Case Report: Cystic fibrosis transmembrane conductance regulator gene heterozygous variation presenting with abdominal pain and hepatopancreatic lesions in a child.

BACKGROUND: Cystic fibrosis (CF) is an autosomal recessive disorder caused by mutations in the cystic fibrosis transmembrane conductance regulator (CFTR) gene, leading to multi-system involvement. CASE REPORT: A 4-year-old girl presented with a 2-day history of abdominal pain. Clinical manifestations included recurrent epigastric pain and vomiting. Physical examination revealed abdominal distension, mild periumbilical tenderness, and hepatomegaly (liver palpable 4 cm below the right costal margin). Laboratory tests showed elevated pancreatic enzymes: serum amylase 607 U/L, lipase 634 U/L, and

A novel PKHD1 missense variant disrupting splicing in a fetus with Caroli disease.

BACKGROUND: Caroli disease (CD) is a rare inherited disorder characterized by dilatation of intrahepatic bile ducts, and prenatal diagnosis of this disease is extremely rare. PKHD1 is the only known causative gene, yet the pathogenicity of most missense variants remains unclear. METHODS: Exome sequencing (ES) was performed on a fetus with clinical features of CD. Candidate variants were validated by Sanger sequencing in the family. The impact of the novel missense variant on pre-mRNA splicing was assessed using minigene assays, and structural modeling of the PKHD1 protein was conducted with A

Transmission of F12-related hereditary angioedema through a sperm donor.

Hereditary angioedema due to F12 pathogenic variants (HAE-FXII) is a rare autosomal dominant disorder characterized by recurrent episodes of angioedema mediated by bradykinin, incomplete penetrance, and marked sex-dependent clinical expression. Most affected individuals carry the recurrent F12 c.983C>A/p.Thr328Lys (T328K) founder variant. Here, we report the first documented transmission of HAE-FXII through sperm donation. The index case was an 18-year-old woman with recurrent estrogen-dependent angioedema, normal C1-inhibitor levels and function, and poor response to conventional antiallergic

Proneness to infections and familial risk of tic disorders.

BACKGROUND: Postinfectious autoimmune processes are hypothesized to be causally implicated in tic disorders, including Tourette syndrome and chronic tic disorder. However, this hypothesis remains controversial. In this nationwide cohort study, we aimed to clarify the mechanisms underlying the association between proneness to infections and tic disorders. METHODS: Using Swedish national registers, we identified 3,886,533 individuals (probands) born between 1970 and 2008 with available data on both biological parents. Probands were linked to six clusters of relatives: parents, full siblings, ma

Family studies in Gaucher Disease: a key resource for early diagnosis and personalized treatment strategies.

Gaucher Disease (GD) is an inherited metabolic disorder caused by mutations in the GBA1 gene, which is responsible for the synthesis of the enzyme glucocerebrosidase (GCase). The clinical manifestations, which are extremely heterogeneous, include splenomegaly, hepatomegaly, anaemia and bone complications. GD is an autosomal recessive condition, meaning that the clinical phenotype manifests itself only in the presence of two mutated alleles, which are inherited from both parents, who are generally healthy and asymptomatic. However, other family members (brothers, sisters, grandparents, uncles,

Host switching via rare sex: Oncorhynchus mykiss as a vector in fusion of three Gyrodactylus salaris genomes into a sexual-clonal population on a landlocked Salmo salar (Monogenea, Platyhelminthes).

The monogenean ectoparasite Gyrodactylus salaris Malmberg, 1957 was detected in 2001 on juveniles of landlocked Salmo salar Linnaeus in three rivers draining to Lake Kuitozero in the White Sea basin. The ancestry of the colony was studied by internal transcribed spacers of the nuclear ribosomal operon (ITS), mitochondrial COI gene and by 12 microsatellite markers. The maternal ancestor was a frozen heterozygotic and heteroplasmic diploid clone RBT marked by a ITS-1068R, spread by rainbow trout and observed in Denmark, Norway, Finland and Russia. The clone combined genomes from two divergent ma

Higher sperm H3K4me3 levels are associated with idiopathic recurrent pregnancy loss.

During fertilization, spermatozoa contribute genetic and epigenetic factors such as chromatin packaged with protamines and histones; DNA methylome, non-coding RNAs, etc. Human sperm chromatin retains 5-15% nucleosomes which can play a key role in embryonic development. Recurrent pregnancy loss (RPL) is a condition mainly attributed to defects in embryo and placenta development. Majority of the known RPL factors are of maternal contribution, while  ~50% RPL cases are termed idiopathic (iRPL). In addition to paternal genetic factors, epigenetic factors via sperm could also be responsible for iRP

High-Fat Diet-Fed Kcnq1 Mutant Mice Have Reduced Pancreatic β-Cell Mass via Gene-Environment Interaction.

BACKGRUOUND: The potassium voltage-gated channel subfamily Q member 1 (KCNQ1) gene has recently received much attention as a candidate susceptibility gene for type 2 diabetes mellitus, especially in Asian populations. We previously reported that Kcnq1 mutant mice exhibit reduced insulin secretion and hyperglycemia due to a decrease in pancreatic β-cell mass. Through in vivo and in vitro analyses, we ascertained that this mechanism is the result of the downregulation of the non-coding RNA 'Kcnq1ot1,' which is expressed in the paternal allele of the Kcnq1 gene region, causing an increase in the

Effects of Loneliness and Subjective Well-Being on Depression in Female Adolescents: A Longitudinal Moderated Mediation Model of the Parental Relationship.

OBJECTIVE: Adolescent depressive symptoms are increasingly recognized as a global concern for youth mental health. Female adolescents consistently report higher levels of depressive symptoms than their male counterparts, yet the psychological mechanisms underlying depressive symptoms remain insufficiently understood. Our longitudinal study examined the relationship between loneliness and depressive symptoms in female adolescents and investigated the mediating effect of subjective well-being (SWB) and the moderating influence of the parental relationship. METHODS: Data were drawn from the Kore

Family misfortune caused by hereditary bias: a reflection on mitochondrial disease diagnosis in a family.

AIM: Assisted reproductive technology (ART) is an invaluable strategy for preventing the inheritance of genetic disorders and promoting the birth of healthy children. Nevertheless, the general public's limited understanding of genetics and low awareness of available services obstruct effective utilization of genetic counseling. Our analysis of a family affected by mitochondrial genetic disease aims to improve public understanding of genetic knowledge and the importance of genetic counseling. METHODS: We gathered comprehensive data on a family with mitochondrial disease and scrutinized the gen

Parental psychiatric disorders and age-specific risk for offspring major depression: Finnish nationwide register-based study.

BACKGROUND: Limited studies have examined the association between the whole range of parental psychopathology and offspring major depression (MD). No previous study has examined this association by age of onset of offspring MD, or restricting to parental psychiatric diagnoses before offspring birth. METHODS: This nested case-control study included 37,677 cases of MD and 145,068 controls, identified from Finnish national registers. Conditional logistic regression models examined the association between parental psychopathology and MD, adjusting for potential confounders. RESULTS: Increased ri

Associations Between Mental Disorders, Personality Disorders, and Filicide-Suicide and Parental Suicide: A Nationwide Cohort Study.

AIM: Filicide-suicide represents a critical intersection of psychiatric and familial crises, yet the literature remains scant on differentiated risk profiles among family members. This study addresses a gap in understanding the specific mental health correlates of filicide-suicide, especially within a comprehensive family context, distinct from traditional parental or child-focused perspectives. METHOD: This study used the Taiwan National Health Insurance Research Database to identify a nationally representative sample of families from 2004 to 2020, with continuous monitoring extending into 2

Mothers' and Fathers' Pregnancy Intentions and Interparental Conflict: Is There Harm in Ambivalency?

Unintended pregnancy represents a critical challenge for family well-being, especially among unmarried parents with low incomes who come from marginalized racial/ethnic backgrounds. This study examines the roles of both maternal and paternal pregnancy intendedness in interparental conflict and violence, focusing on a sample of racially diverse unmarried couples with low income from the Building Strong Families (BSF) project (N = 2,325 couples). Antenatal pregnancy intentions were tested as predictors of destructive conflict and intimate partner violence (IPV) at both 15 months and 3 years post
