Oxygen-glucose-deprived rat primary neural cells exhibit DJ-1 translocation into healthy mitochondria: a potent stroke therapeutic target.

AIMS: DJ-1 is a key redox-reactive neuroprotective protein implicated in regulation of oxidative stress after stroke. However, the molecular mechanism, especially the role of mitochondrial function, by which DJ-1 protects neural cells in stroke remains to be elucidated. The aim of this study was to reveal whether DJ-1 translocates into the mitochondria in exerting neuroprotection against oxidative stress. In particular, we examined DJ-1 secretion from primary rat neural cells (PRNCs) exposed to experimental stroke. METHODS: Primary rat neural cells were exposed to the oxygen-glucose deprivati

A zebrafish model of axenfeld-rieger syndrome reveals that pitx2 regulation by retinoic acid is essential for ocular and craniofacial development.

PURPOSE: The homeobox transcription factor PITX2 is a known regulator of mammalian ocular development, and human PITX2 mutations are associated with Axenfeld-Rieger syndrome (ARS). However, the treatment of patients with ARS remains mostly supportive and palliative. METHODS: The authors used molecular genetic, pharmacologic, and embryologic techniques to study the biology of ARS in a zebrafish model that uses transgenes to mark neural crest and muscle cells in the head. RESULTS: The authors demonstrated in vivo that pitx2 is a key downstream target of retinoic acid (RA) in craniofacial devel

Bazooka mediates secondary axon morphology in Drosophila brain lineages.

In the Drosophila brain, neural lineages project bundled axon tracts into a central neuropile. Each lineage exhibits a stereotypical branching pattern and trajectory, which distinguish it from other lineages. In this study, we used a multilineage approach to explore the neural function of the Par-complex member Par3/Bazooka in vivo. Drosophila bazooka is expressed in post-mitotic neurons of the larval brain and localizes within neurons in a lineage-dependent manner. The fact that multiple GAL4 drivers have been mapped to several lineages of the Drosophila brain enables investigation of the rol

Cdx mediates neural tube closure through transcriptional regulation of the planar cell polarity gene Ptk7.

The vertebrate Cdx genes (Cdx1, Cdx2 and Cdx4) encode homeodomain transcription factors with well-established roles in anteroposterior patterning. To circumvent the peri-implantation lethality inherent to Cdx2 loss of function, we previously used the Cre-loxP system to ablate Cdx2 at post-implantation stages and confirmed a crucial role for Cdx2 function in events related to axial extension. As considerable data suggest that the Cdx family members functionally overlap, we extended this analysis to assess the consequence of concomitant loss of both Cdx1 and Cdx2. Here, we report that Cdx1-Cdx2

The effects of parasympathectomy on serotonin-induced bronchoconstriction in the cat.

We studied the contribution of neural influences, in particular, the nonadrenergic, noncholinergic inhibitory system, on the modulation of airway responses to serotonin (5-HT) in the anesthetized and mechanically ventilated cat. Airway responses were assessed by measurement of pulmonary resistance (RL). Parasympathectomy (PS) caused a variable increase in bronchoconstriction in 9 of 10 cats (n = 10) that had received pretreatment intravenously with atropine (3 mg/kg) and propranolol (2 mg/kg); PS performed following pretreatment with phentolamine (2 mg/kg), propranolol, and atropine led to an

[Somatosensory-visual-vestibular interaction in a case of vermis agenesis].

It is well known that the somatosensory input is important for keep a balance as well as the visual and the vestibular inputs. However the knowledge of somatosensory-visual-vestibular interaction is incomplete at now. We have already made a mathematical model of these interactions by extension of the Raphan-Cohen model (Raphan et al, 1979) for visual-vestibular interaction with a direct and an indirect somatosensory pathway. This indirect somatosensory pathway has access to the common visual-vestibular velocity storage integrative mechanism. So the purpose of the present experiment was to veri

Repetitive Transcranial Magnetic Stimulation Promotes Neural Stem Cell Proliferation and Differentiation after Intracerebral Hemorrhage in Mice.

Repetitive transcranial magnetic stimulation (rTMS) is a physical treatment applied during recovery after intracerebral hemorrhage (ICH). With in vivo and in vitro assays, the present study sought to investigate how rTMS influences neural stem cells (NSCs) after ICH and the possible mechanism. Following a collagenase-induced ICH, adult male C57BL/6 J mice were subjected to rTMS treatment every 24 h for 5 days using the following parameters: frequency, 10 Hz; duration, 2 s; wait time, 5.5 s; 960 trains (500 µV/div, 5 ms/div, default setting). Brain water content and neurobehavioral score were a

Neural circuitry of the adult rat central nervous system after spinal cord injury: a study using fast blue and the Bartha strain of pseudorabies virus.

The distribution of retrogradely and transneuronally labeled neurons in the adult rat brain and spinal cord after contusive mid-thoracic spinal cord injury (SCI) was studied using Fast Blue (FB) and the Bartha strain of pseudorabies virus (PRV), respectively. When FB was injected into the distal spinal cord at 2 days after graded SCI at the 10th vertebral level, labeled neurons were consistently found 7 days later in supraspinal areas that normally project to the spinal cord. The number of FB-labeled neurons decreased as the injury severity increased. An inverse correlation between the number

Targeted quantification of 25 tryptophan metabolites across three metabolic pathways in serum by ultra-high-performance liquid chromatography coupled with quadrupole orbitrap high-resolution mass spectrometry.

Tryptophan metabolism encompasses three interconnected pathways, namely the kynurenine, serotonin, and gut microbiota-mediated indole pathways, which contribute to immune regulation, neural signalling, and host-microbiota interactions. Quantitative analysis of metabolites spanning these pathways remains challenging because of their diverse physicochemical properties, broad concentration ranges, and matrix interference in biological samples. In this study, a targeted analytical method based on ultra-high-performance liquid chromatography coupled with quadrupole Orbitrap high-resolution mass spe

Micro- and Nanoplastics in the Human Brain: Mechanistic Plausibility, Translational Challenges, and Links to Neurological Disease Trends.

The exponential growth in plastic production since the mid-twentieth century has led to the pervasive presence of micro- and nanoplastics (MNPs) across ecosystems and human exposure pathways, coinciding with a rising global burden of neurological disorders. Increasing evidence demonstrates that MNPs are not confined to peripheral tissues but can accumulate even in the human brain, raising concerns about their potential contribution to neurological disease. This structured review synthesizes global trends in plastic production, environmental MNP burden, and human exposure, together with emergin

Expression Signature of lncRNAs and mRNAs in Sevoflurane-Induced Mouse Brain Injury: Implication of Involvement of Wide Molecular Networks and Pathways.

Sevoflurane, one of the most commonly used pediatric anesthetics, was found to cause developmental neurotoxicity. To understand specific risk groups and develop countermeasures, a better understanding of its mechanisms is needed. We hypothesize that, as in many other brain degeneration pathways, long non-coding RNAs (lncRNAs) are involved in the sevoflurane-induced neurotoxicity. Postnatal day 7 (PD7) mice were exposed to 3% sevoflurane for 6 h. To quantify neurotoxicity in these mice, we (1) detected neural apoptosis through analysis of caspase 3 expression level and activity and (2) assessed

Schisantherin A attenuates ischemia/reperfusion-induced neuronal injury in rats via regulation of TLR4 and C5aR1 signaling pathways.

Toll-like receptor 4 (TLR4) and C5aR1 (CD88) have been recognized as potential therapeutic targets for the reduction of inflammation and secondary damage and improvement of outcome after ischemia and reperfusion (I/R). The inflammatory responses which induce cell apoptosis and necrosis after I/R brain injury lead to a limited process of neural repair. To further comprehend how these targets function in I/R state, we investigated the pathological changes and TLR4 and C5aR1 signaling pathways in vitro and in vivo models of I/R brain injury in this study. Meanwhile, we explored the roles of schis

Linking pathways in the developing and aging brain with neurodegeneration.

The molecular and cellular mechanisms, which coordinate the critical stages of brain development to reach a normal structural organization with appropriate networks, are progressively being elucidated. Experimental and clinical studies provide evidence of the occurrence of developmental alterations induced by genetic or environmental factors leading to the formation of aberrant networks associated with learning disabilities. Moreover, evidence is accumulating that suggests that also late-onset neurological disorders, even Alzheimer's disease, might be considered disorders of aberrant neural de

Peptide fingerprinting of folate-responsive proteins in human B lymphoblasts and orofacial clefting.

BACKGROUND: Maternal periconceptional use of folic acid contributes to the prevention of neural crest-related congenital malformations including orofacial clefts. The underlying biological pathways affected by folic acid, however, are still not clarified. In an explorative study, we identify folate-responsive proteins and pathways by advanced proteomic techniques and their possible role in orofacial development in young children. MATERIALS AND METHODS: At 15 months of age, we obtained B lymphoblasts from 10 children with and 10 children without an orofacial cleft. Folate-responsive protein exp

DAB2IP: unifying cardiovascular pathogenesis and cardiovascular brain crosstalk.

Cardiovascular diseases and brain disorders collectively represent a major global health burden. Not only do they frequently occur as comorbidities, but their pathological mechanisms are also intricately linked through a complex network of cardiovascular-brain crosstalk. Cardiovascular diseases (CVDs) and neurological disorders together impose high global health burdens. The DAB2IP gene, initially characterized as a tumor suppressor, serves as a multifunctional regulator and performs roles in both the cardiovascular and neurological systems. In atherosclerosis (AS), DAB2IP suppresses the infla

The Role of the Gut Microbiota in Modulating Signaling Pathways and Oxidative Stress in Glioma Therapies.

Tumors of the central nervous system (CNS), especially gliomas, pose a significant clinical challenge due to their aggressive nature and limited therapeutic options. Emerging research highlights the critical role of the gut microbiota in regulating CNS health and disease. The composition of the gut microbiota is essential for maintaining CNS homeostasis, as it modulates immune responses, oxidative status, and neuroinflammation. The microbiota-gut-brain axis, a bidirectional communication network, plays a pivotal role in cancer and CNS disease treatment, exerting its influence through neural, e

Transcriptome analysis of Xenopus orofacial tissues deficient in retinoic acid receptor function.

BACKGROUND: Development of the face and mouth is orchestrated by a large number of transcription factors, signaling pathways and epigenetic regulators. While we know many of these regulators, our understanding of how they interact with each other and implement changes in gene expression during orofacial development is still in its infancy. Therefore, this study focuses on uncovering potential cooperation between transcriptional regulators and one important signaling pathway, retinoic acid, during development of the midface. RESULTS: Transcriptome analyses was performed on facial tissues defic
