Cyflumetofen disrupts the blood-brain barrier and induces neurotoxicity in zebrafish larvae.

Cyflumetofen is a novel non-systemic acylacetonitrile miticide widely used in agriculture to control various harmful mites. However, its potential safety risks to the nervous system of non-target organisms require further evaluation. Cyflumetofen induces developmental toxicity in zebrafish larvae, manifested as pericardial edema, yolk sac enlargement, and reduced body length. To evaluate the impact on the nervous system, cyflumetofen was found to damage neuronal and motor neuron cells in zebrafish larvae, reduce locomotor activity, and alter brain tissue structure. Additionally, cyflumetofen i

Structural basis of directionality control in large serine integrases.

Large serine integrases catalyze unidirectional insertion of large DNA payloads and, in the presence of a cognate recombination directionality factor (RDF), catalyze unidirectional excision. Understanding how directionality is enforced has been hampered by a lack of structural information. Here, we report 8 structures of SPβ integrase-DNA complexes along the integrative (-RDF) and excisive (+RDF) reaction pathways at resolutions extending to 3.15 Å. These include tetrameric intermediates before and after strand exchange and product-bound dimers for both pathways. We find that recombination-ind

Primary entry route of polycyclic aromatic hydrocarbons into earthworms in contaminated soil: oral ingestion or dermal absorption?

Soil contamination by polycyclic aromatic hydrocarbons (PAHs), which are carcinogenic, mutagenic, and toxic, poses significant ecological risks. Earthworms, the key bioindicator in terrestrial ecosystems, are critically exposed to these pollutants through multiple pathways. However, the dominant route of PAH uptake-whether dermal or oral-remains poorly quantified and visually validated, limiting accurate risk assessment and mechanistic understanding. The study identified dermal absorption as the predominant pathway for phenanthrene (a model PAH) uptake by earthworms (Eisenia fetida), accountin

Activation of a Yap1-mesenchymal-like program in β-cells during diabetes.

A critical driver of type 2 diabetes is the failure of insulin-secreting β cells due to dysfunction and loss of cell number or identity. The signaling pathways that trigger these events remain unclear. Here, we identify an epithelial-to-mesenchymal (EMT)-like program that is activated in β cells to promote their loss with type 2 diabetes. This program is initiated in human and mouse β cells by the EMT transcription factor Yap1. While this protein is normally not present in β cells, Yap1 becomes upregulated in response to oxidative stress and impaired mitochondrial function. Chemical inhibition

Multifaceted performances of α-synuclein in health and neurological diseases.

The misfolding and accumulating α-synuclein (αSyn) is a central pathological hallmark of various neurodegenerative diseases, including Parkinson's disease, dementia with Lewy bodies, and multiple system atrophy. Under physiological conditions, αSyn essential for normal synaptic functions primarily through its regulation of synaptic vesicle trafficking, clustering, and neurotransmitter release. However, in disease states, the accumulation of pathological αSyn disrupts intracellular proteostasis, by impairing the ubiquitin-proteasome system, autophagy-lysosomal degradation pathway and endo-lysos

TWEAK-induced alternative NF-κB signaling regulates Cxcl10 and Mymx to promote myoblast-to-myotube fusion and muscle regeneration.

Myoblast fusion is essential for skeletal muscle growth and repair, yet the upstream signals regulating this process remain incompletely understood. Here, we show that the cytokine TWEAK promotes myotube formation through activation of alternative NF-κB signaling. Transcriptomic profiling of TWEAK-treated myotubes reveals upregulation of Myomixer (Mymx) and the chemokine CXCL10, both critical for fusion. Mechanistically, the NF-κB member RelB directly binds the Mymx and Cxcl10 gene promoters, enhancing their transcription. Genetic or pharmacological disruption of this pathway impairs myoblast

Precision Medicine in Dermatology: MEK Inhibition and MAPK Pathway Modulation for Congenital Melanocytic Nevi.

Congenital melanocytic nevi (CMN) are pigmented skin lesions present at birth with a variable risk of developing into melanoma over time. The mitogen-activated protein kinase signaling (MAPK) pathway plays a critical role in the development and progression of CMN, making it a key target for therapeutic interventions. Precision medicine aims to optimize therapeutic efficacy while minimizing adverse effects by tailoring treatments to individuals' genetic and molecular profiles. This literature review examines the potential of precision medicine in managing CMN, with a specific focus on MEK inhib

Navigating the Continuum: System-Level Requirements for High-Quality Dementia Care Transitions.

The worldwide rise in dementia cases is creating significant pressures on healthcare systems, clinicians, and decision-makers. As dementia does not follow a single predictable pathway, individuals commonly move across different care settings as their needs evolve, requiring coordinated and integrated responses across the healthcare system rather than isolated efforts within single services. This perspective study explores service quality and underlying structural challenges throughout the dementia care pathway with a particular concentration on transitions between various stages. Key barriers

Metabolic reprogramming drives pancreatic β cell neogenesis from α cells.

Loss of functional β cells is a hallmark of diabetes, and restoring β cell mass remains a critical goal in the quest for a specific therapy. One potential strategy is to convert non-β cells in the islet, such as α cells, into insulin-producing cells. Although several compounds have been identified to induce β cell-like features in α cells, none have been successfully translated into clinical applications. In this study, we identify PRC2 inhibitors as potent inducers of β cell-enriched gene expression in α cells, acting through modulation of the AR-ETV1 complex. AR inhibition suppresses glycoge

An updated overview of alkaloids for the prevention and treatment of metabolic dysfunction-associated steatotic liver disease.

Metabolic dysfunction-associated steatotic liver disease (MASLD) has emerged as an increasingly formidable global public health challenge, characterized by a complex pathogenesis and a persistent scarcity of reliable and effective therapeutic interventions. In recent years, the multiple-hit hypothesis has provided novel mechanistic insights into the pathogenesis and progression of MASLD, paving the way for identifying viable therapeutic strategies. Due to their characteristic multi-target and multi-pathway regulatory profiles, natural products align closely with the core tenets of the multiple

A feedback mechanism from prostate cancer cells to macrophages, reinforced by STAT1, regulates tumor progression and resistance to radiotherapy.

The resistance to radiotherapy of prostate cancer is driven by interactions within the tumor microenvironment, particularly between prostate cancer cells and tumor-associated macrophages, however the underlying mechanisms remain poorly understood. In this study, we found that STAT1 enhanced the transcription of critical glycolytic enzymes, leading to an increase in lactate secretion from prostate cancer cells. Then, the lactate was transported to macrophages via the MCT1 transporter, activating the NFκB1 pathway, which subsequently promoted macrophage polarization to the M2 phenotype and activ

Glutathione depletion activates cGAS-STING signaling via oxidative stress in preeclampsia.

Oxidative stress can cause double-strand breaks in DNA in patients with preeclampsia (PE), but whether the cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway is involved in PE remains unclear. Here, we show that glutathione (GSH) metabolism is disrupted in placentas from PE patients, leading to increased oxidative stress and activation of the cGAS-STING pathway. Using metabolomics and and CRISPR-Cas9 generated cGAS/STING knockout human trophoblast cells, we found that GSH depletion elevate reactive oxygen species (ROS) levels in HTR-8/SVneo cells, leading to cellular

Primary cilia-extracellular vesicle crosstalk in Alzheimer's disease: Emerging mechanisms and biomarker potential.

Alzheimer's disease (AD) is a neurodegenerative condition marked by cognitive decline and synaptic issues. Recent studies show primary cilia (PCs), sensory organelles present on the surface of most mammalian cells, act as a critical regulators of brain homeostasis and signaling. PCs act as a signaling hubs for pathways like G protein-coupled receptor, Hedgehog, Wnt, and neuroinflammation. When PCs are depleted or dysfunctional, they impair the processing of amyloid precursor protein, tau phosphorylation, synaptic signaling, and neuron-glia communication, leading to tau tangles, neuroinflammati

Vagus nerve is essential for the anxiolytic effect of YL-IPA08, a potent ligand for the translocator protein (18 kDa).

The vagus nerve serves as a key neural pathway modulating parasympathetic activity, which plays a critical role in mood regulation and has been implicated in anxiety and depression. The translocator protein (18 kDa, TSPO) ligand YL-IPA08 has demonstrated consistent anxiolytic effects in animal studies. Using lipopolysaccharide (LPS)- or cefaclor-induced anxiety models in adult mice with or without vagotomy, we investigated the involvement of the vagus nerve in mediating the effects of YL-IPA08 and its underlying mechanisms. As anticipated, chronic YL-IPA08 administration significantly reversed

Evolutionary Remodeling of Ubiquinone Biosynthesis in Toxoplasma gondii Reveals an Essential Bi-functional Monooxygenase.

Apicomplexan parasites like Toxoplasma gondii harbor a highly divergent mitochondrial proteome, much of which remains uncharacterized despite its essentiality for parasite survival. One such critical pathway is ubiquinone (UQ) biosynthesis. Here, we characterize the UQ synthesis machinery in T. gondii and show that conserved enzymes, TgCoq3 and TgCoq5, are essential for growth and mitochondrial function, forming a multi-protein complex. Using proximity labeling and subcellular fractionation, a strategy suited for detecting proteins of low abundance, we identify TgCoqFAD, a unique FAD-dependent

Metabolic atlas of early human cortex reveals glycolytic remodeling and pentose phosphate pathway control of cell fate transitions.

Cortical development involves rapid progenitor expansion and cell diversification supported by tightly regulated metabolic programs, yet these programs remain largely uncharacterized in human development. Here, we generated a metabolic atlas of the early human cortex using primary tissue and stem cell-derived cortical organoids. We observed dynamic changes in core metabolic functions, including an unexpected increase in glycolysis and pentose phosphate pathway (PPP) activity during late neurogenesis. Manipulation of glucose availability in cortical organoids altered cell-type composition, incr
