelae of SARS-CoV-2 infection (Neuro-PASC). We studied prospectively the first consecutive 200 post-hospitalization Neuro-PASC (PNP) and 1100 non-hospitalized Neuro-PASC (NNP) patients evaluated at our neuro-COVID-19 clini…

The Association of SARS-CoV-2 Infection and COVID-19 Vaccination With Sudden Death: An Explorative Review.

Since its discovery, the novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the causative agent of coronavirus disease 2019 (COVID-19), has become the epicenter of public health concern. This was mainly attributed to the complexity of COVID-19 that resulted in variable disease progression with some developing asymptomatic infections, some suffering mild to moderate infections that resolved without the need for hospitalizations, and a few infected persons developing severe infections that required intensive care unit (ICU) admission and mechanical ventilation. The COVID-19 pand…

Soluble CD13 is a potential mediator of neutrophil-induced thrombogenic inflammation in SARS-CoV-2 infection.

The soluble variant of the ectopeptidase CD13 (sCD13), released from the cell surface by matrix metalloproteinase 14 (MMP14), is a potent pro-inflammatory mediator, displaying chemotactic, angiogenic, and arthritogenic properties through bradykinin receptor B1 (B1R). We revealed a link between sCD13 and amplified neutrophil-mediated inflammatory responses in SARS-CoV-2 infection. sCD13 was markedly elevated in patients with COVID-19 and correlated with disease severity and variants, ethnicity, inflammation markers, and neutrophil extracellular trap formation (NETosis). Neutrophils treated with…
