Cryopreservation of epididymal sperm in tree shrews (Tupaia belangeri).

Cryopreservation of sperm from tree shrews, which are considered primitive primates, would enhance genetic management and breeding programs. Epididymal sperm were surgically harvested from male tree shrews, cryopreserved in two Tes-Tris-based cryodiluents, and used in four experiments. In Experiment 1, there were no significant differences in motility and acrosome integrity among five concentrations of egg yolk in TTE after cooling to 4 °C. However, sperm frozen in TTE containing 20% egg yolk at -172 °C/min had better (P < 0.05) post-thaw motility and acrosome integrity. In Experiment 2, sperm…

Neuronal responses in visual area V2 (V2) of macaque monkeys with strabismic amblyopia.

Amblyopia, a developmental disorder of spatial vision, is thought to result from a cascade of cortical deficits over several processing stages beginning at the primary visual cortex (V1). However, beyond V1, little is known about how cortical development limits the visual performance of amblyopic primates. We quantitatively analyzed the monocular and binocular responses of V1 and V2 neurons in a group of strabismic monkeys exhibiting varying depths of amblyopia. Unlike in V1, the relative effectiveness of the affected eye to drive V2 neurons was drastically reduced in the amblyopic monkeys. Th…

Evolutionary aspects of diet: the omega-6/omega-3 ratio and the brain.

Several sources of information suggest that human beings evolved on a diet that had a ratio of omega-6 to omega-3 fatty acids (FA) of about 1/1; whereas today, Western diets have a ratio of 10/1 to 20-25/1, indicating that Western diets are deficient in omega-3 FA compared with the diet on which humans evolved and their genetic patterns were established. Omega-6 and omega-3 FA are not interconvertible in the human body and are important components of practically all cell membranes. Studies with nonhuman primates and human newborns indicate that docosahexaenoic acid (DHA) is essential for the n…

Mouse models of human T lymphotropic virus type-1-associated adult T-cell leukemia/lymphoma.

Human T-lymphotropic virus type-1 (HTLV-1), the first human retrovirus discovered, is the causative agent of adult T-cell leukemia/lymphoma (ATL) and a number of lymphocyte-mediated inflammatory conditions including HTLV-1-associated myelopathy/tropical spastic paraparesis. Development of animal models to study the pathogenesis of HTLV-1-associated diseases has been problematic. Mechanisms of early infection and cell-to-cell transmission can be studied in rabbits and nonhuman primates, but lesion development and reagents are limited in these species. The mouse provides a cost-effective, highly…

Recombinant Mycobacterium bovis BCG prime-recombinant adenovirus boost vaccination in rhesus monkeys elicits robust polyfunctional simian immunodeficiency virus-specific T-cell responses.

While mycobacteria have been proposed as vaccine vectors because of their persistence and safety, little has been done systematically to optimize their immunogenicity in nonhuman primates. We successfully generated recombinant Mycobacterium bovis BCG (rBCG) expressing simian immunodeficiency virus (SIV) Gag and Pol as multigenic, nonintegrating vectors, but rBCG-expressing SIV Env was unstable. A dose and route determination study in rhesus monkeys revealed that intramuscular administration of rBCG was associated with local reactogenicity, whereas intravenous and intradermal administration of …

Immunization with a synthetic robustoxin derivative lacking disulphide bridges protects against a potentially lethal challenge with funnel-web spider (Atrax robustus) venom.

The venom of male Atrax robustus spiders is potentially lethal to primates. These spiders have been responsible for a number of human deaths. Robustoxin is the lethal toxin in the venom. It is a highly cross-linked polypeptide that has 42 amino acid residues and four disulphide bridges. If these bridges are broken, the resulting polypeptide is non-toxic. Robustoxin was chemically synthesized with all of its eight cysteine residues protected with acetamidomethyl groups in order to avoid formation of disulphide bridges. The resulting derivative was co-polymerized with keyhole limpet haemocyanin.…

Spatial packing, cranial base angulation, and craniofacial shape variation in the mammalian skull: testing a new model using mice.

The hypothesis that variation in craniofacial shape within and among species is influenced by spatial packing has a long history in comparative anatomy, particularly in terms of primates. This study develops and tests three alternative models of spatial packing to address how and to what extent the cranial base angle is influenced by variation in brain and facial size. The models are tested using mouse strains with different mutations affecting craniofacial growth. Although mice have distinctive crania with small brains, long faces, and retroflexed cranial bases, the results of the study indic…

The frequency of lymphocytic gastritis in baboons.

BACKGROUND: In 1985, two independent reports highlighted a novel subtype of chronic inflammation in the gastric mucosa, characterized by the intraepithelial lymphocytic infiltration (ILI) both in the surface and the foveolar epithelium. The disease, subsequently called lymphocytic gastritis (LG) is a rare form of gastritis (0.8%-1.6% of cases), with unclear pathogenesis. More recently, LG was recorded in pigs and in non-human primates. MATERIALS AND METHODS: The frequency of LG (>25 lymphocytes/100 epithelial cells) was assessed in gastric specimens from 92 consecutive baboons, initially file…

Balamuthia amebic encephalitis--California, 1999-2007.

Balamuthia mandrillaris is a free-living ameba that causes encephalitis in humans (both immunocompetent and immunocompromised), horses, dogs, sheep, and nonhuman primates. The ameba is present in soil and likely is transmitted by inhalation of airborne cysts or by direct contamination of a skin lesion. Approximately 150 cases of balamuthiasis have been reported worldwide since recognition of the disease in 1990. Balamuthiasis is difficult to diagnose because 1) the clinical symptoms mimic those of several other types of encephalitis, 2) few laboratories perform appropriate diagnostic testing, …

Assessing flavivirus, lentivirus, and herpesvirus exposure in free-ranging ring-tailed lemurs in southwestern Madagascar.

The ring-tailed lemur (Lemur catta) is an endangered species found in southwestern Madagascar, and understanding infectious disease susceptibility is an essential step towards the preservation of wild and captive lemur populations. Lemurs are primates that are widely dispersed throughout the island of Madagascar and may serve as hosts or reservoirs for zoonotic infections. The aim of this study was to determine the prevalence of antibodies to West Nile virus (WNV), simian immunodeficiency virus (SIV), and herpes simplex virus type 1 (HSV-1) in a population of free-ranging ring-tailed lemur fro…

Influence of perineal swellings on behavior and stress reaction in levonorgestrel implanted Macaca sylvanus females.

OBJECTIVES: Contraceptives are common tools in controlling the reproductive physiology in primates. Levonorgestrel, a synthetic gestagen, can usually enable contraception and does not suppress the extension of perineal swellings. SETTING: This study investigated the effects on intra-sexual behavior and fecal cortisol excretion rates in different swollen Barbary macaque females (N =24) during the non-reproductive phase of the year. The study was conducted on a semi-free population at Affenberg Salem (Germany). RESULTS: The results showed that females with reduced swellings received more aggre…

[Childhood in flux--Part II: Modern times until today].

At the end of the 19th century, the Society for the Prevention of Cruelty to Children was founded in New York, after a barbarous case of neg … ublic awareness arisen. Due to the work of J. Bowlby in the 1950s, it became clear that children of primates need more than air, water and food, namely a relationship between the child and an adult person (attachment). To what degree the basic needs of children are being fulfilled in Western societies today is still a controversial issue.

[The risk of yellow fever in travellers].

Yellow fever is a tropical virus disease characterised by high fever, jaundice, heart and kidney failure, and haemorrhagic diathesis. The causative Flavivirus is endemic in parts of tropical Africa and South America and is transmitted among humans and primates by mosquitoes. The chance that an unvaccinated traveller to West Africa will die of yellow fever is estimated at 1:650 to 1:5000 visitors per month of stay, depending on whether an epidemic occurs. Vaccination with the attenuated yellow fever Asibi 17D virus results in limited virus replication in the body and long-term protection due to…

Animal models for human T-lymphotropic virus type 1 (HTLV-1) infection and transformation.

Over the past 25 years, animal models of human T-lymphotropic virus type 1 (HTLV-1) infection and transformation have provided critical knowledge about viral and host factors in adult T-cell leukemia/lymphoma (ATL). The virus consistently infects rabbits, some non-human primates, and to a lesser extent rats. In addition to providing fundamental concepts in viral transmission and immune responses against HTLV-1 infection, these models have provided new information about the role of viral proteins in carcinogenesis. Mice and rats, in particular immunodeficient strains, are useful models to asses…
