Beta 2-adrenergic receptors contribute to catecholamine-stimulated shortening of action potential duration in dog atrial muscle.

beta 1- and beta 2-adrenergic receptors coexist in atria of humans and a variety of other species. The significance of these receptors and the quantitative extent to which beta 2-adrenergic receptors contribute to the electrophysiologic effects of catecholamines acting at beta-adrenergic receptors have not been determined. In the present study, the beta 1-selective antagonist ICI 89,406 and the beta 2-selective antagonist ICI 118,551 were used to determine the relative densities of beta 1- and beta 2-adrenergic receptors on membranes prepared from dog atria and the contribution that each subty

Influence of transmural pressure on retrograde pressure pulse transmission velocity in the canine superior vena cava.

The influence of intrathoracic and intraluminal pressure on venous pressure pulse propagation velocity in the superior vena cava was investigated in acute canine experiments. The propagation velocity of distinct points of the venous pressure curve was measured under different conditions of preload, during expiratory and inspiratory apnea and during positive-negative pressure respiration. Under control conditions transmission velocities varied from 1.07 m/s to 3.29 m/s. After volume expansion propagation velocities rose significantly and varied from 1.09 m/s to 6.48 m/s. During expiratory apnea

Characterization of the (Ca2+-Mg2+)ATPase purified by calmodulin-affinity chromatography from bovine aortic smooth muscle.

A calmodulin inhibitor, trifluoperazine, suppresses ATP-dependent Ca2+ uptake into microsomes prepared from bovine aortic smooth muscle. From this microsomal preparation which we expected to contain calmodulin-dependent Ca2+-transport ATPase [EC 3.6.1.3], we purified (Ca2+-Mg2+)ATPase by calmodulin affinity chromatography. The protein peak eluted by EDTA had calmodulin-dependent (Ca2+-Mg2+)ATPase activity. The major band (135,000 daltons) obtained after sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) accounted for about 80% of the total protein eluted. This major band was

Responses to noradrenaline of portal vein strips from normotensive and spontaneously hypertensive rats.

1 Responses to noradrenaline of isolated portal vein strips from 5-7 week and 15-17 week Wistar Kyoto (WKY) and spontaneously hypertensive rats (SHR) have been examined. 2 15-17 week SHR tissue exhibited a greater sensitivity (lower ED50) to noradrenaline but only at reduced Ca2+EXT levels. 3 pA2 values for the alpha-adrenoreceptor antagonists WB 4101 and prazosin were significantly lower in 5-7 week SHR consistent with some change at the adrenoreceptor level. 4 Veins from SHR relaxed more rapidly in CA2+-free saline following removal of noradrenaline than did veins from WKY. 6 D600 is a compe

[Effect of antihistaminics on bradykinin action].

On isolated ileum of the guinea pig the antihistamine drugs dimedrol, diprazin (pipolphen), tavegil and suprastin diminished spasmogenic effects of bradykinin and an enhancement of microvascular permeability induced by this polypeptid. The antihistamine drugs display a nonspecific antibradykinin activity. No relationship between the chemical structure and pronounced antibradykinin action of the antihistamines studied was revealed.

Arteriolar tone is determined by activity of ATP-sensitive potassium channels.

The role of ATP-sensitive potassium channels (KATP) in determining resting arteriolar tone and vasodilator reactivity was assessed in superfused, hamster microcirculatory beds studied via intravital microscopy. Under resting conditions, the selective KATP blocker, glibenclamide, produced concentration-dependent vasoconstriction in both the cheek pouch and the cremaster muscle. Concentration-related constriction of cheek pouch arterioles was also observed with tetrapentylammonium, although this agent appeared to have toxic effects on the microcirculation. Glibenclamide (2 microM) abolished arte

Engineering biomimetic coronary artery equivalents for Kawasaki disease modeling and drug screening.

Kawasaki disease (KD) is an acute vasculitis that primarily affects the pediatric population, with coronary artery lesions constituting its most severe clinical sequela. Although investigated for decades, the precise etiology and pathogenesis of this syndrome remain elusive. This critical knowledge gap stems from the absence of robust in vitro platforms capable of accurately recreating the complex pathophysiological dynamics of pediatric coronary arteries. To overcome this challenge, we developed hollow hydrogel fibers using microfluidic coaxial printing, which were then seeded with human coro

A loss-of-function polymorphism in the propeptide of lysyl oxidase exacerbates atherosclerosis.

A single-nucleotide missense polymorphism (rs1800449, R158Q) in the propeptide domain of lysyl oxidase (LOX-PP) is associated with increased risk of coronary artery disease (CAD) independent of changes in plasma lipid levels. Although the enzymatic function of LOX has an essential role for the cross-linking of extracellular matrix proteins in connective tissues, whether and how LOX-PP R158Q contributes to the development of atherosclerosis has not been clearly established. Here, hypercholesterolemia was induced in mice that were WT or homozygous for the LOX-PP R158Q polymorphism by adeno-assoc

Circulating Cell Type Senescence Signatures Reveal High-Resolution Health Status and Trajectories in Human Longitudinal Studies.

Cellular senescence increases in frequency with age and is implicated in age-related pathologies, and identifying circulating biomarkers of senescence holds great diagnostic potential. Circulating senescence signatures are predictive of many age-related traits and diseases, though cell type-specific senescence signatures have not been comprehensively explored. In this study, senescence signatures from the Senescence Catalog (SenCat), including 14 human cell types such as peripheral blood mononuclear cells, renal epithelial cells, vascular smooth muscle cells, among others, are examined for the

MicroRNAs in Cardiovascular Diseases: Molecular Networks of Cellular Homeostasis, Inflammation, and Pathological Remodeling.

Cardiovascular diseases remain the leading cause of morbidity and mortality worldwide, underscoring the need to better define the molecular mechanisms that govern cardiovascular homeostasis and disease progression. Among post-transcriptional regulators, microRNAs have emerged as important modulators of endothelial function, vascular smooth muscle cell plasticity, cardiomyocyte integrity, and cardiac fibroblast activity. This narrative review examines how microRNAs orchestrate molecular networks linking cellular homeostasis to inflammation, oxidative stress, mitochondrial dysfunction, apoptosis

Single-Cell Mapping Identifies Aging-Associated Neutrophil Remodeling Across Multiple Organs.

Aging is accompanied by progressive immune remodeling and functional decline across multiple organs. However, the role of neutrophils and their heterogeneous states in these processes remains incompletely understood. Here we integrated single-cell RNA sequencing datasets from multiple organs of young and aged mice to characterize aging-associated neutrophil remodeling. We identified organ-dependent changes in neutrophil abundance and state composition, together with neutrophil subsets enriched for inflammatory, oxidative stress, complement, and IL-6-related transcriptional programs. In vitro,

Detection of active vasodilatory compounds from hawthorn: Screening and determination of mechanism of action.

Hawthorn is known to reduce arterial blood pressure in humans, but the active component remains unidentified. The vasoactivity of twenty-one substances contained in hawthorn (flavonoids, glycosides, pentacyclic triterpenes) and absorbable metabolites (small phenolic acids) was tested on three types of isolated blood vessels from two species. Isorhamnetin and tamarixetin induced vasodilatation of porcine coronary arteries (EC50 values were 21.7 and 30.9 μM, respectively), rat aorta (16.7 and 23.3 μM), and rat mesenteric arteries (22.2 and 16.8 μM). These values are higher than the typical human

Placental and fetal responses to adrenomedullin deficiency in the rat†.

Adrenomedullin (ADM) is a peptide hormone with a broad spectrum of actions on vascular smooth muscle, endothelial cells, and immune cells. These ADM cellular targets are prominent constituents of the uterine-placental interface that undergo structural and functional transformation during pregnancy. To explore the biology of ADM at the uterine-placental interface, we examined ADM expression using in situ hybridization and generated and characterized an ADM mutant rat model using CRISPR-Cas9-mediated genome editing. At midgestation, Adm was prominently expressed in trophoblast giant cells. Adm e

The role of lactate metabolism in uterine spiral artery remodelling.

Lactate is a key metabolite of glycolysis. Since the proposal of the lactate shuttle hypothesis, accumulating evidence indicates that lactate is not only an essential energy substrate but also a key signalling molecule that regulates cellular functions under both physiological and pathological conditions. Uterine spiral artery (SpA) remodelling is a critical event in successful pregnancy. It involves complex interactions among trophoblasts, vascular smooth muscle cells (VSMCs), decidual immune cells, and endothelial cells (ECs). Similar to the tumour microenvironment, physiological hypoxia at

Methylene Blue for the Treatment of Refractory Hypotension in Polysubstance Overdose.

Methylene blue is commonly used for methemoglobinemia and vasoplegia following coronary artery bypass procedures but has recently been investigated as a treatment for refractory hypotension. One such cause of refractory hypotension is overdose of calcium channel blocker medications. This case presents a patient with polysubstance overdose and refractory hypotension that improved with methylene blue infusion. This 67-year-old female presented to the ED following an intentional toxic ingestion of multiple substances, including metoprolol, amlodipine, mirtazapine, pantoprazole, quetiapine, leveti

Measurement of oxygen consumption rate in mouse aortic tissue.

Thoracic aortic aneurysm and dissection (TAD) is a life-threatening vascular disorder, and smooth muscle cell mitochondrial dysfunction leads to cell death, contributing to TAD. Accurate measurements of metabolic processes are essential for understanding cellular homeostasis in both healthy and diseased states. While assays for evaluating mitochondrial respiration have been well established for cultured cells and isolated mitochondria, no optimized application has been developed for aortic tissue. In this study, we generate an optimized protocol using the Agilent Seahorse XFe24 analyzer to mea

Inhibition of inducible nitric oxide synthase (iNOS) alleviates thoracic aortic aneurysm by regulating mitochondrial dynamics.

Thoracic aortic aneurysm (TAA) is life-threatening once developing to dissection (TAAD) or rupture and currently there is no effective pharmacological treatment. The abnormal activity of inducible nitric oxide synthase (iNOS) has been related with syndromic TAA, but its potential role in non-syndromic TAA is unknown. Here we identified elevations of inducible nitric oxide synthase (iNOS) and its downstream protein kinase G1 (PRKG1) in both human non-syndromic TAA and β-aminopropionitrile (BAPN)-induced mouse model of TAA. The iNOS-specific inhibitor 1400W effectively inhibited the formation of
