Developmental conditions and adult socioeconomic context jointly shape cognitive aging through brain reserve.

OBJECTIVES: Cognitive aging shows substantial inter-individual heterogeneity, which may be shaped by both early-life developmental conditions and adult socioeconomic environments. However, how these life-course factors jointly relate to cognitive aging and the potential neuroanatomical correlates underlying these associations remain unclear. METHODS: Using data from 35,105 UK Biobank participants with birth weight information, comprehensive cognitive assessments, and brain MRI, we examined associations between early-life growth and domain-specific cognitive performance in mid- to late adultho

"Libre de Gluten" or "Gluten-Free"? The Influence of Loanwords on Conceptual Representation.

The expansion of the gluten-free food market is driven by two distinct factors: medical prescriptions for people with celiac disease, and lifestyle adoption among non-celiac consumers. This study examines how the use of the English term "gluten-free" may influence the cognitive conceptualization of the product compared to the term "libre de gluten" among two groups. Methodologically, the cognitive structure of 240 Mexican consumers (120 with celiac disease and 120 without) was evaluated using social representation theory and the word association technique. Findings reveal that the language use

Can socioeconomic status moderate the effect of a conflictive family environment on brain structure and externalizing/internalizing behavior in children and adolescents?

OBJECTIVE: This study examined the effects of socioeconomic status as a possible moderator of the effects of family conflict on externalizing/internalizing behavior and hippocampal and amygdala volume. METHODS: A longitudinal complete-case analysis of 714 children and adolescents (mean age: 11.2 years; 46.2% female) was conducted using data from the Brazilian High-Risk Cohort Study for Psychiatric Disorders in Childhood. At baseline, parents/guardians completed the Family Environment Scale and a socioeconomic status scale. Three years after baseline assessment, the same participants underwent

Neuroticism heterogeneity through item-level associations in resting-state functional connectivity.

Neuroticism is characterized by emotional instability and increased susceptibility to stress-related disorders. While traditionally treated as a unitary construct, growing evidence suggests that neuroticism is heterogeneous in both its genetic basis and its effects on social and health outcomes. To quantify this heterogeneity at the neurofunctional level, we analyzed resting-state functional connectivity (RSFC) in a large sample (n = 33,180) from the UK Biobank dataset. Using machine learning regression analysis, we identified RSFC patterns associated with item-level responses from the neuroti

Loneliness Links Adverse Childhood Experiences to Mortality Risk Across 26 Years.

OBJECTIVES: Adverse childhood experiences (ACEs) are associated with increased mortality risk. Individuals with a history of certain adversity during childhood tend to report higher levels of loneliness in later life. In our preregistered study, we examined whether loneliness mediates the ACEs to mortality risk relation. METHODS: Participants were from the Midlife in the United States Survey (N = 4,963; M [SD] = 46.44 [12.52] years, 53.3% female). Follow-up period spanned 26 years. A comprehensive measure of ACEs was employed consisting of 20 ACEs from 5 categories: physical abuse, emotional

Seminoma Caused by PDE11A Frameshift Variant with Multiple Abnormalities: A Case Report.

INTRODUCTION: Testicular germ cell tumor is a rare malignant tumor predominantly affecting young males. In addition to well-established risk factors like cryptorchidism, genetic factors are increasingly recognized as significant contributors. This case report highlights a patient diagnosed with seminoma associated with multiple abnormalities and explores the genetic basis behind these manifestations. CASE PRESENTATION: A 22-year-old East Asian male presented with left-sided varicocele and recent onset testicular pain. Clinical examination and imaging revealed a smaller left testicle and a tes

Therapeutic targets in aging-related osteoarthritis: A focus on the extracellular matrix homeostasis.

Osteoarthritis (OA) represents a globally prevalent degenerative bone diseases and is the primary contributors to pain and disability among middle-aged and elderly people, thereby imposing significant social and economic burdens. When articular cartilage is in the aging environment, epigenetic modifications, DNA damage and mitochondrial dysfunction lead to cell senescence. Chondrocyte senescence has been identified as a pivotal event in this metabolic dysregulation of the extracellular matrix (ECM). It can affect the composition and structure of ECM, and the mechanical and biological signals t

Environmental Enrichment and Epigenetic Changes in the Brain: From the Outside to the Deep Inside.

The brain plays a vital role in maintaining homeostasis and effective interaction with the environment, shaped by genetic and environmental factors throughout neurodevelopment and maturity. While genetic components dictate initial neurodevelopment stages, epigenetics-specifically neuroepigenetics-modulates gene expression in response to environmental influences, allowing for brain adaptability and plasticity. This interplay is particularly evident in neuropathologies like Rett syndrome and CDKL5 deficiency syndrome, where disruptions in neuroepigenetic processes underline significant cognitive

First-in-class SAM-competitive G9a inhibitor FLAV-27 as a disease-modifying therapy for Alzheimer disease.

Alzheimer's disease (AD) is characterized by a progressive cognitive decline involving a multifactorial pathophysiology, including epigenetic dysregulation. Here, we report the discovery and preclinical validation of FLAV-27, a first-in-class, S-adenosyl-l-methionine (SAM)-competitive, brain-penetrant, and selective inhibitor of the histone methyltransferase G9a. Unlike prior G9a/GLP inhibitors, FLAV-27 exhibits subnanomolar potency, over 30-fold selectivity, and robust central nervous system bioavailability. Structural studies confirm a unique SAM-binding mode that confers superior specificit

What we got wrong about depression and its treatment.

The paradigm is shifting with respect to how we think about depression and its treatment. Some of that shift can be attributed to new findings with respect to its epidemiology and genetics and the rest can be attributed to the incorporation of a new perspective derived from evolutionary theory. In brief, depression is far more prevalent than previously recognized with the bulk of additional cases involving individuals who do not go on to become recurrent. Nonpsychotic unipolar depression (but not bipolar mania which likely is a "true" disease) appears to be an adaptation that evolved to facili

Economic Rationality in Decision-Making Regarding Newborn Screening: A Case Study in Quebec.

Health systems in high-resource countries recognize the importance of making decisions about the services offered to the population based on scientific evidence. Producing this evidence is especially challenging in areas such as newborn care where the frequency of conditions is rare. However, methodological advances in the field of economic evaluation could change how this evidence is used in decision-making. This study aimed to investigate how decision-makers in the Canadian province of Quebec perceive the value of recent advances in economic evaluations for perinatal studies and how these ad

Item Response Theory Analysis and Differential Item Functioning of the Social Appearance Anxiety Scale.

This study examined the item- and scale-level functioning of the Social Appearance Anxiety Scale (SAAS) as well as differential functioning by gender using an item response theory (IRT) analysis. SAAS data collected from 840 college students were analyzed. A graded response model was used to analyze the 16 items comprising the SAAS. The measure was found to be unidimensional in its factor structure, and every item demonstrated high to very high ability to differentiate respondents varying in levels of the underlying trait (i.e., appearance concerns). In addition, we found evidence of different

Epigenetic Modifications and Neuroplasticity in the Pathogenesis of Depression: A Focus on Early Life Stress.

Major depressive disorder (MDD) is a debilitating mental illness, and it is considered to be one of the leading causes of disability globally. The etiology of MDD is multifactorial, involving an interplay between biological, psychological, and social factors. Early life represents a critical period for development. Exposure to adverse childhood experiences is a major contributor to the global burden of disease and disability, doubling the risk of developing MDD later in life. Evidence suggests that stressful events experienced during that timeframe play a major role in the emergence of MDD, le

Drug repurposing strategy II: from approved drugs to agri-fungicide leads.

Epidemic diseases of crops caused by fungi deeply affected the course of human history and processed a major restriction on social and economic development. However, with the enormous misuse of existing antimicrobial drugs, an increasing number of fungi have developed serious resistance to them, making the diseases caused by pathogenic fungi even more challenging to control. Drug repurposing is an attractive alternative, it requires less time and investment in the drug development process than traditional R&D strategies. In this work, we screened 600 existing commercially available drugs, some

Black Women's Maternal Health: Insights From Community Based Participatory Research in Newark, New Jersey.

This study on Black women's maternal health engaged a group of six community members in a community based participatory research project in a state with one of the largest racial disparities in maternal mortality and severe maternal morbidity in the United States. The community members conducted 31 semi-structured interviews with other Black women who had given birth within the past 3 years to examine their experiences throughout the perinatal and post-partum period. Four main themes emerged: (1) challenges related to the structure of healthcare, including insurance gaps, long wait times, lack

Who is vulnerable to energy poverty in China?

Energy poverty has been identified as a problem in China, but research to date does not discuss who experiences it, unlike in other countries. Here, we compared sociodemographic characteristics known to be linked to energy vulnerability in other nations between energy poor (EP) households and non-EP households, using the 2018 survey data from China Family Panel Studies (CFPS). We found that a range of sociodemographic characteristics associated with transport, education and employment, health, household structure, and social security, are disproportionately distributed among five provinces (Ga

Exome sequencing identifies homozygous variants in MBOAT7 associated with neurodevelopmental disorder.

Intellectual disability (ID) is a large group of neurodevelopmental disorders characterized by a congenital limitation in intellectual functioning (reasoning, learning, and problem solving), adaptive behavior (conceptual, social, and practical skills), originated at birth and manifested before the age of 18. By whole exome sequencing of five consanguineous Pakistani families presenting hallmark features of ID, global developmental delay, aggressive and self-injurious behaviors, microcephaly, febrile seizures and facial dysmorphic features, we identified three novel homozygous missense variants

Beluga whale and bottlenose dolphin ACE2 proteins allow cell entry mediated by spike protein from three variants of SARS-CoV-2.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) viruses infect numerous non-human species. Spillover of SARS-CoV-2 into novel animal reservoirs may present a danger to host individuals of these species, particularly worrisome in populations already endangered or threatened by extinction. In addition, emergence in new reservoirs could pose spillback threats to humans, especially in the form of virus variants that further mutate when infecting other animal hosts. Previous work suggests beluga whales (Delphinapterus leucas) and bottlenose dolphins (Tursiops truncatus) may be at risk
