Immunization of mice with recombinant IgA1 protease of Neisseria meningitidis or several structural derivatives thereof protects the animals infected with a variety of deadly pathogens, including N. meningitidis serogroups A, B, and C and 3 serotypes of Streptococcus pneumonia. In sera of rabbits immunized with inactivated pneumococcal cultures, antibodies binding IgA1-protease from N. meningitidis serogroup B were detected. Thus, the cross-reactive protection against meningococcal and pneumococcal infections has been demonstrated in vivo. Presumably it indicates the presence of common epitope

Despite heroic efforts to prevent the emergence of an influenza pandemic, avian influenza A virus has prevailed by crossing the species barriers to infect humans worldwide, occasionally with morbidity and mortality at unprecedented levels, and the virus later usually continues circulation in humans as a seasonal influenza virus, resulting in health-social-economic problems each year. Here, we review current knowledge of influenza viruses, their life cycle, interspecies transmission, and past pandemics and discuss the molecular basis of pandemic acquisition, notably of hemagglutinin (lectin) ac

A house gecko, Hemidactylus turcicus, from an Israeli locality was found infected by Cryptosporidium sp. Infection was recovered in the anterior gut, upon examination by a transmission electron microscope. It comprised from 2.9 x 3.3 microns undivided and 2.6-3.3 x 3.8-3.9 microns dividing meronts, 1.9-2.3 x 2.0-2.6 microns young macrogamonts, and a 3.3 x 3.9 microns sporulated oocyst containing in cross section 4 sporozoites and a residuum. Oocyst dimensions were smaller and apparently not conspecific with the Cryptosporidium sp. reported from the introduced H. turcicus in Texas.

Antibacterial sera activities towards Citrobacter O22 and Salmonella typhimurium were tested with O22, O1, O4, O5, O12 erythrocytic diagnostic agents. Cross activities of O22, O1, O4, O5, O12, and Hi antigens were tested with the same and Hi erythrocytic diagnostic agents in the antibody neutralization test. The findings have confirmed the identity or very close relationship between the tested O antigens. Screening for antigens of the excretions from patients with S. typhimurium and Citrobacter O22 infections has shown that indication of Hi antigen may be considered as the differentiating crit

Porcine deltacoronavirus (PDCoV) is an emerging enteric pathogen that causes severe diarrhea, vomiting, and dehydration in piglets, and a recent human infection report raises concerns about its zoonotic potential. The S1 subunit of the PDCoV-spike protein is the primary target of neutralizing antibodies and is critical for viral attachment and entry. In this study, we generated and characterized eight monoclonal antibodies (mAbs) against S1, two targeting the S1A domain, five targeting the S1B domain, and one binding the S1C domain. Using a genetically engineered luciferase reporter virus, we

BACKGROUND: Infection with HPV 16 and 18, the major causative agents of cervical cancer, can be prevented through vaccination with a bivalent or quadrivalent vaccine. Both vaccines provide cross-protection against HPV-types not included in the vaccines. In particular, the bivalent vaccine provides additional protection against HPV 31, 33, and 45 and the quadrivalent vaccine against HPV31. The quadrivalent vaccine additionally protects against low-risk HPV type 6 and 11, responsible for most cases of genital warts. In this study, we made an analytical comparison of the two vaccines in terms of

Influenza inflicts significant global mortality and morbidity that can be combated by effective immunization. However, the protective efficacy of current vaccines is limited by both the significant antigenic diversity of the viral hemagglutinin protein and the capacity for rapid antigenic change. This necessitates global influenza surveillance efforts, frequent vaccine reformulation and annual readministration. There is, therefore, tremendous interest in the development of novel strategies to elicit broad and durable protection against both seasonal and pandemic infection. This review presents

The objective of this study was to systematically investigate the carriage pattern and load of newly identified methicillin-resistant Staphylococcus aureus (MRSA) colonized or infected patients before any decolonization took place. Cultures of wounds (38%), of sputum (16%) or throat (10%) and of urine (10%) most frequently gave the initial positive MRSA result. Samples from nose, forehead, neck, axilla, and groin were obtained to determine the extent of nasal and extranasal colonization. Fifty-six (69%) of the screened patients proved to be MRSA positive at one or more of these sample sites, a

Zika virus (ZIKV) is a flavivirus that is highly neurotropic causing congenital abnormalities and neurological damage to the central nervous systems (CNS). In this study, we used a human induced pluripotent stem cell (iPSC)-derived blood brain barrier (BBB) model to demonstrate that ZIKV can infect brain endothelial cells (i-BECs) without compromising the BBB barrier integrity or permeability. Although no disruption to the BBB was observed post-infection, ZIKV particles were released on the abluminal side of the BBB model and infected underlying iPSC-derived neural progenitor cells (i-NPs). Intracellular pathogens have numerous strategies for effective dissemination within the host. Many intracellular pathogens first infect leukocytes, which they use as a vehicle to transport them to target organs. Once at the target organ, intracellular parasite Toxoplasma gondii can cross the capillary wall in extracellular form by infecting endothelial cells. However, after egression from leukocytes, extracellular parasites face the risk of host immune attack. In this study, observation of infected mouse organs, using a method that renders tissue transparent, revealed that adhesion of tachyzoi

The wide dissemination of carbapenemase-producing Gram-negatives (CPGNs), including enterobacterial species and non-fermenters, has caused a public health crisis of global dimensions. These organisms cause serious infections in hospitalized patients, and are associated with increased mortality. Cross-transmission is common, and outbreaks may occur in healthcare facilities where the infection control practices are inadequate. CPGNs exhibit extensive drug-resistant phenotypes, complicate therapy, and limit treatment options. Systematic data on therapy are limited. However, regimens combining two

Community-acquired methicillin-resistant Staphylococcus aureus (CA-MRSA) continues to make headlines because of large outbreaks in daycare centers and among members of athletic teams. CA-MRSA infections in children commonly lead to hospitalization. Life-threatening infections, such as necrotizing pneumonitis and brain abscess, can occur. The organism has crossed into hospitals and is now a common cause of hospital-acquired sepsis. Multidrug-resistant strains of MRSA are emerging in Asia, with the resistance based on either a novel gene cassette or a transmissible plasmid. The routine use of an

Where prevalence of geohelminths and schistosomes is high, co-infections with multiple parasite species are common. Previous studies have shown that the presence of geohelminths either promotes or is a marker for greater prevalence and intensity of Schistosoma mansoni infections. Some of this apparent synergy may simply represent shared conditions for exposure, such as poor sanitation, and may not suggest a direct biologic interaction. We explored this question in a study of 13,279 school children in Jequié, Bahia, Brazil, with a survey of demographic characteristics and stool examinations. Cr

In conclusion, there are a number of neurological manifestations of HIV infection, affecting both the central and peripheral nervous systems. Involvement of the CNS may occur very early in the course of infection and manifest itself as an acute aseptic meningitis. HIV encephalopathy is currently the most commonly diagnosed neurologic disorder associated with HIV and may in fact occur as a direct result of HIV infection in the brain. In years to come, HIV encephalopathy may assume epidemic proportions. Thus, nurses and other health care workers will have to be well versed in the major symptoms

BACKGROUND: Tsetse flies (Glossina sp.) are vectors of Trypanosoma brucei subspecies that cause human African trypanosomiasis (HAT). Capturing and screening tsetse is critical for HAT surveillance. Classically, tsetse have been microscopically analysed to identify trypanosomes, but this is increasingly replaced with molecular xenomonitoring. Nonetheless, sensitive T. brucei-detection assays, such as TBR-PCR, are vulnerable to DNA cross-contamination. This may occur at capture, when often multiple live tsetse are retained temporarily in the cage of a trap. This study set out to determine whethe

In grassland agroecosystems, some plant pathogenic bacteria can cause disease in animals. These strains are known as plant and animal cross-kingdom pathogenic bacteria. In this study, we established an alfalfa root infection model and a mouse model via the gavage administration of the Pantoea alfalfae CQ10 (CQ10) bacterial suspension. It was confirmed that the CQ10 strain caused bacterial leaf blight of alfalfa. Mice inoculated with 0.4 mL of 109 cfu/mL bacterial suspension developed clinical symptoms 48 h later, such as diminished vitality, tendencies to huddle, and lack of appetite, includin

Sarcoidosis is a granulomatous disease of unknown etiology, immunologically characterized by a Th1 immune response. Transcriptome-wide expression studies in various types of sarcoid tissues contributed to better understanding of disease mechanisms. We performed a systematic database search on Gene Expression Omnibus (GEO) and utilized transcriptomic data from blood and sarcoidosis-affected tissues in a meta-analysis to identify a cross-tissue, cross-platform signature. Datasets were further separated into training and testing sets for development of a diagnostic classifier for sarcoidosis. A t

Japanese encephalitis virus (JEV) is the major cause of viral encephalitis in South East Asia. It has been suggested that, as a consequence of the inflammatory process during JEV infection, there is disruption of the blood-brain barrier (BBB) tight junctions that in turn allows the virus access to the central nervous system (CNS). However, what happens at early times of JEV contact with the BBB is poorly understood. In the present work, we evaluated the ability of both a virulent and a vaccine strain of JEV (JEV RP9 and SA14-14-2, respectively) to cross an in vitro human BBB model. Using this
