Functional characterization of two cholecystokinin-B/gastrin receptor isoforms: a preferential splice donor site in the human receptor gene.

The cholecystokinin-B and gastrin receptor is encoded by a single gene composed of five exons and spanning over 10 kilobases on human chromosome 11p 15.5-->15.4. Exon 4 has two possible alternative splicing donor sites that seem to be conserved in other species such as the canine, rat, Mastomys, and mouse. They could generate two receptor isoforms (short- and long-form), which differ in their putative third cytoplasmic domain of the serpentine G-protein-coupled receptors. In the present study, we examined whether an alternative splicing is operated in a tissue-specific manner and whether two r

Epidermolysis bullosa dystrophica recessive fibroblasts produce increased concentrations of cAMP within a collagen matrix.

Human dermal fibroblasts grown in tissue culture can be suspended and cultured in collagen lattices. These fibroblast-populated collagen lattices (FPCL) undergo a reduction in size by the process of lattice contraction. Fibroblasts from patients with epidermolysis bullosa dystrophica recessive, EBdr, produce excessive quantities of cAMP. These high concentrations of cAMP may be related to the inability of the EBdr fibroblast to elongate and spread out when incorporated into the collagen matrix. Fibroblasts with these morphologic characteristics are not effective in contracting collagen lattice

A human trophoblastic isozyme (lactate dehydrogenase-Z) associated with choriocarcinoma.

A unique electrophoretic form of lactate dehydrogenase (LDH-Z), formerly observed in a choriocarcinoma cell line (JEG-3) and first-trimester placenta, has been shown to be the same as that produced in hydatidiform mole and term placenta. We have also observed LDH-Z in second-trimester placenta, choriocarcinoma metastasized to the liver, and five of five additional independently derived (from different patients) choriocarcinoma cell lines. The only exception to the production of LDH-Z in a choriocarcinoma was in the cell line BEWO, which was established from the same tumor as JEG-3. Since BEWO

ON THE ORIGIN OF THE HUMAN TREPONEMATOSES (PINTA, YAWS, ENDEMIC SYPHILIS AND VENEREAL SYPHILIS).

A close relationship between the four human treponematoses is suggested by their clinical and epidemiological characteristics and by such limited knowledge of the treponemes as there is at present. No treponeme of this group (except for that of the rabbit) is known other than in man, but the human treponemes probably arose long ago from an animal infection. The long period of infectiousness of pinta suggests that it may have been the earliest human treponematosis. It may have been spread throughout the world by about 15 000 B.C., being subsequently isolated in the Americas when the Bering Stra

Updates in cervical cancer screening: A practical guide for the internist.

Cervical cancer remains a largely preventable malignancy, yet screening gaps persist, particularly in underserved and special populations. Recent advances have expanded screening strategies beyond traditional clinician-collected cytology to include high-risk human papillomavirus (HPV)-based testing and options for cervical and vaginal self-collection. This review summarizes current US screening guidelines, performance characteristics of primary HPV testing and co-testing, and evolving evidence supporting self-collection as a tool to improve access and adherence. Practical considerations for th

The Relationship Between Gut Microbiota and Recurrent Spontaneous Abortion.

Recently, the gut microbiota has been found to be associated with multiple organs and systems in the human body, playing a key role in the occurrence and development of various diseases, such as the gut-brain axis and the gut-liver axis. However, its interaction with miscarriages remains poorly understood. This article reviews the characteristics of gut microbiota and its metabolites in patients with recurrent spontaneous abortion (RSA), the mechanism of gut microbiota inducing RSA, and potential therapeutic strategies. Therefore, it provides a new perspective on the gut microbiota in the path

Diagnosing Rickettsia felis infection with Metagenomic Next-Generation Sequencing (mNGS) in a patient with ankylosing spondylitis: a case report and literature review.

Rickettsia felis, an emerging flea-borne pathogen with global distribution potential, is a neglected cause of undifferentiated febrile illness, although reported human cases remain sparse. The development of molecular diagnostic methods, along with the application of metagenomic next-generation sequencing (mNGS), has improved the diagnostic accuracy of infectious fevers. A case of Rickettsia felis infection was diagnosed by mNGS in a 55-year-old patient with pre-existing ankylosing spondylitis. Five previously reported cases of Rickettsia felis infection were systematically reviewed, with a co

Virus Isolation from Field-Collected Ticks: In Vivo.

As tick-borne viruses such as Powassan virus (POWV) and Deer tick virus (DTV) increase in prevalence, having methodology to isolate circulating strains from field-collected ticks grants an opportunity to better understand viral evolution, to more accurately assess human health risks, and to make research into these viruses more translational. POWV and DTV are genetically similar but distinct emerging tick-borne viruses in North America that can cause severe encephalitic disease with high morbidity and mortality. Research into these viruses typically use prototypical lab strains, which may not

Diethylnitrosamine Induction of Hepatocarcinogenesis in Mice.

Diethylnitrosamine (DEN) is a chemical hepatocarcinogenic agent that triggers a large array of oncogenic mutations after a single injection. Initiated hepatocytes subsequently undergo clonal expansion within a proliferative environment, rendering the DEN model a comprehensive carcinogen. In rodent studies, DEN finds extensive utility in experimental liver cancer research, mimicking several aspects of human hepatocellular carcinoma (HCC), including angiogenesis, metabolic reprogramming, immune exhaustion, and the ability to metastasize. Beyond the wealth of scientific insights gleaned from this

HPV and Penile Cancer: Epidemiology, Risk Factors, and Clinical Insights.

Penile cancer (PC) is a rare malignancy predominantly of squamous cell origin. Approximately 40% of penile tumors are associated with human papillomavirus (HPV) infection. Diagnosing PC remains challenging due to its rarity and variety of clinical presentations. Furthermore, the impact of HPV on the tumor immune microenvironment complicates clinical management, although recent advancements in immune checkpoint inhibitors (ICIs) have shown some efficacy in treating HPV-associated PC. Ongoing research efforts aim to develop oncologic treatments that target HPV-induced cellular modifications. Add

Analysis of Cannabinoids in Biological Specimens: An Update.

Cannabinoids are still the most consumed drugs of abuse worldwide. Despite being considered less harmful to human health, particularly if compared with opiates or cocaine, cannabis consumption has important medico-legal and public health consequences. For this reason, the development and optimization of sensitive analytical methods that allow the determination of these compounds in different biological specimens is important, involving relevant efforts from laboratories. This paper will discuss cannabis consumption; toxicokinetics, the most detected compounds in biological samples; and charact

FGFR3-TACCs3 Fusions and Their Clinical Relevance in Human Glioblastoma.

Oncogenic fusion genes have emerged as successful targets in several malignancies, such as chronic myeloid leukemia and lung cancer. Fusion of the fibroblast growth receptor 3 and the transforming acidic coiled coil containing protein-FGFR3-TACC3 fusion-is prevalent in 3-4% of human glioblastoma. The fusion protein leads to the constitutively activated kinase signaling of FGFR3 and thereby promotes cell proliferation and tumor progression. The subgroup of FGFR3-TACC3 fusion-positive glioblastomas presents with recurrent clinical and histomolecular characteristics, defining a distinctive subtyp

Airway stem cells sense hypoxia and differentiate into protective solitary neuroendocrine cells.

Neuroendocrine (NE) cells are epithelial cells that possess many of the characteristics of neurons, including the presence of secretory vesicles and the ability to sense environmental stimuli. The normal physiologic functions of solitary airway NE cells remain a mystery. We show that mouse and human airway basal stem cells sense hypoxia. Hypoxia triggers the direct differentiation of these stem cells into solitary NE cells. Ablation of these solitary NE cells during hypoxia results in increased epithelial injury, whereas the administration of the NE cell peptide CGRP rescues this excess damage

The importance of intracellular bacterial biofilm in infectious diseases.

Various bacterial species, previously known as extracellular pathogens, can reside inside different host cells by adapting to intracellular modes by forming microbial aggregates with similar characteristics to bacterial biofilms. Additionally, bacterial invasion of human cells leads to failure in antibiotic therapy, as most conventional anti-bacterial agents cannot reach intracellular biofilm in normal concentrations. Various studies have shown that bacteria such as uropathogenic Escherichia coli, Pseudomonas aeruginosa, Borrelia burgdorferi,Moraxella catarrhalis, non-typeable Haemophilus infl
