OBJECTIVES: Group B Streptococcus (GBS) increasingly causes invasive disease in non-pregnant adults, particularly in elderly persons and those with underlying diseases. Combination therapy with penicillin plus gentamicin has been suggested for periprosthetic joint infection. The postulated synergism of this combination is based on experiments with planktonic bacteria. We aimed to assess the efficacy of this combination against sessile bacteria.
METHODS: Four different GBS strains were used. We compared results of MICs with those of minimal biofilm eradication concentrations (MBECs), applied c

The purpose of this investigation was to try to understand the antibacterial mechanism of L-(-)-usnic acid isolated for the first time from fruticose lichen Usnea subfloridana using clinical isolates of methicillin-resistant Staphylococcus aureus (MRSA). The minimum inhibitory concentration (MIC) of L-(-)-usnic acid against the clinical isolates of MRSA and reference strain S. aureus MTCC-96 (SA-96) was in the range 25-50 μg/ml. Treatment of both reference and clinical strains (MRSA-ST 2071) with four-fold MIC concentrations (100-200 μg/ml) of L-(-)-usnic acid reduced the viability of cells wi

Observational studies have long suggested an association between gut microbiota dysbiosis and gastric cancer, yet the causal relevance and underlying biological pathways remain poorly defined. Here, we applied a comprehensive Mendelian randomization (MR) framework to gut microbiota and gastric cancer GWAS summary statistics to identify 14 microbial taxa significantly associated with gastric cancer risk. Sensitivity analyses, including tests for horizontal pleiotropy, heterogeneity, and genetic co-localization, revealed no strong evidence of shared causal variants (H4 < 2%); however, convergent

BACKGROUND: Ottowia massiliensis sp.nov. is a slow-growing and Gram-stain-negative bacillus, first isolated from the stool sample of a healthy volunteer. Due to its slow growth and challenging cultivation, it is easily overlooked in clinical microbiology laboratories. This case report is the first case of its pathogenicity.
CASE PRESENTATION: This case report describes a case of canaliculitis caused by Ottowia massiliensis sp.nov. The patient, an 80-year-old woman, presented with bilateral epiphora and discharge lasting for 4 years, which worsened with redness and swelling over the past week.

SUMMARYImplant-associated infections (IAIs) pose serious threats to patients and can be associated with significant morbidity and mortality. These infections may be difficult to diagnose due, in part, to biofilm formation on device surfaces, and because even when microbes are found, their clinical significance may be unclear. Despite recent advances in laboratory testing, IAIs remain a diagnostic challenge. From a therapeutic standpoint, many IAIs currently require device removal and prolonged courses of antimicrobial therapy to effect a cure. Therefore, making an accurate diagnosis, defining

Governments regulate antimicrobial residues in slaughtered animals with surveillance programs for detecting drugs in food-producing animals. Although initial screening bioassay systems are recognized for their sensitivities to antimicrobial drug groups, none are sensitive to sulfonamides at or near the maximum residue levels (MRLs) in the Codex Alimentarious. We have developed a sulfonamide-sensitive rapid assay using Bacillus stearothermophilus inoculated PM indicator agar containing bromcresol purple and trimethoprim, where the end point is a combination of color change in the agar and zone

Small intestinal bacterial overgrowth (SIBO) is characterized as the increase in the number and/or alteration in the type of bacteria in the upper gastrointestinal tract and accompanies various bowel symptoms such as abdominal pain, bloating, gases, diarrhea, and so on. Clinically, SIBO is diagnosed by microbial culture in duodenum/jejunum fluid aspirates and/or the breath tests (BT) of hydrogen/methane gases after ingestion of carbohydrates such as glucose. The cultural analysis of aspirates is regarded as the golden standard for the diagnosis of SIBO; however, this is invasive and is not wit

Mass spectrometry (MS) has evolved from a specialized research tool into an increasingly indispensable platform in laboratory medicine. Its clinical value is driven by molecular specificity, multiplex quantification, and the ability to measure small molecules, peptides, proteins, drugs, metabolites, and microbial spectral fingerprints in workflows that are becoming more automated and standardized. The field is nevertheless heterogeneous: liquid chromatography-tandem mass spectrometry (LC-MS/MS), matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS), gas ch

OBJECTIVE: Bacterial DNA from oral microorganisms has been found in various cardiovascular tissues, including abdominal aortic aneurysm (AAA) tissue. In addition, experiments in murine models have shown that the oral pathogen Porphyromonas gingivalis can independently contribute to aneurysm formation. However, whether the oral microbial community differs between patients with AAA and those with advanced atherosclerotic disease remains unknown. This study tests the hypothesis that specific bacterial taxa are differentially abundant in patients with AAA compared to those with advanced atheroscle

Ceftazidime was administered intravenously in doses of 1 to 6 g/day to 21 patients with serious Pseudomonas aeruginosa infections (12 pulmonary, 6 septicaemias, 3 urinary tract infections). Mean MIC was 1.0 mg/l. Eighteen (86%) of the 21 patients responded satisfactorily (cured or improved). The selection or emergence of resistant organisms during treatment (mostly Candida, Staphylococcus aureus, and enterococci) was noted in 6 patients. Toxicity was minimal (eosinophilia and reversible mild liver function abnormalities).

Atopic dermatitis (AD) is a chronic relapsing inflammatory skin disease, associated with impaired skin barrier function and an atopic background. Various complicating factors, such as irritants, aeroallergens, food, microbial organisms, contact allergens, sweat, and scratching can induce the development of AD symptoms. Irritants, including soap/shampoo and clothes, can cause itching and eczematous lesions. In addition, young children with AD tend to become sensitized to eggs, milk, or peanuts, while older children and adults more often become sensitized to environmental allergens, such as hous

In this study, the effects of the electric charges and fields on the viability of airborne microorganisms were investigated. The electric charges of different magnitude and polarity were imparted on airborne microbial cells by a means of induction charging. The airborne microorganisms carrying different electric charge levels were then extracted by an electric mobility analyzer and collected using a microbial sampler. It was found that the viability of Pseudomonas fluorescens bacteria, used as a model for sensitive bacteria, carrying a net charge from 4100 negative to 30 positive elementary ch

BACKGROUND: Bacterial vaginosis (BV) is a most common microbiological syndrome. The use of molecular methods, such as multiplex real-time PCR (mPCR) and next-generation sequencing, has revolutionized our understanding of microbial communities. Here, we aimed to use a novel multiplex PCR test to evaluate the microbial composition and dominant lactobacilli in non-pregnant women with BV, and combined with machine learning algorithms to determine its diagnostic significance.
METHODS: Residual material of 288 samples of vaginal secretions derived from the vagina from healthy women and BV patients

Granulomatous amoebic encephalitis (GAE) caused by Acanthamoeba spp. is a rare, nearly universally fatal central nervous system infection that predominantly affects immunocompromised individuals, including solid organ transplant (SOT) recipients. Early diagnosis is essential but often delayed due to nonspecific symptoms and the limited sensitivity of conventional diagnostic tests. We describe the case of a 58-year-old man who presented with fever and diarrhea one month after a simultaneous pancreas-kidney transplant. Despite broad-spectrum antimicrobial therapy, he developed status epilepticus

INTRODUCTION: SGLT2 inhibitors (SGLT2i) are extensively used in India for glycemic control, weight management and cardiorenal protection in individuals with type 2 diabetes (T2DM). SGLT2i promotes glucosuria, which may predispose these patients to asymptomatic bacteriuria (ASB). In this study, we assessed the prevalence, microbial spectrum, and risk factors of ASB in T2DM patients treated with and without SGLT2i at a tertiary centre in Northern India.
MATERIALS AND METHODS: This was a hospital-based, prospective observational study conducted at a tertiary care hospital in North India and incl

Photothermal antimicrobial therapy (PTT) has demonstrated significant potential in eradicating biofilms and reducing bacterial resistance. However, a major drawback of traditional PTT is the lack of selectivity, which reduces the efficiency and can cause substantial damage to healthy cells and tissues. Therefore, improving the precision of photochemical therapy against pathogenic bacteria is critical. This study leverages the high targeting specificity of bacteriophages to develop a bacteriophage-modified, near-infrared-responsive conjugated polymer nanosystem for targeted elimination of bacte
