Recent advances in understanding of male infertility have implicated two major causative factors, oxidative stress and Y chromosome deletions. A major cause of oxidative stress appears to be the high rate of reactive oxygen species generation associated with the retention of excess residual cytoplasm in the sperm midpiece. Other possible causes include the redox cycling of xenobiotics, and antioxidant depletion or apoptosis. Oxidative stress induces peroxidative damage in the sperm plasma membrane and DNA damage in both the mitochondrial and nuclear genomes. Nuclear DNA damage in the germ line

We have constructed a comparative map in mouse of the critical region of human 22q11 deleted in DiGeorge (DGS) and Velocardiofacial (VCFS) syndromes. The map includes 11 genes potentially haploinsufficient in these deletion syndromes. We have localized all the conserved genes to mouse Chromosome (Chr) 16, bands B1-B3. The determination of gene order shows the presence of two regions (distal and proximal), containing two groups of conserved genes. The gene order in the two regions is not completely conserved; only in the proximal group is the gene order identical to human. In the distal group t

A 1580-bp fragment of Staphylococcus aureus plasmid pUB110 encoding resistance to the DNA synthesis inhibitor bleomycin has been cloned and sequenced. A DNA sequence containing an open reading frame of 405 bp was subcloned into several expression vectors and bleomycin resistance was expressed at high level in Escherichia coli under the control of lambda PL promoter. On induction, a ca. 14,000-Da protein was detected by gel electrophoresis. The bleomycin resistance determinant of the gram-positive plasmid pUB110 was compared to that of the enterobacterial transposon Tn5; limited regions of clos

The cloned phoE genes from Enterobacter cloacae and Klebsiella pneumoniae are normally expressed and regulated in Escherichia coli K-12, and their products are correctly assembled into the outer membrane. Differences between the three PhoE proteins were found with binding of two out of ten monoclonal antibodies directed against the cell-surface-exposed part and in pore characteristics, but not in phage receptor function. The DNA sequences of the E. cloacae and K. pneumoniae phoE genes were determined and used to predict the primary structures of the encoded proteins. In the upstream non-coding

Acute respiratory infection (ARI) outbreaks in Beijing caused by human adenovirus B114 (HAdV-B114) have long been misclassified as HAdV-B3. Its origin, evolutionary trajectory and long-term transmission dynamics remain poorly understood. This study aimed to clarify its epidemiological characteristics, genomic variations, recombination features and evolutionary history. Among 51 HAdV outbreaks in Beijing from 2017 to 2024, 32 originally designated as HAdV-B3 were studied. Viral isolation was performed on 267 positive samples, conducted targeted and whole-genome sequencing, and carried out phylo

This study aimed to investigate the current epidemiology and genetic evolution of bovine viral diarrhea virus (BVDV) on cattle farms in Gansu Province, China, between 2021 and 2025. A total of 749 samples from 62 farms across 14 cities and prefectures in Gansu were tested. The overall BVDV positivity rate was 19.89%, determined by amplification of the 5'-UTR and Npro regions. Seven subtypes were identified: BVDV-1a, -1b, -1d, -1m, -1v, -1u, and -2a. BVDV-1u was the predominant subtype (59.06%), followed by BVDV-1v (13.42%). The greatest subtype diversity was observed in Jinchang City and Ganna

BACKGROUND: Given its high mortality and broad societal impact, the COVID-19 pandemic is arguably one of the most consequential public health crises of the twenty-first century. Although previous studies have identified several genes associated with COVID-19 susceptibility, relatively little is known about the genes contributing to severe COVID-19, including their evolutionary histories. In the current study, we analyzed IL-4, TLR2, CCL2, and SLC11A1-immunity genes that have previously been implicated in severe COVID-19 and other immune-related diseases-in globally diverse populations from the

BACKGROUND: Staphylococcus aureus is a major public health threat in Ghana, with the Sequence Type 152 (ST152) lineage emerging as a dominant, Panton-Valentine leukocidin toxin (PVL)-positive lineage. This study investigates the evolutionary dynamics and emergence of methicillin resistance within the ST152 population in Ghana.
METHODS: We analyzed 285 publicly available S. aureus whole-genome sequences (WGS) from Ghana, encompassing isolates of human and animal origin. Comprehensive genomic characterization was performed using the StaphScope pipeline, followed by detailed pangenome and phylog

Cancer arises from oncogenic clones, yet the dynamic mechanisms driving their stepwise evolution toward malignancy remain incompletely understood. Here, we establish the Atlas of Ras-driven Tumors in Drosophila (ART-D), a systematic, cross-species platform that dissects the molecular and phenotypic trajectories of tumorigenesis across ten genetically defined RasV12-driven models. By integrating longitudinal phenotypic profiling, we define three conserved stages of tumor development-initiation, promotion, and progression-distinguished by distinct shifts in tumor burden and tumor-induced cachexi

Viruses within the Bandavirus genus (Phenuiviridae family), phylogenetically divided into SFTSV and Bhanja groups, pose public health risks through cross-species transmission. Despite their significance, a systematic comparison of their infectivity, pathogenicity, and antigenicity is lacking. This study evaluates these key properties for five representative species: Severe fever with thrombocytopenia syndrome virus (SFTSV), Guertu virus (GTV), and Heartland virus (HRTV) from the SFTSV group; and Bhanja virus (BHAV) and Lone Star virus (LSV) from the Bhanja group. The five bandaviruses demonstr

Riemerella anatipestifer (R. anatipestifer), a well-known waterfowl pathogen, increasingly threatens Chinese poultry by spreading to chickens. The genetic differentiation and host adaptation following cross-host transmission remain unclear. Here, we characterized a highly virulent, multidrug-resistant chicken-source strain (SDAU-RA1) and performed comparative genomics with global R. anatipestifer strains to elucidate population structure and evolutionary dynamics. SNP phylogeny revealed significant geographic clustering and dominant clonal groups. Strains from different hosts showed a pattern

OBJECTIVES: Streptococcus suis is an emerging zoonotic porcine pathogen and a leading cause of adult bacterial meningitis in Southeast Asia, associated with raw pork consumption. Most zoonotic S. suis infections globally are caused by strains from lineage CC1 carrying a serotype 2 capsule. However, in Thailand, ∼40% of the reported zoonotic infections are caused by 2 endemic lineages, CC104 and CC233, which also have a serotype 2 capsule. In this study, we aimed to identify the drivers of the emergence and recent evolution of these two lineages.
METHODS: We sequenced the whole genomes of 141

We inoculated clinical materials into deer mice to attempt isolation of Sin Nombre virus. We did not observe productive infection in the natural rodent reservoir. Genomic comparisons between rodent reservoirs and human disease may provide insight into hantavirus evolution and genetic determinants, but reverse zoonosis of Sin Nombre virus appears unlikely.

Global surveillance of Salmonella enterica reveals dynamic evolutionary forces shaping pathogenicity and antimicrobial resistance (AMR), yet the integration of serotyping, multilocus sequence typing (MLST), and phenotypic landscapes remains unexplored. Here, we dissect 935 Salmonella isolates, collected from both clinical and food chain sources, through integrated genomics and phenomics to resolve population structure, spatiotemporal dynamics, and evolutionary drivers. Salmonella Typhimurium (18.7%) and Salmonella Enteritidis (17.1%) dominate the serotype landscape, while MLST uncovers ST34 (2

The loss of the tail is among the most notable anatomical changes to have occurred along the evolutionary lineage leading to humans and to the 'anthropomorphous apes'1-3, with a proposed role in contributing to human bipedalism4-6. Yet, the genetic mechanism that facilitated tail-loss evolution in hominoids remains unknown. Here we present evidence that an individual insertion of an Alu element in the genome of the hominoid ancestor may have contributed to tail-loss evolution. We demonstrate that this Alu element-inserted into an intron of the TBXT gene7-9-pairs with a neighbouring ancestral A

Diarrhea is one of the major public health issues worldwide. Although the infections of individual enteric virus have been extensively studied, elucidation of the coinfection involving multiple viruses is still limited. In this study, we identified the coinfection of human adenovirus (HAdV) and human astrovirus (HAstV) in a child with acute gastroenteritis, analyzed their genotypes and molecular evolution characteristics. The sample was collected and identified using RT-PCR and subjected to whole-genome sequencing on the NovaSeq (Illumina) platform. Obtained sequences were assembled into the c

Patients with Primary immunodeficiency (PIDs) may be infected by Polioviruses (PVs), especially when vaccinated with live Oral Polio Vaccine before diagnosis. They may establish long-term shedding of divergent strains and may act as reservoirs of PV transmission. This study delved into the effect of the genetic evolution of complete PV genomes, from MHC class II-deficient patients, on the excretion duration and clinical outcomes. Stool samples from three PID patients underwent analysis for PV detection through inoculation on cell culture and real-time PCR, followed by VP1 partial sequencing an
