BACKGROUND: Increased production and accumulation of melanin leads to various hyperpigmentation disorders. Melanin synthesis is regulated by melanogenic proteins such as tyrosinase, tyrosinase-related protein (TRP)-1 and -2, and their transcription factors.
OBJECTIVE: In this study, we assessed the effects of PQQ on melanogenic protein expression of murine B16 melanoma cells.
METHODS: We assessed melanin production of PQQ-treated B16 melanoma cells. Furthermore, we investigated the effect of PQQ on the activity of melanogenic enzymes and their expression using Western blot and semi-quantitat

Cdc7 is a serine/threonine kinase that plays essential roles in the initiation of eukaryotic DNA replication and checkpoint response. In previous studies, depletion of Cdc7 by small interfering RNA was shown to induce an abortive S phase that led to the cell cycle arrest in normal human fibroblasts and apoptotic cell death in various cancer cells. Here we report that stress-activated p38 MAP kinase was activated and responsible for apoptotic cell death in Cdc7-depleted HeLa cells. The activation of p38 MAP kinase in the Cdc7-depleted cells was shown to depend on ATR, a major sensor kinase for

Estrogen signaling is required for the proliferation of normal breast epithelial cells. However, prophylactic inhibition of estrogen signaling fails to prevent 56% of human breast cancer cases. The underlying mechanism is not well understood. Aberrant activation of growth factor signaling is known to provide alternative proliferation pathways in breast cells that are fully transformed, but it is not known whether activation of growth factor signaling can substitute for estrogen signaling in causing aberrant proliferation in the normal breast epithelium. Here, we report that in a retrovirus-bas

The frequency and mutation spectra of proto-oncogene activation in hepatocellular neoplasms induced by tetrachloroethylene, trichloroethylene and dichloroacetic acid were examined to help define the molecular basis for their carcinogenicity. H-ras codon 61 activation was not significantly different among dichloroacetic acid- and trichloroethylene-induced and combined historical and concurrent control hepatocellular tumors (62%, 51% and 69% respectively). The mutation spectra of H-ras codon 61 mutations showed a significant decrease in AAA and increase in CTA mutations for dichloroacetic acid-

Cadmium (Cd) and its compounds are well-known human carcinogens, but the mechanisms underlying the carcinogenesis are not entirely understood yet. Aberrant methylation was investigated in order to obtain insight into the DNA repair-related epigenetic mechanisms underlying CdCl(2)-induced malignant transformation of human bronchial epithelial cells (16HBE). Gene expression and DNA methylation were assessed in untreated control cells; 5th, 15th, and 35th passage of CdCl2-treated cells and tumorigenic cells (TCs) from nude mice by using high-performance liquid chromatography, real-time PCR, Weste

N-myc downstream regulated gene 1 (NDRG1) is an important gene regulating tumor invasion. In this study, shRNA technology was used to suppress NDRG1 expression in CaSki (a cervical cancer cell line) and HO-8910PM (an ovarian cancer cell line). In vitro assays showed that NDRG1 knockdown enhanced tumor cell adhesion, migration and invasion activities without affecting cell proliferation. cDNA microarray analysis revealed 96 deregulated genes with more than 2-fold changes in both cell lines after NDRG1 knockdown. Ten common upregulated genes (LPXN, DDR2, COL6A1, IL6, IL8, FYN, PTP4A3, PAPPA, ETV

MicroRNA (miRNA) are a class of highly conserved, small noncoding RNA that emerge as key posttranscriptional regulators in various neoplastic transformations. Our previous study profiling the miRNA transcriptome in Marek's disease virus (MDV)-induced lymphoma revealed many novel and differentially expressed miRNA, including gga-miR-26a, which was downregulated in MDV-infected spleens of chickens. In this study, differential expression of gga-miR-26a between MDV-infected and noninfected spleens at 4, 7, 14, 21, and 28 d postinfection was analyzed by real-time PCR. The results showed gga-miR-26a

Cuproptosis is a recently identified copper-dependent cell death pathway with growing relevance in tumor biology, yet its involvement in thyroid carcinoma (TC) remains poorly understood. In this study, we integrated multi-omics datasets to characterize the functional roles of cuproptosis-related genes (CRGs) in TC progression. Bulk and single-cell transcriptomic datasets from public repositories were analyzed to classify TC into two CRG-based molecular subtypes that showed significant associations with clinicopathological features and immune cell infiltration. A prognostic model derived from C

Previous studies have shown that DARS is highly expressed in patients with myeloproliferative neoplasms (MPN), and these patients have higher disease burdens. However, the mechanism by which DARS promotes the proliferation of MPN cells remains unclear. Here, we explored the tumor-intrinsic role of DARS in human MPN cell models and the associated molecular mechanisms using an integrated multi-omics approach. DARS depletion suppressed the proliferation of MPN cells in vitro and xenograft tumor growth in vivo, induced cell-cycle arrest, and promoted apoptosis. Metabolomic analysis identified 190

BACKGROUND: Malignant melanoma is characterized by marked intratumoral heterogeneity and an immunosuppressive tumor microenvironment (TME), and resistance to immunotherapy remains common. We hypothesized that melanoma contains a poorly differentiated tumor subpopulation characterized by an ETV5-centered transcriptional program and TGF-β-associated intercellular crosstalk, which may contribute to malignant progression and immune evasion. EV-related signaling was explored as a potential, but unvalidated, component of this phenotype.
METHODS: We performed integrative single-cell transcriptomic a

Insulin-like growth factor binding protein 1 (IGFBP1) is documented as a pro- tumorigenic gene in various cancers; nevertheless, its specific function in esophageal carcinoma (ESCA) is unclear. This study used data obtained from The Cancer Genome Atlas (TCGA) to assess IGFBP1 expression, investigating its clinical outcome-related relevance, and explore its correlation in the context of ESCA tumor microenvironment. The Wilcoxon rank-sum test indicated a notable overexpression of IGFBP1 in ESCA compared to normal tissues, which was further validated via immunohistochemistry. Functional enrichmen

Breast cancer is a common malignant tumor with a complex pathogenesis, and while genome-wide association studies (GWAS) have identified multiple risk loci, they often fall short in pinpointing specific functional susceptibility genes. To address this, we collected whole-tissue eQTL data from the GTEx portal and breast cancer GWAS summary data from the Breast Cancer Association Consortium (BCAC) and FinnGen R10 database, employing transcriptome-wide association studies (TWAS) to screen and identify susceptibility genes. Subsequently, spatial transcriptomic sequencing and single-cell RNA sequenc

BACKGROUND: Liquid-liquid phase separation (LLPS) has emerged as a critical mechanism underlying oncogenic signaling and transcriptional regulation; however, its involvement in triple-negative breast cancer (TNBC) remains largely unknown. The pleckstrin homology-like domain family B member 2 (PHLDB2) has been implicated in cytoskeletal organization and cell migration, but its role in phase separation-mediated tumor progression has not been explored.
METHODS: We conducted an integrative multi-omics analysis combining transcriptomic profiling, bioinformatic modeling, and molecular biology exper

INTRODUCTION AND OBJECTIVES: Outcomes for patients with hepatocellular carcinoma (HCC) are generally poor, partly due to significant multidrug resistance. HCC heterogeneity decreases the accuracy of traditional prediction models. This study aimed to evaluate the prognostic significance of the HCC resistome.
MATERIALS AND METHODS: Clinical and transcriptomic data from 371 HCC cases available at TCGA were analyzed. An external dataset (604 patients from 4 cohorts) and 40 HCC samples, in which gene expression was determined by RT-qPCR, were used to validate the prognostic model.
RESULTS: The in

Malignant pleural mesothelioma (MPM) is a rare but aggressive thoracic malignancy primarily linked to asbestos exposure. Despite advances in research, the prognosis of MPM remains poor, and there is a lack of efficient and precise prognostic assessment tools. Its insidious onset, limited treatment options, and high resistance to therapy contribute to poor clinical outcomes and underscore the urgent need for reliable prognostic biomarkers. To address this gap, we integrated multi-omics data to identify a prognostic gene set for MPM and subsequently developed a prognostic model aiming to improve

Melanoma is the most aggressive and lethal form of skin cancer, posing significant challenges for prognosis assessment and treatment. Recently, metabolic reprogramming and epigenetic regulation have gained attention for their roles in cancer progression. The role of the key metabolic enzyme dihydrolipoic acid succinyltransferase (DLST) in cancer is currently unclear. Here we investigate the relationship between DLST expression and prognosis of melanoma patients, further employing bioinformatic tools including single-cell and spatial single-cell analyses to explore its expression and function.

INTRODUCTION: The nuanced roles of neuropilin (NRP) isoforms, NRP1 and NRP2, have attracted considerable scientific interest regarding cancer progression. Their differential expressions across various cancer types are specific to NRP isoforms which are shown in a cancer type-dependent manner. It accounts for the different mechanisms involved, driven by a co-expression of gene-sets associated with overexpressed NRP1 or NRP2. Their different expressions on tumour-associated macrophages (TAMs) with disparate markers are associated with the heterogenous tumour microenvironment (TME) through their

Glioblastoma (GBM) is a highly heterogeneous disease with poor clinical outcomes. To comprehensively dissect the molecular landscape of GBM and heterogeneous macrophage clusters in the progression of GBM, this study integrates single-cell and bulk transcriptome data to recognize a distinct pro-tumor macrophage cluster significantly associated with the prognosis of GBM and develop a GBM prognostic signature to facilitate prior subtypes. Leveraging glioma single-cell sequencing data, we identified a novel pro-tumor macrophage subgroup, marked by S100A9, which might interact with endothelial cell
