Integrative Transcriptomic Analysis Identifies Shared Immune-Fibrotic Transcriptional Programs Across Crohn's Disease and Idiopathic Pulmonary Fibrosis.

Idiopathic pulmonary fibrosis (IPF) and Crohn's disease (CD) share overlapping immune and fibrotic processes, yet their convergent molecular mechanisms remain poorly defined. Here, we performed an integrative transcriptomic analysis of nine public datasets to identify shared transcriptional signatures across IPF and CD. The main discovery and validation analyses were based on bulk transcriptomic datasets and combined differential expression profiling, weighted gene co-expression network analysis, and machine-learning-based feature prioritization. We identified 28 shared disease-associated modu

High dietary fat causes muscle structural breakdown, mitochondrial dysfunction, and contractile deficits in the absence of carnitine palmitoyltransferase 2.

Carnitine palmitoyltransferase 2 (CPT2) deficiency is an inherited autosomal recessive disorder of fatty acid oxidation, which commonly manifests in adolescences and adulthood as muscle weakness and recurrent rhabdomyolysis, limiting physical activity and compromising quality of life. Despite the recognition and avoidance of known triggers such as exercise, fasting, and stress, many patients suffer unexplained periodic episodes of muscle breakdown. While avoidance of fatty meals is recommended, the impact of high-fat consumption alone on muscle biology of CPT-deficient patients is not well def

Size-dependent toxicological effects of microplastics: A review.

Industrialization has accelerated plastic production and emissions, resulting in pervasive environmental plastic pollution. Plastic debris can fragment into microplastics (<5 mm) and nanoplastics, collectively referred to as microplastics and nanoplastics (MNPs). Beyond their ubiquity in ecosystems, MNPs have also been detected in human-relevant matrices (e.g., placenta and breast milk), intensifying concerns about potential organ-specific health risks. Although particle size is increasingly recognized as a critical determinant of MNP toxicity, a cross-system synthesis of size-dependent effect

The race between viral immune evasion and the MHC class I antigen processing pathway.

Viral immune evasion of the major histocompatibility complex class I (MHC-I) antigen processing and presentation (APP) pathway is a centerpiece of the art of deception that enables persistence, reinfection, and severe disease. It does so by blunting peptide-MHC-I (pMHC-I) display, weakening CD8+ cytotoxic T lymphocyte (CTL) surveillance, and balancing the counterpressure imposed by natural killer (NK) cell missing-self responses. Rather than relying on a single trick, viruses deploy coordinated, multinode interference that functionally rewires the APP assembly line. These deceptive strategies

Altered Mitochondrial Base Excision Repair and Mitochondrial DNA Instability in Peripheral Leukocytes of Patients with MASLD.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a multifactorial metabolic disorder that is strongly associated with mitochondrial dysfunction and oxidative stress, which may potentially compromise the integrity of mitochondrial DNA (mtDNA). However, the role of the base excision repair (BER) pathway-the main mechanism responsible for repairing oxidative lesions in mitochondria-and maintaining mtDNA stability in MASLD remains poorly understood. Here, we analyzed total mRNA expression levels of key BER components in whole-blood samples, along with mitochondrial protein level

Unveiling the 12-Hour Ultradian Rhythm: Biological Foundations, Mechanistic Insights, and Potential Applications.

The ~12-h ultradian rhythm (circasemidian) represents an evolutionarily conserved temporal architecture that complements the canonical 24-h circadian clock. Over the past 5 years, mounting evidence has revealed its ubiquity across biological kingdoms, from tidal marine organisms and cyanobacteria to plants, microbiomes, and mammals, including humans, manifesting as intrinsic oscillations in gene expression, metabolism, and behavior that often persist independently of circadian control. In mammals, this rhythm is driven by a cell-autonomous oscillator centered on the XBP1s (X-box binding protei

Extracellular Vesicles in Alzheimer's Disease: Dual Roles in Pathogenesis, Promising Avenues for Diagnosis and Therapy.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by the accumulation of amyloid-β (Aβ) plaques, neurofibrillary tau tangles, chronic neuroinflammation, and synaptic loss, leading to cognitive decline. Extracellular vesicles (EVs)-lipid bilayer nanoparticles secreted by nearly all cell types-have emerged as critical mediators of intercellular communication, playing a complex dual role in both the pathogenesis and potential treatment of AD. This review generally delineates two opposite roles of EVs in pathogenesis and potential treatment of AD. On one hand, EVs

Potential of SERS and proteomics for biomarker detection in cancer cells.

This study investigates the use of quantitative LC-MS/MS-based proteomics and surface-enhanced Raman spectroscopy (SERS) for biomarker detection in classical Hodgkin lymphoma (HL). Two HL cell models with distinct TP53 status were utilized to evaluate the effects of etoposide, a DNA-damaging chemotherapeutic agent, and resveratrol, a polyphenolic compound with known chemosensitizing activity. For SERS, the best performance was achieved by applying logistic regression to classify different treatment conditions and identify discriminative spectral features in the data. Proteomics showed highly r

The TRAP complex (SSR1-SSR4): mechanistic roles and therapeutic opportunities.

BACKGROUND: Esophageal squamous cell carcinoma (ESCC) is a highly aggressive cancer with a poor prognosis, and its molecular mechanisms remain unclear. Our previous research identified the signal sequence receptor subunit delta (SSR4) of the TRAP complex as a potential ESCC biomarker. The TRAP complex, composed of SSR1, SSR2, SSR3, and SSR4, is essential for protein translocation, folding, and quality control, crucial for cellular balance. While individual TRAP subunits have been studied, a comprehensive understanding of their roles in human diseases is lacking. AIM: This review synthesizes c

A Comprehensive Review on the Microbial Signatures and Metabolic Mechanisms Underlying the Gut-Alzheimer's Disease Axis.

Alzheimer's disease (AD) is the most common form of dementia, driven by complex interactions among aging-related biological changes, neuronal degeneration, mitochondrial dysfunction, and environmental factors. Despite extensive research, effective disease-modifying therapies remain unavailable. Increasing evidence highlights the gut-brain axis as an important contributor to AD pathogenesis, particularly through amyloid-producing gut microbes that promote immune activation, neuroinflammation, and cerebral amyloid accumulation. This review summarizes current evidence linking gut microbiota (GM)

Glucocorticoid receptor knockout in mouse Schwann Cell is associated with decreased motor performances and nerve morphological abnormalities.

Glucocorticoid hormones (GC) are major regulators of stress, metabolism, and immunity. Glucocorticoid compounds are frequently used in medicine due to their effective actions as immunomodulators, but they can also present devastating side effects. GC bind to two corticosteroid receptors mediating their effects: the glucocorticoid receptor (GR) and the mineralocorticoid receptor (MR). Despite extensive studies on the role of these receptors in most tissues and organs very little data is available on their role in the peripheral nervous system (PNS). Schwann cells (SC) are the main glial cells o

Neuroendocrine mechanisms leading to novel control techniques of reproduction in livestock: Regulation of the GnRH pulse generator activity.

Pulsatile secretion of gonadotropin-releasing hormone (GnRH) is essential for normal reproductive function in mammals. GnRH pulses from the hypothalamus drive pulsatile gonadotropin release from the anterior pituitary, thereby regulating gonadal activity. Classic studies in rhesus monkeys have demonstrated that intermittent physiological GnRH administration maintains luteinizing hormone and follicle-stimulating hormone secretion, whereas continuous infusion suppresses pituitary responsiveness, establishing the necessity of pulsatile GnRH release. The neural mechanism governing this rhythmic se

Targeting DPP9 attenuates podocyte injury by regulating NRF2 antioxidant signaling.

INTRODUCTION: Podocyte injury is a critical event in the progression of chronic kidney disease (CKD), with oxidative stress as a central pathogenic mechanism. Despite the efficacy of finerenone in mitigating CKD progression, its precise cytoprotective mechanisms within podocytes remain incompletely defined. This study aimed to elucidate the role of dipeptidyl peptidase 9 (DPP9) in podocyte injury and determine whether finerenone exerts its renoprotective effects via DPP9-mediated antioxidant effects. METHODS: Integrated bioinformatic analyses of single-cell and bulk transcriptomic databases (

Senkyunolides: a promising natural compounds for the treatment of migraine headaches.

Migraine, ranked as the second most disabling neurological disorder globally, affects over 1 billion people, imposing a substantial worldwide burden. The complexity of its pathological mechanisms contributes to therapeutic limitations. While existing first-line drugs offer partial efficacy, their utility is often constrained by cardiovascular risks, medication-overuse headaches, low bioavailability, and high treatment costs, necessitating novel therapies that balance efficacy and safety. Notably, senkyunolides, core bioactive compounds derived from traditional anti-migraine herbs like Ligustic

Culturally Adapted Cognitive Behavioural Therapy and First Nations Population: A Role for Nurses?

This perspective paper examines the inconsistent integration of cognitive behavioural therapy (CBT) into the mental health care of First Nations people and considers the implications for mental health nursing practice. Although CBT is widely endorsed as an evidence-based treatment for depression, anxiety, post-traumatic stress disorder and chronic health conditions, its application in First Nations contexts remains uneven, with limited evidence of nurses delivering the intervention. Drawing on national and international literature, the paper explores whether this inconsistency reflects cultura
