The electroneutral Na(+)-(K+)-Cl- cotransport family.

Recently the molecular identification of the major electroneutral sodium-potassium-chloride entry mechanisms present on apical membranes of distal nephron segments of the mammalian kidney, on basolateral membranes of many non-renal epithelial cells and on certain non-epithelial tissues has been achieved. These transporters represent a major pathway for cellular uptake of chloride critical for chloride absorptive and secretory processes and for cell volume regulation following cell shrinkage. In the mammalian kidney, these sodium-coupled chloride cotransporters represent the major target sites

Changes in element concentrations induced by agonist in pig pancreatic acinar cells.

Changes in electrolytes of pig pancreatic acinar cells following application of gastrin-cholecystokinin (CCK) were investigated using the technique of X-ray microanalysis of hydrated and dehydrated sections of freshly frozen pancreas. After stimulation by CCK (10(-9) M), Na and Cl increased significantly in the cytoplasm [Na, from 10 mmol/kg wet wt. (48 mmol/kg dry wt.) to 19 mmol/kg (95 mmol/kg); Cl, from 22 mmol/kg (105 mmol/kg) to 49 mmol/kg (245 mmol/kg)] as well as in the luminal interspace [Na, from 53 mmol/kg (189 mmol/kg) to 65 mmol/kg (283 mmol/kg); Cl, from 65 mmol/kg (232 mmol/kg) t

Labelling and recording from dissociated target-specific rat superior cervical ganglion neurons.

A population of neurons was retrogradely labelled in the superior cervical ganglia (SCG) of the adult rat following the injection of the fluorescent dye Fast blue into the submandibular salivary glands (SMG). The neurons retained the fluorescent label following dissociation and culture. Electrical and chemosensitive properties of the labelled neurons were studied with the whole-cell patch-clamp technique.

Mechanism of acetylcholine action on pacemaker current (i(f)) in canine Purkinje fibers.

We have recently reported in canine Purkinje fibers that acetylcholine (ACh) can reverse the positive voltage shift of the pacemaker current (i(f)) induced by beta-adrenergic stimulation while having no direct action of its own. We have now investigated this effect of ACh on the cyclic adenosine monophosphate (cAMP) cascade in more detail. We find that addition of a membrane permeable analogue of cAMP (8-chlorophenylthio cAMP), 0.5-1 mM, increased the amplitude of i(f). This action was not reversed by 1 microM ACh, implying that ACh acts at a step prior to cAMP action. We then looked at the st

Membrane transport of ions in hypertension.

A variety of disturbances in transmembrane monovalent and divalent cation fluxes has been described in blood cells from hypertensive patients. Other membrane properties, such as fluidity and calcium binding, are also altered. It is now abundantly clear that some of the inconsistencies in this field are due to poor matching of patients and controls. However, even when careful matching is carried out, differences in membrane functions are still seen. It is suggested that these are due to a disturbance in the physicochemical properties of the cell membrane, related to changes in cell membrane pho

Calmodulin antagonists depress calcium and potassium currents in ventricular and vascular myocytes.

Myocytes isolated from guinea pig ventricles or bovine portal veins were voltage clamped with a single patch electrode. The calmodulin antagonists (CaM-A) calmidazolium, trifluoperazine (TFP), and chlorpromazine acted as Ca antagonists; they reduced the calcium inward current ICa in a voltage- and use-dependent way. For ventricular myocytes, 50% effective concentration (EC50) of calmidazolium was 1 microM, and the EC50 for TFP was 2.5 microM. For vascular myocytes, these numbers were 0.3 and 1 microM, respectively. CaM-A moderately retarded the inactivation time course and shifted the ICa avai

beta-Adrenergic treatment of hyperkalemic periodic paralysis.

In a patient with hyperkalemic periodic paralysis, metaproterenol prevented muscular weakness and hyperkalemia in periods of rest after exercise. During a severe attack, the drug rapidly corrected hyperkalemia and seemed to enhance the return of strength. The action of metaproterenol may involve a beta-adrenergic-mediated increase of potassium transport via the sodium-potassium pump.

Batrachotoxinin-A N-methylanthranilate, a new fluorescent ligand for voltage-sensitive sodium channels.

Batrachotoxin and its derivatives have become important tools for the study of membrane excitability by virtue of their effects on voltage-sensitive sodium channels. Recent studies have shown that the 2,4-dimethylpyrrole carboxylate ester normally occurring in the 20-alpha position of batrachotoxin may be replaced by a benzoate moiety without loss of activity. We have now extended this series of active batrachotoxin derivatives and report here the synthesis of batrachotoxinin-A 20-alpha-N-methylanthranilate. The new fluorescent compound is highly toxic (LD50 approx. 15 micrograms/kg, i.p. mous

Transport and control of Ca2+ by pigeon erythrocytes. II. Evidence against a simple feedback control of cell Ca2+ and evidence for the involvement of more than one pool.

Pigeon erythrocytes did not behave as expected from simple feedback mechanisms. The pool size for exchangeable cell Ca2+ was approximately proportional to the A23187-induced apparent 45Ca2+ influx ("J(in,app)") from 0.4 to 14 mumoles/min X l cell water at 184 microM external [Ca2+]. From earlier data, total cell 45Ca2+ was approximately proportional to J(in,app) from 10 to 120 mumoles/l X min. Thus there was no influx range where cell 45Ca2+ was held approximately constant. External [Ca2+] affected Ca2+ pool size independently of its effect of J(in,app). Trifluoperazine did not increase cell 4

A new scorpion toxin with a very high affinity for sodium channels. An electrophysiological study.

The neurotoxic action of toxin gamma from the venom of the Brazilian scorpion Tityus serrulatus (TiTx gamma) has been investigated in cultured mouse neuroblastoma cells (N1E115) using the suction pipette technique. Addition of 14 to 53 nM TiTx gamma to the external solution causes nerve cell membrane depolarization, membrane potential oscillations and spontaneous action potentials within 10 min. None of these effects were observed within 15 min after application of 1 microM toxin IV from Centruroides sculpturatus venom. Under voltage clamp the amplitude of the sodium current evoked by test pul

Properties of the Ca-activated K+ channel in pancreatic beta-cells.

The existence of [Ca2+]i-activated K+-channels in the pancreatic beta-cell membrane is based in two observations: quinine inhibits K+-permeability and, increasing intracellular Ca2+ stimulates it. The changes in K+-permeability of the beta-cell have been monitored electrically by combining measurements of the dependence of the membrane potential on external K+ concentration and input resistance. The changes in the passive 42K and 86Rb efflux from the whole islet have been measured directly. Intracellular Ca2+ has been increased by various means, including increasing extracellular Ca2+, additio

Fluid shift across the placenta: II. Fetomaternal transfer of horseradish peroxidase in the guinea pig.

The fetal side of the isolated guinea-pig chorio-allantoic placenta has been perfused with a fluid containing horseradish peroxidase. Electron micrographs show that protein is transferred across the fetal capillary endothelium via micropinocytosis as well as by transport through intercellular clefts. Under normal hydrostatic pressure no pinocytosis or other signs of fetomaternal protein transfer across the trophoblast have been observed. Instead, macrophages ingest the proteins that have reached the interstitial space between fetal endothelium and trophoblast. When the fetal venous pressure is

[Channel formation properties of black widow venom].

Black widow venom in the concentration 1--10 mkg/ml added on one side of the bilayer of common bovine brain phospholipids induces the formation of conductivity channels with high cation-anion selectivity with the number of cation transfer for K+, Ca2+, Sr2+, Mg2+, Na+, Cs+, Li+ equaling 0.98, 0.97, 0.96, 0.94. 0.88. 0.82, 0.82 correspondingly (at pH = 7.5). At pH less than 3.5 the channels are slightly selective for anions (the number of cation transfer 0.4). Potential-dependence of the channels is found, which is explained by microstructural reconstruction of their protein complex.

Calcium uptake mechanisms affected by some convulsant and anticonvulsant drugs.

An anticonvulsant (phenytoin) and a convulsant barbiturate (5-(2-cyclohexylidene ethyl)-5-ethyl-barbituric acid) (CHEB) reduced depolarization-coupled 45Ca uptake by synaptosomes from rat brain. CHEB reduced uptake to the level of undepolarized control samples. Phenobarbital failed to affect synaptosomal 45Ca uptake. Phenobarbital inhibited mitochondrial 45Ca uptake by 38%. CHEB was even more effective, lowering mitochondrial 45Ca uptake nearly to the levels induced by the mitochondrial inhibitors KCN, ruthenium red and dinitrophenol. Phenytoin, phenobarbital and CHEB did not affect ATP-depend

Specific monosaccharide inhibition of active sodium channels in neuroblastoma cells.

L-Fucose and D-galactose in low concentrations (0.27 or 2.7 mM) inhibited the induction of active Na+ channels in mouse and human neuroblastoma cells when the monosaccharides were added to the culture medium for 4 days with the inducing agent dimethyl sulfoxide. Active Na+ ionophores were determined by measurement of the toxin-stimulated efflux of 86Rb from the cells. At the same time, the amount of a radioactive glycoprotein (Mr 200,000), which was shown previously to be associated with neurite and membrane preparations from cells with active Na+ channels, was decreased.
