Electrodeposited Janus collagen membrane with tunable barrier protection, controlled degradation, and synergistic immunomodulation for bone regeneration in complex defects.

Bone regeneration in complex pathological environments (e.g., infection or diabetes) is particularly challenging. Traditional collagen barrier membranes often degrade quickly and lack multifunctional bioactivity, limiting their effectiveness under such conditions. Here, we describe a collagen-based Janus membrane for advanced guided bone regeneration in these challenging settings. The membrane features an asymmetric bilayer architecture: the soft-tissue side comprises a dense barrier layer formed by electrodeposition, whereas the bone-defect side consists of a porous repair layer. The repair l…

A thiol-ene click chemistry-mediated space-maintaining Janus collagen membrane for GBR with antibacterial, immunomodulatory, and proangiogenic functions.

Resorbable collagen membranes are widely used for guided bone regeneration (GBR) but suffer from premature degradation, inadequate space maintenance, and poor infection and osteoimmune regulation in a bacteria-rich oral environment. Herein, we developed a hydrophobic, antibacterial, anti-inflammatory Janus membrane (HAA-JM) on a clinical collagen platform, directionally engineering soft-tissue and bone-facing sides. Via silane coupling and thiol-ene click chemistry, the outer surface was grafted with perfluoroalkyl groups, enhancing wet mechanical strength and anti-collapse performance and slo…

Repetitive shear loading in fetal bovine cartilage causes depth-dependent alterations in shear mechanical properties and collagen fiber structure.

During joint development, the collagen fiber network undergoes realignment and reorientation, forming the mature structure that is essential for cartilage mechanics. However, recreating this aligned collagen architecture and the shear mechanical properties in engineered cartilage remains challenging. Insights from immature cartilage development suggest that dynamic mechanical loading is necessary for normal joint formation, yet the role of shear loading, a common joint load, on collagen reorganization and changes in mechanical properties is unclear. Additionally, while proteoglycans are known …

Multimodal evaluation of periosteal versus collagen membrane coverage in atelocollagen-associated autologous chondrocyte implantation for large knee cartilage defects.

PURPOSE: Atelocollagen-associated autologous chondrocyte implantation (A-ACI) with periosteal coverage shows favourable outcomes for large knee cartilage defects but is associated with complications such as graft hypertrophy. Collagen membrane coverage remains under-investigated in this technique. This study aimed to evaluate periosteal versus collagen membrane coverage using clinical, radiological, arthroscopic and histological assessments. METHODS: Between 2014 and 2022, patients with ≥4.0 cm2 full-thickness chondral lesions underwent A-ACI using periosteal (n = 13) or collagen membrane (n …

Quantification of intervertebral disc degeneration using dual-energy computed tomography collagen maps: a comparative study with MRI Pfirrmann grading.

To determine the correlation between quantitative HU values derived from DECT collagen maps and the MRI-based Pfirrmann grade. We retrospectively reviewed IVDs in patients who underwent both spinal DECT and MRI within one year. CT scans were acquired using single-source DECT at 80 and 135-kVp with automatic exposure control. Three readers scored IVDs according to Pfirrmann et al. on T2-weighted MRI images. Quantitative evaluation involved placing three standardized regions of interest in each IVD in DECT-based cMaps; in corresponding 135-kVp DECT scans and MRI images. Mean density measurements…

Successful treatment for collagen vascular disease-related respiratory failure with inhaled nitric oxide as a bridging therapy:A case report.

BACKGROUND: Inhaled nitric oxide (iNO) is approved in Japan for perioperative pulmonary hypertension and persistent pulmonary hypertension in newborns;however, its role in non-cardiac respiratory failure is unclear. CASE PRESENTATION: A 47-year-old woman with Sjögren's syndrome developed severe acute type I respiratory failure due to collagen vascular disease. Despite high-FIO2 ventilation, her PaO2/FIO2 ratio remained <80, and extracorporeal membrane oxygenation (ECMO) was considered. Administration of iNO at 10 ppm improved oxygenation, with a rapid increase in PaO2/FIO2 ratio. This respons…

Circulating and matrix-associated type XI collagen epitopes as complementary biomarkers to support stromal-targeting concepts in solid tumors.

Desmoplastic tumors show extensive extracellular matrix (ECM) remodeling driven by cancer-associated fibroblasts (CAFs), contributing to tumor progression and treatment resistance. Type XI collagen is elevated in tumor stroma and linked to poor prognosis. This study examined two complementary type XI collagen epitopes: the circulating biomarker PRO-C11 and the matrix-associated epitope CT11a1, including its potential as a stromal drug-delivery target. PRO-C11 was measured in serum from 78 pancreatic ductal adenocarcinoma patients and correlated with clinical benefit rate and overall survival (…

Collagen-disrupting attIL12 TIL therapy boosts deep T cell infiltration via dual signaling activation and CCKAR reduction in sarcomas.

Tumor-targeted T cell therapies of various types have been booming, but T cell therapy is limited by its inability to penetrate the collagen barrier surrounding tumors. The destruction of tumor collagen is significant because collagen both suppresses T cells and contributes to the formation of the extracellular matrix. Our previously reported cell-surface vimentin (CSV)-targeted and membrane-anchored interleukin 12-armed (attIL12) T cells can reduce collagen production by killing cancer-associated fibroblasts, but fail to reduce collagen expression by tumor cells, resulting in resistance to at…
