Angiogenesis in chronic myeloproliferative diseases.

Increased angiogenic activity has been demonstrated in myelofibrosis with myeloid metaplasia (MMM), chronic myeloid leukemia (CML), and essential thrombocythemia (ET) by both bone marrow microvessel density evaluation and measurement of circulating angiogenic factors. MMM is probably the disease with the more pronounced angiogenesis among myeloproliferative disorders but the significance of this finding remains speculative since the angiogenic activity is not correlated with any of the clinical and laboratory features of the disease. Circulating serum levels of angiogenic factors such as vascu

(31)P NMR of apicomplexans and the effects of risedronate on Cryptosporidium parvum growth.

High-resolution 303.6 MHz (31)P NMR spectra have been obtained of perchloric acid extracts of Plasmodium berghei trophozoites, Toxoplasma gondii tachyzoites, and Cryptosporidium parvum oocysts. Essentially complete resonance assignments have been made based on chemical shifts and by coaddition of authentic reference compounds. Signals corresponding to inorganic pyrophosphate were detected in all three species. In T. gondii and C. parvum, additional resonances were observed corresponding to linear triphosphate as well as longer chain polyphosphates. Spectra of P. berghei and T. gondii also indi

Treatment of acute gastroesophageal variceal hemorrhage.

About 30% to 35% of patients with portal hypertension bleed from gastroesophageal varices and mortality remains high reflecting the challenges of effectively dealing with the bleeding event itself and the problems of underlying liver disease. Careful resuscitation and control of risk of complications is the most essential element of medical therapy (Fig. 2). Use of newer, more effective drug combinations with vasopressin or somatostatin permit control of hemorrhage in the majority of patients with fewer drug-induced complications. Endoscopic sclerotherapy and, more recently, banding therapy pr

Enalapril. A reappraisal of its pharmacology and therapeutic use in hypertension.

Enalapril, an angiotensin converting enzyme (ACE) inhibitor usually administered orally once daily, decreases blood pressure by lowering peripheral vascular resistance without increasing heart rate or output. It is effective in lowering blood pressure in all grades of essential and renovascular hypertension. Patients not responding adequately to enalapril monotherapy usually respond with the addition of a thiazide diuretic (or a calcium antagonist or beta-blocker), and rarely require a third antihypertensive agent. Enalapril is at least as effective as other established and newer ACE inhibitor

Fever, neutropenia and malignancy: a clinical syndrome in evolution.

During a 12-month period, 43 consecutive episodes of new fever in neutropenic cancer patients were evaluated. A two-drug combination of cephalothin, gentamicin or carbenicillin was used empirically for each episode. Overall mortality for this series was 28%. Microbiologic documentation of infection occurred in only nine of 43 episodes but a probable site of infection was observed in 14 others. The lungs were the most common site of infection and pneumonia was associated with a 100% fatal outcome. Despite the large number of febrile episodes with no infectious isolates, response to empiric broa

Bactericidal action of 2-hydroxy-3-(cyclohexylpropyl)-1,4-naphthoquinone on Bacillus megaterium.

The antimalarial drug, 2-hydroxy-3-(cyclohexylpropyl)-1,4-naphthoquinone (NQ), at concentrations of approximately 10(-5)m (3 mug/ml), was bactericidal for the gram-positive bacterium, Bacillus megaterium. Only a few other gram-positive bacteria were sensitive to this drug. All growth inhibitory concentrations of NQ were also bactericidal for B. megaterium, and even resting suspensions of cells were killed. The incorporation of radioactive-labeled leucine, thymidine, uracil, and diaminopimelic acid into protein, deoxyribonucleic acid, ribonucleic acid, and the cell wall polymer was arrested imm

In Vivo Prostate Cancer Modelling: From the Pre-Clinical to the Clinical Setting.

Prostate cancer (PCa) remains one of the most prevalent malignancies in men and a leading cause of cancer-related mortality worldwide. Over the last century, PCa modelling has evolved from basic cell-based to more complex systems. Despite this, the clinical translation of research findings is limited by the constraints of current preclinical models. In this review, rat and zebrafish models are highlighted due to their long-standing and emerging translational relevance, respectively. Rat models have played a pivotal role in understanding carcinogenesis and supporting the preclinical evaluation

Targeted degradation of Influenza a virus nucleoprotein via aptamer-based PROTACs for antiviral therapy.

Influenza A virus (IAV) poses a serious threat to public health due to its high mutability and rapid transmissibility. The viral nucleoprotein (NP), a highly conserved and essential component, has emerged as an ideal target for antiviral therapies. However, its biological function has proven challenging to modulate with conventional drugs, and no NP-targeting therapeutics have reached the market so far. Here, we report the development and application of an aptamer-based proteolysis-targeting chimera (PROTAC) for the targeted degradation of IAV NP. Utilizing the Direct-to-Biology (D2B) platform

Aberrant activation of epigenetic BRD9-DGAT1 axis promotes lipid droplets deposition and ferroptosis resistance in YAP-high prostate cancer.

Aberrant interplay between epigenetics and metabolism contributes to prostate cancer (PCa) progression and represents a formidable challenge limiting the efficacy of drugs. Elucidation of the epigenetic underpinnings of prostate cancer (PCa) could provide promising insights into the drivers of therapy resistance. Through an unbiased siRNA screen of mSWI/SNF family members, which play a significant role in tumorigenesis, we identified Bromodomain containing 9 (BRD9) as an essential gene for PCa growth. Targeting BRD9 abolished PCa colony formation and migration in vitro, and inhibited orthotopi

Pharmacological Management of Peptic Ulcer Bleeding.

Peptic ulcer bleeding (PUB) is the most common cause of non-variceal upper gastrointestinal bleeding, although its incidence is decreasing worldwide. Current epidemiological evidence demonstrates the positive influence of Helicobacter pylori eradication on outcomes, owing to the widespread introduction of proton pump inhibitor (PPI) treatment. However, PUB is still commonly encountered in clinical settings given the increasing indications for nonsteroidal anti-inflammatory drugs, anticoagulants, and antiplatelet agents in the aging population. Regardless of advancements made in the endoscopic

Structural and molecular insights into the binding mechanisms of β-glucosidase-ascorbic acid-rutin/isoquercetin ternary systems.

Elucidating the interaction within the β-glucosidase (BGL)-ascorbic acid (AA)-quercetin glycoside ternary system is essential for advancing the integrated utilisation of Rosa roxburghii Tratt (RRT) resources and BGL in the pharmaceutical and food industries. This study is the first to demonstrate the regulatory role of AA in a ternary system and its influence on BGL conformational changes during substrate binding. Our results show significant molecular aggregation in the BGL-AA-rutin/isoquercetin (Rut/Iso) systems, wherein Rut/Iso induced conformational alterations in BGL. Ternary system forma

Advancements in research on feeding intolerance to enteral nutrition in neurocritical patients undergoing sedation and analgesia.

In the Neurocritical Care Unit (NICU), deep sedation and analgesia are essential for managing critically ill patients to control intracranial pressure and reduce brain stress. However, these drugs-particularly opioids and benzodiazepines-significantly impair gastrointestinal motility and barrier function, increasing the risk of feeding intolerance (FI) to 46.38-62.0% in this population (1). FI manifests as elevated gastric residual volume, abdominal distension, vomiting, or failure to meet enteral nutrition targets within 72 h (2), and is associated with higher infection rates, prolonged hospi

PAX5 modulates vascularization and contributes to neuroendocrine lineage transition during carcinoma progression.

Human carcinomas often gain aggressive characteristics and escape cell type-specific treatment regimens through cryptic shifts in lineage states. However, the underlying mechanisms that govern lineage plasticity in carcinomas are undefined. Here in this study, we found that PAX5, a neural/lymphatic transcription factor, contributed to neuroendocrine (NE) lineage transition. PAX5 was highly expressed in aggressive human NE carcinoma cells and tissues but not in non-NE cancer cells and tissues. Deletion of Pax5 in Rb1fl/fl Trp53fl/fl mice caused a reduction of tumor vessels, loss of NE morpholog

Plant metabolites: potential treatments for ischemic acute kidney injury.

Ischemia-reperfusion injury is a leading cause of acute kidney injury (AKI), which is characterized by high morbidity and mortality, as well as a substantial risk of progression to chronic kidney disease (CKD). Although supportive care has improved considerably, effective disease-modifying therapies are still lacking. Therefore, the development of new therapeutic drugs for ischemic AKI is crucial. Active metabolites derived from medicinal plants, owing to their low toxicity and multi-target characteristics, are promising candidates for ischemic AKI therapy. This study reviews the pathogenesis

Exploring mitochondrial functions and dysfunctions in chondrocytes: toward the identification of novel therapies for osteoarthritis.

Mitochondria are essential organelles primarily described for their vital role in producing energy through oxidative phosphorylation (OxPhos). Due to the hypoxic environment of chondrocytes and their heavy reliance on glycolysis, mitochondrial functions have long been considered of minimal relevance in these cells. However, as major suppliers of energy through the ATP they produce by OxPhos, mitochondria help to regulate the balance between anabolism and catabolism. In osteoarthritis (OA), the most prevalent joint disease, this balance is dysregulated. In addition, correlations between metabol
