Hepatic drug metabolism and adverse hepatic drug reactions.

Drugs and other chemicals are usually metabolized in the liver in the drug-metabolizing enzyme system. The metabolites sometimes bind with cellular macromolecules and injure the cell directly or serve as new antigens to create immunologic injury in a delayed fashion. The immediate or toxic injury is dose-dependent, predictable and zonal in the liver lobule, usually in the central region. Carbon tetrachloride intoxication and acetaminophen overdose are examples of injury resulting from microsomal metabolism. Other injuries related to microsomal metabolism are those produced by vinyl chloride in

SARS-coronavirus-2 replication in Vero E6 cells: replication kinetics, rapid adaptation and cytopathology.

The sudden emergence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) at the end of 2019 from the Chinese province of Hubei and its subsequent pandemic spread highlight the importance of understanding the full molecular details of coronavirus infection and pathogenesis. Here, we compared a variety of replication features of SARS-CoV-2 and SARS-CoV and analysed the cytopathology caused by the two closely related viruses in the commonly used Vero E6 cell line. Compared to SARS-CoV, SARS-CoV-2 generated higher levels of intracellular viral RNA, but strikingly about 50-fold less inf

Accumulation of human-adapting mutations during circulation of A(H1N1)pdm09 influenza virus in humans in the United Kingdom.

UNLABELLED: The influenza pandemic that emerged in 2009 provided an unprecedented opportunity to study adaptation of a virus recently acquired from an animal source during human transmission. In the United Kingdom, the novel virus spread in three temporally distinct waves between 2009 and 2011. Phylogenetic analysis of complete viral genomes showed that mutations accumulated over time. Second- and third-wave viruses replicated more rapidly in human airway epithelial (HAE) cells than did the first-wave virus. In infected mice, weight loss varied between viral isolates from the same wave but sho

Changes in the receptorbinding haemagglutinin protein of wild-type morbilliviruses are not required for adaptation to Vero cells.

We examined the consequences of isolation and adaptation to Vero cells for the receptorbinding haemagglutinin (H) gene of four syncytia-forming isolates of canine distemper virus (CDV) and of a dolphin morbillivirus isolate. A Vero-adapted CDV isolate exhibited biased hypermutation, since 11 out of 12 nucleotide differences to other isolates from the same epidemic were U-C transitions. Most of these transitions appeared to have taken place during in vitro cultivation. Previously, biased hypermutation in morbilliviruses has almost exclusively been described for subacute sclerosing panencephalit

Multi-tissue resource characteristics and comprehensive utilization of medicinal and edible loquat (Eriobotrya japonica): a review.

Loquat (Eriobotrya japonica (Thunb.) Lindl.), a member of the Rosaceae family, exhibits pronounced tissue-specific metabolic specialization that underpins its nutritional value, pharmacological activities, and industrial potential. Contemporary research on loquat has largely focused on single tissues or individual bioactive compounds, whereas comprehensive cross-tissue comparative profiling and mechanistic analyses remain substantially underexplored. This fragmented research paradigm obscures both the tissue-specific distribution of secondary metabolites and the upstream regulatory networks go

Pulsed field ablation: from concepts to technology.

AIMS: Pulsed field ablation (PFA) has emerged as a transformative approach for treating cardiac arrhythmias by exploiting irreversible electroporation to preferentially injure myocardium while limiting collateral damage. Translating this biophysical principle into safe and effective clinical systems requires careful integration of waveform design, catheter engineering, computational and experimental modelling, safety testing, and procedural workflow. OBJECTIVE: This review summarizes the fundamental physical and engineering principles that govern cardiac PFA and examines how these principles

Long-term monitoring of a full-scale parallel-unit downflow hanging sponge system with adaptive capacity for fluctuating wastewater flows in Japan.

This study presents a 30-month full-scale evaluation of a parallel-unit downflow hanging sponge (DHS) system with adaptive downsizing capability at the Susaki Sewage Treatment Plant, Japan. The DHS reactor comprised ten identical units operated in parallel, allowing capacity adjustment by varying the number of active units. The system (primary sedimentation tank → DHS → moving bed biological filter) was monitored under normal, low-temperature (winter), partial downsizing, and sponge sheet installation (upper part of the sponge media-filled segment) conditions. Dichromate-based chemical oxygen

POTN-2: A deep learning-based predictor of HLA-A*02:01-restricted tumor neoantigens.

Tumor neoantigens arise from somatic mutations in malignant cells and are strictly tumor-specific, minimizing autoimmune risks, making them ideal targets for T-cell immunotherapy. However, conventional pan-HLA predictors suffer from reduced accuracy and high false-positive rates for individual HLA subtypes. Our prior POTN model was trained on experimentally validated HLA-A02:01 peptide data. We developed POTN-2, an allele-specific predictor exclusively for HLA-A02:01-restricted tumor neoantigens, by expanding the feature repertoire and algorithmic architecture. We curated a comprehensive datas

Biomarkers of experimental Toxocara canis infection are context-dependent: A systematic review of murine models.

BACKGROUND: Toxocara canis infection is a neglected zoonosis associated with multisystemic involvement and persistent larval infection. Murine models are widely used to investigate host-parasite interactions, but methodological heterogeneity limits biomarker comparison and validation. OBJECTIVE: To characterize experimental murine T. canis infection models and synthesize parasitological, pathological, immunological, molecular, and functional biomarkers and outcomes. METHODS: PubMed/MEDLINE, Embase, Scopus, Web of Science Core Collection, and Virtual Health Library were searched from inceptio

Construction of a survival model for predicting biochemical recurrence of prostate cancer based on propionate metabolism-related genes.

BACKGROUND: About 20-40% of prostate cancer (PCa) develop biochemical recurrence (BCR) after surgery, and propionate metabolism may contribute to tumor progression. BCR remains a major clinical challenge in PCa, as current tools based on histopathology and prostate-specific antigen (PSA) fail to capture the molecular heterogeneity driving the disease. While metabolic reprogramming is known to facilitate post-treatment adaptation, the specific role of propionate metabolism in this context remains largely unexplored. Therefore, this study aimed to systematically investigate propionate metabolism

HistoNeRF: An Accessible and Intelligent Approach for Comprehensive 2D-to-3D Histological Assessment.

Histological analysis is central to biomedical research and diagnostic pathology, yet conventional two-dimensional (2D) sectioning captures only limited aspects of tissue architecture. Critical spatial relationships-such as tumor boundaries, stromal organization, and vascular networks-remain obscured, restricting diagnostic accuracy and biological interpretation. HistoNeRF addresses these limitations by adapting Neural Radiance Fields (NeRF) to reconstruct three-dimensional (3D) tissue volumes from routine histological sections. In this study, 84 toluidine blue (TB)-stained murine ovarian sect

A review of research on crawling soft robots: bionic mechanisms and actuation motion.

Crawling soft robots have attracted widespread attention due to their high adaptability to unstructured environments. However, existing research often focuses on understanding the formation mechanism of their motion capabilities from the perspective of actuation methods or control strategies, resulting in fragmented design logic and a lack of a unified framework. Through comparative analysis across animal, plant, and microbial systems, this paper points out that crawling behavior in different biological systems largely depends on the synergistic effect of morphological deformation and interfac

Intralesional microbial community signatures across histopathologic categories of tongue lesions.

BACKGROUND: The tissue-resident microbiome of oral epithelial lesions remains incompletely characterized, with most prior studies relying on saliva or surface sampling. This study aimed to characterize intralesional microbial communities across histopathologic categories of tongue lesions using formalin-fixed paraffin-embedded (FFPE) tissue. METHODS: This cross-sectional study characterized the intralesional microbiome of 63 formalin-fixed, paraffin-embedded tongue tissues, including fibroma (n = 15), low malignant potential dysplasia (LMP; n = 24), high malignant potential dysplasia (HMP; n

Longitudinal Transcriptomic Remodeling of Adipose Tissue After Bariatric Surgery Revealed by Differential Expression and Explainable Machine Learning.

Background: Bariatric surgery improves metabolic health, but long-term transcriptomic remodeling of white adipose tissue (WAT) after Roux-en-Y gastric bypass (RYGB) remains incompletely defined. This study aimed to characterize longitudinal WAT gene-expression patterns after RYGB and prioritize candidate signatures of post-surgical adaptation using a publicly available dataset. Methods: We analyzed subcutaneous WAT transcriptomic data from women with obesity who underwent RYGB, with samples collected before surgery and at 2 and 5 years after surgery. Differential expression analysis was integr

A systematic framework for assessing conservation effectiveness of migratory marine species: Integrating biomass, habitat quality, and functional traits.

The global expansion of Marine Protected Areas (MPAs) under the "30×30" target necessitates rigorous effectiveness assessments. Traditional evaluations for migratory marine species often rely on static biomass metrics, which fail to capture spatiotemporal mismatches between dynamic migration patterns and static reserve boundaries, deceptively masking structural population crises. Addressing these gaps, we developed a systematic framework integrating biomass, habitat quality, and functional traits. We validated this using the small yellow croaker (Larimichthys polyactis) in the Yellow and East

Unravelling the obesity epidemic among young people in Qatar: a literature review.

BACKGROUND: The prevalence of childhood and adolescent obesity has risen dramatically over recent decades, particularly in rapidly developing countries undergoing significant dietary transition. Qatar now has one of the highest rates of childhood overweight and obesity globally, coinciding with increased availability and consumption of processed and ultra-processed foods (UPFs). Although associations between UPF consumption and obesity are well established, the biological and behavioural mechanisms underpinning this relationship have not previously been synthesised within the context of Qatar.

Marine Bioactive Peptides for Colorectal Cancer Therapy: Mechanisms, Therapeutic Potential, and Translational Challenges.

Colorectal cancer (CRC) is one of the most prevalent gastrointestinal malignancies worldwide and remains a major cause of cancer-related mortality. Although current treatment strategies, including surgery, chemotherapy, radiotherapy, and targeted therapies, have improved patient outcomes, their effectiveness is frequently limited by multidrug resistance, severe adverse effects, tumour recurrence, and restricted patient applicability. Consequently, there is an urgent need to develop novel therapeutic agents with improved efficacy and reduced toxicity. Marine bioactive peptides have emerged as p
