Mechanisms of Deng-Shi-Qing-Mai-Tang in alleviating PM2.5-Induced lung Injury: Network pharmacology, metabolomics, and molecular Target validation.

ETHNOPHARMACOLOGICAL RELEVANCE: Deng-Shi-Qing-Mai-Tang (DSQMT) is a traditional Chinese herbal formula known for treating inflammatory diseases, particularly those affecting respiratory health. Urban air pollution, especially fine particulate matter (PM2.5), induces lung injury primarily through inflammation and oxidative stress. DSQMT's potential to mitigate PM2.5-induced lung damage makes it a promising ethnopharmacological candidate for pollution-related pulmonary disorders. AIM OF THE STUDY: This study aims to uncover the therapeutic mechanisms of DSQMT in alleviating PM2.5-induced lung i…

Maternal exposure to ambient air pollution and risk of congenital heart defects in offspring: A population-based cohort study in Wuhan, China.

BACKGROUND: Existing evidence on maternal ambient air pollutants exposure during pregnancy and the risk of congenital heart defects (CHDs) and its subtypes in offspring is inconsistent, and few studies have focused on preconception exposure. The critical susceptibility window for air pollutants linked to CHDs remains poorly answered. OBJECTIVE: We aimed to explore whether maternal exposure to ambient air pollutants before and after conception were associated with the risk of CHDs and its subtypes, and determined the critical susceptibility windows and potential modifiers for air pollutants ex

Modeling heterogeneity in air pollution mixture effects on birth weight: A spatially varying coefficient approach.

PURPOSE: To extend the existing quantile g-computation framework for studying environmental exposure mixtures to estimate local effects of ambient air pollution mixtures on birth weight. This framework has traditionally been applied to estimate global mixture effects without accounting for spatial heterogeneity. METHODS: First, pregnancy-wide maternal exposure to five common air pollutants is estimated for nearly 1.5 million births occurring in Georgia, USA between 2005 and 2016. Then, a recently developed spatially varying coefficient model based on Bayesian additive regression trees (BART)

Air pollution and depression symptoms in middle-aged and older adults in Los Angeles County.

OBJECTIVE: Long-term exposure to air pollutants may be harmful to the brain, potentially through inducing oxidative stress or inflammation. Few studies of air pollution and depression have been conducted in the United States where this mental health disorder is prevalent among adults. We investigated associations between ambient air pollutants (O3, PM2.5 and NO2) and depression symptoms in middle-aged and older adults (n = 1496) without cardiovascular disease or cognitive impairment in Los Angeles, California. METHODS: Air pollution exposures were assigned to residential addresses using a geo

Association of ambient air pollution with incident rheumatoid arthritis, subsequent respiratory diseases, and death: A multi-state analysis of a prospective cohort.

BACKGROUND: Previous studies have mostly examined the separate effects of air pollution exposure on the onset of rheumatoid arthritis (RA) or respiratory diseases. However, no studies have further explored the impact of air pollution on the progression from a baseline healthy population to the onset of RA, followed by the further complication of respiratory diseases and, ultimately, mortality. METHODS: Based on data from the UK Biobank, we included 340,485 participants without RA or respiratory diseases. Air pollution exposure levels were estimated by land use regression modeling, and multi-s

Biological responses of human tracheobronchial mucociliary pseudo-epithelium repeatedly exposed at the air-liquid interface to PM10 indoor particles.

The PM10 fraction of representative indoor pollution (SRM® 2585) was repeatedly nebulized (1 and 6 μg/cm2/day) on functional mucociliary pseudo-epithelia (NHBE cells differentiated at the Air Liquide Interface, ALI). Four consecutive daily exposures have been performed and the biomarkers were analyzed on day 4, 8 and 11. Exposures at the lower dose induced at day 4 a significant increase in cellular ROS and modulations of the expression of few genes associated with genotoxicity and oxidative stress. IL-6 expression was strongly upregulated accompanied by a delayed increased secretion in the ba

Genetic evidence for the causal effects of air pollution on the risk of respiratory diseases.

BACKGROUND: Epidemiological studies have consistently demonstrated a robust association between long-term exposure to air pollutants and respiratory diseases. However, establishing causal relationships remains challenging due to residual confounding in observational studies. In this study, Mendelian randomization (MR) analysis was used to explore the causal and epigenetic relationships between various air pollutants and common respiratory diseases. METHODS: We utilized a two-sample Mendelian randomization (TSMR) approach to explore the impact of PM2.5, PM2.5-10, PM10, NO2, and NOX on the inci

Effects of long-term exposure to outdoor air pollution on COVID-19 incidence: A population-based cohort study accounting for SARS-CoV-2 exposure levels in the Netherlands.

Several studies have linked air pollution to COVID-19 morbidity and severity. However, these studies do not account for exposure levels to SARS-CoV-2, nor for different sources of air pollution. We analyzed individual-level data for 8.3 million adults in the Netherlands to assess associations between long-term exposure to ambient air pollution and SARS-CoV-2 infection (i.e., positive test) and COVID-19 hospitalisation risks, accounting for spatiotemporal variation in SARS-CoV-2 exposure levels during the first two major epidemic waves (February 2020-February 2021). We estimated average annual

Air pollution, genetic factors, and chronic rhinosinusitis: A prospective study in the UK Biobank.

BACKGROUND: Chronic rhinosinusitis (CRS) is a prevalent upper respiratory condition that manifests in two primary subtypes: CRS with nasal polyps (CRSwNP) and CRS without nasal polyps (CRSsNP). While previous studies indicate a correlation between air pollution and CRS, the role of genetic predisposition in this relationship remains largely unexplored. We hypothesized that higher air pollution exposure would lead to the development of CRS, and that genetic susceptibility might modify this association. METHODS: This cohort study involving 367,298 adult participants from the UK Biobank, followe

Long-term exposure to air pollution, road traffic noise and greenness, and incidence of myocardial infarction in women.

BACKGROUND: Emerging evidence shows that long-term exposure to air pollution, road traffic noise, and greenness can each be associated with cardiovascular disease, but only few studies combined these exposures. In this study, we assessed associations of multiple environmental exposures and incidence of myocardial infarction using annual time-varying predictors. MATERIALS AND METHODS: In a population-based cohort of 20,407 women in Sweden, we estimated a five-year moving average of residential exposure to air pollution (PM2.5, PM10 and NO2), road traffic noise (Lden), and greenness (normalized

Maternal PM2.5 exposure is associated with preterm birth and gestational diabetes mellitus, and mitochondrial OXPHOS dysfunction in cord blood.

Maternal exposure to fine particulate matter (PM2.5) is associated with adverse pregnancy and neonatal health outcomes. To explore the mechanism, we performed mRNA sequencing of neonatal cord blood. From an ongoing prospective cohort, Air Pollution on Pregnancy Outcome (APPO) study, 454 pregnant women from six centers between January 2021 and June 2022 were recruited. Individual PM2.5 exposure was calculated using a time-weighted average model. In the APPO study, age-matched cord blood samples from the High PM2.5 (˃15 ug/m3; n = 10) and Low PM2.5 (≤ 15 ug/m3; n = 30) groups were randomly selec…

Gestational and Postpartum Exposure to PM2.5 Components and Glucose Metabolism in Chinese Women: A Prospective Cohort Study.

Pregnant women are physiologically prone to glucose intolerance, while the puerperium represents a critical phase for recovery. However, how air pollution disrupts glucose homeostasis during the gestational and early postpartum periods remains unclear. This prospective cohort study conducted an oral glucose tolerance test and measured the insulin levels of 834 pregnant women in Guangzhou, with a follow-up for 443 puerperae at 6-8 weeks postpartum. Residential PM2.5 and five chemical components were estimated by an established spatiotemporal model. The adjusted linear model showed that an IQR i

Circulating metabolomics revealed novel associations between multiple ambient air pollutants exposure and chronic obstructive pulmonary disease incidence: Evidence from a prospective cohort study.

The mechanisms underlying relationships between ambient air pollution and chronic obstructive pulmonary disease (COPD) risk remained largely uncertain. In this study, we aim to evaluate whether metabolic signature comprising multiple circulating metabolites can characterize metabolic response to the multiple air pollution; and to assess whether the identified metabolic signature contribute to COPD risk. A total of 227,962 participants with complete data were included from the UK biobank study. Concentrations of nitrogen dioxide (NO2), nitrogen oxides (NOx), and particulate matter (PM2.5 and PM…
