Integrated Single-Cell RNA Sequencing and Proteome-Wide Mendelian Randomization Identifies Therapeutic Targets for Psoriasis and Associated Complications.

BACKGROUND: To identify new targets for protein-based treatments in psoriasis and evaluate the possible negative impacts of these druggable proteins. METHODS: We carried out an extensive analysis of the entire set of proteins in the blood (proteome-wide) to determine if there are causal links between certain blood proteins and the likelihood of developing psoriasis. The proteins were selected from the UK Biobank Pharma Proteomics Project (UKBPPP) database, which includes genetic data for 2,940 different blood proteins. We obtained the cis-expression quantitative trait locus (cis-eQTL) of drug

Pathogenic FANCC Variants Are Associated with Accessory Breasts in a Sub-Saharan African Multiplex Family.

Accessory breasts denote the formation of extra breast tissue along the milk line, and are known to be more prevalent among Black and Asian populations, affecting both genders. This first-ever study aimed to determine the genetic aetiology of accessory breasts in a multiplex family, where all female siblings present with bilateral accessory breasts. The study also ascertained secondary findings (SFs) responsible for comorbidities. Clinical data and saliva samples were obtained from all family members. Ultrasound and histopathology confirmed the diagnosis. Whole-exome sequencing was conducted o

Sex chromosome identification and genome curation from a single individual with SCINKD.

In most animal species, the sex determining pathway is typically initiated by the presence/absence of a primary genetic cue at a critical point during development. This primary genetic cue is often located on a single locus-referred to as sex chromosomes-and can be limited to females (in a ZZ/ZW system) or males (in an XX/XY system). One trademark of sex chromosomes is a restriction or cessation of recombination surrounding the sex-limited region (to prevent its inheritance in the homogametic sex). This may lead to-through a variety of mechanisms-higher amounts of genetic divergence within thi

Fundamental origins of neural tube defects with a basis in genetics and nutrition.

Neural tube defects (NTDs) are leading congenital malformations. Its global prevalence is one in 1000 pregnancies and it has high morbidity and mortality. It has multiple risk factors like genetic errors and environmental stressors like maternal malnutrition and in utero exposure to pollutants like chemicals. The genetic program determines neural tube development based on timely expression of many genes involved in developmental signaling pathways like BMP, PCP and SHH. BMP expression defines ectoderm. SOX represses BMP in ectoderm and convertes to the neuroectoderm. Subsequently, PCP molecule

Amplified fragment length polymorphism genotyping of Trichophyton indotineae indicates possible zoonotic transmission.

The anthropophilic dermatophyte Trichophyton interdigitale and its counterpart T. mentagrophytes are phylogenetically closely related species. In India, the most common endemic dermatophyte species belongs to the T. indotineae. The internal transcribed spacer genotype VIII within this species complex was recently renamed as T. indotineae based on its rapid emergence in India and its elevated virulence and terbinafine resistance. While humans are considered a source of T. interdigitale infection, animals are considered a source of T. mentagrophytes. For T. indotineae it is not known whether inf

Dengue Virus and the Host Immune System: A Battle of Immune Modulation, Response and Evasion.

Dengue virus (DENV) is a major global health concern, with pathogenesis driven by complex interactions between the virus, host genetics, and immune responses. Key determinants of disease severity include antibody-dependent enhancement (ADE), cross-reactive T cells, anti-NS1 antibodies, autoimmunity, and genetic predisposition, with the NS1 protein and its antibodies strongly implicated in severe dengue. This review highlights recent advances in our understanding of how DENV impacts host immune responses at cellular, molecular, and genetic levels. We particularly focus on how the virus interact

Mutational analysis differentiating sporadic carcinomas from colitis-associated colorectal carcinomas.

BACKGROUND: Ulcerative colitis (UC) is a chronic inflammatory bowel disease (IBD) that is associated with increased risk of developing colitis-associated carcinoma (CAC). The genetic profile of CACs is fairly similar to the sporadic colorectal carcinomas (sCRCs), although showing certain differences in the timing and sequence of alterations that contribute to carcinogenesis. Also, both cancer types typically show a strong histological resemblance, which complicates the pathologists' diagnosis. Due to the different clinical consequences, it is of utmost importance to categorize the correspondin

A Titin Truncating Variant Causing a Dominant Myopathy With Cardiac Involvement in a Large Family: The Exception That Proves the Rule.

BACKGROUND: Titin truncating variants (TTNtvs) have been repeatedly reported as causative of recessive but not dominant skeletal muscle disorders. OBJECTIVE: To determine whether a single heterozygous nonsense variant in TTN can be responsible for the observed dominant myopathy in a large family. METHODS: In this case series, all available family members (8 affected and 6 healthy) belonging to a single family showing autosomal dominant inheritance were thoroughly examined clinically and genetically. RESULTS: All affected family members showed a similar clinical phenotype with a combination

Comparison Between Electroporation at Different Voltage Levels and Microinjection to Generate Porcine Embryos with Multiple Xenoantigen Knock-Outs.

In the context of xenotransplantation, the production of genetically modified pigs is essential. For several years, knock-out pigs were generated through somatic cell nuclear transfer employing donor cells with the desired genetic modifications, which resulted in a lengthy and cumbersome procedure. The CRISPR/Cas9 system enables direct targeting of specific genes in zygotes directly through microinjection or electroporation. However, these techniques require improvement to minimize mosaicism and low mutation rates without compromising embryo survival. This study aimed to determine the gene edi

Rett syndrome in Ireland: a demographic study.

BACKGROUND: Rett syndrome (RTT) is a rare neurodevelopmental condition associated with mutations in the gene coding for the methyl-CpG-binding protein 2 (MECP2). It is primarily observed in girls and affects individuals globally. The understanding of the neurobiology of RTT and patient management has been improved by studies that describe the demographic and clinical presentation of individuals with RTT. However, in Ireland, there is a scarcity of data regarding individuals with RTT, which impedes the ability to fully characterize the Irish RTT population. Together with the Rett Syndrome Assoc
