The EAAT1 aspartate/glutamate transporter is dispensable for acute myeloid leukemia cell growth and response to therapy.

Acute myeloid leukemia (AML) is an aggressive malignancy of hematopoietic stem and progenitor cells characterized by profound metabolic dysregulation. Pyrimidine biosynthesis has emerged as a critical metabolic dependency in AML, but clinical translation has been hampered by unacceptable toxicity of current pyrimidine synthesis inhibitors. Since aspartate is an essential nutrient for pyrimidine biosynthesis, we investigated the role of aspartate import via the excitatory amino acid transporter 1 (EAAT1) in AML. We found that EAAT1 is broadly expressed across AML cell lines and patient samples,

UPP1 in Cancer: Context-Dependent Roles in Metabolic Adaptation and Treatment Response.

Uridine phosphorylase 1 (UPP1) catalyzes the reversible phosphorolysis of uridine to uracil and ribose-1-phosphate, thereby linking pyrimidine salvage to central carbon metabolism. In cancer, UPP1 is functionally relevant when UPP1-linked pathway engagement and a perturbation-dependent phenotype converge within a defined cellular, nutrient/substrate, molecular, or treatment setting. The direction of effect is determined by the dominant biochemical or signaling output, whereas the strength of UPP1 dependence may be attenuated by compensation through related nucleoside-metabolic pathways. DRP1-mediated mitophagy facilitates oral squamous cell carcinoma tumor cell survival under glucose restriction.

Oral squamous cell carcinoma (OSCC) can progress within a nutrient-limited microenvironment, especially under low-glucose conditions. However, the mechanisms enabling tumor cells to cope with glucose limitation, particularly those that counteract reactive oxygen species (ROS) accumulation, remain largely unknown. Here, we showed that dynamin-related protein 1 (DRP1)-mediated mitophagy promoted OSCC cell survival under glucose restriction. High expression of DRP1 was observed in OSCC tumor cells and identified as an indicator of poor prognosis. Either expression knockdown or pharmacological inh

Seasonal Effect of a Small Hydropower Plant on Benthic Biofilm Microbial Community Structure and Predicted Functional Potential.

Benthic biofilm microbial communities play a critical role in riverine primary production, nutrient cycling, and organic matter decomposition, yet their responses to hydrological alterations due to small hydropower plants (SHPs) remain poorly understood, particularly with respect to seasonal dynamics. This study conducted a one-year monitoring survey across four river sections (upstream, reservoir, dewatered reach, and downstream) in the Oujiang River Basin, Zhejiang Province, China, covering all four seasons. The results showed that the reservoir section exhibited distinct bacterial compositi

Reconfiguring biofortification strategies to transform food systems and address micronutrient deficiency of the 21st century.

Despite achieving substantial progress in caloric food security, micronutrient deficiencies remain a major global health challenge, highlighting a persistent disconnect between crop productivity and nutritional quality. Crop biofortification has emerged as a promising strategy to address this gap by enhancing the intrinsic nutrient content of widely consumed foods to meet dietary requirements through complementary roles of staple crops and horticultural species in improving diet quality. In this review, we reposition biofortification within a broader nutrition-sensitive food system framework a

Multi-omics analysis reveals Protein Kinase A-associated regulatory remodeling during adaptation of Trichoderma reesei to lignocellulosic substrate.

The filamentous fungus Trichoderma reesei is a major industrial source of holocellulolytic enzymes, and its response to complex carbon sources is regulated by nutrient-sensing mechanisms, including the cyclic adenosine monophosphate (cAMP)-protein kinase A (PKA) signaling pathway. Here, we integrated transcriptomics, quantitative proteomics, and phosphoproteomics to analyze PKAc1-associated responses in the parental strain QM9414 and a Δpkac1 strain cultivated under glucose or sugarcane bagasse conditions. Deletion of pkac1 was associated with altered growth-related phenotypes and reduced extr

Dissecting Cancer Metabolism and Therapeutic Resistance Using In Vitro Platforms.

Understanding cancer metabolism and its adaptations is critical for developing novel therapeutic strategies. In vitro models remain indispensable tools for dissecting tumor biology and testing metabolism-based therapies. Traditional 2D monolayer cultures provide controlled, cost-effective, and reproducible platforms for mechanistic studies, genetic manipulation, and high-throughput drug screening. Advances in 3D models, such as spheroids, organoids, and microfluidic tumor-on-a-chip systems, try to come closer to a natural tumor in order to recapitulate cellular heterogeneity, nutrient and oxyg

Cysteine, methionine, and pantothenic acid remodel the Saccharomyces cerevisiae transcriptome and volatile sulfur compound metabolome during alcoholic fermentation.

Yeast nitrogen and vitamin nutrition are fundamental levers for managing wine quality, yet the specific mechanisms linking nutrient availability to sulfur aroma formation remain poorly understood. This study explored the impact of methionine, cysteine, and pantothenic acid (vitamin B5)-nutrients with direct, pivotal roles in sulfur metabolic pathways-on the Saccharomyces cerevisiae transcriptome and VSC metabolome during fermentation. Our findings reveal that methionine and cysteine catabolic routes act as isolated compartments; the genes responsible for bridging these two pathways remain tran…

Yeast strain development and process intensification in high-gravity fermentation.

High- and very-high-gravity (HG/VHG) fermentation increases substrate loading and product titers, thereby improving fermenter utilization and potentially reducing water use and downstream processing requirements. Initially developed for brewing and fuel ethanol production, these approaches are now applied more broadly in food, beverage, and bioproduct manufacturing. This MiniReview summarises operational definitions and industrial drivers of HG/VHG fermentation and examines the associated constraints in rheology, mass and heat transfer, osmotic and ethanol stress, nutrient availability, and ox

Sex-specific placental adaptations associated with fetal growth restriction: a review.

Fetal growth restriction (FGR) is a major obstetric complication linked to placental insufficiency, increasing the risk of perinatal morbidity, mortality, and long-term health issues. The placenta is central to fetal development and maternal-fetal communication; its dysfunction leads to impaired fetal development. Numerous experimental FGR models exist, from rodents to large animals, but most studies, despite changes to some federal funding requirements, have historically overlooked the impact of fetal sex on placental development, function, and adaptation. Recent evidence indicates that male

PPE57 Cooperates with MmpL3 to Mediate Bacterial Lipid Transport and Promote Persistent Infection of Mycobacterium tuberculosis.

Mycobacterium tuberculosis (M. tb) possesses a unique, lipid-rich cell envelope that is critical for virulence, drug resistance, and persistence. Here, we identify the PPE family protein PPE57 as a key regulator of mycolic acid transport and host lipid exploitation. PPE57 physically interacts with the essential lipid transporter MmpL3, promoting trehalose monomycolate (TMM) translocation, enhancing trehalose dimycolate (TDM) synthesis, and increasing cell wall lipid content. Site-directed mutagenesis identified G175 as a critical residue for PPE57-MmpL3 binding. Deletion of PPE57 reduces cell

Ready-to-Use School Meals in Northern Ghana Are a Viable Alternative to Traditional School Meals.

School feeding provides nutrition, brings order to the school day, and enhances student participation. School feeding in low-income countries is often sporadic due to coordination challenges among multiple stakeholders. To assess the reliability of school feeding in Mion district, a food-insecure area in northern Ghana, Project Peanut Butter (PPB) studied ready-to-use school meals (RUSMs) and micronutrient-fortified home-grown school food (HGSF). The school meals were initially provided daily in elementary schools and then extended to junior high schools. The key elements of functional program
