Loss of H3K27me3 in meningiomas.

BACKGROUND: There is a critical need for objective and reliable biomarkers of outcome in meningiomas beyond WHO classification. Loss of H3K27me3 has been reported as a prognostically unfavorable alteration in meningiomas. We sought to independently evaluate the reproducibility and prognostic value of H3K27me3 loss by immunohistochemistry (IHC) in a multicenter study. METHODS: IHC staining for H3K27me3 and analyses of whole slides from 181 meningiomas across three centers was performed. Staining was analyzed by dichotomization into loss and retained immunoreactivity, and using a 3-tiered scori

Kinesin-7 CENP-E regulates cell division, gastrulation and organogenesis in development.

Kinesin-7 CENP-E motor protein is essential for chromosome alignment and kinetochore-microtubule attachment in cell division. Human CENP-E has recently identified to be linked with the microcephalic primordial dwarfism syndromes associated with a smaller head, brain malformations and a prominent nose. However, the roles of CENP-E in embryonic development remain largely unknown. In this study, we find that zebrafish CENP-E inhibition results in defects in early zygote cleavage, including asymmetric cell division, cell cycle arrest and the developmental abnormalities. We also demonstrate that CE

Epigenetics and bone diseases.

Owing to the development of new technologies, the epigenome, a second dimensional method for genome analysis has emerged. Epigenetic mechanisms, including DNA methylation, histone modifications and noncoding RNAs, regulate gene expression without changing the genetic sequence. These epigenetic mechanisms normally modulate gene expression, trans-generational effects and inherited expression states in various biological processes. Abnormal epigenetic patterns typically cause pathological conditions, including cancers, age-related diseases, and specific cartilage and bone diseases. Facing the rap

The role of post-translational modifications in hearing and deafness.

Post-translational modifications (PTMs) are key molecular events that modify proteins after their synthesis and modulate their ultimate functional properties by affecting their stability, localisation, interaction potential or activity. These chemical changes expand the size of the proteome adding diversity to the molecular pathways governing the biological outcome of cells. PTMs are, thus, crucial in regulating a variety of cellular processes such as apoptosis, proliferation and differentiation and have been shown to be instrumental during embryonic development. In addition, alterations in pr

New perspectives of valproic acid in clinical practice.

INTRODUCTION: Valproic acid (VPA) has been used in clinical practice as an anticonvulsant for more than four decades. Its pharmacokinetics and toxicity are thus well documented. VPA is also a potent class-selective histone deacetylase (HDAC) inhibitor at nontoxic therapeutic concentrations. New areas of application for VPA are currently opening up in clinical practice. AREAS COVERED: The authors discuss VPA and how it may serve as an effective drug for cancer therapy. This is due to its ability to induce differentiation of a number of cancer cells in vitro and also to decrease tumor growth an

Cardiac autophagy: good with the bad.

Stress-induced hypertrophic growth of the myocardium is a pathogenetic milestone in the progression of heart failure. Some evidence suggests that suppression of pathological cardiac hypertrophy per se is a viable target for therapeutic intervention, and cardiomyocyte autophagy is an attractive mechanism for consideration as a means of controlling the hypertrophic response. However, although considerable insights have been gleaned in the molecular mechanisms governing cardiomyocyte autophagy, many details critical to rational targeting of the response remain unknown. Among them, mechanisms unde

Expanding cofactor repertoire of protein lysine methyltransferase for substrate labeling.

Protein lysine methyltransferases (PKMTs) play crucial roles in normal physiology and disease processes. Profiling PKMT targets is an important but challenging task. With cancer-relevant G9a as a target, we have demonstrated success in developing S-adenosyl-L-methionine (SAM) analogues, particularly (E)-hex-2-en-5-ynyl SAM (Hey-SAM), as cofactors for engineered G9a. Hey-SAM analogue in combination with G9a Y1154A mutant modifies the same set of substrates as their native counterparts with remarkable efficiency. (E)-Hex-2-en-5-ynylated substrates undergo smooth click reaction with an azide-base

Antihistone antibodies in rheumatoid arthritis and Felty's syndrome.

Although antihistone antibodies are frequently detected in patients with systemic lupus erythematosus, they appear to be uncommon or present only at low levels in rheumatoid arthritis (RA) patients. We used an enzyme-linked immunosorbent assay to study sera from 42 patients with RA, and we confirmed that antihistone antibodies mainly occurred at low levels in these patients. In contrast, 83% of 24 patients with Felty's syndrome had antihistone antibodies, often at high levels. Thus, antihistone antibodies may be a common autoimmune abnormality in patients with Felty's syndrome but not in other

HDAC1 Represses NRF2 Transcription via H3K27ac Remodeling to Aggravate Neomycin-Induced Cochlear Hair Cell Damage.

Aminoglycoside-induced hearing loss is closely associated with oxidative stress and mitochondrial dysfunction in cochlear hair cells, but the upstream epigenetic mechanisms remain incompletely understood. This study investigated whether histone deacetylase 1 (HDAC1) contributes to neomycin-induced cochlear injury through transcriptional repression of NRF2. In HEI-OC1 cells, neomycin increased apoptosis, reactive oxygen species accumulation, mitochondrial depolarization, and mitochondrial superoxide production, accompanied by HDAC1 upregulation and NRF2 suppression. HDAC1 silencing attenuated t

T cell receptor signaling induces expression of lysine demethylase KDM6B to maintain Treg homeostasis.

Tregs expressing forkhead box P3 (FOXP3) play crucial roles in maintaining immune tolerance and tissue integrity. EZH2, a histone H3 lysine 27 (H3K27) methyltransferase, is known as a key regulator of Treg identity and suppressive function upon activation. Here, we demonstrate that the H3K27 lysine demethylase KDM6B, which catalyzes the opposing reaction to EZH2, is also required for Treg identity and function after activation. Treg-specific deletion of Kdm6b impaired tissue Treg fate and function. KDM6B was upregulated after T cell antigen receptor signaling in Tregs and contributed to the re

Design, one-pot synthesis, and structural characterization of novel 2-ureido-6-nitrobenzamide derivatives as MTDLs targeting HDAC8/epigenetic-kinase networks.

Modulating intersecting pathological pathways via multi-target directed ligands (MTDLs) is an active frontier in drug discovery. Herein, we report the design, one-pot synthesis, and characterization of a novel series of functionalized 2-ureido-6-nitrobenzamide derivatives 4-8 in 72-82% yields. Sequential nitration and imidization of phthalic anhydride 1, followed by sulfonylation, afforded the N-cyclic imide 2 and its sulfonyl ester building block 3. Aminolysis of 3 using various amines and hydrazines furnished target compounds 4-8 via a base-catalyzed Lossen rearrangement/addition-nucleophile

Design, Synthesis, and Biological Evaluation of Novel PAK1/HDAC10 Dual Inhibitors That Activate Antitumor Immunity for Triple-Negative Breast Cancer Treatment.

Triple-negative breast cancer (TNBC) remains a clinical challenge due to the lack of druggable targets, an immunosuppressive tumor microenvironment (TIME), and the limited efficacy of immune checkpoint inhibitors (ICIs). Herein, we report the first rational design, synthesis, and evaluation of dual PAK1/HDAC10 inhibitors to concurrently suppress oncogenic signaling and influence the TIME. Optimization yielded YDH-704, which displays nanomolar potency against PAK1 and HDAC10, excellent selectivity for HDAC10, and negligible off-target kinase activity. Mechanistically, YDH-704 coinhibits PAK1 on

Gut-Derived Sodium Butyrate Attenuates Liver Fibrosis by Inhibiting Aerobic Glycolysis via the HDAC3/c-Myc Signaling Axis.

Liver fibrosis remains a major global health challenge, driving interest in bioactive metabolites from natural sources as potential therapies. Sodium butyrate (NaB), a gut microbiota-derived short-chain fatty acid with notable anti-fibrotic activity, shows promise, although its mechanisms remain unclear. This study aims to explore the molecular mechanisms through which NaB attenuates liver fibrosis. This study integrates bioinformatics and network pharmacology to predict the molecular targets through which NaB exerts its anti-fibrotic effects. Candidate targets were validated through molecular

Sepsis-Induced Exosomal Transfer of MAFB mRNA Reprograms Hepatocytes via a miR-155-Jarid2-H3F3A Epigenetic Cascade.

Exosomes carry bioactive macromolecules driving sepsis pathogenesis, but the mechanisms underlying macrophage-to-hepatocyte communication during systemic inflammation remain poorly understood. We investigated how sepsis-induced macrophage exosomes are involved in remote intercellular crosstalk with hepatic cells via transcription factor-encoding mRNA cargo. Human monocytic THP-1 macrophages were stimulated with lipopolysaccharide (LPS), followed by exosome isolation, recipient cell uptake verification, and high-throughput RNA sequencing cargo analysis. Senkyunolide A interrupts TRAF6-HDAC3 interaction to epigenetically suppress c-MYC and attenuate cholestatic liver injury.

UNLABELLED: Introduction Cholestatic liver diseases are highly prevalent and lack effective treatment, ultimately progressing to end-stage liver diseases. Our recent study indicates that the interplay between c-MYC and lncRNA H19 exacerbates the ductular reaction during cholestasis. OBJECTIVE: This study aims to unveil the underlying mechanisms of the protective effects of senkyunolide A (SenA) on cholangiocyte overproliferation in cholestatic liver diseases. METHODS: Through comprehensive characterization using RNA sequencing, CHIP analysis, protein truncation, amino acid mutation or deleti

MOF promotes cisplatin resistance in lung cancer cells by enhancing WSTF acetylation.

Cisplatin-based chemotherapy is the first-line treatment for lung cancer. However, cisplatin resistance (CR) remains a major challenge, leading to treatment failure. A key driver of CR is enhanced DNA damage repair. Although males absent on the first (MOF) participate in DNA repair, their specific role in mediating CR remains unclear. In this study, CR models were established in PC9 and A549 lung cancer cell lines. Our results showed that high expression of Williams syndrome transcription factor (WSTF) in lung cancer cells was associated with CR. WSTF knockdown inhibited proliferation and prom
