Penalizing Parenthood and Perpetuating Poverty: Black Parents' Perspectives on Wisconsin's Birth Cost Recovery Policy.

Social safety-net policies and programs are often a double-edged sword—they provide necessary resources, but also punish poor Americans. In Wisconsin, one policy that exemplifies this pattern is Birth Cost Recovery, a state-enforced cost recovery policy through which unmarried, non-custodial fathers are required to repay Medicaid-covered delivery expenses. Supporters assert that Birth Cost Recovery promotes paternal responsibility and increases the sustainability of safety-net programs, but opponents contend it has detrimental effects on families. Yet few studies have explored Birth Cost Recov

Observations and investigation of STR genotype inconsistencies due to mutations, silent alleles, and uniparental disomy in South African populations.

The fundamental concept of parentage and kinship testing is that a child inherits DNA in accordance with the principles of Mendelian inheritance. A child receives half of its DNA from each parent. However, sometimes the observed genotype patterns between a child and its purported parents do not fully align with Mendelian inheritance, which can result in a false conclusion regarding parentage. Mutations, silent alleles, and uniparental disomy (UPD) are examples of events that can produce non-Mendelian results. This study examines STR profiles from South African parentage testing data to assess

Ubiquinol in Fertility and Reproduction: A Conditionally Essential Nutrient for Critical Early-Life Stages.

Background/Objectives: Infertility is a multifactorial condition with an etiopathology that remains largely unclear. Although substantial evidence implicates oxidative stress (OS) as a key contributor to both male and female infertility, targeted strategies for OS-mediated reproductive dysfunction are still not well defined and require further investigation. Ubiquinol is the reduced form of Coenzyme Q10 involved in mitochondrial bioenergetics. It can be synthesized by humans endogenously or provided by dietary sources-typically egg yolks, oily fish, organ meats, and in smaller amounts in nuts

Genomic Imprinting, Epigenetic Dysregulation, and Neuropsychiatric Mechanisms in Prader-Willi Syndrome: A Multi-Level Integrative Review.

Prader-Willi syndrome (PWS) is a rare imprinting-related neurodevelopmental disorder caused by loss of paternally expressed genes within the chromosome 15q11-q13 region, including SNORD116, MAGEL2, and NDN. It provides a natural model for examining how genomic imprinting disruptions shape neural development and psychiatric vulnerability. This review synthesizes current evidence to clarify the mechanistic pathways linking imprinting defects and epigenetic dysregulation to neuropsychiatric outcomes in PWS. Published studies-including patient-derived induced pluripotent stem cell (iPSC) models, a

Joint effects of IVF and cancer history on birth outcomes.

PURPOSE: To examine how live birth rates, preterm birth, and major birth defects are affected by parental cancer history and conception method. METHODS: IVF births were identified by linking the Society for Assisted Reproductive Technology Clinic Outcomes Reporting System to birth certificates in three States. For each IVF-conceived birth, the subsequent 10 naturally conceived births (from birth certificates) created the comparison group. Parental cancer history was identified by linkage to state cancer registries. Preterm birth (PTB) was defined from birth certificates; birth defects (BD) fr

Distinct Involvement of X-Inactivation in Organogenesis.

X-inactivation is the process of dosage compensation to balance X-linked gene expression levels between females (XX) and males (XY). One of the 2 X-chromosomes is randomly selected for inactivation in each cell during embryogenesis and remains inactive throughout life. Therefore, females have 2 types of cells: those with paternal X-chromosome activation and those with maternal X-chromosome activation. However, it remains unclear how X-inactivation is involved in palate and tooth development. Ofd1 is located on the X-chromosome. We found cleft palate in all heterozygous Ofd1 mutant mice (Ofd1fl

Fathers' experiences of caring for children with developmental disabilities.

BACKGROUND: Although caregiving for children with developmental disabilities has been widely examined, fathers' experiences remain relatively underexplored. This qualitative descriptive study explored fathers' caregiving experiences in the context of children with developmental disabilities, focusing on psychological, social, economic, and systemic dimensions of caregiving and fathers' coping and adaptation processes. DESIGN AND METHODS: A qualitative descriptive design was used. Data were collected through semi-structured, in-depth interviews with 19 fathers of children diagnosed with develo

Integrative forensic genomics resolves trace DNA in a major transnational drug trafficking case.

Illicit drug source tracing increasingly combines forensic toxicology, analytical chemistry, and genomics, yet connecting packaging or production to specific individuals remains challenging when touch DNA is limited, degraded, or contaminated. Here, we applied an integrated forensic genomics approach to a major transnational maritime cocaine trafficking case. Touch DNA recovered from multilayer packaging underwent 80 Mb SNP capture targeting autosomal, Y-chromosomal, and mitochondrial loci, with whole-genome sequencing used for comparison. Despite low DNA amounts and varying microbial contamin

Perceived Erectile Dysfunction in a Single Young Male With Multiple Associated Psychological Factors.

Erectile dysfunction (ED) is a common presentation in primary care; while psychogenic ED is well-described globally, cases in sexually inexperienced young men where moral incongruence, pornography-related guilt, and childhood trauma are predominant contributors are rarely documented. ED in young males is increasingly recognized as multifactorial, with biopsychosocial complexity, and it can be assessed even in the absence of partnered sexual activity using masturbation-based evaluation. We present the case of a 24-year-old single male with perceived sexual dysfunction: difficulty in sustaining

Perinatal smoking in mothers and fathers: Longitudinal associations with infant birth outcomes in the Triple B Pregnancy Cohort Study.

BACKGROUND AND AIMS: Maternal smoking in pregnancy is a risk factor for poorer infant health outcomes, yet knowledge on impacts of smoking in the preconception and periconception periods is limited, particularly in fathers. This study examined the association between parental smoking at preconception, periconception and across pregnancy, with infant birth outcomes. METHODS: Pregnant women and fathers from the longitudinal Triple B Pregnancy Cohort Study (1582 mothers; 805 fathers) were assessed across the perinatal period, and infants assessed at birth. Maternal and paternal smoking was asses

Simultaneous Prenatal cfDNA Screening of Aneuploidy, Recessive Single-Gene Conditions, and Fetomaternal Blood Compatibility.

BACKGROUND: The provision of prenatal genetic screening in 3 core clinical contexts-recessive conditions, aneuploidy, and serological incompatibility-involves multiple tests across reproductive partners. This complexity limits utilization and impairs adherence to guideline-recommended care, particularly in carrier screening, where male partners are frequently not tested when a female carrier is identified. Here, we describe the analytical validation of a fetomaternal integrated recessive, serological, and trisomy genetic screen (FIRSTGENE), a single assay that harnesses in silico fragment-leng

Genotype-based management of fertilization and cleavage failure: case-series evidence from DNAH10, DNAH12, CFAP47 , and PLCZ1.

Fertilization and early-cleavage failure are the major causes of assisted reproductive technology failure, but genotype-guided care is underused. We conducted whole-exome sequencing with Sanger confirmation in 20 men from couples with unexplained fertilization and cleavage failure after in vitro fertilization or intracytoplasmic sperm injection (ICSI). Then, further sperm transmission electron microscopy, immunofluorescence, and, where appropriate, embryo quantitative parental contamination test (qPCT) and kinship analysis were integrated in selected cases. Six novel variants were found in fou

A real-time PCR-based noninvasive prenatal RHD screening assay optimized for population-specific RHD allelic spectra in China.

BACKGROUND: Prenatal RhD screening enables risk stratification for RhD-associated hemolytic disease of the fetus and newborn (HDFN) and supports rational,evidence-based use of prophylactic anti-RhD immunoglobulin. However, noninvasive prenatal RhD screening remains challenging in the Chinese population, primarily due to population-specific RHD allelic diversity and the limited adaptability of current assay designs. METHODS: We developed and optimized a real-time PCR-based assay for noninvasive prenatal RHD genotyping, specifically designed to accommodate the spectrum of clinically relevant RH

Allele-specific CRISPR perturbation of the imprinted Dlk1-Dio3 domain reveals regulation of BMP-NOTCH-VEGF signaling in embryonic organogenesis.

The Dlk1-Dio3 imprinted domain is essential for mammalian development, yet its role in coordinating multi-organ embryogenesis is not fully understood. Here we generated four distinct mouse models via CRISPR/Cas9-mediated insertion of a transcriptional termination cassette within the Dlk1-Dio3 domain, encompassing homozygous (HOMO), maternally inherited (MK), paternally inherited (PK), and wild-type alleles (WT), to elucidate parent-of-origin-specific effects. Integrative transcriptomic analyses across multiple scales revealed allele-specific dysregulation of imprinted genes and altered epigene

The presence of microplastics in human semen and the protective role of nicotinamide mononucleotide against polystyrene nanoplastics-induced reproductive and early embryonic toxicity.

Nano(micro)plastics (NMPs) are ubiquitous environmental pollutants that pose significant potential risks to male reproductive health. However, their size-dependent effects on human sperm and the underlying mechanisms remain poorly characterized, and effective interventions are lacking. This study utilized both human and mouse models to evaluate the reproductive hazards of polystyrene nanoplastics (PS-NPs), while also investigating the protective role of nicotinamide mononucleotide (NMN) as a potential intervention. Using laser direct infrared spectroscopy (LDIR), we detected microplastics (MPs

Immune tolerance: Parallels between pregnancy and kidney transplantation.

Pregnancy represents a unique immunological paradox in which the maternal immune system must tolerate the semi-allogeneic fetus expressing paternal antigens. The success of pregnancy depends on a highly coordinated network of immune tolerance mechanisms, encompassing the placental barrier, chemokine-guided leukocyte migration, restricted alloantigen presentation, regulatory T cells, tolerogenic natural killer cells, immune checkpoint molecules, and both hormonal and metabolic adaptations. In contrast, achieving long-term acceptance of transplanted organs-referred to as "operational tolerance"-

Global research trends on the impact of obesity on male infertility: a bibliometric analysis.

BACKGROUND: The parallel rise in global obesity and the decline in male fertility have driven research into their causal relationship. While obesity is recognized as a systemic metabolic disorder, the mechanisms by which it impairs male reproduction remain complex. Tracking progress in this field can inform future research directions. METHODS: On January 10, 2026, a literature search on the topic of Obesity and Male Infertility was conducted using two databases, including Web of Science and Scopus. Key publication information, including complete records and references, was extracted from both

Biallelic DAW1 variants reveal a tissue-specific role in heterotaxy without primary ciliary dyskinesia.

Defects in motile cilia cause a range of disorders, including heterotaxy (HTX), congenital heart disease (CHD), and primary ciliary dyskinesia (PCD). Although these conditions often co-occur, the genetic and mechanistic bases for tissue-specific manifestations remain poorly understood. Here, we identify compound heterozygous variants in DAW1, a dynein arm assembly factor, in a proband with an HTX/laterality defect spectrum, presenting with complex CHD without extracardiac organ malposition and no evidence of PCD. Whole-genome sequencing revealed a maternally inherited canonical splice-site var
