Red blood cells (RBCs), formerly viewed as mere oxygen transporters, are now acknowledged as dynamic biological entities with intricate molecular characteristics which impact their deformability, lifespan, and interactions within the vascular system. Progress in molecular hematology has elucidated how modifications in RBC proteins, lipids, ion channels, and adhesion molecules influence physiological adaptability, pathogenic mechanisms, and transfusion results. Inherited and acquired illnesses, such as sickle cell disease, thalassemia, malaria, human immunodeficiency virus (HIV), and metabolic

Integrating high-resolution spatial transcriptomics with metabolomics is essential for linking cellular identity to metabolic state, yet same-section, single-cell alignment is limited by incompatible substrates, chemistries, and imaging. We introduce OpenFISH, an open-source, low-cost, modular spatial transcriptomics platform and a matrix-assisted laser desorption ionization mass spectrometry imaging (MALDI-MSI)-compatible workflow. Through optimized tissue handling and a guided registration pipeline, OpenFISH enables cell-type-aware co-mapping of transcripts and metabolites. We demonstrate it

Lipids are vital cellular components with structural, storage, signaling, and defensive functions, and lipid metabolism is critical to strawberry quality. However, the regulatory mechanism underlying lipid metabolism during strawberry development and ripening remains unclear. Although LEAFY COTYLEDON 2 (LEC2) regulates lipid metabolism, its function in strawberries is unknown. This study explored the role of FaLEC2 in strawberries. FaLEC2 is highly expressed during strawberry development. FaLEC2 overexpression delayed ripening, promoted the accumulation of phosphatidylcholines (PCs), including

Snake venom phospholipase A2 (SVPLA2) is a key toxic component of Naja atra (N. atra) venom, yet its systematic impact on macrophage metabolism remains elusive. Using non-targeted metabolomics, combined with multivariate statistics and KEGG pathway enrichment, this study delineated the metabolic reprogramming of RAW 264.7 macrophages induced by SVPLA2 in N. atra venom and assessed the reversal effects of the specific inhibitor varespladib. SVPLA2 was associated with a a triple metabolic phenotype in macrophages: massive production of histamine and arachidonic acid derivatives, activating infla

CLN5 Batten disease, caused by biallelic mutations in CLN5, is a rare, early-onset neurodegenerative lysosomal storage disorder that has no cure and lacks validated biomarkers, hindering accurate diagnosis and assessment of therapeutic response. We recently identified CLN5 as the synthase of bis(monoacylglycero)phosphate (BMP), an endolysosomal phospholipid crucial for lysosome function and lipid catabolism. This suggested BMP and its precursor lysophosphatidylglycerol (LPG) as clinically relevant biomarkers. It also prompted in vivo confirmation of CLN5 as the biologically relevant lysosomal

Acute lung injury (ALI), which can have a mortality rate approaching 40%, lacks effective therapies. Macrophage pyroptosis represents a pro-inflammatory event and a potential therapeutic target. Here, we report a single-atom immunomodulator, a potassium-based single-atom catalyst (K-SAC) with K-N4 sites, that suppresses macrophage pyroptosis for ALI therapy through antioxidant catalysis. Constructed from biocompatible potassium, the most abundant intracellular metal in humans, K-SAC displays potent superoxide dismutase- and catalase-like activities, with the underlying mechanisms revealed by d

Loss-of-function variants of the CHKB gene cause an autosomal recessive disease described as an early onset congenital megaconial (large peripheral mitochondria) muscular dystrophy. CHKB encodes choline kinase β, the first enzyme in the biochemical pathway for synthesis of the major membrane phospholipid phosphatidylcholine. Chkb -/- mice recapitulate the human disease with affected skeletal muscle displaying a decrease in strength, myofiber atrophy, megaconial mitochondria, fat accumulation within muscle cells, and an increase in muscle injury. Here, we assessed the therapeutic potential of a

Ferroptosis is a form of regulated cell death driven by iron-dependent phospholipid peroxidation and has emerged as a key mechanism of neuronal injury across a broad spectrum of neurological disorders. MicroRNAs (miRNAs), which function primarily as post-transcriptional regulators of gene expression, are increasingly recognized as important modulators of the regulatory networks governing ferroptosis and as potential therapeutic targets in these conditions. In this review, we synthesize current advances in miRNA-mediated regulation of ferroptosis in neurological disorders. We first outline the

Late-life depression involves neurodegeneration, inflammation, metabolic disruption, and psychosocial stress. Lipid metabolism has drawn increasing attention in this field. This review examines three interconnected aspects: peripheral metabolic disturbance, potential central lipid alterations, and dietary lipid regulation. Peripherally, adverse lipid profiles and lipidomic shifts correlate with depressive symptoms, cognitive decline, and cardiometabolic load. Mechanisms include inflammation, membrane remodeling, mitochondrial dysfunction, and oxidative stress. Centrally, the disorder may invol

Resistance to apoptosis, metabolic plasticity, and redox adaptation are major contributors to cancer progression and treatment failure. Ferroptosis and cuproptosis have therefore emerged as metal-dependent forms of regulated cell-death programs with potential relevance for tumours that survive conventional therapy. Ferroptosis is driven by iron-dependent phospholipid peroxidation when glutathione peroxidase 4 (GPX4)-dependent and parallel antioxidant systems fail, whereas cuproptosis depends on mitochondrial copper engagement of lipoylated tricarboxylic-acid-cycle proteins, lipoylated-protein

L-theanine, a natural amino acid in tea, exhibits potential neuroprotective effects. However, its impact on depression via the microbiota-gut-brain axis remains unclear. Here, L-theanine alleviated reserpine-induced depression-like behaviors in rats, improving anhedonia, despair, and cognitive deficits, while reducing serum IL-1β, IL-6, and TNF-α, restoring hippocampal BDNF, and mitigating neuronal damage. Multi-tissue non-targeted metabolomics (serum, brain, colon, feces) revealed that L-theanine reversed phospholipid and bile acid disturbances and restored key neuroprotective metabolites. Ta

Plasma membranes (PMs) exhibit asymmetry between their two leaflets in terms of phospholipid headgroups, unsaturation, and resulting membrane properties, such as packing and fluidity. Lateral heterogeneity, including the formation of lipid domains, is another crucial aspect of PMs with significant biological implications. However, the nature and even the existence of lipid domains in the two leaflets of PMs remain elusive, hindering a complete understanding of the significance of lipid asymmetry. Using coarse-grained molecular dynamics simulations of the asymmetric PM, we find that the outer l

Colorectal cancer is a malignant tumor of the colon or rectum, with approximately 150,000 new cases each year. Current treatment strategies, such as surgery, chemotherapy, radiotherapy, and immunotherapy, face challenges ranging from cancer recurrence, drug resistance to significant toxicity. Therefore, these patients urgently need more effective treatments. Ferroptosis, a novel form of cell death characterized by iron-dependent lipid peroxidation, has emerged as a promising new approach for treating colorectal cancer. Inactivation of phospholipid hydroperoxide glutathione peroxidase (GPX4) or

Serine Hydroxymethyltransferase 1 (SHMT1) plays a pivotal role in one-carbon metabolism, facilitating the production of SAM. In this study, dysregulation of one-carbon metabolism is reported in both Parkinson's disease (PD) patients and animal models, characterized by significantly downregulated expression of SHMT1. Astrocyte-specific conditional knockout of Shmt1 decreased SAM level, exacerbated motor dysfunction, and dopaminergic neuronal loss in a PD mouse model. While SAM is conventionally generated through the one-carbon cycle, the data indicate that, despite significant alterations in SH

Gastric cancer (GC) remains the leading cause of cancer deaths worldwide. GC development is a multistep and multifactorial process, and the molecular characterization of the multistage progression of gastric lesions to GC is poorly understood. Induction of ferroptosis driven by iron-dependent phospholipid peroxidation ameliorates the malignant progression of GC. Here, we found that solute carrier family 47 member 1 (SLC47A1) promoted GC progression by regulating ferroptosis. Clinically, SLC47A1 was elevated during the progression of gastritis to GC, and its high expression was associated with

Silver nanoparticles (AgNPs) are used in a variety of different fields due to their excellent antimicrobial potential. Despite clear advantages, concerns about their toxicity have arisen, also concerning biogenic nanoparticles. Simultaneously, global healthcare is facing a problem of spreading antimicrobial resistance towards existing antibiotics. Using combined therapies involving AgNPs and antibiotics seems to be a promising solution to the above problems. The aim of this study was to evaluate the enhancement of the effectiveness of AgNPs, ciprofloxacin, and gentamicin against Staphylococcus

Tongue squamous cell carcinoma (TSCC) is a major subtype of head and neck cancer with limited treatment regimens. This paper examined phospholipid scramblase 1 (PLSCR1) and forkhead box protein A1 (FOXA1) expression patterns and function in TSCC progression. Abnormally high expression of PLSCR1 was screened by the GEO datasets, and PLSCR1 expression in TSCC cells was verified. The upstream mechanism of abnormally elevated PLSCR1 was investigated by bioinformatics analysis, and FOXA1 expression in TSCC cells was detected. The interaction between FOXA1 and the PLSCR1 promoter in cells was detect

Vancomycin, a glycopeptide antibiotic, is typically administered intravenously (IV) for severe Gram-positive bacterial infections. While the oral route offers higher patient adherence, it is limited by poor mucosal transport, restricting its use to intestinal infections. To address these challenges, this study explores the encapsulation of vancomycin into archaeosomes -phospholipid-based nanocarriers incorporating archaeal lipids, which exhibit exceptional stability in the gastrointestinal environment and interact specifically with enterocytes. Previous work demonstrated that lipid nanocarrier
