Divergent immunomodulatory properties of milk proteins from Gannan yak, cattle-yak, and Holstein cattle: Insights from a comparative proteomics and murine model study.

While Gannan yak milk and Gannan cattle-yak milk are unique livestock resources in Gansu Province, China, the specific functional properties of their milk proteins remain to be fully elucidated. To address this, we employed tandem mass tag (TMT) quantitative proteomics to analyze the differentially expressed proteins (DEPs) among Gannan yak milk proteins (YMP), Gannan cattle-yak milk proteins (CYMP), and Holstein milk proteins (HMP). A total of 648 proteins were quantitatively analyzed. Compared with HMP, the DEPs in YMP and CYMP were primarily enriched in immune-related pathways such as the C…

The Possible Significance of Proteomics in Understanding Molecular Mechanisms of Progressive Supranuclear Palsy, Corticobasal Degeneration, Multiple System Atrophy, and Dementia with Lewy Bodies.

Atypical Parkinsonisms are a diverse group of diseases associated with multiple pathologies, including synucleinopathies and tauopathies. Atypical Parkinsonisms include progressive supranuclear palsy, corticobasal degeneration, multiple system atrophy, and dementia with Lewy bodies. The examination of these diseases is complicated due to their overlapping clinical manifestations. Hence, tools enabling reliable supplementary assessment of atypical Parkinsonisms are needed. The most common methods involve neuroimaging; however, these evaluations generally involve basic magnetic resonance imaging

[Investigation on antidepressant mechanism of Piper laetispicum extract based on quantitative proteomics and zebrafish model of depression induced by chronic unpredictable mild stress].

This study aimed to evaluate the antidepressant activity of Piper laetispicum extract and preliminarily explore its underlying mechanism using a zebrafish model of depression combined with quantitative proteomics. A chronic unpredictable mild stress(CUMS) paradigm was employed to establish the depression model. AB strain zebrafish were randomly assigned into four groups: control, model, P. laetispicum, and fluoxetine, with 15 fish in each group. Except the control group, all groups were subjected to CUMS for 2 weeks. Intervention of P. laetispicum extract or fluoxetine lasted for 3 consecutive…

Quantitative proteomics analysis of papillary thyroid carcinoma reveals protein S, clusterin, and leucine-rich α-2-glycoprotein 1 as potential prognostic protein biomarkers.

Papillary thyroid carcinoma (PTC) is the most common type of thyroid cancer. The primary challenge is identifying patient subgroups with PTC and choosing the most effective treatment approach. To explore the differentially expressed proteins (DEPs) between high and low recurrent-risk PTCs, we collected 15 tissues comprising high (n = 7) and low (n = 8) recurrent-risk groups from PTC. The samples were detected by tandem mass tag labeling proteomics. Using The Cancer Genome Atlas data on thyroid cancer, prognosis-related DEPs were identified. Furthermore, an immunohistochemistry stain of 53 case…

Inflammation and epithelial-mesenchymal transition in a CFTR-depleted human bronchial epithelial cell line revealed by proteomics and human organ-on-a-chip.

Cystic fibrosis (CF) is a genetic disease caused by mutations in the CF transmembrane conductance regulator (CFTR) gene, leading to chronic, unresolved inflammation of the airways due to uncontrolled recruitment of polymorphonuclear leukocytes (PMNs). Evidence indicates that CFTR loss-of-function, in addition to promoting a pro-inflammatory phenotype, is associated with an increased risk of developing cancer, suggesting that CFTR can exert tumor-suppressor functions. Three-dimensional (3D) in vitro culture models, such as the CF lung airway-on-a-chip, can be suitable for studying PMN recruitme

Proteomics analysis reveals the antagonistic interaction between high CO2 and warming in the adaptation of the marine diatom Thalassiosira weissflogii in future oceans.

While it is known that warming and rising CO2 level might interactively affect the long-term adaptation of marine diatoms, the molecular and physiological mechanisms underlying these interactions in the marine diatom Thalassiosira weissflogii on an evolutionary scale remain largely unexplored. In this study, we investigated the changes in metabolic pathways and physiological responses of T. weissflogii under long-term ocean acidification and/or warming conditions (∼3.5 years), integrating proteomics analyses and physiological measurements. Our findings reveal that proteins involved in central …

Simple method for isolation and culture of primary buffalo (Bubalus bubalis) endometrial epithelial cells (pBuEECs) and its characterization using high throughput proteomics approach.

Early embryonic mortality resulting from insufficient interaction between the embryo and the uterus leads to the failure of pregnancy in livestock animals. Thus, it is imperative to comprehend the multifaceted process of implantation at molecular levels, which requires synchronized feto-maternal interaction. The in-vitro models serve as valuable tools to investigate the specific stages of implantation. The present study was undertaken to develop a simple method to isolate and culture the primary buffalo endometrial epithelial cells (pBuEECs), followed by proteome profiling of the proliferating

Plasma proteomics analysis of Chinese HIV-1 infected individuals focusing on the immune and inflammatory factors afford insight into the viral control mechanism.

BACKGROUND: Long-term non-progressors (LTNPs) with HIV infection can naturally control viral replication for up to a decade without antiretroviral therapy (ART), but the underlying mechanisms of this phenomenon remain elusive. METHODS: To investigate the relevant immune and inflammatory factors associated with this natural control mechanism, we collected plasma samples from 16 LTNPs, 14 untreated viral progressors (VPs), 17 successfully ART-treated patients (TPs), and 16 healthy controls (HCs). The OLINK immune response panel and inflammation panel were employed to detect critical proteins, a

Integration of proteomics with prospective birth cohort to elucidate early life origins of cardiometabolic diseases: rationale, study design, lab assay, and quality control.

There is growing evidence that the plasma proteome provides insights into personal health status at different stages of life. However, limited data are available on high-throughput proteomic studies in pediatric populations, especially, using prospective birth cohorts. We launched a proteomics study in 990 children from a US predominantly urban, low-income, multi-ethnic prospective Boston Birth Cohort (BBC, referred as "BBC proteomics study"), which aimed to leverage proteomics to investigate the biological pathways underlying the link between preterm birth and child long-term cardiometabolic …

Differential proteomics of circulating extracellular vesicles of placental origin isolated from women with early-onset preeclampsia reveal aberrant innate immune and hemostasis processes.

PROBLEM: Early-onset preeclampsia (EOPE) is a severe gestational hypertensive disorder with significant feto-maternal morbidity and mortality due to uteroplacental insufficiency. Circulating extracellular vesicles of placental origin (EV-P) are known to be involved in the pathophysiology of EOPE and might serve as an ideal reservoir for its specific biomarkers. Therefore, we aimed to characterize and perform comparative proteomics of circulating EV-P from healthy pregnant and EOPE women before delivery. METHOD OF STUDY: The EV-P from both groups were isolated using immunoaffinity and were cha…
