Loss of PINK1 impairs mitophagy and accelerates ovarian aging independent of Parkin.

PINK1 and Parkin are central regulators of mitophagy, a quality-control process essential for mitochondrial homeostasis and implicated in aging. However, their specific roles in ovarian physiology remain unclear. Here, we show that Pink1 deletion in mice leads to decreased ovarian weight, diminished ovarian reserve, and reduced oocyte quality, accompanied by increased granulosa cell apoptosis, accelerated ovarian ageing, and impaired fertility. Pink1 deficiency also compromises ovulation efficiency, increases oocyte cytoplasmic fragmentation, and disrupts meiotic spindle assembly, resulting in

Levan-derived oligosaccharides (LOS) prime barley against fungal pathogens through asparagine metabolism.

Overuse of chemical pesticides motivates green disease control strategy. Levan-derived oligosaccharides (LOS) are immune elicitors, but its structure information and underlying regulatory mechanisms remain unclear. In this study, LOS produced from sucrose using LevB1SacB fusion enzyme, was purified by CSR-1Na resin, and characterized by HPLC, ATR-FTIR and 1D/2D NMR. Foliar LOS spraying (0.5‰) enhanced barley resistance to Rhizoctonia solani and Pyrenophora graminea, reducing necrotic area by 60.5% and 22.4%, but had little effect on the hemibiotrophic pathogen Bipolaris sorokiniana of spot blo

PHB2 ameliorates ferroptosis and aortic aneurysm/dissection through NEDD4L-dependent ubiquitination of NCOA4.

BACKGROUND: Aortic aneurysm/dissection (AAD) is a catastrophic vascular emergency with limited therapeutic options and poorly understood molecular underpinnings. Ferroptosis, an iron-dependent form of regulated cell death, emerged as a crucial driver of vascular degeneration although its pathological mechanisms in AAD remain largely undefined. METHODS: We integrated transcriptomic datasets to identify key dysregulated genes in AAD. PHB2 expression was examined by immunohistochemistry, immunofluorescence, and Western blotting in human tissues, murine models, and isolated vascular smooth muscle

Non-canonical double-stranded DNA break repair in Pseudomonas aeruginosa includes LigD-independent non-homologous end joining.

Pseudomonas aeruginosa is a highly adaptive Gram-negative pathogen that is commonly managed with fluoroquinolone (FQ) antibiotics. Even without heritable resistance mutations, FQ-susceptible cells of P. aeruginosa can survive treatment in a phenomenon termed antibiotic persistence. Current models of stationary-phase FQ persistence established with other bacterial species posit that persister survival is dependent on DNA double-stranded break (DSB) repair via RecA-mediated homologous recombination (HR). In P. aeruginosa, however, the effects of RecA loss on FQ persistence vary depending on the

ACSL5 Mediates Adaptation to the Palmitic Acid-Enriched Pulmonary Microenvironment to Enhance Metastatic Breast Cancer Cell Survival and Lung Metastasis.

UNLABELLED: Solid tumors frequently preferentially metastasize to specific organs. Metabolites within metastatic niches have emerged as critical regulators of organotropic metastasis. In this study, we found that palmitic acid (PA) accumulated in both premetastatic and macrometastatic lung niches. Lung-preferential metastatic breast cancer (LM-BC) cells secreted exosomal USP47 that was taken up by lung-resident alveolar type II epithelial cells and enhanced fatty acid synthesis via YAP activation, resulting in PA enrichment and subsequent lung metastasis. ACSL5 in LM-BC cells facilitated PA ad

New mechanistic insight into SARM1 activation: TRIM32-mediated ubiquitination is a key lever that actuates SARM1's catalytic action, leading to axon degeneration and cell death.

Sterile alpha and TIR motif-containing protein 1 (SARM1) is an enzyme that cleaves nicotinamide adenine dinucleotide (NAD+) and plays a role in disrupting neural circuits through axon degeneration and cell death. SARM1 is activated by changes in the nicotinamide mononucleotide (NMN)/NAD+ ratio and by various post-translational modifications, but its complete regulatory mechanism remains poorly understood. Here, we report that tripartite motif-containing protein 32 (TRIM32) activates SARM1 through specific ubiquitination triggered by anticancer drug treatment. TRIM32 promotes the attachment of

Dysfunctional TRIM31 of POMC Neurons Provokes Hypothalamic Injury and Peripheral Metabolic Disorder under Long-Term Fine Particulate Matter Exposure.

Particulate matter ≤2.5 µm (PM2.5) elevates risks of neurological and chronic metabolic diseases, but the underlying mechanisms linking PM2.5-induced central nervous system (CNS) injury to metabolic dysfunction remain unclear. Hypothalamic pro-opiomelanocortin-expressing (POMC+) neurons regulate systemic metabolic homeostasis, and tripartite motif-containing protein 31 (TRIM31) modulates inflammation and metabolism. Here, we investigated whether TRIM31 in POMC+ neurons mediates PM2.5-induced hypothalamic injury and peripheral metabolic disorders in mice subjected to 24-week PM2.5 exposure. TRI

Molecular recognition of substrate intermediate and inhibitors by tryptophanyl-tRNA synthetase from Mycobacterium tuberculosis.

Protein biosynthesis represents a key target for anti-tubercular drug design. Tryptophanyl-tRNA synthetase (TrpRS) is an essential enzyme in Mycobacterium tuberculosis (M.tb) that is involved in translation. Here, we investigate how TrpRS recognizes the substrate-intermediate versus inhibitors and find essential physicochemical features relevant for inhibitor design. We performed systematic molecular dynamics simulations for 24 microseconds across eight TrpRS states, including the apo form, the substrate intermediate, and six indolmycin-based inhibitor-bound states. The substrate-intermediate

Systemic AAV8-SIL1-TAT therapy improves motor function in a mouse model of Marinesco-Sjögren syndrome.

Marinesco-Sjögren Syndrome (MSS) is a rare genetic disorder characterized by cerebellar ataxia, congenital cataracts, and progressive myopathy. Approximately 60% of cases result from SIL1 gene mutations, causing endoplasmic reticulum stress and neuromuscular degeneration. We investigated AAV8-mediated SIL1 gene replacement combined with TAT peptide-mediated protein delivery in the woozy (Sil1wz) mouse model. Thirty-two female Sil1wz mice received either AAV8-SIL1-TAT vector (5 × 1012 genome copies) or saline intravenously at 4 weeks of age. The construct enabled liver-produced SIL1 protein upt

Transient Trim25 Knockdown Enhances Transfection Efficiency in Two Teleost Fish Cell Lines.

Efficient transgene expression following nucleic acid transfection remains difficult in many fish cell lines, particularly those derived from marine species. Although extracellular delivery systems have been extensively optimized, intracellular factors that limit the expression of delivered nucleic acids remain poorly understood in teleosts. In mammals, TRIM25 (tripartite motif-containing protein 25) participates in cytoplasmic exogenous RNA surveillance. Here, we examined whether its teleost homolog contributes to the low reporter expression observed after transfection. Transient siRNA-mediat

An astrocytic cellular model of Lafora disease to study polyglucosan accumulation and inflammation.

Lafora disease (LD) is a devastating form of progressive myoclonus epilepsy characterized by the accumulation of insoluble forms of glycogen [polyglucosan bodies (PGBs)] in the brain and peripheral tissues. It has been proposed that the accumulation of PGBs is pathogenic. Several mouse models of LD have been generated to study the relationship between PGBs and the pathophysiology of LD. However, the use of LD mice is difficult and time consuming; thus, more amenable cellular systems would be desirable. We recently described a cellular model based on the culture of primary postnatal astrocytes

TRIM9 regulates the proliferation and apoptosis of dermal papilla cells by activating the Wnt/β-catenin signaling pathway to improve androgenetic alopecia.

BACKGROUND AND PURPOSE: Dermal papilla cells (DPCs) are signaling hubs for hair follicle growth and play a critical role in hair growth in patients with androgenetic alopecia (AGA). TRIM9, a tripartite motif (TRIM)-family E3 ubiquitin ligase, regulates cell differentiation, proliferation, and apoptosis. This study aimed to investigate the effects of TRIM9 on the proliferation and apoptosis of DPCs. METHODS: AGA mouse and cellular models were induced by dihydrotestosterone (DHT) treatment to conduct experimental investigations. The damage to human dermal papilla cells (hDPCs) and mouse dorsal

AJUBA Attenuates Pathological Cardiac Hypertrophy by Enhancing NEDD4-Mediated Ubiquitination and Degradation of DVL2.

Cardiac hypertrophy, a response to pathological stimuli, eventually progresses to heart failure (HF). AJUBA is a multifunctional protein expressed in various human tissues, serving as a critical modulator of cellular processes and signalling pathways involved in cardiac hypertrophy and remodelling. It plays significant roles in cellular signalling, gene transcription, and cytoskeletal organisation, modulating cardiac growth, inflammation, and cell survival. However, the precise function of AJUBA in cardiac hypertrophy remains unclear. In the present study, we examined its function as a novel r

Residue 188 of the Hexon protein governs FAdV-4 pathogenicity by activating PINK1/Parkin-mediated mitophagy.

Fowl adenovirus serotype 4 (FAdV-4) infection causes significant economic losses to the global poultry industry. Viruses often hijack host cellular machinery to facilitate their replication; however, the mechanisms by which FAdV-4 manipulates host pathways remain poorly defined. Mitochondria, the central hubs for energy metabolism and innate immunity in hepatocytes and cardiomyocytes, are critical targets for viral manipulation, yet their role in FAdV-4 pathogenesis remains unexplored. Here, we demonstrated that FAdV-4 infection caused direct mitochondrial damage and induced PINK1/Parkin-depen

Schisandrol A in Fufang Muniziqi Granules suppresses the abnormal expression of MHC-II in renal tubular epithelial cells to treat lupus nephritis through disrupting the interaction between STUB1 and PPARγ.

OBJECTIVE: In lupus nephritis (LN), renal tubular epithelial cells (RTECs) can acquire an abnormal MHC-II antigen-presenting cell-like phenotype and thereby amplify local immune responses. This study aimed to evaluate the therapeutic effects of the Uyghur medicine Fufang Muniziqi Granules (FMG) in LN, determine whether FMG acts by suppressing aberrant MHC-II expression in RTECs, and identify its potential targets and molecular mechanisms. METHODS: We treated spontaneous LN MRL/lpr mice with different doses of FMG. We assessed systemic immune activation by measuring the spleen index, autoantib
