Design and Synthesis of Ibuprofen-Based Self-Deliverable Hybrid Gel Systems: Stabilization of Silver Nanoparticles, Antibacterial and Self-Healing Properties.

The reported molecule gel-forming component N1,N3,N5-tris(4-aminophenyl)benzene-1,3,5-tricarboxamide (TABTA) has been synthesized and characterized using various spectroscopic tools and techniques. The hybrid gel systems were fabricated using TABTA with ibuprofen and G8, which act as silver nanoparticle synthesis and stabilization templates. All gels were fabricated using the minimum critical gelation (MGC) method, and the interactions among the gelator components forming hybrid gel matrices were examined by using FT-IR and PXRD spectroscopy. G8-TABTA forms metallogels with Ag+, Fe3+, Fe2+, Co

Enhanced Antimicrobial Activity and Low Phytotoxicity of Acoustically Synthesized Large Aspect Ratio Cu-BTC Metal-Organic Frameworks with Exposed Metal Sites.

Metal-organic frameworks (MOFs) have recently been shown to be effective antimicrobial agents, particularly if they comprise pathogenicidal metal ions. Nevertheless, the accessibility of these active metal sites to the pathogen, and hence the MOFs' antimicrobial activity itself, is often poor since the metal nodes are usually embedded deep within its three-dimensional (3D) structure. We show that a unique copper-based (copper(II)-benzene-1,3,5-tricarboxylate) MOF, whose quasi-two-dimensional (quasi-2D) swordlike structure facilitates exposure of the metal ions along its surface, exhibits enhan

Pharmacotherapy for Bordetella pertussis infection. I. A synthesis of laboratory sciences.

There is considerable history and practice experience both with laboratory susceptibility testing for Bordetella pertussis and clinical treatment. This two-part narrative review provides a synthesis of the laboratory and clinical sciences as they apply to this bacterium and the clinical consequences of treating infection. It is generally held that antibiotic susceptibility testing for B. pertussis is not sufficiently standardised, but there has not been an urgent need to consolidate the same given the lack global experience with major resistance profiles. Experience in China, however, has prov

Ten years of external quality assessment (EQA) of Neisseria gonorrhoeae antimicrobial susceptibility testing in Europe elucidate high reliability of data.

BACKGROUND: Confidence in any diagnostic and antimicrobial susceptibility testing data is provided by appropriate and regular quality assurance (QA) procedures. In Europe, the European Gonococcal Antimicrobial Susceptibility Programme (Euro-GASP) has been monitoring the antimicrobial susceptibility in Neisseria gonorrhoeae since 2004. Euro-GASP includes an external quality assessment (EQA) scheme as an essential component for a quality-assured laboratory-based surveillance programme. Participation in the EQA scheme enables any problems with the performed antimicrobial susceptibility testing to

A Narrative Review of Airway and Gut Microbiota in Chronic Obstructive Pulmonary Disease: Clinical Associations, Methodological Heterogeneity, and Translational Priorities.

Chronic obstructive pulmonary disease (COPD) is a heterogeneous disorder in which exacerbation susceptibility, persistent inflammation, disease progression, and treatment response are not fully explained by spirometry. Culture-independent studies have associated airway and gut microbial features with clinically relevant COPD phenotypes, but findings are highly sensitive to sampling site, low biomass, contamination control, sequencing depth and platform, bioinformatic workflow, microbial-load quantification, medication exposure, disease state, and host or environmental confounding. Methodologic

Effects of high hydrostatic pressure on bacterial growth on human ossicles explanted from cholesteatoma patients.

BACKGROUND: High hydrostatic pressure (HHP) treatment can eliminate cholesteatoma cells from explanted human ossicles prior to re-insertion. We analyzed the effects of HHP treatment on the microbial flora on ossicles and on the planktonic and biofilm states of selected isolates. METHODOLOGY: Twenty-six ossicles were explanted from cholesteatoma patients. Five ossicles were directly analyzed for microbial growth without further treatment. Fifteen ossicles were cut into two pieces. One piece was exposed to HHP of 350 MPa for 10 minutes. Both the treated and untreated (control) pieces were then

Investigation of the physiological relationship between the cyanide-insensitive oxidase and cyanide production in Pseudomonas aeruginosa.

Pseudomonas aeruginosa is an opportunistic pathogen which demonstrates considerable respiratory versatility, possessing up to five terminal oxidases. One oxidase, the cyanide-insensitive oxidase (CIO), has been previously shown to be resistant to the potent respiratory inhibitor cyanide, a toxin that is synthesized by this bacterium. This study investigated the physiological relationship between hydrogen cyanide production and the CIO. It was found that cyanide is produced in P. aeruginosa at similar levels irrespective of its complement of CIO, indicating that the CIO is not an obligatory ele

Gut dysbiosis and jejunal injury in olfactory dysfunction: Experimental characterization and genetic epidemiological evidence.

BACKGROUND: Olfactory dysfunction (OD) is increasingly recognized not only as a sensory impairment but also as a condition associated with neurodegeneration and gut microbiota dysbiosis. However, the relationship and biological mechanisms linking the gut microbiota to olfactory function remain poorly understood. METHODS: We established a thiamazole-induced OD mouse model and evaluated olfactory function (buried food test), olfactory epithelial and jejunal histology, and gut microbiota by 16S rRNA sequencing. We further performed a two-sample Mendelian randomization (MR) analysis using GWAS (g

Impact of compost application on the spread of antibiotic resistance: emerging risks to public and aquatic health in Sri Lanka.

Composting is becoming an increasingly popular practice in the global circular bio-economy. Municipal Solid Waste (MSW) compost specially derived from chicken manure and animal litter poses a risk of spreading antibiotic-resistant contaminants. The objectives of the present study were the isolation of Amoxicillin (AMX), Cloxacillin (CLOX), Tetracycline (TET), and Ciprofloxacin (CIP) resistant bacteria from MSW and commercially available compost samples in Western Province, the determination of Minimum Inhibitory Concentration (MIC), Multiple Antibiotic-Resistant (MAR) indexes, and the detectio

Contribution of viable but non culturable cells and small colony variants in antibiotic insusceptibility and therapeutic failure against S. aureus and P. aeruginosa biofilm co-infections.

P. aeruginosa and S. aureus are often co-isolated from biofilm-associated infections, such as those afflicting cystic fibrosis (CF) patients. Biofilms, along with the interspecies interactions, play a significant role in fostering antibiotic insusceptibility, contributing to infection chronicity. Previously, we showed that S. aureus adopts a viable but non-culturable (VBNC) state in biofilms with P. aeruginosa. Here, we aimed to gain insight into the impact of VBNC and phenomena such as phenotypic switching on antimicrobial treatment and vice-versa. Single- and dual-species biofilms of two iso

Large-scale testing of antimicrobial lethality at single-cell resolution predicts mycobacterial infection outcomes.

In vitro antibiotic testing is important for guiding therapy and drug development. Current methods are focused on growth inhibition in bulk bacterial populations but often fail to accurately predict treatment responses. Here we introduce Antimicrobial Single-Cell Testing (ASCT), a large-scale live-cell imaging approach that quantifies bacterial killing in real time at single-cell resolution. By tracking over 140 million mycobacteria and analysing ~20,000 time-kill curves, we identify key determinants of antibiotic killing and its clinical relevance. For Mycobacterium tuberculosis, we found tha

Beyond plasma: defining the translation value of epithelial lining fluid exposure profiles for the treatment of pneumonia.

BACKGROUND: Plasma pharmacokinetic/pharmacodynamic (PK/PD) targets are a surrogate of pulmonary exposures for pneumonia, despite potential differences in exposures. This study defined free plasma and pulmonary epithelial lining fluid (ELF) PK/PD targets for different antibiotics against Klebsiella pneumoniae (KP) in a preclinical pneumonia model, and assessed the predicted cfu/lung response of human exposures relative to MIC breakpoints. METHOD: Secondary analyses were performed on previously published in vivo cfu/lung data generated using 17 KP after administration of different dosing regime

Ionophore PBT2 as a novel approach to combat antibiotic-resistant Helicobacter pylori.

Helicobacter pylori colonizes the gastric mucosa of around half of the world's population and is a major cause of chronic gastritis, peptic ulcer disease, and gastric cancer. Current therapies are becoming increasingly ineffective due to the rapid spread of antibiotic resistance, creating an urgent need for new treatment options with distinct mechanisms of action. Drug repurposing offers a practical and cost-effective approach to address this gap. PBT2 is an 8-hydroxyquinoline derivative originally developed for the treatment of neurodegenerative diseases and has more recently been shown to po
