Effects of Extreme Heat Exposure on Heatstroke and Liver Injury in Mice: The Role of PPARα.

BACKGROUND: Liver injury is a frequent complication of heatstroke and constitutes a direct cause of death. However, only a few studies examined the mechanism underlying heatstroke-induced liver injury. OBJECTIVE: We aimed to evaluate the role of peroxisome proliferator-activated receptor α (PPARα) in heatstroke-induced liver injury and to explore the potential mechanisms. METHODS: Male C57BL/6N mice were subjected to a control (22 ± 1 °C) or extreme heat temperature (39.5 ± 0.5 °C) to induce a heatstroke-associated liver injury animal model. PPARα agonist, ferroptosis inhibitor, and AAV8-media

Time-resolved urinary proteomics reveals heme-associated oxidative stress responses in neonatal hypoxic-ischaemic encephalopathy.

BACKGROUND: Neonatal hypoxic-ischemic encephalopathy (HIE) triggers systemic oxidative stress and redox imbalance, contributing to multi-organ injury. Urine is a noninvasive matrix for longitudinal profiling of molecular responses, yet time-resolved proteomic studies in HIE are limited. METHODS: We performed longitudinal SWATH-MS proteomic profiling of urine from term neonates with moderate-to-severe HIE treated with therapeutic hypothermia (n = 16) and non-asphyxiated controls (n = 19) at six time points during the first eight days of life. Proteins identified in ≥ 80% of samples were quanti

Linagliptin and/or α-pinene attenuate cisplatin nephrotoxicity in rats via modulating phosphatase and tensin homolog-induced kinase 1/Parkin/microtubule-associated proteins 1A/1B light chain 3II/SQSTM1/sequestosome-1-assisted mitophagy.

Acute kidney injury is considered the most harmful and prevalent type of cisplatin-induced nephrotoxicity. In the current research, the potential prophylactic effects of linagliptin and/or α-pinene were investigated on cisplatin-induced functional, biochemical, and structural changes in rat kidneys. Forty Sprague-Dawley male rats were separated into five groups (n = 8): (1) normal group; (2) model group (cisplatin 7.5 mg/kg, i.p., single dose); (3) linagliptin group (3 mg/kg, p.o. for 18 days); (4) α-pinene group (50 mg/kg, p.o. for 18 days); and (5) linagliptin + α-pinene group (linagliptin 3

Probiotics and palmitoylethanolamide (PEA) for osteoarthritic pain: individual effects in a multiple baseline design study.

BACKGROUND/OBJECTIVES: Osteoarthritis (OA) is a leading cause of chronic pain. As healthcare services struggle to meet demand to treat chronic pain, many individuals self-manage their symptoms. Probiotics and palmitoylethanolamide (PEA) may be promising therapeutic options because of their anti-inflammatory properties. PEA is an endogenously produced N-acylethanolamine, considered "endocannabinoid-like" due to its structural similarity to endocannabinoids. While it has non-direct influence on the endocannabinoid system, it primarily acts via non-cannabinoid pathways, most notably through activ

Metabolomic Insights into HCF-1-dependent Regulation of Liver Homeostasis.

BACKGROUND: Host cell factor-1 (HCF-1) is a conserved transcriptional and epigenetic coregulator known to influence hepatocyte proliferation, liver regeneration, and lipid metabolism. While its role in transcriptional control and chromatin dynamics has been explored, the metabolic consequences of HCF-1 loss remain incompletely understood. OBJECTIVE: To investigate the metabolic alterations triggered by hepatocyte-specific HCF-1 loss in murine liver and their potential relevance to metabolic-associated fatty liver disease (MAFLD). METHODS: Using Alb-Cre-ERT2 tg ; Hcfc1 hepKO/Y mice, we perfor

Sub‑chronic dietary exposure to environmentally relevant doses of di‑(2‑ethylhexyl) adipate (DEHA) induces hepatic steatosis in female mice via disrupting the PPARγ pathway and inducing endoplasmic reticulum stress: In vivo and in vitro evidence.

Di-(2-ethylhexyl) adipate (DEHA), initially regarded as a safer alternative to Di-(2-ethylhexyl) phthalate (DEHP) phthalate plasticizer, is widely used in food packaging and consumer products. Despite its high detection levels in food and established human exposure, the health impacts of DEHA at environmentally relevant doses remain poorly understood. The main aim of this study was to investigate whether sub-chronic dietary exposure to DEHA disrupts metabolism homeostasis in female mice, and to clarify the underlying mechanism through combined in vivo and in vitro models. In this study, 27 fou

Gut microbiota modulation by novel synbiotic improves production performance and ovarian function in aged laying hens via gut-ovary axis.

The management of oxidative stress and ovarian dysfunction associated with aged laying hens, which is highly involved in gut microbiota, has been suggested as a feasible approach to improve production performance. Here, we investigated the effects of a novel synbiotic (Bacillus amyloliquefaciens + inulin, BAI), a gut microbiota regulator, on the improvement of production performance in aged laying hens, and dissected the underlying mechanisms using multi-omics analysis. Our findings showed that, compared to the control, high-dosage BAI supplementation significantly improved production performa

PM10 Disrupts Mitochondrial Homeostasis in Corneal Epithelial Cells: Protective Effects of SKQ1.

Airborne particulate matter with a diameter of <10 μm (PM10) can damage the corneal epithelium by inducing oxidative stress, disrupting the NRF2 antioxidant pathway, and triggering epithelial barrier dysfunction and inflammation. However, the role of mitochondria in mediating PM10-induced damage remains unexplored. This study investigated the impact of PM10 on mitochondrial homeostasis in both immortalized human corneal epithelial cells (HCE-2) and the mouse corneal epithelium, as well as the protective effects of SKQ1. For in vivo assessment, female C57BL/6 mice were exposed to either control

Complex I Modulator BI4500 Reduces MASH by Limiting Oxidative Stress and Reprogramming Lipid Metabolism via AMPK in MCD Rats.

BACKGROUND: Metabolic-dysfunction-associated steatotic liver disease (MASLD) is a multifactorial liver disease in which mitochondrial dysfunction, oxidative stress, and inflammation play key roles in driving the progression toward metabolic dysfunction-associated steatohepatitis (MASH) and hepatocellular carcinoma (HCC). Dysfunctional mitochondria generate excess reactive oxygen species (ROS), impair antioxidant defenses, activate pro-inflammatory pathways and hepatic stellate cells, and perpetuate liver injury. Mitochondrial Complex I is a major ROS source, particularly under conditions of dy

Combined effects of high-fat diet feeding and Streptococcus agalactiae infection on lipid metabolism, antioxidant status, and immune response in tilapia (Oreochromis niloticus).

High-fat diet (HFD) and Streptococcus agalactiae are common pathogenic factors affecting tilapia health, yet their combined effects and underlying mechanisms are not well understood. To address this, we conducted a comprehensive evaluation of the potential response mechanisms in tilapia subjected to both factors. Tilapia were fed normal (NC) or high-fat diet (HFD) for 90 days, then challenged with S. agalactiae. At 48 h post-infection, blood, liver, and spleen samples were collected for biochemical parameter analysis and gene expression profiling. The results indicated that the combined treatm

Kudzu resistant starch effectively ameliorates non-alcoholic fatty liver disease by protecting the gut microbiota intestinal barrier and suppressing inflammatory responses.

BACKGROUND: Non-alcoholic fatty liver disease (NAFLD), including non-alcoholic steatohepatitis, may lead to a cocktail of disease consequences, for instance, type II diabetes, liver fibrosis, and even hepatocarcinogenesis. The complex pathogenesis of NAFLD results in a shortage of an effective drug for its treatment. Currently, a healthy diet and regular exercise are considered one of the most effective and safe ways to mitigate NAFLD as well as other types of metabolic disorders. Kudzu-resistant starch (KRS) has developed as a potential dietary supplement attributed to its prebiotic propertie

Emodin Enhances Rosiglitazone's Therapeutic Profile by Dual Modulation of SREBP1-Mediated Adipogenesis and PPARγ-Driven Thermogenesis.

Background/Objectives: Rosiglitazone (RSG), a potent PPARγ agonist for type 2 diabetes mellitus (T2DM), induces adverse adipogenic effects that limit clinical use. We investigated whether emodin (1,3,8-trihydroxy-6-methylanthraquinone, EMO), a natural anthraquinone, mitigates RSG-induced complications while enhancing its insulin-sensitizing benefits in severe obesity. Methods: Male ob/ob mice with established obesity and diabetes were treated for 4 weeks with RSG (10 mg kg-1 day-1), EMO (200 or 400 mg kg-1 day-1) or their combination. Metabolic profiling, organ function, and adipose histology

Maternal, fetal, and neonatal toxicity and potency estimates of perfluorohexane sulfonate (PFHxS) from oral maternal exposure in the Sprague-Dawley rat.

Perfluorohexane sulfonate (PFHxS) is a legacy, long-chain per- and polyfluoroalkyl substance (PFAS) frequently detected in human serum and environmental media across the globe. Previously published developmental studies in rats indicated a general lack of effects in common endpoints like maternal and offspring body and liver weights at doses up to 50 mg/kg/d. To facilitate mixture-based studies of PFAS co-exposure, individual dose response parameters are necessary for model predictions. To address these data needs we exposed timed-pregnant Sprague-Dawley rat dams to PFHxS (3-125 mg/kg/d) via o

Molecular and cellular mechanisms of pentadecanoic acid.

Pentadecanoic acid (C15:0) is an odd-chain fatty acid, the β-oxidation of which yields propionyl-CoA that replenishes succinyl-CoA and tricarboxylic acid cycle flux; higher circulating levels are associated with reduced type 2 diabetes, cardiovascular disease, metabolic dysfunction-associated steatotic liver disease, and mortality. Summarize the cellular and molecular mechanisms underlying these associations. A comprehensive literature search (2000-2025) identified studies of C15:0's mechanistic actions in vitro and in vivo, and multi-omics studies focused on receptor binding, signaling cascad
