The ovarian Growing Teratoma Syndrome (GTS) is a rare condition among patients with primary Non-Seminomatous Germ Cell Tumours (NSGCT) presenting with enlarging masses during or after appropriate chemotherapy in the context of normalized serum markers. Several modes of dissemination are suggested, with the most frequent site of metastasis being the peritoneum. We report a case of a young patient with primary ovarian mixed NSGCT, who presented with Growing Teratoma Syndrome not only in the peritoneum but also within a trocar site after an initial surgery consisting in the laparoscopic morcellat

A 33-year-old man visited our hospital complaining of painless left scrotal swelling. Serum alphafetoprotein (AFP) and human chorionic gonadotropin-beta (HCG-β) were within normal range. Computed tomography revealed a heterogeneous tumor in the left scrotum and para-aortic lymph node swelling. We diagnosed the tumor as stage IIA testicular cancer and performed left high inguinal orchiectomy. Histopathological diagnosis was mature teratoma with no associated malignant germ cell tumor. The patient was followed-up without adjuvant chemotherapy because the size of para-aortic lymph nodes was remar

We present a 52-year-old gentleman with an unusual cause of progressive dysphagia, namely due to extrinsic lower oesophageal compression from a cystic mass of the posterior mediastinum. Cystic masses in adults are uncommon, and there is a wide differential diagnosis. This includes neoplastic, such as germ cell tumour (cystic teratoma), and non-neoplastic aetiologies. The later include foregut duplication cysts, lymphatic malformations, infective (hydatid), simple mediastinal cysts or pseudocysts. Management is principally surgical with complete excision, or alternatively, in cases of benign cy

INTRODUCTION: Seminoma comprises approximately 50% of testicular germ cell tumors. Renal metastases are infrequent, and are usually recognized at necropsy.
CLINICAL CASE: A 24 years-old man with history of left radical orchiectomy due to classical seminoma (stage I), and adjuvant radiotherapy, showed elevated levels of beta-human chorionic gonadotropin and lactate dehydrogenase in the eleven month follow-up. Computed tomography showed a 9 × 8 cm lobulated, heterogeneous tumor in the left kidney. The histopathological and immunohistochemical assay demonstrated a classical metastatic seminoma.

Sacrococcygeal teratomas and their malignant counterparts (germ cell tumors) are the most common solid tumors in neonates. Prompt diagnosis is essential because the frequency of malignant transformation increases from 10-20 % in neonates to 67 % in patients over 2 months of age. Cross-sectional imaging has largely replaced surgical exploration for staging these tumors and assessing their response to chemotherapy. Radiologists must be familiar with changes in the imaging findings of these tumors during and after treatment so that they can advise clinicians regarding the efficacy of therapy and

To examine a possible influence of estrogen receptor variants on the genetic susceptibility to germ cell cancer we have analyzed the allelic frequencies of three polymorphisms within the estrogen receptor gene in testicular cancer (n = 454) and control populations (n = 672). There were no differences in allelic frequencies in cancer patients compared to controls. Subgroup analyses did not indicate differences in allele frequencies for any of the polymorphisms in those individuals most likely to be predisposed to testicular cancer (patients with a history of maldescent of testicles and/or infer

Twenty-five patients with nonseminomatous germ cell carcinoma of the testis underwent CT and nuclear magnetic resonance (NMR) of the retroperitoneum followed by radical retroperitoneal lymph node dissection for surgical proof of metastatic disease. Computed tomography correctly predicted the presence or absence of adenopathy in 88% and assigned the correct stage in 84%. Nuclear magnetic resonance had comparable figures of 84 and 80%. Computed tomography appeared superior to NMR in detecting other abdominal abnormalities, although these were few in number. Nuclear magnetic resonance is nearly e

By multiple transcriptome datasets (TCGA, GSE59102, GSE25727, GSE27020 and GSE65858) and multi-omics (RNA-seq, SNP, CNV, DNA methylation) in-depth analysis, we found that cancer germline antigen (CGA) family/genes MAGEB2 is involved in the imitation, progression and prognosis in LC as well as correlated positively with lymphatic metastasis and negatively with DNA methylation. Then, in vitro experiment verified that MAGEB2 expression renders significant alteration in LC tissues and cells via immunohistochemical (IHC), qRT-PCR and western blot (WB), and up-regulation of MAGEB2 expression could f

γδ T cells play important roles in immune responses by rapidly producing large quantities of cytokines. Recently, γδ T cells have been found to be involved in tissue homeostatic regulation, playing roles in thermogenesis, bone regeneration and synaptic plasticity. Nonetheless, the mechanisms involved in γδ T-cell development, especially the regulation of TCRδ gene transcription, have not yet been clarified. Previous studies have established that NOTCH1 signaling plays an important role in the Tcrg and Tcrd germline transcriptional regulation induced by enhancer activation, which is mediated th

The role of telomere biology and telomerase activation in skin cancers has been investigated in melanoma and basal cell carcinoma but limited evidence is available for cutaneous squamous cell carcinoma (cSCC). We will review the current knowledge on the role of telomere and telomerase pathway in cSCC pathogenesis. At the somatic level, both long and short telomere lengths have been described in cSCC. This telomere dichotomy is probably related to two different mechanisms of tumour initiation which determines two tumour subtypes. Telomere shortening is observed during the invasive progression f

Toxic agents can interfere with the male reproductive system at many targets. One of the major unresolved questions concerning male infertility is identification of its molecular origins. Clinical and animal studies indicate that abnormalities of spermatogenesis result from exposure to three toxic metals (lead acetate, cadmium chloride, and arsenic trioxide), but the effects on primary spermatocyte DNA of the male rat after chronic exposure to these metals have not been identified. The aims of this study were to analyze, in three independent experiments, the DNA damage induced by lead (Pb), ca

The effect of the nephrotoxic substance, N-(3,5-dichlorophenyl)succinimide, on the histological pattern and incidence of kidney tumors induced by streptozotocin in rats was studied. In groups administered streptozotocin alone or in combination with nicotinamide, the histological pattern and the incidence of kidney tumors in rats were similar; renal cell tumors developed in 1 of 9 rats (11.1%) and in 2 of 17 rats (11.8%), respectively. However, post-treatment with N-(3,5-dichlorophenyl)succinimide increased the induction of epithelial tumors by streptozotocin, and embryonal cell tumors were als

CFTR gene mutations are responsible for Cystic Fibrosis. For half a decade, a triple therapy has been available for patients carrying the most frequent mutation: p.F508del. Among classified mutations, intronic mutations are rare, and no therapeutic strategies have yet been developed for such patients. We genome edited the parental iPSC line PCIi033-A to introduce two intronic mutations: A.s.Cas12a for c.1585-1G>A (PCIi033-A-8) and SpCas9 for c.1680-886A>G (PCIi033-A-9). Both cell lines have normal morphology and karyotype, conserved pluripotency, and differentiate into the three germ layers. O

Immature teratoma of the ovary is a rare condition primarily affecting young women and constitutes a significant proportion of ovarian cancer cases in adolescents and young adults. It is commonly diagnosed at stage I, though advanced-stage disease is not infrequent. Accurate diagnosis and a fertility-sparing approach are central to the management of this condition, as many affected women retain, or have yet to develop, a desire for childbearing. However, due to the rarity of this disease, management is often guided by retrospective data derived from subanalyses of studies on malignant ovarian

Bortezomib-induced peripheral neuropathy (BIPN) is a frequent, dose-limiting side effect in multiple myeloma (MM) patients treated with the proteasome inhibitor bortezomib (BTZ). To study the pathophysiology of pain in BIPN, we generated three induced pluripotent stem cell (iPSC) clones from human dermal fibroblasts (hdF) of a MM patient who developed painful BIPN and carried two homozygous risk variants (rs2839629 and rs915854). All clones exhibited typical iPSC morphology, expressed pluripotency markers, possessed normal karyotypes, and differentiated into all three germ layers. This patient

The adverse effects of obesity on male reproductive function are mainly manifested as the abnormal development of the reproductive system, decrease of testosterone level, decline of sperm quality, and impact on the health of offspring, while its regulatory mechanism is far from being clarified. This paper expounds the influence of obesity on the male reproductive system in the aspects of population epidemiology and animal experiments, presents an overview on the action mechanisms of obesity from the perspectives of the hypothalamus-pituitary-testis axis, blood-testis barrier, inflammatory reac

Growing teratoma syndrome (GTS) is an uncommon post-chemotherapy event in non-seminomatous germ cell tumors and presents as enlarging masses composed of mature teratoma despite normalized markers. As systemic therapy is generally ineffective, treatment focuses on surgical excision. We report the case of a 30-year-old male with an immature teratoma of the right testis who, after completing chemotherapy with normalized tumor markers, developed enlarging lymph node masses histologically confirmed as mature teratoma, consistent with GTS. This case highlights the importance of maintaining a high in

Swyer syndrome is a rare congenital condition that serves as a risk factor for developing germ cell tumors. The condition belongs to the group of 46, XY disorders of sexual development, is characterized by complete gonadal dysgenesis (CGD) and is mostly manifested as delayed puberty and primary amenorrhea during adolescence. Individuals with Swyer syndrome are known to be phenotypically female with normal internal and external female genitalia at birth. 46, XY GD involves a high risk of gonadoblastoma development with malignant potential such that the onset is greatest at or after the event of

Long QT syndrome (LQTS) is an inherited cardiovascular disorder characterized by electrical conduction abnormalities leading to arrhythmia, fainting, seizures, and an increased risk of sudden death. There are over 15 genes involved in causing LQTS, including SNTA1. Here we generated two human-induced pluripotent stem cell (iPSC) lines from two LQT patients carrying a missense mutation in SNTA1 (c.1088A > C). Both lines showed normal morphological properties, expressed pluripotency markers, showed a normal karyotype profile, and had the ability to differentiate into the three germ layers, makin
