p16High immune cells control disease tolerance as a defense and health span-extending strategy.

Host survival during infection has traditionally been attributed to pathogen clearance, yet increasing evidence supports a complementary mechanism known as disease tolerance, which limits tissue damage without directly affecting pathogen burden. Here, we identify p16High immune cells as critical mediators of disease tolerance. We show that the FDA-approved BNT162b2 mRNA COVID-19 vaccine rapidly induces p16High immune subsets in mice and humans. These cells are required for protection against lipopolysaccharide-induced endotoxin shock, bacterial sepsis, and ionizing irradiation. Mechanistically

When Neutrophils Go Rogue: Dysregulated Inflammation in Airway Diseases and Implications for Therapy.

Failure to resolve the neutrophilic inflammation during the innate immune response results in neutrophil accumulation in the airways. These cells exhibit prolonged survival, increased necrosis, and impaired phagocytic capacity. Paradoxically, despite reduced pathogen-clearing ability, neutrophils release serine proteases that drive the destruction of airway tissue. Their persistence in the lung chronically promotes a cycle of tissue damage, remodeling, and inflammation, leading to progressive pulmonary dysfunction. Recent discoveries in neutrophil dysregulation open up the possibility of devel

Total ear canal ablation for the treatment of auricular hemangiosarcoma in a cat: case report.

Ear neoplasms are growths that commonly affect light-coated, unpigmented felines exposed to prolonged sunlight. Another factor associated with these growths is the constant tissue trauma caused by intense pruritus in cases of chronic otitis. Any tissue making up the structure of the pinna can undergo changes and initiate pathogenic cell proliferation. In this context, cutaneous hemangiosarcomas may arise from alterations in the ear's vascular endothelium, albeit less frequently. This report describes the case of a 12-year-old, white, mixed-breed cat with a history of prior pinnectomy and local

FluxAtlas is a pan-cancer metabolic flux atlas that identifies tumor-selective dependencies and prognostic biomarkers.

Metabolic reprogramming is a hallmark of cancer, yet dynamic metabolic flux has been difficult to study systematically. Here, we present FluxAtlas, a pan-cancer atlas of metabolic flux generated from genome-scale metabolic modeling of over 10,000 tumors across 28 The Cancer Genome Atlas (TCGA) cancer types. By integrating enzyme constraints and nutrient diffusion limits, we reveal conserved and tissue-specific metabolic rewiring, including alterations in bile acid recycling, urea metabolism, and amino acid biosynthesis. We identify a bile acid-associated program that remodels glutathione homeo

A tumor profiling resource for ovarian cancer: insights into chemotherapy-driven heterogeneity and personalized treatment strategy.

In women with high-grade serous ovarian cancer, chemotherapy remains the primary standard treatment, despite growing recognition of the disease as highly heterogeneous. Here, we examine the feasibility and clinical utility of comprehensive multimodal molecular profiling to inform treatment decisions. We analyze blood, single-cell and bulk tumor tissue, and malignant ascites using up to eleven technologies (DNA, RNA, protein, and functional assays) within a four-week turnaround time. Hypothetical treatment recommendations are altered for 76% of patients, and multi-omics-guided maintenance thera

Anatomical dynamics define cancer cachexia subtypes and identify systemic inflammation as a marker of lethal wasting.

Cancer cachexia is a wasting syndrome that remodels the anatomy of the patient. How this remodeling unfolds across tissues, whether it defines distinct disease states, and how these states relate to underlying biology remain unknown. We used longitudinal computed tomography imaging from 4,516 patients to quantify evolution of muscle, adipose, and organs during cachexia. Across two independent institutional cohorts, unsupervised analysis identified three reproducible anatomical subtypes of cachexia, including an inflammatory Type A marked by progressive hepatosplenic enlargement and inferior su

Low-density lipoprotein cholesterol drives multinucleated giant cell formation in response to mycobacterium bovis Bacille Calmette-Guérin.

Macrophages play a central role in tuberculosis, both in bacterial persistence and tissue pathology. Body weight and a dysregulated lipid metabolism profoundly affect disease progression and survival. Yet, the relationship between lipid metabolism and mycobacterial immunity is still poorly understood. Accordingly, this study investigated the influence of cholesterol and lipoproteins on macrophage responses to mycobacteria, in particular the formation of multinucleated giant cells (MGC), which are key components of granulomas and involved in the containment of mycobacteria. We found that low-de

Partial amputation regrowth of P4 distal digit in an eastern grey kangaroo (Macropus giganteus): a case report.

This case report describes distal regrowth of the fourth hind digit (P4) following traumatic autoamputation in a young adult eastern grey kangaroo (Macropus giganteus). Injury to P4 is generally considered to carry a poor prognosis due to its critical role in weight-bearing and propulsion during hopping. Following loss of the distal phalanx, the nail bed remained intact and healing progressed without surgical intervention. Sequential observations documented epithelialisation, preservation of the nail bed and subsequent formation of a keratinised nail structure. The animal maintained normal loc

Renal-clearable fluorescent probe for early detection of pancreatic cancer in living murine models.

Pancreatic cancer (PC) is a highly aggressive and lethal malignancy with extremely poor patient survival rates. The lack of biomarkers and deep tissue detection barriers has presented long-standing and unsolved issues, severely hindering the early diagnosis of PC even in living rodent models. Here, we report a renal-clearable leucine aminopeptidase (LAP)-activatable fluorescence probe, overturning the probe's pharmacokinetics from the liver to kidney pathway (90% of the injection dosage). This strategy reduces interference in the liver and enables LAP as a biomarker for PC detection in murine

Role of Açaí (Euterpe oleracea) in Modulating the Immune Response During Experimental Oral Infection with Trypanosoma cruzi.

Oral infection is now the main route of Chagas disease transmission in endemic countries, with açaí (Euterpe oleracea) being the primary food involved in Brazil. However, the role of açaí in parasite-host interaction remains largely unexplored. This study assessed the effect of açaí inoculum on experimental Trypanosoma cruzi infection. BALB/c mice were orally infected with metacyclic trypomastigotes in RPMI or açaí. No difference in survival was observed. Tissue parasite load showed higher gastric parasitism in the RPMI group on day 5 after infection. Proteomic analysis of the açaí group revea

Multi-Omics and -Organ Insights into Energy Metabolic Adaptations in Early Sepsis Onset.

Systemic metabolic dysregulation in sepsis critically impacts patient survival. To better understand its onset, untargeted serum metabolomics and lipidomics are analyzed from 152 presymptomatic patients undergoing major elective surgery, and identified key metabolites, including serine and aminoadipic acid, that differentiate postoperative uncomplicated infection from sepsis. Using single-nucleus RNA sequencing data from an in vivo mouse model of sepsis, tissue-independent down-regulation and tissue-specific differences of serine and energy-related genes including key module roles for the mito

Infectious bacteria, but not the microbiota, induce a NOX-ATM-cytokine pathway that controls epithelial turnover.

While the gut microbiota is essential for maintaining intestinal homeostasis, infection with pathogenic microbes induces epithelial stress and accelerates tissue renewal. Cytokine production and intestinal stem cell (ISC) activity are coordinated by stress-responsive pathways that link damage to inflammation and repair. In this study, we demonstrate that pathogenic microbes, but not innocuous microbiota, trigger DNA damage response characterized by an atypical accumulation of γH2av in aged enterocytes. γH2av accumulation occurred without detectable DNA breaks and required both the Ataxia Telan

Multidisciplinary treatment for primary cardiac angiosarcoma: a case report and overview of the literature.

BACKGROUND: Primary cardiac angiosarcoma is rare and highly invasive to myocardial tissue and prone to metastasis. Surgery is an effective treatment, but it is difficult and risky. Postoperative therapy could improve survival rate, but specific regimens are controversial. CASE PRESENTATION: We herein demonstrated the management experience of a patient with primary cardiac angiosarcoma at 19 weeks of pregnancy, which can be used as a reference for other patients. She underwent surgical R1 resection during pregnancy, but the fetus aborted spontaneously 4 h after surgery. Under the care of our mu

Synergistic Effect of 5-Fluorouracil and Amphotericin B in Murine Paracoccidioidomycosis: Immune Modulation and Enhanced Fungal Clearance.

Paracoccidioidomycosis (PCM) is a systemic fungal infection that primarily affects the lungs. Previous studies have shown that 5-fluorouracil (5-FU), a chemotherapeutic agent, reduces pulmonary myeloid-derived suppressor cells (MDSCs), thereby stimulating immune responses in PCM. This study aimed to evaluate the efficacy of the combined 5-FU and AmB therapy in a murine model of PCM. C57BL/6 mice were infected with Paracoccidioides brasiliensis and treated with AmB and/or 5-FU. We found that the 5-FU and AmB combination therapy led to improved disease control, as evidenced by reduced fungal bur

Dehydrated Human Amnion-Chorion Membrane as a Bioactive Scaffold for Dental Pulp Tissue Regeneration.

The dehydrated human amnion-chorion membranes (dHACMs) derived from the human placenta have emerged as a promising biomaterial for dental pulp regeneration owing to their unique biological and structural properties. The purpose of this review is to explore the potentials of dHACMs in dental pulp tissue engineering, focusing on their ability to promote cellular proliferation, differentiation, angiogenesis, and neurogenesis. dHACMs are rich in extracellular matrix proteins and growth factors such as TGF-β1, FGF2, and VEGF. They also exhibit significant anti-inflammatory and antimicrobial propert

Leptin haploinsufficiency exerts sex-dependent partial protection in SOD1G93A mice by reducing inflammatory pathways in the adipose tissue.

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disorder characterized by significant metabolic disruptions, including weight loss and hypermetabolism in both patients and animal models. Leptin, an adipose-derived hormone, displays altered levels in ALS. Genetically reducing leptin levels (Lepob/+) to maintain body weight improved motor performance and extended survival in female SOD1G93A mice, although the exact molecular mechanisms behind these effects remain elusive. Here, we corroborated the sexual dimorphism in circulating leptin levels in ALS patients and in SOD1G93A mic

NELL1 variant protein (NV1) modulates hyper-inflammation, Th-1 mediated immune response, and the HIF-1α hypoxia pathway to promote healing in viral-induced lung injury.

Research underscores the urgent need for technological innovations to treat lung tissue damage from viral infections and the lasting impact of COVID-19. Our study demonstrates the effectiveness of recombinant human NV1 protein in promoting a pro-healing extracellular matrix that regulates homeostasis in response to excessive tissue reactions caused by infection and injury. NV1 achieves this by calibrating multiple biological mechanisms, including reducing hyperinflammatory cytokine levels (e.g., IFN-γ, TNF-α, IL-10, and IP-10), enhancing the production of proteins involved in viral inactivatio

Enhancing 7-dehydrocholesterol suppresses brain ferroptosis and tissue injury after neonatal hypoxia-ischemia.

Neonatal hypoxic-ischemic brain injury (HIBI) results in part from excess reactive oxygen species and iron-dependent lipid peroxidation (i.e. ferroptosis). The vitamin D precursor 7-dehydrocholesterol (7-DHC) may inhibit iron-dependent lipid peroxidation. Primary neurons underwent oxygen and glucose deprivation (OGD) injury and treatment with 7-DHC-elevating medications such as cariprazine (CAR) or vehicle. Postnatal day 9 mice underwent sham surgery or carotid artery ligation and hypoxia and received intraperitoneal CAR. In neurons, CAR administration resulted in significantly increased cell

Regulatory mechanisms governing GLI proteins in hedgehog signaling.

The Hedgehog (Hh) signaling pathway is critical for regulating cell growth, survival, fate determination, and the overall patterning of both vertebrate and invertebrate body plans. Aberrations in Hh signaling are associated with congenital abnormalities and tumorigenesis. In vertebrates, Hh signaling depends uniquely on primary cilia, microtubule-based organelles that extend from the cell surface. Over the last 2 decades, studies have demonstrated that key molecules regulating Hh signaling dynamically accumulate in primary cilia via intraflagellar transport systems. Moreover, through the prima
