Resolution Failure in Periodontal Diseases: Dysregulated Pro-resolving Mechanisms in Chronic Inflammation and Tissue Breakdown.

Periodontitis is a highly prevalent chronic inflammatory disease characterized by irreversible destruction of the tooth-supporting tissues. Although classically interpreted as the consequence of excessive inflammation that drives microbial dysbiosis, accumulating experimental and clinical evidence indicates that periodontitis can be more precisely described as a disorder of failed inflammatory resolution. In periodontal tissues-constantly exposed to microbial challenge-resolution is not a terminal event but a constitutive biological requirement essential for maintaining tissue homeostasis. Thi

Uncommon intraoral source of generalized tetanus: Case report and review of intensive care strategies.

INTRODUCTION: Tetanus is an acute disease caused by the tetanospasmin toxin produced by Clostridium tetani, with a high mortality particularly in developing countries. Although preventable through primary immunization and booster vaccination, cases in adults with uncertain immunization status remain common. Intraoral wounds are uncommonly reported as portals of entry. CASE PRESENTATION: A 54-year-old male developed generalized tetanus after manipulating a tooth with a toothpick, causing bleeding. Initial dental pain and oral numbness progressed to trismus, opisthotonus, and risus sardonicus.

Rationalization Progress of Novel Drug Delivery System for Intra-periodontal Pockets Against Periodontitis.

Periodontitis (PD) is a pathological condition that results in chronic swelling in the tissue around a tooth, which results in advanced periodontal structural injury to the encircling soft and hard tissues with eventual exfoliation and movement of teeth. It affects around 60% of the world's population, indicating a relatively high prevalence. Therefore, the discovery of efficient therapeutic interventions for dental disorders is a primary goal of the health sciences, and periodontitis is a significant public health problem. Currently, perioceutics plays a revolutionary role in periodontal ther

Photodynamic therapy in non-surgical treatment of periodontitis.

Periodontitis is a chronic inflammation disease that, apart from caries, remains the main cause of tooth loss, that lead to a deterioration of the quality of life. Non-surgical treatment is the basic form of periodontal therapy, regardless of the stage of the disease. Novel supportive treatment methods are constantly implemented to improve the effects of non-surgical treatment, including photodynamic therapy that leads to a selective photosensitization of bacteria with the simultaneous participation of a photosensitizer applied into the periodontal pocket. The aim of the study is to evaluate t

Foot orthosis design for children with Charcot-Marie-Tooth and impact on gait.

BACKGROUND: Charcot-Marie-Tooth (CMT) is a progressive disease resulting in distal sensory deficit and muscular weakness. As the disease progresses, most children develop a cavovarus foot deformity. Foot orthoses (FO) are widely prescribed in CMT to support the foot and improve gait, but there is a lack of guidelines for their conception. The aim of this pilot study was to report the methodology used for the design of FO (FOmax) based on an evaluation of foot deformities and to assess its effects on gait in children with CMT. METHODS: This study included 11 children with CMT. Participants wer

On-Site Visualized Periodontitis Screening via Dual-Signal H2S Probe.

Periodontitis is a destructive disease that causes irreversible damage to periodontal supporting tissues, posing a serious threat to patients' oral health. Given the insidious nature of periodontal tissue destruction, early diagnosis of periodontitis is crucial. Salivary hydrogen sulfide (H2S) has been identified as a potential biomarker for periodontitis monitoring. However, existing detection methods struggle to precisely locate the source of H2S release within the oral cavity, making it difficult to accurately identify affected areas and significantly hindering early diagnosis and treatment

From Disease to Illness: Reframing Periodontitis Through an Anthropological Lens.

While periodontitis is globally recognized as a significant public health problem, its common definition as a plaque-based inflammatory condition is incomplete. Disease progression, personal experience, and treatment are shaped by social, economic, and structural forces largely invisible in clinical practice and policy. A lens from medical anthropology helps us see periodontitis as more than a clinical diagnosis; it is a lived experience, deeply entangled with a person's social world. The physical reality of inflammation translates into profound emotional distress-from the shame and stigma of

The Ighmbp2 -R604X mouse recapitulates the severe SMARD1 clinical symptoms of aspiration, respiratory and feeding deficits.

Spinal muscular atrophy with respiratory distress type 1 (SMARD1) and Charcot Marie Tooth type 2S (CMT2S) are due to mutations in immunoglobulin mu binding protein two (IGHMBP2). We generated the Ighmbp2 -R604X mouse (R605X-humans) to understand how alterations in IGHMBP2 function impact disease pathology. The IGHMBP2-R605X mutation is associated with patients with SMARD1 or CMT2S. The impact of this mutation is substantial, Ighmbp2 R604X/R604X mice have a decreased lifespan (6 days) and weight, and failure to thrive consistent with SMARD1 symptoms. Significant respiratory changes were present

AI-assisted phenotyping in a zebrafish hypophosphatasia model enables early and precise detection of skeletal alterations.

Hypophosphatasia (HPP) is a rare genetic disorder mainly affecting bone and tooth mineralization in patients due to ALPL gene mutations. Understanding genotype-phenotype correlations in HPP remains challenging due to different severities and the disease's heterogeneity. To address this, we established a novel zebrafish animal model (alplwue7), which mimics severe HPP disease forms. To bypass limitations in human-based phenotypic classification of skeletal alterations in this transgenic line, we developed and trained an artificial intelligence (AI) model capable of image-based classification wi

One Year Follow-Up of a 4-Year-Old Caucasian Girl Diagnosed with Stage IV Grade C Localized Periodontitis.

Stage IV grade C localized periodontitis (pre-puberal localized aggressive periodontitis/LPP), an extremely rare form of periodontal disease, occurs in otherwise healthy individuals (no signs of dental plaque/calculus) due a hyper-aggressive auto-immune response to high periodontopathic bacteria levels. Methods: A 4-year-old Caucasian girl with unusually high mobility of the deciduous lower left canine and localized gingival inflammation was misrecognized by multiple clinicians (initially diagnosed with hypophosphatasia, genetic and metabolic disorders, all turning negative), over a period of

Chapter 2: Epidemiology of Coronal Caries.

This chapter summarizes the clinical data on coronal caries distribution and sociodemographic drivers in children, adults, and older people, at a global level. Mapping global caries prevalence showed an extreme variation on the prevalence, revealing still high rates in several countries. The disease is presented in each group as prevalence by age and mean number of affected teeth. Differences in the prevalence of dental caries in developed and developing countries may be due to not only wide variations in the age groups studied but also ethnic, cultural, geographic, and developmental differenc

A literature review of local and systemic considerations for endodontic treatments in older adults.

OBJECTIVES: The aim of this literature review was to summarise the clinical important findings on the endodontic treatment outcome in older patients (≥60 years old) with pulpal/periapical disease considering local and systemic factors from a body of knowledge that is heterogeneous in methods or disciplines. BACKGROUND: Due to the increasing number of older patients in the endodontic practice, and the current trend for tooth preservation, the need for clinicians to have a better understanding of age-related implications that may influence the required endodontic treatment in older adults to re

Upside-Down Preference in the Forskolin-Induced In Vitro Differentiation of 50B11 Sensory Neurons: A Morphological Investigation by Label-Free Non-Linear Microscopy.

In this study, we revealed a peculiar morphological feature of 50B11 nociceptive sensory neurons in in vitro culture related to the forskolin-induced differentiation of these cells growing upside-down on cover glass supports. Multi-photon non-linear microscopy was applied to monitor increased neurite arborization and elongation. Under live and unstained conditions, second harmonic generation (SHG) microscopy could monitor microtubule organization inside the cells while also correlating with the detection of cellular multi-photon autofluorescence, probably derived from mitochondria metabolites.

Portrait of Dysferlinopathy: Diagnosis and Development of Therapy.

Dysferlinopathy is a disease caused by a dysferlin deficiency due to mutations in the DYSF gene. Dysferlin is a membrane protein in the sarcolemma and is involved in different functions, such as membrane repair and vesicle fusion, T-tubule development and maintenance, Ca2+ signalling, and the regulation of various molecules. Miyoshi Myopathy type 1 (MMD1) and Limb-Girdle Muscular Dystrophy 2B/R2 (LGMD2B/LGMDR2) are two possible clinical presentations, yet the same mutations can cause both presentations in the same family. They are therefore grouped under the name dysferlinopathy. Onset is typi

Expanding the phenotype of males with OFD1 pathogenic variants-a case report and literature review.

Pathogenic variants in the OFD1 gene have been classically associated with the Orofaciodigital syndrome type 1 in females, a condition previously considered to be X-linked dominant with male embryonic lethality. However, an increasing number of males with pathogenic OFD1 variants who survived beyond the neonatal period have now been reported in the literature. Although each new report has added to the ever-broadening spectrum of clinical findings seen in males, many questions about genotype-phenotype correlations and disease mechanism remain. Herein, we describe a 9-year-old male child with a

WARS1 and SARS1: Two tRNA synthetases implicated in autosomal recessive microcephaly.

Aminoacylation of transfer RNA (tRNA) is a key step in protein biosynthesis, carried out by highly specific aminoacyl-tRNA synthetases (ARSs). ARSs have been implicated in autosomal dominant and autosomal recessive human disorders. Autosomal dominant variants in tryptophanyl-tRNA synthetase 1 (WARS1) are known to cause distal hereditary motor neuropathy and Charcot-Marie-Tooth disease, but a recessively inherited phenotype is yet to be clearly defined. Seryl-tRNA synthetase 1 (SARS1) has rarely been implicated in an autosomal recessive developmental disorder. Here, we report five individuals w
