Impact of recurrent laryngeal nerve lesion on oropharyngeal muscle activity and sensorimotor integration in an infant pig model.

The successful performance of a swallow requires dynamic integration between a wide range of sensory inputs and muscle activities to produce … pharyngeal swallow. Taken with previous biomechanical results, these patterns suggest an intricate brain stem sensorimotor integration that occurs as part of a swallow. In particular, the execution of oral motor function is changed as a result of this simple lesion. NEW & NOTEWORTHY Damage to the recurrent laryngeal nerve compromises swallowing despite an a …

PET and MRI detection of early and progressive neurodegeneration in spinocerebellar ataxia type 36.

BACKGROUND: The spinocerebellar ataxias (SCAs) form a clinically, genetically, and pathological heterogeneous group of autosomal-dominant de … ellar hemisphere. In the ataxic stage, hypometabolism spread to both cerebellar hemispheres and the brain stem. MRI was normal in asymptomatic and preataxic individuals and showed superior cerebellar vermis atrophy early in the ataxic stage, diffuse cerebellar atrophy some years into the disease course, and a pattern of olivopontocerebellar atrophy in the ol …

ATXN2-AS, a gene antisense to ATXN2, is associated with spinocerebellar ataxia type 2 and amyotrophic lateral sclerosis.

OBJECTIVE: Spinocerebellar ataxia type 2 (SCA2) is a neurodegenerative disease caused by a CAG repeat expansion in the gene ataxin-2 (ATXN2). ATXN2 intermediate-length CAG expansions were identified as a risk factor for amyotrophic lateral sclerosis (ALS). The ATXN2 CAG repeat is translated into polyglutamine, and SCA2 pathogenesis has been thought to derive from ATXN2 protein containing an expanded polyglutamine tract. However, recent evidence of bidirectional transcription at multiple CAG/CTG disease loci has led us to test whether additional mechanisms of pathogenesis may contribute to SCA2

Therapeutic vaccination against autologous cancer stem cells with mRNA-transfected dendritic cells in patients with glioblastoma.

BACKGROUND: The growth and recurrence of several cancers appear to be driven by a population of cancer stem cells (CSCs). Glioblastoma, the most common primary brain tumor, is invariably fatal, with a median survival of approximately 1 year. Although experimental data have suggested the importance of CSCs, few data exist regarding the potential relevance and importance of these cells in a clinical setting. METHODS: We here present the first seven patients treated with a dendritic cell (DC)-based vaccine targeting CSCs in a solid tumor. Brain tumor biopsies were dissociated into single-cell su

Long-term MR follow-up of cerebral lesions in neuro-Behçet's disease.

To study the long-term evolution of cerebral lesions in neuro-Behçet's disease, MRI was carried out on 12 patients, with follow-up from 1.5 to 6 years (mean 3.5 years). On the first MRI, 66 lesions in all were found; each patient had 1-10 lesions (mean 5.5). There were 30 (46%) lesions in the brain stem, 18 (27%) in the basal ganglia region and 18 (27%) in the periventricular white matter. Of these 22 (33%) were small, 31 (47%) medium-size and 13 (20%) large lesions. On the last MRI, 60 lesions were found: each patient had 1-10 lesions (mean 5). At this time 31 lesions (52%) were in the brain …

Short- and long-term results of microvascular anastomosis in the vertebrobasilar system, a critical analysis.

During the past five years, 22 patients were operated on and 21 occipital artery-caudal, loop-posterior, inferior cerebellar artery anastomoses were completed. All but one patient had suffered either TIAs of the vertebrobasilar system or brain stem stroke with complete or partial recovery. All the patients had significant atherosclerotic occlusive disease of both vertebral arteries and bilateral hypoplasia of the posterior communicating arteries. Three patients died shortly after surgery because of brain stem infarction and myocardial infarction. In one of these patients no anastomosis was per…

3D organoids containing endothelial and neural cells generation by serial inductions of differentiation on human iPSC-derived embryoid bodies.

3D brain organoids have been widely used as a tool to study human brain development and disorders. Although endothelial cells play important roles in the brain development and pathogenesis in neurological disorders, most 3D brain organoids lack inherent endothelial cells and need either the addition of endothelial cells or to be transplanted to animals to reconstitute such vascular structures, likely missing the developmental interactions of endothelial cells and other cells in the human brain. In order to reconstitute a 3D organoid mimicking the in vivo neural and endothelial cells developmen

Static or microfluidic? A comparative pilot study of an in vivo-in vitro validated human iPSC-derived blood-brain barrier model for neurotoxicity hazard assessment.

Blood-brain barrier (BBB) permeation determines human brain exposure to chemicals and is therefore a prerequisite for neurotoxicity. However, the lack of toxicokinetic BBB models remains a critical uncertainty in regulatory in vitro risk assessment, which was highlighted in the new EU roadmap towards phasing out animal testing. Human induced pluripotent stem cell (hiPSC)-derived BBB models combined with clinical positron emission tomography (PET) data provide translational benchmarks for evaluating whether in vitro permeability data predict human brain exposure. Microphysiological systems (MPS

Melatonin Promotes Neurogenesis via the JAK2/STAT3 Pathway in Hypoxic-Ischemic Neonatal Rats.

Neonatal hypoxic-ischemic brain damage (HIBD) is a leading cause of neurological dysfunction and long-term disability in newborns. Lactate accumulation and metabolic disturbances after brain injury inhibit neurogenesis, while the restorative capacity of endogenous neural stem cells (NSCs) is essential for neural reconstruction. Melatonin (Mel) alleviates neonatal brain injury, but its effects on NSCs proliferation and migration remain unclear, and the visualization methods for dynamic monitoring of metabolic changes are inadequate. In this study, a neonatal rat model of HIBD was established, a

Vascularized glioblastoma-on-a-chip reveals microglia-mediated regulation of tumor invasion and stemness.

Glioblastoma multiforme (GBM) is the most aggressive primary brain tumor in adults, characterized by rapid progression and poor prognosis. Central to GBM pathobiology are glioma stem cells (GSCs), which preferentially localize within the perivascular niche (PVN) and sustain tumor growth, invasion, and therapeutic resistance through dynamic interactions with the surrounding PVN microenvironment. Notably, microglia (MG), the brain-resident immune cells, represent a substantial fraction of the GBM PVN and play a pivotal role in establishing an immunosuppressive tumor microenvironment (TME). Targeting DNA Repair and Survival Signaling in Diffuse Intrinsic Pontine Gliomas to Prevent Tumor Recurrence.

Therapeutic resistance remains a major obstacle to successful clinical management of diffuse intrinsic pontine glioma (DIPG), a high-grade pediatric tumor of the brain stem. In nearly all patients, available therapies fail to prevent progression. Innovative combinatorial therapies that penetrate the blood-brain barrier and lead to long-term control of tumor growth are desperately needed. We identified mechanisms of resistance to radiotherapy, the standard of care for DIPG. On the basis of these findings, we rationally designed a brain-penetrant small molecule, MTX-241F, that is a highly select…

Three-dimensional modeling of human neurodegeneration: brain organoids coming of age.

The prevalence of dementia and other neurodegenerative diseases is rapidly increasing in aging nations. These relentless and progressive diseases remain largely without disease-modifying treatments despite decades of research and investments. It is becoming clear that traditional two-dimensional culture and animal model systems, while providing valuable insights on the major pathophysiological pathways associated with these diseases, have not translated well to patients' bedside. Fortunately, the advent of induced-pluripotent stem cells and three-dimensional cell culture now provide tools that

Multicellular Crosstalk Between Exosomes and the Neurovascular Unit After Cerebral Ischemia. Therapeutic Implications.

Restorative strategies after stroke are focused on the remodeling of cerebral endothelial cells and brain parenchymal cells. The latter, i.e., neurons, neural precursor cells and glial cells, synergistically interact with endothelial cells in the ischemic brain, providing a neurovascular unit (NVU) remodeling that can be used as target for stroke therapies. Intercellular communication and signaling within the NVU, the multicellular brain-vessel-blood interface, including its highly selective blood-brain barrier, are fundamental to the central nervous system homeostasis and function. Effects of human umbilical cord blood CD34+ cell transplantation in neonatal hypoxic-ischemia rat model.

Perinatal brain injury can cause death in the neonatal period and lifelong neurodevelopmental deficits. Stem cell transplantation had been proved to be effective approach to ameliorate neurological deficits after brain damage. In this study we examine the effect of human umbilical cord blood CD34+ cells on model of neonatal rat hypoxic-ischemic brain damage and compared the neuroprotection of transplantation of CD34+ cells to mononuclear cells from which CD34+ cells isolated on neonatal hypoxic-ischemia rat model.
