Lumacaftor-Ivacaftor in Pediatric Patients With Cystic Fibrosis and Advanced Liver Disease: A Pilot Study.

PURPOSE: Lumacaftor-ivacaftor (LI) is a cystic fibrosis transmembrane conductance regulator (CFTR) modulator for patients with cystic fibrosis homozygous for the F508del-CFTR variant. All CFTR modulators, including LI, are metabolized in the liver, and their use in advanced cystic fibrosis-related liver disease (ACFRLD) is not recommended due to concerns of worsening liver impairment, despite potential benefits for nutrition and lung function. METHODS: A pilot study was conducted at the Queensland Children's Hospital, Australia, to describe the pharmacokinetics of LI and its metabolites (ivac

Effectiveness of a near-peer teaching model in an otolaryngology bootcamp for medical students: a pilot study.

BACKGROUND: Studies show that near-peer, simulation-based training can help develop technical skills. We sought to examine the effectiveness of near-peer instruction in otolaryngology emergencies by assessing student knowledge and procedural confidence before and after a 2-hour workshop. METHODS: We designed a pilot study for an otolaryngology workshop led by a second-year resident and 2 third-year medical students. The workshop consisted of lectures for anatomy and procedural overview, followed by hands-on simulation stations. Participants completed pre- and post-workshop quizzes and surveys

Comparative Evaluation of APAFIX and 10% Formalin for Human Histologic Tissue Preservation: A Pilot Study.

Formalin has long served as the standard fixative in histopathology; however, its limitations-such as antigen masking, nucleic acid degradation, and carcinogenicity-have prompted the search for safer alternatives. APAFIX, a formalin-free fixative, was developed to maintain diagnostic integrity while improving safety in routine tissue handling. This pilot study was conducted to compare the efficiency of the APAFIX solution with formalin in preserving human histologic tissue architecture and cellular morphology as assessed by routine Hematoxylin and Eosin (H and E) staining. Ten human biopsies f

Influence of brief carbon dioxide inhalation on acute exercise performance and recovery: A pilot study.

Brief inhalation of carbon dioxide (CO2) has been proposed to counteract transient hypocapnia by enhancing oxygen unloading, vasodilation, and sympathetic modulation. Despite these potential ergogenic mechanisms, CO2 has rarely been investigated in the exercise context. The present study investigated whether brief CO2 inhalation (iCO2) influences performance and recovery during high-intensity interval exercise. Nine healthy adults completed two randomized, single-blinded crossover trials of repeated cycling intervals at 85% peak work rate until volitional exhaustion or cadence dropped below 60

Low-Supervision Asynchronously Monitored Home Rehabilitation for Adhesive Capsulitis: A Pilot Randomized Study.

BACKGROUND: Adhesive capsulitis often requires prolonged rehabilitation, and home exercise performed without supervision is difficult to document objectively. This pilot randomized trial evaluated the preliminary clinical effects and system-recorded engagement of a low-supervision, asynchronously monitored home rehabilitation system used as an adjunct to conventional outpatient rehabilitation. METHODS: Thirty adults with adhesive capsulitis were randomly assigned 1:1 at a university-affiliated outpatient rehabilitation department. Both groups received conventional outpatient rehabilitation tw

Phytoscreening the homes of pregnant residents for volatile organic compounds: a pilot study in metro Detroit.

Exposure to anthropogenic volatile organic compounds (VOCs) has been associated with preterm birth, yet the spatial distribution of VOC exposures within urban environments remains poorly characterized, particularly for vapor intrusion pathways. Plants have recently been used as a cost-effective, non-invasive tool to "phytoscreen" for belowground contaminants. Here, we evaluated the use of phytoscreening within a birth cohort in metro Detroit, Michigan, USA by (a) assessing participant willingness to allow sampling in residential front yards, (b) quantifying VOC concentrations across plant tiss

Vulvovaginal Collagen Injection as a Regenerative Strategy in Genitourinary Syndrome of Menopause: Results of a Pilot Study.

Background/Objectives: Genitourinary syndrome of menopause (GSM) is a common and under-diagnosed condition that significantly affects the quality of life of post-menopausal women. Conventional treatments, especially those based on estrogens, have limitations, which has prompted the search for alternative therapies in the field of regenerative medicine. In this context, intradermal injectable collagen, with regenerative and analgesic properties, could represent an innovative option. This study aims to evaluate the efficacy and safety of multipoint intradermal injections of collagen (MD-Tissue)

The effect of bariatric surgery on patient reported outcomes of heart failure in women: a pilot study.

BACKGROUND: Heart failure (HF) with preserved ejection fraction (HFpEF) disproportionately affects women, with obesity serving as a significant risk factor. Metabolic and bariatric surgery (MBS) has been associated with reduced risk of HF development and may reverse cardiac geometry and metabolic abnormalities that contribute to impaired cardiac function through weight loss dependent and independent mechanisms. However, the impact of MBS on early patient reported outcomes of HF in female patients remains understudied. OBJECTIVE: The purpose of this pilot study was to determine the early impac

Closed-loop temporomandibular joint arthrocentesis: pilot of a technique for effective synovial fluid collection.

Temporomandibular joint (TMJ) arthrocentesis is a common minimally invasive method used for both diagnosis and treatment. However, traditional lavage techniques often cause excessive dilution of synovial fluid, reducing the accuracy of subsequent biochemical tests. This technical note introduces a closed-loop arthrocentesis technique aimed at maximizing synovial fluid recovery while maintaining protein levels and sample quality. The procedure employs a closed-loop system that prevents fluid leakage and enables controlled and efficient collection of synovial fluid. In this pilot study, synovial

A soft tissue-sparing positioning guide for the DCIA flap in mandibular reconstruction: a pilot study.

BACKGROUND: Precise placement of bone grafts is critical for functional and aesthetic outcomes in mandibular reconstruction. Conventional positioning guides typically require extensive stripping of periosteum and surrounding soft tissues, which may increase surgical trauma. This pilot study aimed to assess the feasibility and preliminary clinical performance of a novel soft tissue-sparing positioning guide for mandibular reconstruction. METHODS: This single-arm case series feasibility study included five female patients with segmental mandibular defects. Preoperative computed tomography data

Developing and Piloting a Standardized Thermography Protocol to Expedite Rheumatology Referrals for Possible Inflammatory Arthritis.

OBJECTIVE: Inflammatory arthritis, especially rheumatoid arthritis (RA), imposes substantial morbidity that diminishes quality of life and escalates health care costs when left untreated. Timely diagnosis and treatment are pivotal. However, limited access to rheumatologists underscores the importance of triaging referrals based on symptom severity. We developed a protocol for using infrared thermography (IRT) to detect joint inflammation and implemented it in an academic primary care clinic as a screening tool for rheumatology referrals. MATERIALS AND METHODS: We enrolled people with RA, oste

Multichannel Optimization Strategy for Functional Electrical Stimulation in Grasp Gesture Restoration: A Pilot Study.

Grasping difficulties profoundly diminish the quality of life in individuals affected by stroke or spinal cord injury. Multi-channel functional electrical stimulation (FES) enables precise, targeted muscle activation through selectively activated electrodes, offering a promising approach for motor restoration in neurorehabilitation. However, the requirement for gesture-specific calibration limits the scalability of the system to the diverse and highly coordinated hand functions essential for daily living. We present a biophysically informed modeling approach that selects optimal multichannel s
